Membrane Topology of Helix 0 of the Epsin N-terminal Homology Domain

  • Specific interaction of the epsin N-terminal homology(ENTH) domain with the plasma membrane appears to bridge other related proteins to the specific regions of the membrane that are invaginated to form endocytic vesicles. An additional $\alpha$-helix, referred to as helix 0 (H0), is formed in the presence of the soluble ligand inositol-1,4,5-trisphosphate [$Ins(1,4,5)P_3$] at the N terminus of the ENTH domain (amino acid residues 3-15). The ENTH domain alone and full-length epsin cause tubulation of liposomes made of brain lipids. Thus, it is believed that H0 is membrane-inserted when it is coordinated with the phospholipid phosphatidylinositol-4,5-bisphosphate [$PtdIns(4,5)P_2$], resulting in membrane deformation as well as recruitment of accessory factors to the membrane. However, formation of H0 in a real biological membrane has not been demonstrated. In the present study, the membrane structure of H0 was determined by measurement of electron paramagnetic resonance (EPR) nitroxide accessibility. H0 was located at the phosphate head-group region of the membrane. Moreover, EPR line-shape analysis indicated that no pre-formed H0-like structure were present on normal acidic membranes. $PtdIns(4,5)P_2$ was necessary and sufficient for interaction of the H0 region with the membrane. H0 was stable only in the membrane. In conclusion, the H0 region of the ENTH domain has an intrinsic ability to form H0 in a $PtdIns(4,5)P_2$-containing membrane, perhaps functioning as a sensor of membrane patches enriched with $PtdIns(4,5)P_2$ that will initiate curvature to form endocytic vesicles.

  • A simple method to measure the elastic modulus E and Poisson's ratio v of diamod-like carbon (DLC) films deposited on Si wafer was suggested. Using the anisotropic etching technique of Si we could make the edge of DLC overhang free from constraint of Si substrate. DLC film is chemically so inert that we could not on-serve any surface damage after the etching process. The edge of DLC overhang free from constraint of Si substrate exhibited periodic sinusoidal shape. By measuring the amplitude and the wavelength of the sinu-soidal edge we could determine the stain of the film required to adhere to the substrate. Since the residual stress of film can be determine independently by measurement of the curvature of film-substrate com-posite we could calculated the biaxial elastic modulus E/(1-v) using stress-strain relation of thin films. By comparing the biaxial elastic modulus with the plane-strain modulus E/(1-{{{{ { v}^{2 } }}) measured by nano-in-dentation we could further determine the elastic modulus and Poisson's ratio independently. This method was employed to measure the mechanical properties of DLC films deposited by {{{{ { {C }_{6 }H }_{6 } }} rf glow discharge. The was elastic modulus E increased from 94 to 169 GPa as the {{{{ { V}_{ b} / SQRT { P} }} increased from 127 to 221 V/{{{{ {mTorr }^{1/2 } }} Poisson's ratio was estimated to be abou 0.16∼0.22 in this {{{{ { V}_{ b} / SQRT { P} }} range. For the {{{{ { V}_{ b} / SQRT { P} }} less than 127V/{{{{ {mTorr }^{1/2 } }} where the plastic deformation can occur by the substrate etching process however the present method could not be applied.

Development of Sports Brassiere Pattern Using 3D Shaping Technology (3차원 쉐이핑 기술을 활용한 스포츠 브래지어 개발)

  • This study used 3D technology to develop a multi-functional sports brassiere with increased comfort and fit that can be worn as a base layer during exercise or as underwear. A 75A size industrial lingerie figure was used to develop a standard pattern. 3D tools for scanning and pattern making, such as Vivid 910, Geomagic Design X, 2C-AN and Yuka CAD were used. The sports brassiere was designed as a tank top style with dual structure and linings attached to a pad utilized with a sport brassiere mold cup. 3D outer and lining's pattern was differently developed in consideration of the body's curvature with pad's shape and structure. Shoulder and neck part reduction rates were adjusted to increase the neck areas fit that considered the nude pattern's structure due to uncomfortableness felt by wearers who were uncomfortable with the neck areas fit on existing brand products. The reduction rate was also set differently on each part. For example, the reduction rate on outer side panel was set strongly to increase the breast's volume. Two products, developed by a 3D sports brassiere and previously released product, were worn on 8 subjects in their 20's to evaluate fit, comfort, and purchase preferences. The evaluation proved that newly developed 3D products were superior to comparative products. The results of the clothing pressure measurement indicate that the newly developed sports brassiere's front part had less pressure on upper bust and shoulder areas compared to comparative products as well as showed less pressure on the back side, which shows improved wearing comfort compared to comparative products.

Body Pressure Distribution and Textile Surface Deformation Measurement for Quantification of Automotive Seat Design Attributes (운전자의 체압 분포 및 시트변형에 대한 정량화 측정시스템)

  • Proper seat design is critical to the safety, comfort, and ergonomics of automotive driver's seats. To ensure effective seat design, quantitative methods should be used to evaluate the characteristics of automotive seats. This paper presents a system that is capable of simultaneously monitoring body pressure distribution and surface deformation in a textile material. In this study, a textile-based capacitive sensor was used to detect the body pressure distribution in an automotive seat. In addition, a strain gauge sensor was used to detect the degree of curvature deformation due to high-pressure points. The textile-based capacitive sensor was fabricated from the conductive fabric and a polyurethane insulator with a high signal-to-noise ratio. The strain gauge sensor was attached on the guiding film to maximize the effect of its deformation due to bending. Ten pressure sensors were placed symmetrically in the hip area and six strain gauge sensors were distributed on both sides of the seat cushion. A readout circuit monitored the absolute and relative values from the sensors in realtime, and the results were displayed as a color map. Moreover, we verified the proposed system for quantifying the body pressure and fabric deformation by studying 18 participants who performed three predefined postures. The proposed system showed desirable results and is expected to improve seat safety and comfort when applied to the design of various seat types. Moreover, the proposed system will provide analytical criteria in the design and durability testing of automotive seats.

  • There are prior studies that describe in detail the classification and shape of 26 species of Korean flounder, but it is difficult to make them out and they are used interchangeably or misused. It is intended to present the taxonomic standards of Pleuronichthys cornutus and Pleuronichthys japonicus and use them as tourism education materials using aquatic resources. Morphological comparison was made through counting and measurement. The two fish species were generally flattened in an oval shape with a low body height and a wide body width, and the anal fin developed and sized similar to the dorsal fin, and the caudal fin was widely spread out. Among the hyoid bones, in three parts of the coccyx, the main part, the ischial part, and the cardinal process, there were differences in the degree of curvature, the open angle, and the shape of the ischial part and the lateral process, which corresponded to the representative classification characteristics. In addition, it will be expected that it will serve as a basis as a fishery and marine education material for marine tourism that classifies and categorizes the characteristics as a genus of flounder and categorizes the characteristics, and reduces misuse of scientific names and names in the future.

An automatic rotating annular flume for cohesive sediment erosion experiments: Calibration and preliminary results

  • Flows of water in the environment (e.g. in a river or estuary) generally occur in complex conditions. This complexity can hinder a general understanding of flows and their related sedimentary processes, such as erosion and deposition. To gain insight in simplified, controlled conditions, hydraulic flumes are a popular type of laboratory research equipment. Linear flumes use pumps to recirculation water. This isn't appropriate for the investigation of cohesive sediments as pumps can break fragile cohesive sediment flocs. To overcome this limitation, the rotating annular flume (RAF) was developed. While not having pumps, a side-effect is that unwanted secondary circulations can occur. To counteract this, the top and bottom lid rotate in opposite directions. Furthermore, a larger flume is considered better as it has less curvature and secondary circulation. While only a few RAFs exist, they are important for theoretical research which often underlies numerical models. Many of the first-generation of RAFs have come into disrepair. As new measurement techniques and models become available, there is still a need to research cohesive sediment erosion and deposition in facilities such as a RAF. New RAFs also can have the advantage of being automatic instead of manually operated, thus improving data quality. To further advance our understanding of cohesive sediment erosion and deposition processes, a large, automatic RAF (1.72 m radius, 0.495 m channel depth, 0.275 m channel width) has been constructed at the Hydraulic Laboratory at Chungnam National University (CNU), Korea. The RAF has the ability to simulate both unidirectional (river) and bidirectional (tide) flows with supporting instrumentation for measuring turbulence, bed shear stress, suspended sediment concentraiton, floc size, bed level, and bed density. Here we present the current status and future prospect of the CNU RAF. In the future, calibration of the rotation rate with bed shear stress and experiments with unidirectional and bidirectional flow using cohesive kaolinite are expected. Preliminary results indicate that the CNU RAF is a valuable tool for fundamental cohesive sediment transport research.

Measurement of Residual Stress Distribution in the Depth Direction of Annealed Materials of Lapped Bearing Steel Using Weighted Averaging Analysis Method (가중평균 해석법을 이용한 래핑된 베어링강 어닐링재료의 깊이방향에 대한 잔류응력분포 측정)

  • This paper reports the results of an experimental examination using X-rays to test annealing materials for lapped bearing steel (STB2), to confirm the validity of the weighted averaging analysis method. The distribution behavior for the α𝜓-sin2𝜓 diagram and the presence or absence of differences in the peak method, half-value breadth method, and centroid method were investigated. When lapping the annealed bearing steel (STB2) material, a residual stress state with a non-directional steep gradient appeared in the surface layer, and it was found that the weighted averaging analysis method was effective. If there is a steep stress gradient, the sin2𝜓 diagram is curved and the diffraction intensity distribution curve becomes asymmetric, resulting in a difference between the peak method, half-value breadth method, and centroid method. This phenomenon was evident when the stress gradient was more than 2~3 kg/mm2/㎛. In this case, if the position of the diffraction line is determined using the centroid method and the weighted averaging analysis method is applied, the stress value on the surface and the stress gradient under the surface can be obtained more accurately. When the stress gradient becomes a problem, since the curvature of the sin2𝜓 diagram appears clearly in the region of sin2𝜓 > 0.5, it is necessary to increase the inclination angle 𝜓 as much as possible. In the case of a lapping layer, a more accurate value can be obtained by considering 𝜎3 in the weighted averaging analysis method. In an isotropic biaxial residual stress state, the presence or absence of 𝜎3 can be determined as the presence or absence of strain for sin2𝜓≈0.4.

Influence of Detailed Structure and Curvature of Woven Fabric on the Luminescence Effect of Wearable Optical Fiber Fabric (직물의 세부 구조 및 굴곡이 웨어러블 광섬유의 발광 효과에 미치는 영향)

  • The two main requirements of wearable optical fiber fabrics are that they must presuppose a high degree of flexibility and they must maintain the luminance effect in both flat and bent conformations. Therefore, woven optical fiber fabrics that satisfy the above conditions were developed by both weaving and by using computer embroidery. First, we measured the brightness of the wearable optical fiber fabric in the flat state at a total of 10 measurement points at intervals of 1 cm. Second, the wearable optical fiber fabric was placed horizontally on the forearm, where three-dimensional bending occurs, and the luminance values were recorded at the same 10 measurement points. For the woven fabric in the flat state, the maximum, minimum, average, and standard deviation luminance values were $5.23cd/m^2$, $2.74cd/m^2$, $3.56cd/m^2$, and $1.11cd/m^2$, respectively. The corresponding luminance values from the bent forearm were $7.92cd/m^2$ (maximum), $2.37cd/m^2$ (minimum), $4.42cd/m^2$ (average), and $2.16cd/m^2$ (standard deviation). In the case of the computer-embroidered fabric, the maximum, minimum, average, and standard deviation luminance values in the flat state were $7.56cd/m^2$, $3.84cd/m^2$, $5.13cd/m^2$, and $1.04cd/m^2$, respectively, and in the bent forearm state were $9.6cd/m^2$, $3.63cd/m^2$, $6.13cd/m^2$, and $2.26cd/m^2$, respectively. Therefore, the computer-embroidered fabric exhibited a higher luminous effect than the woven fabric because the detailed structure reduced light-loss due to the backside fabric. In both types of wearable optical fiber fabric the luminance at the forearm was 124% and 119%, respectively, and the light emitting effect of the optical fiber fabric was maintained even when bent by the human body. This is consistent with the principle of Huygens, which defines the wave theory of light, and also the Huygens-Fresnel-Kirchhoff principle, which states that the intensity of light increases according to the magnitude of the angle of propagation of the light wavefront (${\theta}$).

A study of Curved Dosimeter for Flattening Filter Free Beam Quality Assurance Evaluation using Curved Dosimeter in Radiotherapy (Flattening Filter Free Beam의 정도관리를 위한 곡면선량계 가능성 연구)

  • Radiation therapy using flattening filter free beam can prevent beam attenuation caused by flattening filter and can improve treatment efficiency. However, accurate dose control is not established for nonuniform iso dose distributions. In this study, curved dosimeter based on photoconductive material $HgI_2$ was fabricated and its reproducibility and linearity were evaluated at 6 MV photon energy to verify its performance. In order to show the usefulness of the curved measurement, the signals measured on the flat substrate and the curved substrate were compared in the flattening filter free beam using the acrylic filter. As a result, the reproducibility of the unit cell dosimeter was evaluated as SE 0.613%, and the linearity was evaluated as R-sq 0.9999. The usability evaluation of the array curve dosimeter demonstrated its usefulness by indicating a curvature error rate of 11.073%p and a reduced error rate.

Development of Embedded Type Sensor Module for Measuring Stress of Concrete Using Hetero-core Optical Fiber (헤테로코어 광섬유를 이용한 콘크리트 응력 측정용 매립형 센서모듈의 개발)

  • In this study, in order to directly evaluate the prestress of the PSC structure, a new sensor module based on the measurement of the deformation of concrete was proposed using hetero-core optical fibers and performance tests were performed. In a hetero-core optical fiber, optical loss occurs when a specific part of the transmission path is bent, and the amount of optical loss changes linearly according to the magnitude of the curvature. In order to confirm the measurement performance of the sensor module and the applicability of the optical fiber, the sensor module was deformed and the light passing through the optical fiber was converted into wattage and measured. It can be seen that the light passing through the optical fiber has a linearity of 0.9333 in relation to the deformation while generating the maximum deformation of 0.5 mm at a rate of 0.12 mm/min in a cylindrical concrete specimen with a diameter of 15 cm and a height of 35 cm in which the sensor module is embedded. Based on the results of this experiment, it is judged that it is possible to directly evaluate the prestress of a PSC structure by embedding a sensor module using a hetero-core optical fiber in the structure and measuring the compression deformation in concrete. It is judged that it can be used as useful data for the development of a sheath tube integrated sensor module to be applied to be applied to the girder model experiment.
