Attenuating Effects of N-acetylcysteine (NAC) against Fish Parasiticide-induced Toxicity in Carp Cyprinus carpio (잉어(Cyprinus carpio)에서 어류 구충제에 대한 N-acetylcysteine(NAC)의 독성 저감 효과)

  • This study examined whether N-acetylcysteine (NAC), a glutathione precursor, could attenuate toxic effects of three fish anti-parasitic agents, trichlorfon, hydrogen peroxide ($H_2O_2$) and formalin, all of which are known to exert side effects through free radical production. Common carp Cyprinus carpio were fed with NAC (approx. 50 mg/kg/day) for 3 consecutive days prior to anti-parasite bathing for a 24 hr period. Mortality rates were examined during this 24 hr bathing period, and selected hematological and biochemical parameters were also assessed at the termination of anti-parasite exposure. The mortality rates and plasma glucose elevations caused by all three anti-parasitics were significantly reduced by NAC pretreatment. Trichlorfon, but not $H_2O_2$ or formalin, elevated plasma levels of aspartatetransaminase (AST) and alanine-transaminase (ALT), and these elevations were attenuated by NAC. There was no change in hematocrit values in any treatment. The results provide evidence for the attenuating effects of NAC against toxicity caused by anti-parasite agents that act through free radical-producing properties. The results found in this study also suggest that NAC may be administered to fish to minimize toxicity in fish parasiticide use.

Growth Response to a GH-Autotransgenesis in Common Carp Cyprinus carpio

  • Autotransgenic manipulation with a growth hormone (GH)-construct is a potential approach to improving the growth rates of farmed fish. Here, we present the generation of GH-autotransgenic common carp Cyprinus carpio carrying a transgene comprised of the carp homologous GH gene and a ${\beta}$-actin regulator. Autotransgenic carp showed similar viability to their non-transgenic siblings. Early growth characteristics of founder autotransgenic carp up to 50 days postfertilization were highly variable among individuals; i.e., some fish exhibited significant growth depression, while others showed dramatic acceleration of growth, achieving greater than sixfold increases in body weight relative to their non-transgenic counterparts. Stimulated growth performance became more notable with age and many transgenic individuals of the largest class reached 5 kg within 8 or 9 months, which is at least 10 times heavier than the average body weight of communally grown non-transgenics. Four of six founder transgenic males were successful in passing the transgene to their $F_1$ offspring with frequencies ranging from 19 to 36%. Growth stimulations were also persistent in all $F_1$ progeny groups examined.

  • Monogeneans(Phylum Platyhelminthes) have been known as common parasites onto the skin and gills of cultured freshwater fishes. Infestation with these parasites result in a great loss in aquaculture industry. Some classification studies on these parasites have mostly been conducted in Japan but rarely in Korea. For the purpose of classifying monogenean parasites infesting two Korean freshwater fishes, carp(Cyprinus carpio) and loach(Misgurnus anguillicaudatus), samples captured from March 1998 to April 2000 were examined. Here we report that three different species of Monogeneans, Diplozoon nipponicum, Dactylogyrus extensus from carp and Gyrodactylus paralatus from loach, respectively. Of these the latter two parasites have never been identified in Korea.

First report of Aeromonas veronii infection in farmed Israeli carp Cyprinus carpio in Korea

  • In May 2007, mass mortality of Israeli carp, Cyprinus carpio L., was occured on a pond farm located in Jeollabukdo Province, Korea. The mortality rates reached up to 2% of the total fish in the farm per day. Typical clinical signs were abdominal distension, reddish foci on the skin, enteritis, liver congestion and enlarged spleen and kidney. On the basis of biochemical characteristics and 16S rDNA sequence, the causative bacteria isolated from affected carp were identified as Aeromonas veronii. Histologically, degeneration of hepatocytes and congestion in sinusoids were observed in the liver. Spleen showed hemorrhage and the destruction of the sheathed tissues. In kidney, necrotized renal tubules and glomerular destructions were observed. Intestinal tissues revealed necrotized and severe hemorrhage. Mass hemorrhage was observed in muscles. This is the first report that A. veronii caused mortality in cultured Israeli carp in Korea.

First report of gill thelohanellosis from common carp (Cyprinus carpio) fingerling in Korea

  • Myxosporeans are widespread cnidarian endoparasites in marine and freshwater ecosystems and several species were reported to be a threat to cultured fish causing serious diseases with mass mortality. In the present study, we found a myxosporean species in the genus Thelohanellus from the gills of the cultured common carp (Cyprinus carpio) fingerling for the first time in Korea. The morphological observation showed 500 ㎛ ~ 1 mm size, oval to circular shaped plasmodia containing spores which are pyriform at the anterior end and round at the posterior end (average size 20.1 ㎛ × 9.1 ㎛), with 5 to 6 turns of a single polar filament located in the polar capsule with an average size of 10 ㎛ × 4.6 ㎛. The 18S rRNA sequence was closest to the sequence of T. wangi among Thelohanellus species infecting gills but was not completely identical. Based on the morphological characteristics and molecular analysis results, we classified the present myxosporean parasite as Thelohanellus sp., temporarily. The prevalence and infection intensity of Thelohanellus sp. in the common carp fingerling were very high, which was thought to be the main cause of high mortality.

  • Mirror carp, Cyprinus carpio nudus (weight 26.2±3.1 g, length 11.8±0.5 cm) were exposed to waterborne total ammonia nitrogen (TAN) at 0, 2, 4, 8, 16, 32, 64 and 128 mg TAN/L. The lethal concentration 50 (LC50) of mirror carp, C. carpio nudus exposed to waterborne ammonia was 60.38 mg TAN/L. The red blood cell (RBC) count was significantly reduced by ammonia exposure, whereas there was no significant changes in the hemoglobin concentration and hematocrit value. In the plasma components, glucose, total protein, aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by ammonia exposure. The results of this study suggest that the ammonia exposure to C. carpio nudus affects the survival rates, hematological parameters and plasma components as toxicity.

  • Bukholderia pyrrocinia CAB08106-4 has an anti-fungal effect on Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp. It is environmentally friendly microbial product that prevents and controls a variety of phytopathogens including Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp. The aim of this study was to assess the environmental toxicity using Cyprinus carpio and Daphnia magna. Bukholderia pyrrocinia CAB08106-4 ($1.0{\times}10^9cfu/mL$) was adminatrated to Cyprinus carpio and Daphnia magna according to the toxicity test guideline for peciticide. $LC_{50}$ of Bukholderia pyrrocinia CAB08106-4 is over $6.67{\times}10^4cfu/mL$ in Cyprinus carpio and Daphnia magna and no adverse effect was observed. Based on these results, we concluded that Bukholderia pyrrocinia CAB08106-4 has no toxiciy for Cyprinus carpio and Daphnia magna.

  • This study was conducted to investgate the effect of fish meal analogue (FMA) on the growth of Israeli carp, Cyprinus carpio averageing 43g. Leather meal, meat and bone meal, feather meal, squid liver powder, poultry by-product and blood meal were used for ingredient of FMA. Four experimantal diets were formulated to contain 36% crude protein and 15.4 kJ available enery g-1. The percentage of follows ; Diet 1, 100% 3, 60% WFM + 40% FMA (40% FMA) ; Diet 4, 40% WFM + 60% FMA (60% FMA). In this expriment, fingerling Israeli carp averaging 43 g were assigned to each diet treatment. When the experiment was terminated, weigth gain (WG), feed conversion (FC), specific growth rate (SGR) and protein efficiency ratio (PER) were measured respectively. In the experiment, WG, FC, SGR and PER of Diet 2 were not significantly different from those of the control diet (P0.05). And WG, FC and SGR of Diet 3 were not significantly of the control Diet. In Diet 4, WG, FC and PER were not significantly different from those of the control Diet, but SGR was lower than that fo the control Diet. According to the results from this study, it might be possible to replace fish meal protein by FMA up to 60% in fingerling Israeli carp diets.

Changes of Plasma Cortisol Concentration to High Water Temperature on Common Carp (Cyprinus carpio) (고수온에 따른 잉어(Cyprinus carpio)의 혈장 코티졸 농도 변화)

  • The changes of plasma cortisol concentration to high water temperature on common carp, Cyprinus carpio were studied. 20℃, 24℃ and 28℃ groups showed significantly increased plasma cortisol levels until 6 hours, and decreased plasma cortisol levels after 24 hours. Cortisol levels of 20℃ and 24℃ groups decreased to values close to the cortisol content before the experiment. However, 32℃ group showed significantly increased plasma cortisol levels until 24 hours, and maintained plasma cortisol level until 48 hours. The Cumulative mortality rates of 20℃, 24℃ and 28℃ groups increased until 12 hours, and maintained until 48 hours. Cumulative mortality rate of 32℃ group increased continuously until the end of the experiment. Using the results of this study, it could be used as a method to confirm the risk of mass perish in a specific area through hormone analysis.

  • In this study, cinnamon essential oil (CEO) was formulated as emulsifiable concentrate (EC) and a granule. For the evaluation of their negative effects on the ecosystem, acute toxicities against Cyprinus carpio was determined in a static condition. The formulations were made using CEOs extracted by 3 different methods (steam distillation (SD), solvent extraction and supercritical fluid extraction (SFE)) and were tested to obtain $LC_{50}$ values. Among the ECs, EC including CEO extracted by SFE showed highest acute toxicity against C. carpio. Among the granules, a granule including CEO extracted by SD showed highest acute toxicity against C. carpio. Nevertheless, $LC_{50}$ of EC and a granule formulation with CEOs was higher than toxicity level III of pesticide standardized by Korea rural development administration. These results were similar to those using zebrafishes. Chronic toxicities were not found for 45 days in zebrafishes until $500{\mu}gL^{-1}$ level of EC formulation including CEO obtained by the SD. Based on these results, EC formulation of CEOs may be considered to be used as environmental-friendly natural insecticides in accordance with the standards.
