Larvae Growth and Biochemical Composition Change of the Pacific Oyster Crassostra gigas, Larvae during Artificial Seed Production (참굴 Crassostrea gigas 인공종묘생산 시 유생의 성장과 체성분 변화)

  • A nutritional demand of oyster, Crassostrea gigas larva as part of research for improving of utilization of microalgae being used for the artificial oyster seed production. The change of body growth and biochemical compositions of larvae were investigated during larvae rearing in hatchery. The larvae were cultured in 60 M/T tank and fed mixture 6 different phytoplankton species, Isochrysis galbana (30%), Cheatoceros gracilis (20%), Pavlova lutheri (20%), Phaeodactylum triconutum (10%), Nannochryis oculata (10%) and Tetraselmis tetrathele (10%). The initial feeding amount was $0.3{\times}10^4cells/mL$ at three times a day to D-shaped larva and the feeding amount had been increased 30% gradually every two day since the larvae were raising. The larvae were developed from D shape to pediveliger stage for 12 days. The daily growth of shell length and hight were $5.8{\sim}30.8\;{\mu}m$ and $8.7{\sim}31.4\;{\mu}m$, respectively and weight gains were changed from D shape to pediveliger as follow: wet weight was $0.52{\sim}15.0\;{\mu}g/larva$, dry weight was $0.2{\sim}6.5\;{\mu}g/larva$, and ash free dry weight was $0.1{\sim}8.5\;{\mu}g/larva$. The larvae growth pattern shown a logarithmic phase from D shape to umbone stage but after that stage shown a exponential growth aspect. The crude protein, crude lipid and nitrogen free extract (NFE) of larvae during rearing periods were analyzed as $6.1{\sim}10.6%$, $0.6{\sim}1.1%$ and 1.0-2.7%, respectively. And the total amino acid contents of the larvae during rearing periods were in order as glutamic acid $1.26{\sim}2.24%$, aspartic acid $0.97{\sim}1.70%$, and methionine $0.12{\sim}0.33%$. Of the total fatty acid in the analyzed larvae, the saturated fatty acid (SSAFA) was decreased from 54.3% (D shaped larvae) to 17.1 % (pediveliger) as larvae development but the total mono-unsaturated fatty acid (${\Sigma}MOFA$) and Poly-unsaturated fatty acid (${\Sigma}PUFA$) were increased from 29.9% and 7.8% to 40.6% and 45.6%, respectively. By the way the each fatty acid of the larvae were composed of palmitic acid $9.89{\sim}36.95%$, oleic acid $12.17{\sim}32.29%$, linoleic acid $1.96{\sim}33.55%$, EPA $2.17{\sim}11.58%$ and DHA $1.95{\sim}4.51%$. As a result of this study, the larvae of oyster were demanded a various nutrients for healthy growth and the feeding control, expecially after umbone stage larvae are a rapidly growing time, is very important for success of artificial seed production.

  • The maturity of gonads, early development of the fertilized egg, spawning period, and morphogenesis of larvae in Limnoperua fortunei were studied from October, 1981 to November, 1982 as on attempt to identify its life cycle. 1) Active motility of its sperm was observed at late May, and matured egg could be found at late June. 2) It was estimated that the spawning was occured from late August. This species was belong to the short-term breeder because it finished its spawning within 20 days. 3) It has free-living trochophore and D-shaped larva stage. The shell lengths of early, middle, and D-larval stage were $140.0{\mu}m$, $167.6{\mu}m$ and $210.0{\mu}m$, respectively. The shell heigths of each stage were $97.3{\mu}m$, $137.6{\mu}m$ and $178.2{\mu}m$, respectively.

  • This study was performed to find out the optimal kind of additive and concentration of ethylene glycol (EG) as the cryoprotectant on the D-shaped larvae of arkshell, Scapharca broughtonii. The larvae in straws was carried in the programed freezer set uo at 0$^{\circ}C$, frozen to -12$^{\circ}C$ by the freezing rate of -1$^{\circ}C$/min, held for 10 minutes, seeded at -12$^{\circ}C$, finally refrozen to -35$^{\circ}C$ by the freezing rate of -1$^{\circ}C$/min and then directly plunged into liquid nitrogen. The survival of larvae frozen in 1.0 M and 2.0 M ethylene glycol added with 0.5 M sucrose were high (50.5 ${\pm}$ 1.3% and 51.9 ${\pm}$ 1.7%, respectively) in the early D-shaped larvae cryopreserved in 1 M and 2 M ethylene glycol diluted with 0.2 M and 0.5 M fructose, glucose and sucrose. In the late D-shaped larvae were cryopreserved according to five concentrations of ethylene glycol added with 0.5 M sucrose, the survival of larvae frozen in 2.0 M ethylene glycol was the highest as 51.9 ${\pm}$ 1.7%. The morphological differences in cells between unfrozen and frozen-thawed D-shaped larvae were not found by the scanning and transmission electron microscopy.

  • We examined the effects of serotonin on the spawning response, sperm motility, and D-shaped larva production in Eastern Gooeyduck clam Panopea sp. based on the sperm densities at fertilization and washing after mixing the eggs and sperm. The highest spawning induction was found showed in females and males injected with 1 mL of 2 mM serotonin. The spawning responses in females and males were higher at concentrations greater than 1 mM and 0.75 mM, respectively. Regarding the activities of sperm in sea water after serotonin injection, the sperm showed activity at >90.0% until 120 mins. We also examined the effects of sperm concentration at the fertilization and washing times after mixing the eggs and sperm. We confirmed that washing within 1 minute at a concentration of 1,500 sperms/mL or less can prevent egg destruction by polyspermy and secure a large number of D-phase larvae. These results should be useful for developing the aquaculture process for Eastern Gooeyduck clam, Panopea sp.

Spawning Inducement, Egg Development and Early Larval Rearing of Ark Shell (Tegillarca granosa) (L.) (꼬막 (Tegiilarca granosa) (Linngeus)의 산란유발 및 난 발생과 초기 유생 사육)

  • Spawning induction, egg development and larval growth of ark shell (Tegillarca granosa) (L.) were investigated. The most effective method of spawning induction was steady temperature increasing from$4^{\circ}C\;to\;28^{\circ}C$ with irradiation of sea water by UV after T. granosa was exposed to air at $4^{\circ}C$ Optimum condition for larval roaring was under the 32.4 psu and two temperature $regimes:\;28{\pm}1^{\circ}C\;and \;25{\pm}1^{\circ}C$. Fertilized eggs was demersal isolated eggs, and egg diameter was $60{\mu}$. D-shaped larvae appear about 20 hr after hatching with $94.1{\mu}$ in shell length and $86.7{\mu}$ in shell height. Ten days were required from hatching to umbo larva stage, of a mean shell length $125.2{\mu}$. On 25th day, the larva grew to $450{\mu}$ in shell length and began to settle on the bottom. Effect of temperature between $25^{\circ}C$ (control group) and $28^{\circ}C$ on larval growth was not different. Survival rate of larvae settled on the bottom was about $19{\%}$ in both temperatures conditions $(25^{\circ}C\;and\;28^{\circ}C)$.

Biological Studies on Aquaculture for Resources Enhancement of Protothaca jedoensis I. Egg Development and Larva Reared (살조개, Protothaca jedoensis 자원조성을 위한 양식생물학적 연구 I. 난발생과 유생사육)

  • In order to obtain the aquaculture fundamental data far resources enhancelnent of the Protothaca jedoensis, the egg development and larva growth were investigated at different conditions such as water temperature, phytoplankton and density. Water temperature, at which P. jedoensis egg successfully completed development, ranged from $15{\~}30^{\circ}C$. The required time from fertilization to D-shaped larva was 39.7 hours at $15^{\circ}C$, 31.2 hours at $20^{\circ}C$, 26.8 hours at $25^{\circ}C$ and 26.2 hours at $30^{\circ}C$ P. jedoensis. In regard to water temporature, growth and survival rates of larvae were high at $30^{\circ}C$. In growth and survival rates of larvae with various rearing densities, the highest aver-age growth and survival rates were 4${\~}$6 ind./ml When larvae were fed mixed phytoplankton, such as Isochrysis galbana, Pavlova lutheri and Chaetoceros calcitrans, their growth and survival rates were the high among the groups. In growth and survival rates of larvae with various rearing food concentrations, the highest average growth and survival rates were $218{\mu}m$, and $45\%$ at the food concentration of $1{\times}10^4$ cells/ml, respectively.

  • Water temperature, at which Spisula sachdinensis egg successfully completed development ranges from 10 to $25^{\circ}C$. Within this range, the development duration is reduced with increasing temperature. The relationships between temperature (WT, $^{\circ}C$) and the required time (t: hour) from egg to each developmental stage are described as follows: 2-cell : 1/t = 0.0554 WT -0.3769; 8-cell : 1/t = 0.0210 WT -0.1142; Morula : 1/t = 0.0125WT -0.0712; Trochophore : 1/t = 0.0039 WT -0.0203; D-shaped larva : 1/t = 0.0024W T -0.0108. Biological minimum temperature for the egg development is 5.5$^{\circ}C$.

  • Infection rates of oyster ovarian parasite, Marteiliodes chungmuensis and productivity of the oyster shellstock infected with the parasite were investigated at the main seed collection areas in the southern coast of Korea where the extreme bad seed collection of oyster occurred in 1992 and 1993 to evaluate the cause of the bad seed collection. Additionally, the bacterial flora of the sea water and oyster lana were examined to identify the shellfish larva pathogenic bacteria like Vibrio sp. and Pseueomonas sp. In August 1992 to September 1993, infection rate of oyster ovarian parasite, M. chungmuensis at Tongyong, Kyongsangnam province, and Yosu, Chollanam province where the bad seed collection occurred, were $11.8\~100\%$ and $14.3\~100\%$, respectively. But the parasite was not detected in the shellstock collected at Daechon, Chungchongnam province. While a virus-like particle was identified in the cytoplasm of the egg infected by the parasite. The parasite infected egg was not able to fertilize completely. Uninfected egg in the gonad contaminated by the parasite could be able to fertilize but showed an abnormal development till D-shaped larva and then, died of necrosis after D-shaped lana. And some lana developed from low lipid content egg could not develop to the spat and died after the early umbo stage. The predominant bacteria in the oyster lana collected at bad seed collection areas were Pseudomonas sp. and Pseudomonas like bacteria and the occupancy rates were $53.3\~87.1\%$.

  • We studied to find out the effect of water temperature on the egg development of pearl oyster, Pincata fucata martensii and Pacific oyster, Crassostrea gigas. The optimum water temperatures for egg development were $20{\~}25^{\circ}C$ in P. fucata martensii and $15{\~}25^{\circ}C$ in C. gigas. The required time from fertilization to D-shaped lana was $41.7\;hours\;at\;20^{\circ}C$ and 27.5 hours at $25^{\circ}C$ in P. fucata martensii, and 35.3 hours at $15^{\circ}C$, 26.3 hours at $20^{\circ}C$ and 17.6 hours at$ 25^{\circ}C$ in C. gigas, respectively. Artificial Spawning, Larval and Spat Developments of the Bay Scallop, Argopecten irradians (해만가리비, Argopecten irradians의 인공산란과 유생 및 치패의 발생)

  • Artificial spawning, larval and spat developments of the bay scallop, Argopecten irradians, which was transplanted from China on 16 August 1996, were investigated monthly until August 1997 in the Deukyang Bay, Jangheung-gun, Jeollanam-do, Korea. Sufficient amount of cultured microalgae supplemented seawater were supplied as food (6 kinds of phytoplankton) for fully grown adult individuals at 17.1-23.2$^{\circ}C$ for 44 days. A total of 45,320,000 eggs were spawned by way of 2 times of artificial spawning inductions such as exposure stimulus to the air and thermal shock (with water temperature) on 29 January and 31 January in 1997. Artificially fertilized eggs were developed to D-shaped larvae (77.5 ${\times}\;63.8\;{\mu}m)$ and metamorphosed to larvae (191.8 ${\times}\;181.2\;{\mu}m)$ 181.2 m) in the attached larval stage on the collectors. A total of 110,000 spats (average 3.04 mm in shell length) were produced at 22.8-26.3$^{\circ}C$ and 31.0-34.4 psu in the indoor rearing tank from 14 February through 7 May in 1997. In case of Argopecten irradians, if the attached larvae in the attachment stage are detached from the collector, they could not live. Accordingly, it is assumed that survival (%) of the attached larvae of A. irridians showed very low because of weak power attached to the collector due to the small number of the byssuses of the attached larva, not the short attachment period by the byssus as seen in other scallops such as Argopecten balloti.
