Two New Eisenia Species from South Korea Similar to E. koreana and Comparable to Eisenoides from USA (Oligochaeta: Lumbricidae)

  • New Korean lumbricids are described: Eisenia gaga Blakemore sp. nov. from remote Gageodo Island and E. sindo Blakemore, sp. nov. from an island at Incheon. Both are comparable to Eisenia koreana (Zicsi, 1972) from near Pyongyang, North Korea. A remarkable yet previously unrecognized similarity to American earthworm Eisenoides carolinensis (Michaelsen, 1910) is discussed, but synonymy is rejected on tenuous morphological grounds (form of nephridial vesicle bladders) with taxonomy supported by objective molecular data (mtDNA cytochrome c oxidase subunit 1 barcodes).

  • A great deal of effort has been put into the identification of Alexandrium tamarense/fundyense/catenella complex by understanding correlation between morphological and subcellular characteristics. To date, the most promising tool for the study of these species is sequence analyses of rRNA genes that have been useful for various organisms' taxonomy and phylogeny, and its application such as in situ hybridization. Four unrecorded species of macrofungi from South Korea

  • In 2022, our investigation focused on exploring the macrofungal diversity in South Korea. Through the examination of morphological characteristics and ITS sequences of the rDNA, we successfully identified four unrecorded species (Laccaria striatula, Leotia atrovirens, Malvipezia emileia, Xerula strigosa). Detailed morphological descriptions, accompanied by photographs and molecular phylogenetic analyses were performed.

  • 본 연구는 3개의 색소체 matK, trnL-F, rbcL DNA 염기서열과 1개의 핵 ITS DNA 염기서열을 근거로 한국산 Aloe 3종 A. arborescens, A. vera 그리고 A. saponaria 등과 하나의 변이종 알로에의 유전적 상관관계를 알고자 수행되었다. 전체 2,420 bp 서열이 증폭되었다. 두 개의 삽인-결실(indel)이 trnL 지역에서 확인되었고, 또한 여러 개의 종 특이적인 염기자위가 전체 29개의 최소변이 정보지역(parsimonious informative site)에서 확인되었다. 조사된 한국산 4 종간에는 148 염기변이 지역이 있었으며, 세계산 Aloe 종들을 포함한 비교해서는 170개의 변이지역 중 75개의 최소변이 지역이 확인되었다. UPGMA를 이용한 phenogram에서 새로운 변이종 알로에는 A. vera와 가장 가깝게 유집되었다. 변이종 알로에는 아직까지 보고된 어떤 종류의 Aloe속내 종과 형태적 및 유전적으로 일치하지 않았다. 조사된 알로에 종들의 유집분석 결과는 기존의 연구결과와 일치하였다.

한국산 흰명아주와 근연종의 세포분류학적 연구 (Cytotaxonomical Study of the Chenopodium album and its Related Species in Korea)

  • 한국산 흰명아주(Chenopodium album var. album), 명아주(var. centrorubrum), 가는명아주(var. stenophyllum)를 대상으로 aceto-orcein에 의한 염색체 수와 모양을 조사하였고, 45S rDNA 유전자를 이용한 FISH (fluorescence in situ hybridization) 방법을 수행하여 세포유전학적 유연관계를 고찰하였다. 체세포 염색체 수는 흰명아주와 명아주는 모두 2n = 6x = 54개인 반면에 가는명아주는 2n = 4x = 36으로 뚜렷이 구별되었으며, 기본염색체수는 x = 9 개였다. 명아주속의 염색체에서 45S rDNA의 위치를 알아보기 위한 FISH 결과는 흰명아주의 경우 8개의 signal이, 가는명아주에서는 2개의 signal이 관찰되어 종간 차이를 보였으며, 모두 염색체 말단부위에서 관찰되었다. 염색체의 수와 형태, 45S rDNA를 이용한 FISH 결과는 명아주가 흰명아주에 통합되지만, 가는명아주와는 뚜렷이 구별됨을 지지해 주었다.

Phylogenetic relationships of Iranian Allium species using the matK (cpDNA gene) region

  • Allium L. is one of the largest genera of the Amaryllidaceae family, with more than 920 species including many economically important species used as vegetables, spices, medicines, or ornamental plants. Currently, DNA barcoding tools are being successfully used for the molecular taxonomy of Allium. A total of 46 Allium species were collected from their native areas, and DNA was extracted using the IBRC DNA extraction kit. We used specific primers to PCR amplify matK. DNA sequences were edited and aligned for homology, and a phylogenetic tree was constructed using the neighbor-joining method. The results show thymine (38.5%) was the most frequent and guanine (13.9%) the least frequent nucleotide. The matK regions of the populations were quite highly conserved, and the amount of C and CT was calculated at 0.162 and 0.26, respectively. Analysis of the nucleotide substitution showed C-T (26.22%) and A-G (8.08%) to have the highest and lowest percent, respectively. The natural selection process dN/dS was 1.16, and the naturality test results were -1.5 for Tajima's D and -1.19 for Fu's Fs. The NJ dendrogram generated three distinct clades: the first contained Allium austroiranicum and A. ampeloprasum; the second contained A. iranshahrii, A. bisotunense, and A. cf assadi; and the third contained A. rubellum and other species. In this study, we tested the utility of the matK region as a DNA barcode for discriminating Allium. species.

  • An unrecorded Antrodia species was collected in South Korea and based on morphological characteristics, the species was identified as Antrodia sitchensis. To confirm its affinity within the polypores, the phylogenetic relationships of A. sitchensis and allied species were established using large subunit rDNA sequences.

ITS 염기서열에 의한 한국산 미나리아재비속 미나리아재비절의 분류학적 검토

  • 한국산 미나리아재비속 미나리아재비(Acris Schur)절에 속하는 미나리아재비(Ranunculus japonicus)와 근연종인 산미나리아재비(R. acris var. nipponicus) 및 바위미나리아재비(R. crucilobus)의 실체와 분류학적 한계를 파악하기위해 속, 종간 규명에 많이 이용하고 있는 핵리보좀(ribosomal) DNA의 internal transcribed spacer 구간의 염기서열을 분석하였다. 본 연구는 6개의 군외군을 포함하여 총 18개의 DNA 재료(accessions)의 정열된 염기서열들을 바탕으로 bootsrap을 포함한 maximum parsimony와 maximum likelihood 분석법에 의한 계통수로 평가하였다. 연구 결과 Acris절에 속하는 미나리아재비, 산미나리아재비 및 바위미나리아재비는 단계통군으로 나타났으며 특히 미나리아재비(R. japonicus)와 산미나리아재비(R. acris var. nipponicus)는 같은 분계조를 형성하였다. 이와 달리 바위미나리아재비는 미나리아재비와 산미나리아재비에서 분지된 결과를 보여, 한라산 해발 1500m이상의 높은 지역에 분포하는 바위미나리아재비는 미나리아재비의 아종(R. japonicus Thunb. subsp. chrysotrichus (Nakai) Y. N. Lee, comb. nud.)으로 처리하기보다는, 독립된 고유종인 R. crucilobus H. L$\acute{e}$v.으로의 처리를 지지하였다.

Phytopythium and Pythium Species (Oomycota) Isolated from Freshwater Environments of Korea

  • Oomycetes are widely distributed in various environments, including desert and polar regions. Depending upon different habits and hosts, they have evolved with both saprophytic and pathogenic nutritional modes. Freshwater ecosystem is one of the most important habitats for members of oomycetes. Most studies on oomycete diversity, however, have been biased mostly towards terrestrial phytopathogenic species, rather than aquatic species, although their roles as saprophytes and parasites are essential for freshwater ecosystems. In this study, we isolated oomycete strains from soil sediment, algae, and decaying plant debris in freshwater streams of Korea. The strains were identified based on cultural and morphological characteristics, as well as molecular phylogenetic analyses of ITS rDNA, cox1, and cox2 mtDNA sequences. As a result, we discovered eight oomycete species previously unknown in Korea, namely Phytopythium chamaehyphon, Phytopythium litorale, Phytopythium vexans, Pythium diclinum, Pythium heterothallicum, Pythium inflatum, Pythium intermedium, and Pythium oopapillum. Diversity and ecology of freshwater oomycetes in Korea are poorly understood. This study could contribute to understand their distribution and ecological function in freshwater ecosystem.
