In vitro Biological Activities of Anthocyanin Crude Extracts from Black Soybean (In vitro 실험에서 검정콩 안토시아닌 조추출물의 효능 분석)

  • This study was carried out to investigate the antioxidative and anti-inflammatory activity of crude anthocyanin compounds extracted from black soybean. The crude anthocyanin compounds were extracted with 80% methanol and concentrated to powder. The most abundant compound isolated from the extract was C3G(cyanidin-3-glucoside). The superoxide dismutase (SOD) assay was conducted to assess the antioxidative activity of the crude extract. SOD, which catalyzes the dismutation of the superoxide anion into hydrogen peroxide and molecular oxygen, is one of the most important antioxidative enzymes. The black soybean anthocyanin extracts inhibited more than 90% of the superoxide radical at a concentration of 0.1% and 100% at a concentration of 0.5%, indicating that this extract displayed excellent antioxidative activity. To assess the anti-inflammatory activity of the extract, a NO(Nitric oxide) production assay in RAW 264.7 cells was performed. NO is an important physiological messenger and effector molecule in many biological systems, including immunological, neuronal and cardiovascular tissues. In this assay, the anthocyanin extracts showed a high anti-inflammatory potential, where the inhibitory potency for NO production was similar to the positive control, particularly for EGCG(epigallocatechin-3-gallate), which is known to have excellent anti-inflammatory activity. Thus, it can be concluded that the anthocyanin extracts from black soybean have distinctive pharmaceutical activities and may be used as an excellent source materials to supplement the health benefits of various food products.

A Major Antioxidative Components and Comparison of Antioxidative Activities in Black Soybean (검정콩의 주요 항산화 원인물질 및 항산화 효과의 비교)

  • Contents of isoflavone, phenolic acids, tocopherol, and anthocyanin in black soybean (Glycine max) were measured by HPLC. To compare antioxidative activities of main black soybean components, antioxidative effects of the same levels of commercial standard components were measured by Trolox equivalent antioxidant capacity assay (TEAC). Most effective component was gentisic acid followed by anthocyanin, p-coumaric acid, ferulic acid, genistein, syringic acid, and daidzein. TEAC assay results revealed genistein in isoflavone, gentisic acid in phenolic acids, p-tocopherol in tocopherol, and anthocyanin showed highest antioxidative and synergistic acitivities, with anthocyanin showing strongest synergy effect.

Antioxidant Activity and Cytotoxicity on Human Cancer Cells of Anthocyanin Extracted from Black Soybean (검정콩 안토시아닌의 항산화 및 암세포독성)

  • Anthocyanin pigments in soybean seed coat were D3G (Delphinidin-3-glucoside), C3G (Cyanidin-3- glucoside) and Pt3G (Petunidin-3-glucoside), which have been known potential roles in the prevention and treatment of chronic diseases. Anthocyanin contents in seed coat of blck soybean were significantly different according to soybean variety, C3G content showed the highest value in all materials and its variation was also wide. Antioxidant activity of each pigment was analyzed by DPPH and TEAC methods in which D3G and C3G showed high activity. And this study was carried out to investigate the effects of anthocyanin to human cancer cells. Cytotoxity were analyzed by MTT assay after anthocyanin pigments treated on leukemia (Jurkat T) and adenocarsinoma (MCF-7) cells. It showed decrement of cell numbers as anthocyanin concentration is increasing. ${EC}_50$ range of anthocyanin concentrations were $100{\sim}250\;ug/mL$ and $100{\sim}250\;ug/mL$ in Jurkat T and MCF-7 cell, respectively. D3G showed higher cytotoxicity than other pigments in Jurkat T cell whereas activity of C3G was high in MCF-7 cell. It is believed that supplementation of human diets with soybean anthocyanin markedly reduces human cancer mortality rates.

Antioxidant Contents and Antioxidant Activities of Hot-Water Extracts of Aronia (Aronia melancocarpa) with Different Drying Methods (건조방법에 따른 아로니아(Aronia melancocarpa) 열수 추출물의 항산화 성분 함량 및 항산화 활성)

  • This study determined the antioxidant levels and activities of hot water aronia extracts by different drying methods such as sun drying, sun drying after steam treatment, freeze-drying, and oven drying. The total polyphenol content, calculated as gallic acid equivalent, was the highest in the freeze-dried sample (910 mg), followed by sun-dried after steam treatment (779 mg), sun-dried (769 mg), and oven-dried (757 mg) samples. Similar patterns were observed for the total flavonoid and anthocyanin contents. Freeze-dried aronia samples contained the highest polyphenol, flavonoid, and anthocyanin contents as compared to the samples dried by other methods. All antioxidant activities were found to increase in a dose-dependent manner. For the hot water-extracted freeze-dried aronia powder (200 mg/mL), the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radicalscavenging activities were 65.5% and 61.7% and the hydroxyl and superoxide anion radical-scavenging activities were 50.5% and 52.1%, respectively. These results suggest that comparatively, freeze-drying is a better method for preserving the bioactive components and the antioxidant activities of aronia.

The effect of citric acid and enzyme inactivation treatment on C3G stability and antioxidant capacity of mulberry fruit jam (구연산 및 효소 불활성화 처리가 오디잼의 C3G 안정성과 항산화능에 미치는 영향)

  • We have developed processing methods improved cyanidin-3-glucoside(C3G) stability in mulberry fruit's jam. In the processing of mulberry jam, 3 minutes blanching using microwave inhibited the C3G destruction by maintaining the antioxidant capacity and inactivation of enzymes related to pigment's stability. Also citric acid increased C3G stability in the mulberry fruit's jam. As the adding sugar, sucrose was considered the more desirable sugar than oligosaccharide or xylitol. It showed high C3G content and antioxidant capacity compared to the two treatment groups. In conclusion, when processing mulberry fruit's jam, the addition of citric acid and the enzyme's inactivation treatment were considered effective treatment for the C3G stability and antioxidant ability.

  • In ornamental crops, the color and shape of flowers are one of the important traits. Generally, flower colors are determined by accumulating pigments such as carotenoids, flavonoids, and betalains. Among them, flavonoids are responsible for broad ranges of colors. Chrysanthemums are one of the most popular ornamental crops in the world, and there have been many efforts to change their flower color. In chrysanthemum flowers, cyanidin-based anthocyanin confers pink or red color, whereas terpenoid-based carotenoids are mainly responsible for yellow and green colors. However, blue colored chrysanthemums do not occur in nature. To date, there have been attempts to obtain blue or violet-colored chrysanthemum flowers through the introduction of a novel gene for accumulating delphinidin-based anthocyanins, while other studies have reported changing endogenous metabolites through the reconstruction of flavonoid biosynthesis. Since various transcription factors are involved in the regulation of flavonoid biosynthesis, it is important to understand not only the structural genes, but also the transcription factors required for the modification of flavonoid-based flower color. Therefore, in this paper, we describe the flavonoid biosynthetic pathway and its regulation, and review previous studies on the change in flower color through modification of flavonoid biosynthesis. This effort could be an important milestone in successfully achieving the modification of chrysanthemum flower color by means of plant biotechnology.

Variation of Anthocyanin and Protein Contents in Glycine max L. (Merr) (Soybean) Germplasms from Korea

  • Soybean (Glycine max L. (Merr) is commonly consumed and found in major foods including soymilk, soy sauce, tofu, and soy sprout in Korea and east Asia. In addition, it is common to cook the whole seeds with rice. Soybean is known to have ranges of health benefits including antiaging, anticancer, neuroprotective and antidiabetic taken either as supplement or dietary food. Anthocyanins and flavonoids in G. max are found to be the main contributors to such wide arrays of health benefits. Due to increasing economic values of soybean, development of specialty soybean cultivars is becoming an area of interest worldwide. In this study, 746 black soybean accessions from National Agrobiodiversity Center were characterized as part of an attempt to identify important germplasms of G. max. Seed coats of each accession were analyzed for their total anthocyanin, cyanidin 3-O-Glucoside (C-3-O-G), delphinidin 3-O-glucoside (D-3-O-G), petunidin-3-O-glucoside (Pt-3-O-G), and their whole seeds for crude protein contents. HPLC was used to determine and quantify the anthocyanin compositions while crude protein was determined using Kjeldahl method by Kjeltec auto-analyzer (Kjeltec 8400, Foss, Sweden). Accessions were grouped according to their anthocyanins and protein contents; the mean content of which were correlated to agronomic traits including maturity date, one hundred seed weight, cotyledon color and seed lust color. The results indicated that the total anthocyanin content (TAC) ranged from 273.77 to 6250.52 mg/100 g, with mean value of 1853.03 mg/100 g while the crude protein content (CPC) being between 33.43 and 47.51%, with mean value of 40.81%. The highest number of accessions (45.97%) showed TAC between 1000~1900 mg/100 g while 30.96% of accessions showed CPC between 41~43%. Among the 746 accessions considered, 11 (IT142935, 175818, 175855, 177191, 177209, 177211, 177214, 177216, 177218, 177220, 177274) of them showed TAC above 4000 mg/100 g. C-3-O-G was found to be the major contributor to TAC showing strong correlation. Accessions with green cotyledon color showed high mean TAC compared to those having yellow cotyledon color, and accessions with dull seed lust color showed high mean TAC than those having shiny seed lust color. One hundred seeds weight and maturity date showed positive correlation with all anthocyanin contents, except for Pt-3-O-G in the latter case. The overall result of the present study could be used as background for developing new black soybean cultivars and breeds with high anthocyanin and protein contents. The result depicted that many of the accessions could be used as potential parental lines.

Genetic Characteristics and Anthocyanin Content of Basil(Ocimum basilicum L.)

  • Basil(Ocimum basilicum L.) is an annual herb that grows wild in hot and humid regions such as tropical Asia and Africa. This study was carried out for resource discovery, propagation, and DB construction of aromatic plants. In order to evaluate the genetic characteristics, 28 kinds of basil seeds were supplied from the Center for Genetic Resources. For basil characteristics, planting date, plant height, growth shape, leaf length, leaf shape, petiole color, petiole, stem color, flower color, after harvest, Ml length including roots, flower length, stem thickness and biological weight were measured. For anthocyanin analysis, only basil with purple color was selected, anthocyanins were extracted with 60% ethanol containing 1% citric acid, a standard quantitative curve was prepared with cyanidin-3-glucoside, and absorbance was measured at 525 nm. Basil planting started around June 16, and the flowering period lasted for a total of 18 days from July 19 to August 5. On August 6, when flowering was completed, morphological characteristics including flower color were measured at the site by an object. The measured basil plant length was 26.6-59.6 cm, leaf length 2.0-7.9 cm, leaf width 0.7-5.2 cm, and petiole 1.3-3.9 cm. The growth pattern of basil was generally straight, medium in shape. The color of the leaves was observed evenly by mixed green, purple, purple and green, and the recesses in the shape of the leaf were also conspicuously observed. A total of three basil flower colors were observed: white, light pink(149P 10P/84), and purple(37V 2.5RP 4/12). During the second growth survey after harvest, the total length including the basil roots is 26.7-52.0 cm, the flower length is 8.2-29.3cm, the stem thickness is 7.1-15.9mm, the number of stem nodes is 3-12 nodes, and the total weight of the basil was measured to be about 218-1540 g. There are a total of 16 types of basil expressed in purple, and the anthocyanin content was measured to be 73.74 ~ 380.72 mg/100g.

Variation of Anthocyanin, and Isoflavone Contents in Korean Black Soybeans Grown at Different Latitudinal Locations (위도가 다른 재배지역에서 생육한 검정콩의 안토시아닌 및 이소플라본 함량 변이)

  • This experiment was conducted to investigate the variation of anthocyanin, and isoflavone contents in five cultivars and two lines of Korean domestic black soybeans grown at different latitudinal locations, a high latitude, Suwon ($37^{\circ}$16'N) and a low latitude, Milyang ($35^{\circ}$30'N). Delphinidin-3-glucoside (D3G) contents of anthocyanin in Geomjeongkong # 3 and Ilpumgeomjeongkong, cyanidin-3-glucoside (C3G) content in Milyang # 113, petunidin-3-glucoside (Pt3G) contents in Milyang # 113, and Ilpumgeomjeongkong, and total anthocyanins in Milyang # 113 were highest among the seven black soybean cultivars and lines. D3G, C3G, and total anthocyanins in Geomjeongkong # 3, C3G, and total anthocyanins in Ilpumgeomjeongkong grown at high latitude were higher compared to low latitude. Daidzein, glycitein, genistein, and total isoflavone contents in Geomjeongkong # 4 were highest among the seven cultivars and lines. Daidzein contents of isoflavone in Geomjeongkong # 3, Milyang # 112, and Milyang # 113 grown at high latitude were higher compared to low latitude. Glycitein contents in Geomjeongkong # 3, and # 4 grown at high latitude were higher compared to low latitude, while it in Milyang # 113 grown at low latitude was higher compared to high latitude. Genistein contents in most black soybeans except Milyang # 113, and total isoflavone contents in Geomjeongkong # 4 and Cheongjakong grown at low latitude were higher compared to high latitude. The variations of anthocyanin except Pt3G and isoflavone contents seemed to be affected by environmental conditions like different latitude than the genotype because they showed the significant interaction between cultivars and locations.

Effect of LED Light Wavelength on Lettuce Growth, Vitamin C and Anthocyanin Contents (LED광 파장이 상추생육과 비타민 C 및 안토시아닌 함량에 미치는 영향)

  • In this study, we analyzed the growth characteristics of red lettuce under Light-emitting diode (LED) light environment as well as the change of vitamin C and anthocyanins of lettuce. We made five monochromatic light treatments (red 647 nm, 622 nm, blue 463 nm, 450 nm, White), six mixed red (R) and blue (B) light treatments (R : B = 9 : 1, 8 : 2, 7 : 3, 6 : 4, 5 : 5) and red + white, and three light treatments made according to photoperiod of LED with lighting sources ratio of red : blue (R : B = 8 : 2(18/06 h, 15/09 h, 12/12 h)). It was composed of totally 14 control beds. As a result, the red lettuce the most developed leaf height, leaf numbers and fresh weight under red single light, root length and leaf developed when grown under blue single light. Therefore, red light were related to above part of the lettuce, blue light were related to the growth of the underground part of lettuce. Case of the mixed light, leaf height, leaf numbers, fresh/dry weight of above and underground part were highest red + white light and root length and chlorophyll content were highest under red 7 : 3 blue light. Result of growing investigation by photoperiod, the red lettuce were considered to be most effective in 15/09 h (on/off). The content of anthocyanins; the single light source, mixed light and light irradiation period were highest under blue light (463 nm), red 7 : 3 blue and 18/06 h (on/off) light irradiation, respectively. The vitamin C showed the lowest content of $1.26mg{\cdot}L^{-1}$ under the white light, but showed the greatest content of $3.02mg{\cdot}L^{-1}$ for the control group.
