마우스에서 울금 에탄올 추출물의 단회 경구투여 독성에 관한 연구 (Acute Oral Toxicity Study of Ethanol Extract of Curcuma longa L. in Mice)

  • 울금(Curcuma longa L., turmeric)은 생강과(Zingiberaceae)에 속하는 다년생 초본 식물로 전통적으로 한약재, 향신료 및 식용으로 사용되어 왔다. 강력한 항산화 작용, 울금 에탄올 추출물의 항산화 활성, 항돌연변이성, 항염증, 항암성 및 항균성 효과 등이 보고되고 있다. 그러나 울금에 대한 생리활성 연구는 이미 많이 이루어져 있는 반면 울금에 대한 독성평가는 이루어지지 않았다. 본 연구는 울금 에탄올 추출물에 대한 안정성을 확보하기 위하여 단회경구투여 독성시험을 BALB/c 마우스를 이용하여 실시하였다. 울금 에탄올 추출물은 0, 20, 200, 2,000 mg/kg 농도로 경구 투여하였으며, 14일간 관찰 후 희생시켰다. 울금 에탄올 추출물 투여 후 운동성, 비정상적 임상증상, 부검 소견상, 체중의 유의적인 변화는 관찰되지 않았다. 혈액 생화학적 측정과 다양한 장기의 무게에서도 유의적인 변화를 관찰할 수 없었다. 이들 결과로 미루어볼 때 울금 에탄올 추출물의 단회 투여에 따른 치사량은 2,000 mg/kg 이상을 상회할 것으로 추정되며 급성독성에 어떠한 유해성이 없다는 것을 의미한다.

잔소리졸 함유 껌의 치태 및 치은염 억제 효과 (Effect of Curcuma Xanthorrhiza containing gum on reduction of the plaque and the gingivitis)

  • The purpose of the present study was to determine the effect of a chewing gum containing 70% xylitol and 0.23% Curcuma xanthorrhiza extract to remove dental plaque and reduce gingivitis when used as a supplement to daily toothbrushing for 3 weeks. The study group consisted of 75 adults with moderate gingvitis. Participants were divided into 3 groups(contol 1 group - Gum base, contol 2 group - 70% Xylitol, experiment group - 70% xylitol and 0.23% xanthorrhizol) and instructed to chew the study gum for 3 times daily for 3 weeks in addition to regular daily toothbrushing. Chewing xylitol/xanthorrhizol gum significantly reduced plaque index with significant difference by week 3(p<0.01). Gingival index and bleeding on probing were decreased in xy/ Curcuma xanthorrhiza extract group by 35.9% and 31.65% each in the same period. No adverse effects on the oral tissues were observed in any of the participants for the duration of the study. In conclusion, regular use of a chewing gum containing 70% xylitol and 0.23% Curcuma xanthorrhiza extract appears safe and effective for the removal of dental plaque and reduction of gingivitis when used in conjunction with daily toothbrushing.

갈근강황추출물의 갱년기 증상 개선에 대한 유효성 및 안전성 평가 (Efficacy and Safety of Extract Obtained from Pueraria thunbergiana Bentham and Curcuma longa L. on the Improvement of the Menopausal Symptoms)

  • Objectives: This clinical trial is performed to investigate the effect of improving the menopausal symptoms and the safety of extract obtained from Pueraria thunbergiana Bentham and Curcuma longa L. in menopausal women Methods: This study was randomized, double blinded, placebo-controlled clinical trial. 110 women with menopausal symptoms were assigned to treatment group (n=55) and control group (n=55). The results of this study were evaluated through changes in Kupperman Index score, Menopause-specific Quality of Life Questionnaire (MENQOL) score, degree of hot flashes and sweating, Body Mass Index(BMI), laboratory test results including female hormones. Results: In the treatment group compared to before baseline, there was significant difference in the Kupperman Index total score, score of each item in the Kupperman Index, MENQOL total score, hot flush and sweating visual analogue scale (VAS), hot flush score, and hot flush duration at one time. However, there was also a significant change compared to baseline in the control group, there was no significant difference between the treatment group and the control group. There was no significant change in female hormone levels in both group, and there were no adverse events associated with the clinical trial product. Conclusions: This clinical trial showed that extract obtained from Pueraria thunbergiana Bentham and Curcuma longa L. was helpful in improving menopausal symptoms in menopausal women without significant side effects.

전립선 비대유도 쥐의 전립선 조직에서 울금 급여에 따른 염증인자의 변화 (Effect of Hot Water Extract from Curcuma longa L. on Inflammatory Mediators in Prostate Tissue of BPH-Induced Rats)

  • 본 연구에서는 테스토스테론으로 전립선 비대를 유도한 래트에 울금(Curcuma longa L.)의 열수추출물을 다양한 농도로 급여하여 그에 따른 염증인자의 변화를 검토하였다. 조직학적 분석에서 울금 열수추출물(CL) 급여에 따라 전립선비대가 완화되는 것을 검토할 수 있었으며, 염증성 사이토카인인 TNF-${\alpha}$, IL-6 및 IL-$1{\beta}$이 전립선 비대 유도군에서 그 발현이 현저히 증가하였으나, 각 농도의 CL 군에서는 농도의존적으로 감소하였다. 또한, CL 급여에 따라 finasteride enzyme인 COX-2와 염증 전사인자인 NF-${\kappa}b$의 활성화의 감소를 확인하였다. 이와 같은 결과는 전립선 비대에서 울금 열수추출물이 항염증 효과로써 전립선 비대의 유발 및 발전을 억제시킬 수 있다는 것을 부분적으로나마 설명할 수 있는 기초 자료가 될 수 있다고 사료된다.

울금(鬱金) 추출물의 미백 활성 및 진피 섬유구조 단백질에 미치는 효과 (Effect of Curcuma longa L. Extract on the Melanin Accumulation and Expression of Skin Fibril Proteins)

  • Objectives : In this study, various biological effects of Curcuma longa L. have been studied, however, beneficial effect of Curcuma longa L. in skin health remain still unclear. In this study, Curcuma longa L. water extract (CLE) was prepared. Inhibitory effect of CLE on melanin accumulation of B16F10 cells and expression levels of skin fibril-related proteins of human skin fibroblasts (HSF) were evaluated. Methods : The cytotoxic effect of CLE in B16F10 cells and HSF were examined by MTT assay. Inhibitory effect of CLE on the ${\alpha}-MSH-$ and IBMX-induced melanin accumulation and tyrosinase activity were evaluated in B16F10 cells. The expression levels of connective tissue growth factor (CCN2), Smad2, procollagen $1{\alpha}2$, collagen $1{\alpha}2$, and fibronectin in CLE-treated HSF were analyzed by western blotting. Results : The CLE treatment (concentrations 10 to $400{\mu}g/ml$) for 72 h did not affect to the B16F10 viability. However, 200 and $400{\mu}g/ml$ of CLE treatment for 24 h showed cytotoxic effect in HSF. Therefore, the concentrations 10, 50, and $100{\mu}g/ml$ of CLE were chosen in this study. The CLE treatment for 72 h dose dependently and significantly suppressed melanin accumulation and tyrosinase activity of B16F10 cells. In addition, the CLE treatment up-regulated expression levels of skin fibril-related proteins such as CCN2, Smad2, procollagen $1{\alpha}2$, collagen $1{\alpha}2$, and fibronectin. Conclusions : In conclusion, these results suggest that the CLE could be used as a natural material for skin health.

Hypoglycemic effect of a polyherbal aqueous extract in experimentally induced diabetic rats

  • The present study was carried out to investigate the hypoglycemic effect of a polyherbal aqueous extract (Curcuma longa Linn., Emblica officinalis Gaertn., Trigonella foenum-graecum Linn., Enicostemma littorale Blume) in alloxan-induced diabetic rats. Short term experiments showed a decrease in blood glucose levels at $2^{nd}\;hr$ of administration of the aqueous extract in alloxan-induced diabetic rats with increase in serum insulin levels. The extract did not show any effect on blood glucose or serum insulin levels in normoglycaemic rats. Treatment with the extract (1.5 g dry plant equivalent extarct/100 g body weight/day) for 20 days in diabetic rats showed a significant decrease in blood glucose and glycosylated haemoglobin levels and an increase in serum insulin levels. The aqueous extract also showed an enhanced glucose-induced insulin release at 11.1 mM glucose from isolated rat pancreatic islets. The extract did not show any toxicity at the particular dose used.

천연추출물 Curcuma xanthorriza oil 함유치약의 치태 및 치은염 억제효과 (Suppressive effect of Curcuma xanthorrhiza oil on plaque and gingivitis)

  • To find out the suppressive effect of natural extract Curcuma xanthorrhiza on $IL-1{\beta}$ and MMP-2 derived from periodontal ligament cells through in vitro study and to confirm its effect on plaque and gingivitis through clinical study, Curcuma xanthorrhiza containing toothpaste was used and following results were produced. 1. In vitro study, type IV collagenase MMP-2 production was inhibited dose-dependently in the group treated with Curcuma xanthorrhiza compared to the control group. 2. In vitro study, the production of $IL-l{\beta}$ which is one of the inflammatory mediators associated with periodontitis was inhibited dose-dependently in the group treated with Curcuma xanthorrhiza. 3. On the third week, the plaque index of the groups treated with or without Curcuma xanthorrhiza containing toothpastes were both increased significantly compared to the baseline(p<0.05). 4. On the third week, the gingival index of the group treated with Curcuma xanthorrhiza containing toothpaste was not significantly different from baseline. However, the group treated without Curcuma xanthorrhiza containing toothpaste showed a significant increase of gingival index at shielded area(p<0.05). 5. The gingival index of the group without Curcuma xanthorrhiza containing toothpaste showed a significant increase in the sites without tooth brushing when compared to sites with tooth brushing(p<0.05). However. there was no significant difference for the group with Curcuma xanthorrhiza containing toothpaste in sites either with or without tooth brushing. 6. The Bleeding on probing for the group without Curcuma xanthorrhiza containing toothpaste showed no significant difference even when tooth brushing was done. However, for the group with Curcuma xanthorrhiza containing toothpaste, bleeding on probing was significantly reduced compared to baseline when tooth brushing was done(p<0.05).

4가지 한방 소재(행인, 호장근, 자초, 강황)의 항산화활성에 관한 연구 (Study on Anti-oxidant Activity of Four Kinds of Korea Herb Medicine Materials)

  • There was an increasing interest that herbal medicine and natural material extracts were proved processes of antioxidant, cosmeceutical activity and the other effects. The aim of this study was to assess the antioxidant of extraction of four kinds from Prunus armeniaca L., Reynoutria elliptica, Curcuma aromatica, Lithospermum erythrorhizon. RE (Reynoutria elliptica) and CA (Curcuma aromatica) have good electron donating ability. The water and ethanol extract of RE at a 100 ppm concentration showed over 70%, the water extract at 500 ppm concentration showed 83% and the ethanol extract at 100 ppm concentration showed 86% of CA. Xanthine oxidase inhibition activity of the water extract of LE (Lithospermum erythrorhizon) at a 1,000 ppm concentration showed over 44%, on the other hand, RE showed in all lowest effect and there was no inhibition activity of a couple more extracts. In the measurement of nitrite scavenging activity, all extracts showed highly scavenging activity. Especially the water and ethanol extract of RE showed over 99% at 500 ppm, also LE showed over 40% at 10 ppm concentration.

울금(Curcuma longa L.) 추출물의 산화억제 및 질소산화물 소거활성 (Evaluation of Oxidation Inhibition and Nitrogen Oxide Scavenging Activity from Curcuma longa L. Extracts)

  • 울금(Curcuma longa L.)의 산화억제 및 질소산화물 소거활성 등 기능성 소재로서 활용 가능성을 검토한 결과, 프로안토시아니딘(proanthocyanidin) 함량은 $69.000{\pm}2.737mg$ catechin equivalents (CE)/g dry weight으로 확인되었으며, 증류수(distilled water, DW), 70% 에탄올 및 노르말 부탄올(n-butanol)의 3가지 용매를 사용한 추출 수율은 DW (17.11%), 70% 에탄올(15.26%), 노르말 부탄올(4.12%) 순으로 관찰되었다. 총 플라보노이드(total flavonoid) 함량은 DW, 70% 에탄올 및 노르말 부탄올에서 각각 0.032, 0.512 및 2.221 mg quercetin equivalents (QE)/g의 함량으로 나타났고, 노르말 부탄올에서는 유의적인 차이를 보이며 높게 관찰되었다(p<0.05). Nitric oxide (NO) 라디칼 소거 활성은 농도 별(0.2~0.8 mg/mL) DW에서 15.64~26.20%, 70% 에탄올 10.52~20.76%, 노르말 부탄올에서 13.39~69.92%로 확인되었다. Nitrite ($NO_2$) 소거활성은 DW 및 70% 에탄올과 비교하였을 때 노르말 부탄올에서 강한 $NO_2$ 소거활성을 보였다. ${\beta}$-carotene 탈색 저해활성은 DW에서 8.81~25.93%, 70% 에탄올 1.20~20.20%, 노르말 부탄올 12.08~43.93%로 나타났다. 지질과산화 저해활성은 DW, 70% 에탄올 및 노르말 부탄올에서 각각 5.60~27.54%, 37.78~50.79% 및 41.79~46.39%로 동정되었다. 이에, 천연 항산화제 등 기능성 소재로서의 활용 가능성이 있을 것으로 사료된다.

Inhibitory Effect of Standardized Curcuma xanthorrhiza Supercritical Extract on LPS-Induced Periodontitis in Rats

  • Periodontitis, which is a severe inflammatory disease caused by endotoxins secreted from oral pathogens, destructs gingival tissue and alveolar bone. Curcuma xanthorrhiza, commonly called Java turmeric, has been shown to possess anti-bacterial and anti-inflammatory activities. The present study evaluated the inhibitory effect of C. xanthorrhiza supercritical extract (CXS) standardized with xanthorrhizol on lipopolysaccharide (LPS)-induced periodontitis in an animal model. LPS was topically injected into the periodontium of Sprague-Dawley rats to induce periodontitis and CXS (30 and $100mg{\cdot}kg^{-1}{\cdot}day^{-1}$) was orally administered after day 12. Histologically, CXS inhibited the collapse of gingival tissue by preventing cell infiltration. CXS significantly downregulated the expression of matrix metalloproteases (MMPs) and inflammation-related biomarkers, such as nuclear factor-kappa B ($NF-{\kappa}B$) and interleukin-1 beta ($IL-1{\beta}$) in gingival tissue. CXS also improved bone remodeling by downregulating osteoclastic transcription factors, such as nuclear factor of activated T-cells c1 (NFATc1), tartrate-resistant acid phosphatase (TRAP), and cathepsin K. In addition, CXS upregulated osteoblast differentiation-related markers, alkaline phosphate (ALP) and collagen type I alpha (COLA1). Thus, CXS can ameliorate periodontitis by inhibiting inflammation and improving bone remodeling.
