Improving Texture and Storage Stability of Chinese-Style Pork Jerky by the Addition of Humectants

  • Chinese-style pork jerky with different levels (3, 6 and 9%) of glycerol or sorbitol were prepared. Moisture content, water activity ($a_w$) and shear value decreased with the addition of glycerol or sorbitol. During storage at $28^{\circ}C$ thiobarbituric acid (TBA) value of all samples declined with the addition of glycerol or sorbitol during storage time. After storage at room temperature ($28^{\circ}C$) for 180 days, the volatile basic nitrogen (VBN) of all treated products were less than the control group. In addition, the mold and yeast growth were inhibited slightly with the addition of glycerol or sorbitol. The absorption isotherm of pork jerky with different levels of glycerol at $25^{\circ}C$ had an aw above 0.75, and moisture contents at 6 and 9% which were higher than the control group. The moisture content was less than the control group with the addition of sorbitol. The chewiness desirability score of pork jerky with 6% glycerol was higher than the other treatments. However, no differences in hardness desirability score due to treatments were detected by sensory panelists.

Fed-Batch Sorbose Fermentation Using Pulse and Multiple Feeding Strategies for Productivity Improvement

  • Microbial oxidation of D-sorbitol to L-sorbose by Acetobacter suboxydans is of commercial importance since it is the only biochemical process in vitamin C synthesis. The main bottleneck in the batch oxidation of sorbitol to sorbose is that the process is severely inhibited by sorbitol. Suitable fed-batch fermentation designs can eliminate the inherent substrate inhibition and improve sorbose productivity. Fed-batch sorbose fermentations were conducted by using two nutrient feeding strategies. For fed-batch fermentation with pulse feeding, highly concentrated sorbitor (600g/L) along with other nutrients were fed intermittently in four pulses of 0.5 liter in response to the increased DO signal. The fed-batch fermentation was over in 24h with a sorbose productivity of 13.40g/L/h and a final sorbose concentration of 320.48g/L. On the other hand, in fed-batch fermentation with multiple feeds, two pulse feeds of 0.5 liter nutrient medium containing 600g/L sorbitol was followed by the addition of 1.5 liter nutrient medium containing 600g/L sorbitol at a constant feed rate of 0.36L/h till the full working capacity of the reactor. The fermentation was completed in 24h with an enhanced sorbose productivity of 15.09g/L/h and a sorbose concentration of 332.60g/L. The sorbose concentration and productivity obtained by multiple feeding of nutrients was found to be higher than that obtained by pulse feeding and was therefore a better strategy for fed-batch sorbose fermentation.

백나무로부터 분리된 quercetin-3-O-$\alpha$-L-rhamnopyranoside의 알도스 환원효소 및 솔비톨 억제효과 (Inhibitory Effect of quercetin-3-O-$\alpha$-L-rhamnopyranoside from Chamaecyparis obtuse on Aldose Reductase and Sorbitol Accumulation)

  • Taxifolin-3-O-$\beta$-D-xylopyranoside and quercetin-3-O-$\alpha$-L-rhamnopyranoside were isolated from an EtOAc-soluble extract of the leaves of Chamaecyparis obtuse. Quercetin-3-O-$\alpha$-L-rhamnopyranoside was found to possess a potent inhibitory activity of human recombinant aldose reductase in vitro, its $IC_{50}$ value being $11.5\;{\mu}M$. Kinetic analysis showed that quercetin-3-O-$\alpha$-L-rhamnopyranoside exhibited uncompetitive inhibition against DL-glyceraldehyde. Also, quercetin-3-O-$\alpha$-L-rhamnopyranoside suppresses sorbitol accumulation in rat lens under high glucose conditions, demonstrating the potential to prevent sorbitol accumulation in vivo. These results suggest that this compound may be a promising agent in the prevention or treatment of diabetic complications.

고정화균체 반응기에서 L-Sorbose 연속생산

  • The conversion of D-sorbitol to L-sorbose by Gluconobater suboxydans was analyzed, and continuous production of L-sorbose was carried out in immobilized cell reactors. L-Sorbose production by high densities of resting cells was more effective than by conventional batch fermentations. Sorbitol dehydrogenase, an enzyme converting D-sorbitol to L-sorbose, did not suffer from substrate inhibition, but from product inhibition. When L-sorbose production was carried out with Ca-alginate-immobilized cells, about 60 g/l of L-sorbose was obtained. On the other hand, when the corn steep liquor (CSL) concentration of medium was reduced to 0.08%, 80 g/l of L-sorbose was obtained. Outgrowth inside the immobilized carriers was thought to block the pores of the carriers so that substrate could not easily diffuse through the carriers. Continuous production of L-sorbose was well accomplished in a bubble column reactor, and 6. 5 g/l.h of productivity and 81.2% of yield were obtained at a substrate feeding rate of 0.08h$^{-1}$ under the optimum conditions with carrier volume of 55% and aeration rate of 3 vvm.

Empirical Force-Field 방법에 의한 D-Sorbitol 의 구조와 에너지에 관한 연구 (The Structure and Energy of D-Sorbitol from an Empirical force-Field)

  • Empirical force-field 방법을 중성자 회절 방법으로 분자구조가 밝혀진 D-sorbitol, CH_2OH\;(CHOH)_4\;CH_2OH$에 적용하여 비교 연구하였다. C-C결합 길이는 계산값과 실험값이 0.009${\AA}$ 내에서 C-O 결합 길이는 0. 023${\AA}$내에서 일치하는 좋은 결과를 얻었다. C-C-C 와 C-C-O 결합 각도는 각각 $2.3^{\circ}$$1.9^{\circ}$이내에서 일치하였으나 crystal packing force의 영향을 많이 받고 있는 torsion angle은 상당한 차이가 있었다. C(1)-C(2)-C(3)-C(4) torsion angle을 규칙적으로 변화시키면서 steric 에너지를 연구한 결과 분자의 최저 에너지는 ${\phi}$=+$90^{\circ}$부근에서 나타나 회절법에 의한 결정 구조와는 다른 결과를 얻었다. 그러나 C-O, O-H, O-lone-pair dipole들 간의 상호작용을 무시하면 ${\phi}$=-$60^{\circ}$부근에서 최저에너지를 나타내어 결정구조와 일치 하였다. Empirical force-field 방법의 dipole 상호작용에 관한 에너지항을 개선하면 더 좋은 결과를 얻을 수 있을 것으로 예측 한다.

Modified sorbitol MacConkey agar for the rapid isolation of Escherichia coli O157:H7

  • Unlike most Escherichia coli strains, E coli O157 : H7 didn't ferment sorbitol within 24h of incubation and showed a negative reaction for $\beta$-glucuronidase. We developed a new medium for the rapid isolation of E coli O157 : H7 using sorbitol MacConkey agar with cefixime, potassium tellurite and 4-methylumbelliferyl-${\beta}$-D-glucuronide (MUG) as a primary plating medium. The addition of $20{\mu}g/ml$ of vancomycin in enrichment broth for E coli O157 : H7 inhibited lots of Gram positive bacteria. Three strains (10.3%) of 29 non-O157 E coli strains and 3 strains (8.3%) of 36 Salmonella spp were inhibited at the $0.05{\mu}g/ml$ of cefixime and 23 strains (79.3%) of 29 non-O157 E coli strains and 12 strains (33.3%) of 36 Salmonella spp were inhibited at the $2.0{\mu}g/ml$ of potassium tellurite. But none of the E coli O157 : H7 was affected at these concentration. The addition of MUG at $100{\mu}g/ml$ level to sorbitol MacConkey agar with cefixime and potassium tellurite (CTM-SMAC) aided in the rapid isolation of E coli O157 : H7 from samples by checking sorbitol-negative and $\beta$-glucuronidase negative phenotypes simultaneously. In conclusion, inoculation of a positive in the O157 screening test from enrichment broth on CTM-SMAC appeared to be a rapid, cost-effective and sensitive method for the isolation of E coli O157 : H7.

배당화 메타크릴레이트와 아크릴에리트를 이용한 하이드로겔의 합성: I. 솔비탄 아크릴레이트의 화학.효소적 합성에 관한 연구 (Hydrogel Synthesis using Glycosyl Methacrylate and Acrylate: 1. A Study on Chemo-Enzymatic Synthesis of Sorbitan Acrylate)

  • 당알콜인 D-sorbitol을 화학 촉매 p-toluenesulfonic acid(p-TSA)를 이용하여 탈수반응을 수행하여 고리구조의 1,4-sorbitan을 제조 후 효소적으로 고정화 리파제 Novozym 435를 이용하여 아크릴산과 에스테르화 반응을 수행한 결과 아래와 같은 결론을 얻었다. D-Sorbitol을 p-TSA를 화학촉매로 하여 탈수반응을 통한 고리화 반응의 생성물인 1,4-sorbitan의 최대수율을 얻을 수 있는 최적조건은 1% (w/w)의 p-TSA를 이용하여 200 mmHg의 감압조건에서 13$0^{\circ}C$에서 150분 반응 후 약 90%의 1,4-sorbitan을 얻을 수 있었으며, 부산물과 미반응의 D-sorbitol의 양을 최소화시킬 수 있었다. 화학적 촉매반응의 생성물로 얻은 1,4-sorbitan을 고정화 리파제 (Novozym 435)를 이용하여 반응매질 t-butanol에서 아크릴산과의 에스테르화 반응을 수행한 결과, 5$0^{\circ}C$에서 고정화효소는 최대 활성을 보였으며, 최적 첨가량은 3% (w/v)이었다. 또한 초기농도가 반응의 전환율에 미치는 영향을 조사한 결과 1,4-sorbitan 50 8/L의 초기농도에서 55.8%의 최대 전환율을 얻었으며, 63.5%의 반응수율을 보인 1:3의 몰비가 최적으로 조사되었다.

Enhancing the Biological Control of Rice Seedling Disease by Adding Specific Carbon Sources into the Bacillus cereus D324 Formulation in Water-Seeded Rice

  • Utilization of carbon sources by Bacillus cereus D324, a biological control agent, and Pythium species, which causes rice seedling disease, was studied with the objective of increasing the efficacy of biological control by providing the biological control agent with specific beneficial carbon sources. D-galactose, D-sorbitol, and D-mannitol were poor carbon sources for Pythium spp. growth but were good for B. cereus D324 growth. Growth in a growth chamber of rice seeds coated with B. cereus D324 amended with specific carbon sources, such as D-galactose and D-sorbitol, showed significantly enhanced seedling emergence compared to seeds coated only with B. cereus D324. Field trials showed that both seedling emergence and yield increased, when the above specific carbon sources were added to B. cereus D324 in seed coating formulations. This result indicated that amending seed coating formulations with specific carbon sources could significantly increase seedling emergence and yield in the field.

가공조건이 alginate 필름의 물리적성질에 미치는 영향 (Effects of processing conditions on physical properties of alginate film)

  • 생분해성으로서 가식성인 alginate 필름을 가공하기 위한 기초자료로써 $3\%$ $Na_2CO_3$ 추출시간 5시간으로 추출한 alginate로 만든 alginate 필름의 수증기 투과율과 장력성에 영향을 미치는 alginate농도와 가소제의 종류별 첨가량에 관한 조건을 측정한 결과는 다음과 같다. Alginate의 농도가 증가할수록 인장강도는 증가하였으나 수증기 투과율과 신장도는 감소하였다. 반면에 가소제의 첨가량이 증가할수록 수증기 투과율과 신장도는 증가하였으나 인장강도는 감소하였다. 가소제의 종류별에 따른 수증기 투과율은 glycerol, polyethylene glycol 및 sorbitol 첨가 필름 순으로 높게 나타났으며, 인장강도는 polyethylene glycol, sorbitol, glycerol 첨가 필름 순서이었으나 신장도는 그 반대되는 결과였다. 가소제의 종류와 필름의 색은 sorbitol 첨가 필름의 색이 가장 자았고, 황색도는 glycerol 첨가 필름이 가장 높았고, 적색도는 polyethylene glycol 첨가 필름이 가장 높았다. 그리고 전체적인 색의 차이에서는 polyethylene glycol 첨가 필름이 가장 높았다.

  • 고온에서 고농도 기질을 발효할 수 있는 효모를 육성하고자 8%의 NaCl 내성을 갖은 고온발효성인 Saccharomyces cerevisiae D-71 과 Ca-pantothenate 를 절대요구하는 내삼투압성인 Zygosaccharomyces rouxii SR-S를 친주로 하여 원형질체 형성과 융합조건을 검토하였다. Sacch. cerevisiae D-71은 YEPG 배지에 12시간 배양한 세포를 0.1mM EDTA와 0.5M sorbitol를 함유한 인산완충액(pH7.5)에 현탁시켜 1%의 2-mercaptoethanol로 10분간 전처리한 후 Zymolyase-20T 4.0mg/$m\ell$를 가하여 3$0^{\circ}C$에서 50분간 반응시켰을 때 96% 이상의 수율로 윈형질체가 형성되었고 Zygosacch. rouxii SR-S는 YEMEPG 배지에 24시간 배양한 세포를 0.1mM EDTA와 0.5~l.0M mannitol을 함유한 인산완충액(pH7.0)에 현탁시켜 1%의 2-mercaptoethanol로 10분간 전처리 한 후 Zymolyase-20T 4.0mg/$m\ell$를 가하여 3$0^{\circ}C$에서 120분간 반응시켰을 때 95% 정도의 수율로 원형질체가 형성되었다. 시험효모의 원형질체를 CaCl$_2$와 Sorbitol 및 40% 의 PEG 4000을 함유한 융합배지에 1 : 1로 혼합한 후 35$^{\circ}C$에서 20분간 처리하여 융합시킨 다음 3%의 한천을 함유한 최소배지에 중층하여 5일간 배양하였을 때 9.1$\times$$10^{-6}$의 융합빈도수를 보였다. 또한 주사형전자현미경으로 구형의 원형질체와 융합재생된 타원형의 세포를 관찰할 수 있었다.
