In Vitro Antifungal Activity and Mode of Action of 2',4'-Dihydroxychalcone against Aspergillus fumigatus

  • 2',4'-Dihydroxychalcone (2',4'-DHC) was identified from a heat shock protein 90 (Hsp90)-targeting library as a compound with Hsp90 inhibitory and antifungal effects. In the presence of 2',4'-DHC ($8{\mu}g/mL$), radial growth of Aspergillus fumigatus was inhibited 20% compared to the control, and green pigmentation was completely blocked. The expression of the conidiation-associated genes abaA, brlA, and wetA was significantly decreased (approximately 3- to 5-fold) by treatment with 2',4'-DHC. The expression of calcineurin signaling components, cnaA and crzA, was also significantly reduced. The inhibitory effects of 2',4'-DHC on metabolic activity and mycelial growth were significantly enhanced by combination treatment with itraconazole and caspofungin. Docking studies indicated that 2',4'-DHC bind to the ATPase domain of Hsp90. These results suggest that 2',4'-DHC act as an Hsp90-calcinurin pathway inhibitor.

DHC Characteristics of M11 Pressure Tube in Wolsong Unit 1

  • Delayed hydride cracking (DHC) velocity and threshold stress intensity factor for DHC ($K_{IH}$) tests in the radial direction on M11 pressure tube material in Wolsong unit 1 were carried out following the Atomic Energy Canada Limited (AECL) standard test procedure in order to identify the effect of undercooling on DHCV and to acquire the $K_{IH}$ data. The results showed that $K_{IH}$ 's were 8.8$\pm$0.8 MPa√m in the back offcut and 11.4$\pm$0.7 MPa√m in the front offcut. The fact that $K_{IH}$ in the front offcut is about 20% higher than that in the back offcut is attributed to the microstructural difference between the materials of the front and back ends. $K_{IH}$ 's in M11 pressure tube appeared to be higher than the values from the tubes made of double melted ingot reported earlier. This can be interpreted by the fact that very small amounts of Chlorine (Cl) and Phosphorus (P) are contained in the ingot and that the content of the harmful elements in the M11 pressure tube is equivalent to that made of a quadruple melting process. DHC velocities at 25$0^{\circ}C$ in the front offcut in the radial direction are measured to be 5~8$\times$10$^{-8}$ m/s. The results show that the prior thermal history change the DHC velocity significantly. This effect was confirmed by the experiment of undercooling prior to the DHC tests.DHC tests.

On the effect of temperature on the threshold stress intensity factor of delayed hydride cracking in light water reactor fuel cladding

  • Delayed hydride cracking (DHC) was first observed in pressure tubes in Canadian CANDU reactors. In light water reactors, DHC was not observed until the late 1990s in high-burnup boiling water reactor (BWR) fuel cladding. In recent years, the focus on DHC has resurfaced in light of the increased interest in the cladding integrity during interim conditions. In principle, all spent fuel in the wet pools has sufficient hydrogen content for DHC to operate below $300^{\circ}C$. It is therefore of importance to establish the critical parameters for DHC to operate. This work studies the threshold stress intensity factor ($K_{IH}$) to initiate DHC as a function of temperature in Zry-4 for temperatures between $227^{\circ}C$ and $315^{\circ}C$. The experimental technique used in this study was the pin-loading testing technique. To determine the $K_{IH}$, an unloading method was used where the load was successively reduced in a stepwise manner until no cracking was observed during 24 hours. The results showed that there was moderate temperature behavior at lower temperatures. Around $300^{\circ}C$, there was a sharp increase in $K_{IH}$ indicating the upper temperature limit for DHC. The value for $K_{IH}$ at $227^{\circ}C$ was determined to be $2.6{\pm}0.3MPa$ ${\surd}$m.

  • Unstable cosmetic active ingredients could rapidly break down in chemical and photochemical process. Therefore, it has become a very important issue to encapsulate active ingredient for the stabilization. 7-Dehydrocholesterol (7-DHC), a precursor of vitamin $D_3$, has been shown to increase levels of protein and mRNA for heat shock protein in normal human epidermal keratinocytes. However, topical dermal application of 7-DHC is restricted due to its poor solubility and chemical unstability. In this study, 7-DHC was incorporated into nano-emulsion (NE), solid lipid nano-particle (SLN), and chitosan coated solid lipid nano-particle (CASLN), respectively. In order to prepare NE and SLN dispersion, high-pressure homogenization at temperature above the melting point of lipid was used Hydrogenated lecithin and polysorbate 60 were used as stabilizer for NE and SLN. CASLN was prepared by high speed homogenizing after adding chitosan solution to the SLN dispersion and showed positively charged particle properties. Decomposition rate of 7-DHC in NE, SLN and CASLN was studied as a function of time at different temperature. Differential scanning calorimetry (DSC) and X-ray diffraction (XRD) studies were performed to characterize state of lipid modification. It appeared that CASLN is the most effective to stabilize 7-DHC and may be used for a useful topical dermal delivery system.

Antitumor Activity of 23, 24-dihydrocucurbitacin D Isolated from Bryonia alba L.

  • The cytotoxic and antitumor activity of 23,24-dihydrocucurbitacin D (dhc D) isolated from Bryonia alba L. was examined. Our results showed that dhc D strongly inhibited the viability of the cultured cancer cells, A-549, COLO 205, SK-MEL-2, and L121O. It also exhibited effective antitumor activity in ICR mice, which had been intra peritoneally implanted with sarcoma 180 ascites tumor cells. The dhc D also strongly inhibited the viability of immortalized macrophages RAW 264.7, but not normal peritoneal macrophages. These results indicate that dhc D has antiproliferative effect on cancer cells and it may more sensitive on immortalized cells than normal cells.

A Study on the Characteristics of Delayed Hydride Cracking in Zr-2.5Nb Pressure Tube with the Heating-up and Heat-treatment (열처리 및 가열방식에 따른 Zr-2.5Nb 압력관의 수소지연균열 특성에 관한 연구)

  • The objective of this study was to obtain a better understanding of the delayed hydride cracking (DHC) of Zr-2.5Nb alloy. The DHC model has some defects: first, it cannot explain why the DHC velocity (DHCV) becomes constant regardless of an applied stress intensity factor, even though the stress gradient is affected by the applied stress intensity factor at the notch tip. Second, it cannot explain why the DHCV has a strong dependence on the method of approaching the test temperature by a cool-down or a heating-up, even under the same stress gradient, and third, it cannot predict any hydride size effect on the DHC velocity. The DHC tests were conducted on Zr-2.5Nb compact tension specimens with the test temperatures reached by a heating-up method and a cool-down method. Crack velocities were measured in hydrided specimens, which were cooled from solution-treatment temperatures at different rates by being furnace-cooled, water-quenched, and liquid nitrogen-quenched. The resulting hydride size, morphology, and distributions were examined by optical metallography. It was found that fast cooling rates, which produce very finely dispersed hydrides, result in higher crack growth rates. This different DHC behavior of the Zr-2.5Nb tube with the cooling rate after a homogenization treatment is due to the precipitation of the $\gamma$-hydrides only in the water-quenched Zr-2.5Nb tube. This experiment will provide supporting evidence that the terminal solid solubility of a dissolution (TSSD) of $\gamma$-hydrides is higher than that of $\delta$-hydrides.

A Correlation of Striation Spacing and DHC Velocity in Zr-2.5Nb Tubes (Zr-2.5Nb 압력관에서 Striation Spacing과 DHCV의 관계)

  • The objective of this study is to elucidate what governs delayed hydride cracking (DHC) in Zr-2.5Nb tubes by correlating the striation spacings with DHCV(DHC Velocity). To this end, DHC tests were conducted on the compact tension specimens taken from the Zr-2.5Nb tubes at different temperatures ranging from 100 to $300^{\circ}C$ with a 3 to 6 data set at each test conditions. The compact tension specimens were electrolytically charged with 27 to 87 ppm H before DHC tests. After DHC tests, the striation spacings and DHCV were determined with the increasing the test temperature and yield strength. The striation spacing and DHCV increased as a function of yield $strength^2$ and the temperature. Since the plastic zone size ahead of the crack tip can be represented by ${\sim}(K_{IH}/{\sigma}_{Y})^2$, we conclude that the striation spacing is governed by the plastic zone size which in turn determines a gradient of hydrogen concentration at the crack tip. The relationship between the plastic zone size and the striation spacing was validated through a complimentary experiment using double cantilever beam specimens. Two main factors to govern DHCV of Zr-2.5Nb tubes are concluded to be hydrogen diffusion and a hydrogen concentration gradient at the crack tip that are controlled by temperature and yield strength, respectively. The activation energy of DHCV in the Zr-2.5Nb tubes is discussed on the basis of temperature dependency of hydrogen diffusion and the striation spacing.

7α,25-Dihydroxycholesterol-Induced Oxiapoptophagic Chondrocyte Death via the Modulation of p53-Akt-mTOR Axis in Osteoarthritis Pathogenesis

  • This study aimed to exploring the pathophysiological mechanism of 7α,25-dihydroxycholesterol (7α,25-DHC) in osteoarthritis (OA) pathogenesis. 7α,25-DHC accelerated the proteoglycan loss in ex vivo organ-cultured articular cartilage explant. It was mediated by the decreasing extracellular matrix major components, including aggrecan and type II collagen, and the increasing expression and activation of degenerative enzymes, including matrix metalloproteinase (MMP)-3 and -13, in chondrocytes cultured with 7α,25-DHC. Furthermore, 7α,25-DHC promoted caspase-dependent chondrocyte death via extrinsic and intrinsic pathways of apoptosis. Moreover, 7α,25-DHC upregulated the expression of inflammatory factors, including inducible nitric oxide synthase, cyclooxygenase-2, nitric oxide, and prostaglandin E2, via the production of reactive oxygen species via increase of oxidative stress in chondrocytes. In addition, 7α,25-DHC upregulated the expression of autophagy biomarkers, including beclin-1 and microtubule-associated protein 1A/1B-light chain 3 via the modulation of p53-Akt-mTOR axis in chondrocytes. The expression of CYP7B1, caspase-3, and beclin-1 was elevated in the degenerative articular cartilage of mouse knee joint with OA. Taken together, our findings suggest that 7α,25-DHC is a pathophysiological risk factor of OA pathogenesis that is mediated a chondrocyte death via oxiapoptophagy, which is a mixed mode of apoptosis, oxidative stress, and autophagy.
