Inhibitory effects of grapefruit seed extract(DF-100) on growth and toxin production of Penicillium islandicum (자몽종자 추출물(DF-100)이 Penicillium islandicum생육 및 독소 성분 skyrin생합성에 미치는 저해효과)

  • The anthraquionone mycotoxin, skyrin, is produced by Penicillium islandicum. DF-100 which was extracted from grapfruit seed extract and is a natural organic complex inhibited the biosynthesis of skyrin by Penicillium islandicum. This study was carried out to determine the potential of DF-100 to support Penicillium islandicum and skyrin production. DF-100 inhibited the growth of the fungus at 750ppm or less and caused complete inhibition of skyrin production at 500ppm or less. DF-100 appears to block the incorporation of emodinathrone into skyrin and an enzymatic step in the skyrin biosynthetic pathway which lies before skyrinanthrone.

  • Chicken skins or carcasses inoculated with Salmonella typhimurium were exposed to 0.1 or 0.5% grapefruit seed extracts (DF-100) for 1 or 3 min to evaluate antibacterial activity of DF-100 and its possible application in proultry processing. The numbers of live salmonellae on chicken skins were reduced by 0.8-1.2 logs/cm2 with 0.5% DF-100. Dipping chicken carcasses into 0.5% DF-100 for 3 min reduced salmonelae by 4.3 logs/carcass. Scanning electron microscopy showed that DF-100 killed the cells attached but did not detach cells from the skin. No odor or changes in the color of chicken skin were detected after DF-100 treatment.

Optimization of Membrane Separation Process for the Production of Dietary Fibers from Tangerine Peels (감귤 과피 유래 식이섬유 생산을 위한 막분리 공정 최적화)

  • Dietary fibers (DF) have been used as functional food components due to the various health promoting activities. Dietary fibers have been separated from the peels of Korean tangerine by employing ultrafiltration (UF) membranes. Optimum conditions in a batch type ultrafiltration unit using YM100 (molecular weight cut-off, MWCO=100,000), YM 10 (MWCO=10,000) and YM1 (MWCO=1,000) membranes were : transmembrane pressure 7.5 psi, temperature of the peel extracts $35^{\circ}C$, and pH of the peel extract 3.0, respectively. The flux in YM 10 membrane unit was higher than that in YM 10 or YM 1 membrane unit. However, YM 100 membrane was superior to YM 10 or YM 1 membrane with respect to the recovery of the retentate and the contents of DF The contents of DF in the tangerine peel extract, in the 170 mesh retentate, and in the YM 100 retentate were shown to be 33.4%, 18.5% and 8.4% based on dry matter, respectively. Most dietary fibers were recovered at the separation stages of 170 mesh and YM 100.

Effects of Additives on the Quality and Storagebility of soondae (순대 제품의 품질 및 저장성에 미치는 첨가제의 영향)

  • Soondae is one of the traditional food in Korea. In order to develop the quality of Soondae, Mulberry leaf, safflower, DF-100 and Chitosan were added and their effect on the property change of Soondae during storage periods were investigated. There was no significant difference in proximate compositions among samples. TBA value of all sample increased as storage periods and TBA value of Soondae with DF-100 was lowest during storage periods. The content VBN of Soondae with was lowest during the storage periods. Microbiology was inhibited by adding the DF-100 and Chitosan at Soondae. Adding Chitosan of Soondae was highest point ati Sensory test. 18:1 content of lipid acid composition was the highest among samples.

Effect of Dietary Fiber Levels on The Production Parameters in Colored Broiler Chicks (식이섬유 수준에 따른 유색육용계의 능력과 도체특성)

  • This experiments was conducted to determine the effect of dietary fiber(DF) levels with diets of isocaloric (3,010 kcal/1kg diet) and isoprotein(21% of diet) on colored brolier chicks(Juk jun hatchery farm). Four groups of 6 chicks 4 replication. were fed corn-soy meal diets containing 0% (DF, 5%), 2%(DF, 6%), 4%(DF, 7%), and 6%(DF, 8%) of dehydrated alfalfa meal(AM) from 21 to 42 days of age. The addition of dietary fiber levels to the broiler chicks diets did noty affect body weight, feed efficiency, energy efficiency and protein efficiency, but affect feed intake of chicks fed 6% of fiber. The level of fiber diets did not affect gizzard weight of female(1.45g/100g BW) and male(1.25g/100g BW), but affect small intestine length of female. When comparing two the dietary fiber levels, 5% and 8%, the contents of the total serum cholesterol was slightly higher in chicks fed 5% of fier than that of 8% without significant. However, total serum cholesterol levels of female(121mg/dl) and male(119mg/dl) were not influenced by increased dietary fiber levels. The liver weight was not influenced by increasing of dietary fiber levels (female 2.03g and male 2.05g/100g BW). The yields of dressed weight(72% of shrunk body weight), skin weight)11% of dressed weight) were not influenced by increased dietary fiber.

  • For the analytical method of PCDD/Fs in blood, which have been issued recently, SRM 1589a of NIST(National Institute of Standards and Technology) was used and a practical analytical method of PCDD/Fs in blood was presented through comparison of methods of Canada and Japan. The proposed method used alkali-digestion extraction for removal of the lipid effectively using two capillary columns. The limit of quantification of TeCDD/DF and PeCDD/DF was 1 pg/g-lipid, HxCDD/DF and HpCDD/DF was 2 pg/g-lipid, OCDD/DF was 4 pg/g-lipid. With consideration the range of detected concentration, calibration standards were presented as $CS_L$(0.1~1), $CS_1/2$(0.25~2.5), $CS_1$(0.5~5.0), $CS_2$(2~20), $CS_3$(10~100).

Prevention of Quality Changes in the Cultured Wild Ginseng During Storage (산삼배양근의 저장 중 품질변화 억제)

  • Physicochemical changes were investigated for the shelf-life extension of cultured wild-ginseng roots during storage with various pre-treatments with blanching, CAMICA-SD and DF-100 and treatments with citric acid and vitamin C. The pH of cultured wild-ginseng roots showed the range of $6.06{\sim}6.42$ at $10^{\circ}C$, but showed higher ranges of $6.08{\sim}6.91$ and $6.08{\sim}8.68$ at 20 and $30^{\circ}C$, respectively. Browning index (a/b) was increased with increasing storage temperature, and the index at 10 and $30^{\circ}C$ were 0.405 and 0.469 after 2 weeks, respectively. Browning index and viable cell number of CAMICA-SD pre-treatment showed little changes compared to pre-teatment with blanching or DF-100. When the cultured wild-ginseng roots were treated with 1.0% citric acid and 0.2% DF-100 after pre-treatments with CAMICA-SD, viable cell number was slightly increased to $4.9{\times}10^2CFU/g$ for 3 weeks storage at $10^{\circ}C$. The mixture of citric acid and DF-100 was also used to prevent the growth of microbiology and to reduce browning reaction, especially enzymatic browning reaction. The mixture might effectively extend shelf life of the cultured wild-ginseng roots.

A study on pressurizer cutting scenario for radiation dose reduction for workers using VISIPLAN

  • The operations in the design lifecycle of a nuclear power plant targeted to be decommissioned lead to neutron activation. Operations in the decommissioning process include cutting, decontamination, disposal, and processing. Among these, cutting is done close to the target material, and thus workers are exposed to radiation. As there are only a few studies on pressurizers, there arises the need for further research to assess the radiation exposure dose. This study obtained the specifications of the AP1000 pressurizer of Westinghouse and the distribution of radionuclide inventory of a pressurizer in a pressurised water reactor for evaluation based on literature studies. A cutting scenario was created to develop an optimal method so that the cut pieces fill a radioactive solid waste drum with dimensions 0.571 m × 0.834 m. The estimated exposure dose, estimated using the tool VISIPLAN SW, in terms of the decontamination factor (DF) ranged from DF-0 to DF-100, indicating that DF-90 and DF-100 meet the ICRP recommendation on exposure dose 0.0057 mSv/h. At the end of the study, although flame cutting was considered the most efficient method in terms of cutting speed, laser cutting was the most reasonable one in terms of the financial aspects and secondary waste.

PLAQUE ADHESION ON THE SURFACES OF VARIOUS COMPOSITE RESIN (수종 복합레진에 대한 치태 부착도 비교)

  • The surface characteristics of restoration such as surface roughness and droplet contact angle are important part for the process of bacterial adhesion. The purpose of this study is to compare plaque adhesion by measuring roughness, droplet contact angle, and amount of accumulated plaque on the surfaces of composite resins. Four kinds of composite resins, Z-100(Z1), Durafil(DF), Filtek supreme(FS), Clearfil AP X(CA) were used. Ten samples were divided into unpolished and polished group. Surface roughnesses and droplet contact angles were measured by profilometer and goniometer. Plaque weight gains are measured. The results were as follows: 1. The experimental group were rougher than the control group. Surface roughnesses were decreased in the following order; (Z1, DF, CA)>FS in the control group, and CA>Z1>(FS, DF) in the experimental group(P<0.05). 2 The control group showed larger contact angle than the experimental group. Contact angles were decreased in the following order; CA>(FS, DF, Z1) in the control group, and (CA, DF)>(FS, Z1) in the experimental group(P<0.05). 3. The experimental group showed more much plaque than the control group. The amounts of plaque accumulation in vitro were decreased in the following order; Z1>(DF, FS)>CA in the control group, and Z1>FS>(CA, DF) in the experimental group. The latter showed more much plaque than the former(P<0.05). 4. There were stronger correlation between plaque deposition and contact angle (P<0.05) than that of plaque deposition and surface roughness.

Characterization of an Amylase-sensitive Bacteriocin DF01 Produced by Lactobacillus brevis DF01 Isolated from Dongchimi, Korean Fermented Vegetable

  • A DF01 strain that inhibits tyramine-producing Lactobacillus curvatus KFRI 166 was isolated from Dongchimi, a traditional Korean fermented vegetable, and identified as Lactobacillus brevis by biochemical analysis and reverse transcriptase sequencing of 16S rRNA. The antimicrobial compound produced by L. brevis DF01 was secreted at a maximum level of 640 AU/mL in late exponential phase in MRS broth, and its activity remained constant during stationary phase. The activity of bacteriocin DF01 was totally inactivated by $\alpha$-chymotrypsin, pronase E, proteinase K, trypsin, and $\alpha$-amylase, but not by catalase, which indicates the compound was glycoprotein in nature. The activity was not affected by pH changes ranging from 2 to 12 or heat treatment (60, 80, and $100^{\circ}C$ for 30 min), but was reduced after autoclaving. Bacteriocin DF01 had bacteriolytic activity and a molecular weight of approximately 8.2 kDa, as shown by tricine-SDS-PAGE analysis. Therefore, bacteriocin DF01 can be used in the manufacture of fermented meat products due to its inhibition of tyramine-producing L. curvatus and non-inhibition of L. sake, which is used as a starter culture for meat fermentation.
