The Effect of 50% dietary restriction on Protein and Nucleic acid Metabolism of Albino rats (식이제한이 흰쥐의 체내대사에 미치는 영향)

  • Elucidation of the metabolic pathway due to 50% dietary restriction carried out in this study. Seventy male and female wealning rats, weighed $43{\pm}2g$ were divided into seven groups, 10 rats each. Twenty rats, ten males and ten females were sacrificed every three weeks after 50% dietary restriction for whole length of the experiment, nine weeks. Pair-feeding was employed in this study. According to the increment of the dietary restricted period, the body and organ weights were decreased. Especially liver and spleen were mostly shrinked in their weights, and brain was the most stable organ in account of dietary restricted effect. In comparison nitrogen retention between restricted and unrestricted groups, the former showed lower than the later but tubulated into the rate of Nitrogen retention per gram of body weight, reverse was true in this respect. In regardness of the experimental organs, spleen revealed the most fast change and the brain the most slow change their content of RNA and DNA in account of the 30% dietary restriction. Hematological investigation did not show any anemic conditions in both restricted and unrestricted groups. Also serum albumin contents A/G ratio, did not effect due to 50% dietary restrictions.

Taxonomic Characteristics of Nitrogen-Fixing Oligotrophic Bacteria from Forest Soil (산림토양으로부터 분리한 저영양성-질소고정세균의 분류학적 특성)

  • Many isolates from different forest soil layers did not show appreciable growth on full strength of the conventional nutrient broth (NB medium) but grow on its 100-fold dilution (DNB medium). These isolates were divided into four types according to organic nutrient concentration in the growth medium from $1^{-1}\;to\;10^{-4}$dilution of normal NB medium. Oligotrophic bacteria were type II and type IV which grew in $10^{-4}$ dilution of NB (1 mg C/l) medium. Sixty strains were isolated for obligate oligotrophic bacteria. Chemotaxonomic and phylogenetic characteristics of eleven isolates of acetylene-reducing (nitrogen-fixing) oligotrophic bacteria from forest soil were investigated. They showed similar characteristics: the cellular fatty acid mainly consisted of straight-chain unsaturated $C_{18:1}$ (60-84% of total fatty acids). Ubiquinone Q-10 and a high guanine plus-cytosine content(61-64 mol%) were found. Eleven isolates of nitrogen-fixing oligotrophic bacteria were found to be closely related by full 16S rDNA sequence simility and many common taxonomic traits. Analysis of full 16S rDNA sequences of eleven isolates indicated that they were more closely related to Bradyrhizobium (similarity values: 98.1-98.8%), Agromonas, Nitrobacter, and Afipia.

  • To improve the starch fermentation ability of yeast, hybrids were introduced by protoplast fusion of S. cerevisiae and S. diastaticus. The protoplasts of parental auxotrophic cells were fused in the presence of 10 mM CaCl$_2$and 30% of polyethyleneglycol (M.W 4, 000). The frequencies of fusant formation varied depending upon the strains used and were 3.51$\times$10$^{-4}$ to 5.04$\times$10$^{-4}$ for the regenerated protoplasts. The strains capable of extensive starch hydrolysis produce only 10% to total fusants. The 4 strains were finally selected by the results of starch fermentation and genetic stability test. The DNA content and cell volume of the fusants were greater than those of the parental strains.

  • Protoplast susion between Zygosaccharmoyces rouxii M-12 and Saccharimyces cerevusuare M-43 were investigated for breeding of a new brewing yeat strain for soy sauce. Auxotrophic mutants of Zygosaccharomyces rouxii ZRM-83 ($Met^-,\;Thr^-$) and Saccharomyces cerevisiae SCM-46 ($Lys^-,\;Arg^-$) were selected by treatment of 3.0% ethylmethane sulfonate and nutritional complementary method. Protoplast of both strains were more effective by treatment of 0.05mg/ml zymolase 20T for 60min. Fusion effeciency was much higher by treatment of 30% PEG 6,000 for 30min and fusion frequencies were $10^{-4}{\sim}10^{-5}$. These fusants originated from two protoplasts had properties of big cell size and much DNA content.

유류오염 토양에서 유류성분의 생분해와 무기오염 물질 함량의 관계성에 대한 예비 연구

  • 유류오염 토양내 유류의 분해와 무기오염물질의 함량과의 관계성을 알아보기위해 부산 문헌지구의 유류 오염 토양 profile중 유류가 비교적 많은 두 층과 유기물이 다량 함유된 제주도 토양을 경유로 오염시킨 토양을 선택하여 유류가 분해 정도와 Pb, Zn, Cd, Ni의 함량을 측정 비교하였다. 토양 채취후 즉시 유류를 추출한 것, 15일간 별도의 미생물 영양원을 가하지 않고 습도와 통풍만을 행하여 토양내 존재하는 미생물들에 의하여만 유류가 분해되도록 한 것, $105^{\circ}C$에서 24시간 열처리를 행하여 미생물에 의한 분해보다는 물리적인 방법으로 비교적 강하게 처리한 것 세가지 경우에서 토양에서 추출한 유류를 분석한 결과 normal alkane이 서서히 상당량 분해되는 양상을 보여주었다. 토양내 Pb, Zn, Cd, Ni를 분석한 결과 부산 시료의 경우 두 층(B2, B3)모두 Cd은 검출한계 이하로, Ni의 경우 극소량의 함량을 보여주었다. Zn의 경우 B2는 28~29ug/g, B3는 12~14ug/g으로 세가지 경우 별 차이 없는 양상을 보여 주었다. 하지만, Pb의 경우 B2에서는 그 함량이 유류의 normal alkane의 분해와 함께 서서히 증가하는 경향을 확연히 보여주었으며, B3의 경우도 B2와 유사한 변화 양상을 보여주었다. 제주도 토양의 경우 Pb는 검출되지 않았고, Zn은 소량 검출되었다. Ba, Ni 함량은 normal alkane 분해에 따라 다소 증가하였다. 따라서, 본 예비실험을 통하여 유류오염 토양에서 유류의 분해와 일부 무기이온사이에는 관련성을 갖고 있으며, 이는 유류 분해정도를 파악하는 지시자로써 특정 무기 오염물질을 이용할 수 있을 가능성이 있으므로 좀더 이들 관계성에 대한 연구가 진행될 필요성이 있다고 판단된다.고 과학적으로 분석할 수 있는 방법이 될 수 있을 것으로 기대된다. 의미를 되새기는 것으로 짧은 연구를 시작하겠다. 등은 활성 값이 70% 이상으로 퇴적물 독성이 상대적으로 낮았다. 이중나선 DNA 함량은 28.4 % - 49%로 대조군에 비해서 감소가 크다. 대부분의 정점이 대조군의 30% 내외로 정점 간의 차이는 크지는 않다. 그러나 다른 측정자료와 같이 정점 22에서 18%로 최소치를 나타내고, 정점 2, 12에서 20% 내외의 값을 보인다. 종합적으로 볼 때 오염물질의 유입이 크고, 광양제철 인근 정점 들이 모두 다른 정점에 비해서 낮아서, 퇴적물 독성이 높은 정점으로 조사되었다.hiwo의 광합성 능력은 낮은 농도들에서는 대조구와 유사하였으나, 5 $\mu\textrm{g}$/l의 높은 농도에서는 초기에 매우 낮은 광합성 능력을 보이다가 시간이 경과하면서 대조군보다 더 높은 경향을 나타냈다. 이러한 결과는 식물플랑크톤이 benso[a]pyrene의 낮은 농도에서 노출될 때는 이 물질을 탄소원으로 사용할 가능성이 있음을 시사한다. 본 연구의 결과들은 연안해역에 benso[a]pyrene과 같은 지속성 유기오염물질이 유입되었을 때 내정여부에 따라 식물플랑크톤 군집내 종 천이와 일차생산력에 크게 영향을 미칠 수 있음을 시사한다.TEX>5.2개)였으며, 등급별 회수율은 각각 GI(8.5%), GII(13.4%), GIII(43.9%), GIV(34.2%)로 나타났다.ments of that period left both in Japan and Korea. The Role of Nitric Oxide on the Growth Regulation of Chinese Cabbage (Brassica campestris L.) Primary Leaves (배추 (Brassica campestris L.) 제 1엽의 생장조절에 대한 Nitric Oxide의 역할)

  • The possible role of nitric oxide (NO)-induced cell division was investigated to explain the physiologycal effects of a NO donor, sodium nitroprusside (SNP) on the growth of primary leaves in chinese cabbage seedling plants. Exogenous treatment of SNP to chinese cabbage plants for 8 days at different concentrations (0, 200, 500 and 1000 ${\mu}M$) affected the leaf growth in a concentration-dependent manner, showing a maximum growth at $200\;{\mu}M$. In accordance with leaf growth responses, the chlorophyll and soluble protein contents increased strongly to 142% and 134% of control at $200\;{\mu}M$ SNP, respectively. However, a very little decrease in chlorophyll and a 13%> decrease in protein were observed at $1000\;{\mu}M$ SNP. In addition, the content of DNA and RNA also increased maximumly to 142% and 139% of the control at $200\;{\mu}M$ SNP, respectively, whereas they decreased to 80% and 84% of the control at $1000\;{\mu}M$ SNP. With respect to the development of enzymes related to cell wall synthesis, $200\;{\mu}M$ SNP led to the maximum activities in both phenylalanine ammonia-lyase (212% of the control) and guaiacol peroxidase (134% of the control). However, the activities of both enzymes were not modified significantly at $1000\;{\mu}M$ SNP. In conclusion, these results suggest that the enhancement of leaf growth in chinese cabbage plants by SNP at the effective concentration was probably due to the NO ability in the induction of cell division.

Ethanol Induced Leucocytic and Hepatic DNA Strand Breaks Are Prevented by Styela clava and Styela plicata Supplementation in Male SD Rats (알코올로 인한 흰쥐의 백혈구 및 간 DNA 손상에 미치는 미더덕과 오만둥이 분말의 보충섭취 효과)

  • In this study, the ability of Styela clava or Styela plicata to reduce ethanol-induced hepatotoxicity and hepatic and leucocytic DNA damages was evaluated. Twenty four male SD rats were given 25% ethanol containing water (ad lib, p.o.) and divided into 3 groups; ethanol treated control group (EtOH), ethano1+3% S. clava (EtOH+SC), and ethano1+3% S. plicata (EtOH+SP). After 6 weeks, the supplementation of S. clava reduced the plasma ALT, ALP and LDH activities significantly (p<0.05), while S. plicata induced significant decrease in the plasma LDH activity only. The comet assay was employed to quantify the alcohol-induced DNA damage in rat hepatocytes and leucocytes. A significant protective effect on hepatic and leucocytic DNA damages was observed in S. clava or S. plicata supplemented groups compared to the EtOH control group. The hepatic DNA damage was correlated positively with plasma ALP and LDH activities. These results demonstrated that S. clava or S. plicata supplementation protected alcohol-induced hepatic and leucocytic DNA damage.

Antioxidative and Antigenotoxic Effect of Omija (Schizandra chinensis B.) Extracted with Various Solvents (다양한 용매에 의해 추출된 오미자의 항산화능과 항유전독성 효과)

  • The purpose of this study was to evaluate antioxidant and antigenotoxic effects of Omija (Schizandra chinensis B.) extracted with various solvents (acetone, ethanol, and methanol). The total polyphenol content (TPC) of methanol extract (ME), ethanol extract (EE) and acetone extract (AE) from Omija were 1183.3, 1009.4, and 747.3 mg/100 g (garlic acid equivalents: GAE), respectively. Antioxidant effects of the Omija extracts was measured by DPPH radical-scavenging activity (RSA) and superoxide dismutase (SOD)-like activity. The $IC_{50}$ for DPPH RSA was in the order of EE $(1411.1\;{\mu}g/mL)$=AE $(1462.0\;{\mu}g/mL)$>ME $(1585.0\;{\mu}g/mL)$. The $IC_{50}$ for SOD-like activities was the highest in ME $(905.7\;{\mu}g/mL)$=EE $(970.3\;{\mu}g/mL)$>AE $(1579.4\;{\mu}g/mL)$. The antigenotoxic effect of Omija on DNA damage induced by $H_2O_2$ in human leukocytes was evaluated by comet assay. $H_2O_2$ induced DNA damage was effectively protected by all of the Omija extracts. Aectone extract of Omija showed the highest antigenotoxic effect ($IC_{50}$ value of AE is $14.6\;{\mu}g/mL$) followed by EE, and ME (21.4 and $34.4\;{\mu}g/mL$), respectively. As a result, we propose that Omija (Schizandra chinensis B.) can serve as a new natural source enriched with potent antioxidant and antigenotoxic agents.

Antioxidative Activity of Extracts from Wisteria floribunda Flowers (등나무 꽃 추출물의 항산화 활성)

  • The antioxidant activities of Wisteria floribunda flowers (WFF) were evaluated. The samples were prepared by extracting separately two different colored flowers (purple and white) with four different solvents (methanol, ethanol, acetone, and water). The antioxidant properties were evaluated by determining total phenolic contents (TPC), radical scavenging activity (RSA), and reducing power (RP). Water extract from purple WFF and ethanol extract of white WFF showed the highest total phenol contents (491 and 787 ${\mu}M$ gallic acid equivalents), respectively. Water extracts of purple and white WFF also showed higher RSA. In the case of RP, ethanol extract of purple WFF, methanol and water extracts of white WFF showed relatively higher values. The 200 ${\mu}M$ $H_2O_2$ induced oxidative DNA damage in human leukocytes was significantly inhibited with WFF extracts excluding ethanol and acetone extracts of purple flowers. These results suggest that W. floribunda flowers have significant antioxidative activity and protective effect against oxidative DNA damage.

  • Formaldehyde responsive protein(FRP) 1 belongs to the family of universal stress protein(USP) and is known to respond to stress caused by fumigation of gaseous volatile organic compounds(VOCs) such as formaldehyde and toluene. However, the molecular function of this protein is not well understood at cellular and molecular level. In this study, loss of function mutant of FRP1 generated by T-DNA insertion(frp1-4) has been isolated from Arabidopsis thaliana and the function of FRP1 was characterized. The loss-of-function mutant of FRP1 appeared slight growth defects with shorter stem and rosette leaves compared to wild type. In addition, the damage caused by exogenous VOCs was more severe in frp1-4 than in control. Therefore, Arabidopsis FRP1 seems to be the protein involved not only in the growth and development of plant but also the stress resistance against toxic volatile organic compounds.
