Uric acid transport across the basolateral membrane of renal proximal tubules was studied in rabbit kidney cortical slices. Uric acid uptake was greater under $O_2$ atmosphere compared to under $N_2$ atmosphere, and was increased with $Na^{2+}$ concentration in incubation medium. Uric acid inhibited PAH uptake but not TEA uptake and did trans-stimulated PAH efflux. PAH also inhibited uric acid uptake. Uric acid uptake was inhibited by harmaline, ouabin, SITS, DIDS and pyrazinoic acid. The inhibition of PAH uptake by these inhibitors also was reasonably comparable to that of uric acid uptake. These results suggest that uric acid was transported across the basolateral membrane of renal tubule by a carrier-mediated process which was by a common transport system with PAH in rabbit.
The aim of the current study was to evaluate the transport system in EMT-6 cell for the uptake of the methylmercury(MeHg). Several inhibitors ere used to test used to test which potential transport system might be involoved in MeHg uptake. Probenecid was used to test the organic transport system, valinomycin for testing the effect of the membrane potential, cytochalasin B for testing the facilitated diffusive D-glucose transport system and colchicine for testing the microtubule system. Ouabain for evaluating active transport system, 4',4-diisothiocyano-2',2-stilbenedisulfonic acid(DIDS) the Cl- ion transport system and verapamil for the $Ca^{2+}$ transprot system. Significantly, MeHg decreased the synthesis of nitric oxcide(NO) and intracellular ATP in ENT-6 cells. In the condition of ouabain containing with MeHg decreased the production of NO and intracelluar ATP. In the treatment of inhibitors, ouabain showed protective effect against cytotoxicity of MeHg but ather inhibitors not showed protective effects. The protective effects of ouabain against the cytotoxicity of MeHg deoended on the concentration of added ouabain to the culture medium for MET-6 cells. These result showed that the uptake of MeHg might be involved in the active transport system. Active transports system seems to share similarities with the transport systems for the uptake of MeHg when using MeHg and MeHg-glutathione complex.x.
The caudal subnucleus of the spinal trigeminal nucleus (medullary dorsal horn; MDH) receives direct inputs from small diameter primary afferent fibers that predominantly transmit nociceptive information in the orofacial region. Recent studies indicate that reactive oxygen species (ROS) is involved in persistent pain, primarily through spinal mechanisms. In this study, we aimed to investigate the role of xanthine/xanthine oxidase (X/XO) system, a known generator of superoxide anion ($O_2{^-}$), on membrane excitability in the rat MDH neurons. For this, we used patch clamp recording and confocal imaging. An application of X/XO ($300{\mu}M/30mU$) induced membrane depolarization and inward currents. When slices were pretreated with ROS scavengers, such as phenyl N-tert-butylnitrone (PBN), superoxide dismutase (SOD), and catalase, X/XO-induced responses decreased. Fluorescence intensity in the DCF-DA and DHE-loaded MDH cells increased on the application of X/XO. An anion channel blocker, 4,4-diisothiocyanatostilbene-2,2-disulfonic acid (DIDS), significantly decreased X/XO-induced depolarization. X/XO elicited an inward current associated with a linear current-voltage relationship that reversed near -40 mV. X/XO-induced depolarization reduced in the presence of $La^{3+}$, a nonselective cation channel (NSCC) blocker, and by lowering the external sodium concentration, indicating that membrane depolarization and inward current are induced by influx of $Na^+$ ions. In conclusion, X/XO-induced ROS modulate the membrane excitability of MDH neurons, which was related to the activation of NSCC.
Mammalian gastric smooth muscles generate spontaneous rhythmic contractions which are associated with slow oscillatory potentials (slow waves) and spike potentials. Spike potentials are blocked by organic $Ca^{2+}-antagonists,$ indicating that these result from the activation of L-type $Ca^{2+}-channel.$ However, the cellular mechanisms underlying the generation of slow wave remain unclear. Slow waves are insensitive to $Ca^{2+}-antagonists$ but are blocked by metabolic inhibitors or low temperature. Recently it has been suggested that Interstitial Cells of Cajal (ICC) serve as pacemaker cells and a slow wave reflects the coordinated behavior of both ICC and smooth muscle cells. Small segments of circular smooth muscle isolated from antrum of the guinea-pig stomach generated two types of electrical events; irregular small amplitude (1 to 7 mV) of transient depolarization and larger amplitude (20 to 30 mV) of slow depolarization (regenerative potential). Transient depolarization occurred irregularly and membrane depolarization increased their frequency. Regenerative potentials were generated rhythmically and appeared to result from summed transient depolarizations. Spike potentials, sensitive to nifedipine, were generated on the peaks of regenerative potentials. Depolarization of the membrane evoked regenerative potentials with long latencies (1 to 2 s). These potentials had long partial refractory periods (15 to 20 s). They were inhibited by low concentrations of caffeine, perhaps reflecting either depletion of $Ca^{2+}$ from SR or inhibition of InsP3 receptors, by buffering $Ca^{2+}$ to low levels with BAPTA or by depleting $Ca^{2+}$ from SR with CPA. They persisted in the presence of $Ca^{2+}-sensitive$$Cl^--channel$ blockers, niflumic acid and DIDS or $Co^{2+},$ a non selective $Ca^{2+}-channel$ blocker. These results suggest that spontaneous activity of gastric smooth muscle results from $Ca^{2+}$ release from SR, followed by activation of $Ca^{2+}-dependent$ ion channels other than $Cl^-$ channels, with the release of $Ca^{2+}$ from SR being triggered by membrane depolarization.
During depolarization, extrusion of $Ca^{2+}$ from sarcoplasmic reticulum through forward-mode $Na^+-Ca^{2+}$ exchange was studied in the rat ventricular myocytes patch-clamped in whole-cell configuration. In order to confine the $Ca^{2+}$ responses in a micro-domain by limiting the $Ca^{2+}$ diffusion time, rat ventricular myocytes were dialyzed with high (14 mM) EGTA. $K^+$ current was suppressed by substituting KCl with 105 mM CsCl and 20 mM TEA in the pipette filling solution and by omitting KCl in the external Tyrode solution. $Cl^-$ current was suppressed by adding 0.1 mM DIDS in the external Tyrode solution. During stimulation roughly mimicking action potential, the initial outward current was converted into inward current, $47{\pm}1%$ of which was suppressed by 0.1 mM $CdCl_2.$ 10 mM caffeine increased the remaining inward current after $CdCl_2$ in a cAMP-dependent manner. This caffeine-induced inward current was blocked by $1\;{\mu}M$ ryanodine, $10\;{\mu}M$ thapsigargin, 5 mM $NiCl_2,$ or by $Na^+\;and\;Ca^{2+}$ omission, but not by $0.1\;{\mu}M$ isoproterenol. The $I{\sim}V$ relationship of the caffeine-induced current elicited inward current from -45 mV to +3 mV with the peak at -25 mV. Taken together, it is concluded that, during activation of the rat ventricular myocyte, forward-mode $Na^+-Ca^{2+}$ exchange extrudes a fraction of $Ca^{2+}$ released from sarcoplasmic reticulum mainly by voltage-sensitive release mechanism in a micro-domain in the t-tubule, which is functionally separable from global $Ca^{2+}{_i}$ by EGTA.
Ginsenosides, major active ingredients of Panax ginseng, are known to regulate excitatory ligand-gated ion channel activity such as nicotinic acetylcholine and NMDA receptor channel activity. However, it is not known whether ginsenosides affect inhibitory ligand-gated ion channel activity. We investigated the effect of ginsenosides on human recombinant $GABA_A$ receptor (${\alpha}_1{\beta}_1{\gamma}_{2s}$) channel activity expressed in Xenopus oocytes using a two-electrode voltage-clamp technique. Among the eight individual ginsenosides examined, namely, $Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$ and $Rg_2$, we found that Rc most potently enhanced the GABA-induced inward peak current ($I_{GABA}$). Ginsenoside Rc alone induced an inward membrane current in certain batches of oocytes expressing the $GABA_A$ receptor. The effect of ginsenoside Rc on $I_{GABA}$ was both dose-dependent and reversible. The half-stimulatory concentration ($EC_{50}$) of ginsenoside Rc was 53.2$\pm$12.3 $\mu$M. Both bicuculline, a $GABA_A$ receptor antagonist, and picrotoxin, a $GABA_A$ channel blocker, blocked the stimulatory effect of ginsenoside Rc on $I_{GABA}$. Niflumic acid (NFA) and 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS), both $CI^{-1}$ channel blockers, attenuated the effect of ginsenoside Rc on I$I_{GABA}$. This study suggests that ginsenosides regulated $GABA_A$ receptor expressed in Xenopus oocytes and implies that this regulation might be one of the pharmacological actions of Panax ginseng.

Over the past decades, there have been numerous studies for approaches to build and operate electronic government systems. Among those studies, work breakdown structure(WBS) has been gained attention increasingly. As a general system engineering techniques. WBS is a kind of methods to engineer and manage electronic government systems effectively. WBS is to organizes tasks and data items for management and engineering for electronic government systems. In this paper, the authors proposed a conceptual WBS model for management and engineering electronic government systems. Also, the authors conducted a case study based on the conceptual WBS model. The results of this study provide useful insights and guidelines for applying the WBS to management and engineering for electronic government systems. The authors believed that the conceptual WBS models will be widely used to management and engineering an electronic government systems.

In this paper, we propose BCON, a service that allows individuals to store personal contents safely, and reliably guarantee their ownership of contents, certifying their identities with DIDs(Decentralized identifiers). DID technology, which supports decentralized identification service based on a blockchain that cannot forgery or alter data, allows users to selectively provide their information, controlling personal information and reinforcing their sovereignty over their identity. BCON stores information about the content specified by a user on the blockchain and Authenticates the user's identity based on DID technology. It also provides functions for the user to safely upload and download the user's content to a distributed database. BCON consists of the content service verifier, the content storage service, the content management contract, and the user application, administrating rhe DID registry for Authority management.

Smart grid that supports efficient energy production and management is used in various fields and industries. However, because of the environment in which services are provided through open networks, it is essential to resolve trust issues regarding security vulnerabilities and privacy preservation. In particular, the identification information of smart meter is managed by a centralized server, which makes it vulnerable to security attacks such as device stolen, data forgery, alteration, and deletion. To solve these problems, this paper proposes a blockchain based authentication protocol for a smart meter. The proposed scheme issues an unique decentralized identifiers (DIDs) for individual smart meter through blockchain and utilizes a random values based on physical unclonable function (PUF) to strengthen the integrity and reliability of data. In addition, we analyze the security of the proposed scheme using informal security analysis and AVISPA simulation, and show the efficiency of the proposed scheme by comparing with related work.
Traditional authentication systems typically involve users entering their username and password into a centralized identity management system. To address the inconvenience of such authentication methods, a decentralized identity authentication system based on Distributed Identifiers(DID) is proposed, utilizing decentralized identity technology. The proposed system employs QR code scanning for login, enhancing security through the use of blockchain technology to ensure the uniqueness and safety of user identities during the login process. This system utilizes DIDs and integrates the InterPlanetary File System(IPFS) to securely manage organizational members' identity information while keeping it private. Using the distributed identity authentication system proposed in this study, it is possible to effectively manage the security and personal identity of organization members. To improve the usability of the system proposed in this study, research is needed to expand it into a solution.
