The purpose of this study is to investigate the state projects related to psychological sentiment for child and youth in Korea, examine the current status of art treatment-related projects for child and youth, and seek ways to improve them, focusing on projects supported by the Ministry of Health and Welfare, the Ministry of Gender Equality and Family, and the Ministry of Culture, Sports and Tourism. The results examined through this study were firstly, mental health promotion projects conducted in the public sector in Korea are mainly carried out on consignment to affiliated organizations, and are promoted not only for diagnosis, mediation, and therapeutic intervention but also for care and prevention. Second, psychological and emotional support projects are being attempted with various approaches such as reading, playing, gardening, and art, and art therapy is actively used as a part of art programs. In addition, it is suggested that the government should provide efficient welfare and utilize it as a common goal through linkage and cooperation with other ministries for the continuous and stable implementation of state-led psychological support projects. In addition, it is hoped that this research will be used as a research material to explore specific strategies for effectiveness of national policy projects on the overall psychological sentiment of child and youth.
Public data has been transformed from provider-oriented information disclosure to a form of personalized information sharing centered on individual citizens since government 3.0. As a result, the government is implementing policies and projects to maximize the value of public data and increase reuse. This study analyzes the issues related to public data in the news and seeks the status of government agencies and government projects by issue. We conducted semantic analysis on domestic online news and public agency bidding information including public data and conducted the work of linking major key words derived with social and economic values inherent in public data. As a result, major issues related to public data were divided into broader access to public data, growth of new technology, cooperation and conflict among stakeholders, and utilization of the private sector, which were closely related to transparency, efficiency, participation, and innovation mechanisms. Also major agencies of four issues include the Ministry of Strategy and Finance and Seoul, Ministry of Culture, Sports and Tourism and Gyeonggi-do, Ministry of Trade, Industry and Energy and Incheon, and Ministry of Land, Infrastructure and Transport and Gyeongsangbuk-do. Most of the issues are being led by the government.
A thermotolerant yeast strain, Saccharomyces cerevisiae KNU5377 (abbreviated as KNU5377, was exposed to inorganic acids including sulfuric, nitric and hydrochloric acid. As a stressor, each inorganic acid is very easily dissociated in water, resulting in lowering environmental pH. When compared with a reference S. cerevisiae ATCC24858, KNU5377 could overcome such a severe condition containing a final 0.4% concentration of sulfuric acid or nitric acid to grow at the overnight culture, but this reference could not. Additionally, this strain showed a surprisingly strong tolerance by surviving despite of exposure to the regime of 0.35% of hydrochloric acid for over 90 min and also to 0.6% of sulfuric acid for 30 min. On the contrary, both strains could not survive against a final 0.45% concentration of nitric acid. This strain KNU5377 could produce ethanol of 3% in 2 days by using the fermentation medium containing a final 0.3% concentration of sulfuric arid. Moreover, change into a final 0.2% concentration of sulfuric acid caused this strain to enhance fermentation productivity up to about 4.5% even at $40^{\circ}C$. In exposure to a final 0.2% of sulfuric acid for 60 min, trehalose was most accumulated within 30 min in KNU5377, and this suggested a cellular defense system led by this disaccharide was profitable for this strain to lead to no morphological changes.
This study analysed the components and characters of welfare rights through discussions of the concepts and types of universal rights, and discussions of human rights and citizenship rights. Economic rights are composed of the right of labor, intervension of labor market, job security, and capital control. cultural environmental rights are composed of the right of culture and environment. And welfare rights has several characters. First, it is natural rights which is bestowed on the citizens or people in modern civil societies. it is samely characterised as liberties and political rights. second, it has the same values like other rights such as lberties and political rights. Or it is more important, because it is necessary for other rights. Third, it is not the objective being which is constant, but it is changed, formed and constructed as total rights with human rights and citizenship rights. Fourth, it is truely rights, but is simultaniously accompanied by obligations. But the obligations is unconditional like as other rights. Endly, levels of welfare on the welfare rights must be modicum rather than minimum. The meaning of modicum level is uncertain, but it aims to the entire participation of peoples as citizen and social integration. And it has to aim to the prevention of heridity and continuity of inequality.

Despite the significance of external environmental factors in differentiation, putative factors involved in differentiation of Aspergillus nidulans have not yet been fully understood. A sporulation-specific proteome analysis of A. nidulans in the present study revealed that the expression levels of more than 2,400 proteins were affected under conditions inducing sporulation (0.6 M KCl) compared with normal conditions. Among the proteins with predicted functions, two targets, AN1342 and AN9419, were functionally analyzed using targeted deletion strains and phenotypic observations. For AN1342, because the deletion of the corresponding open reading frame caused a reduction in stalk length during asexual development and in pigment production in liquid culture, the gene was designated as sspA ($\underline{s}hort$$\underline{s}talk$ & $\underline{p}igment$). Deletion of the AN9419 gene, which is predicted to encode alanyl-tRNA synthetase, led to severe growth defects due to alanine auxotrophy and abolishment of asexual reproduction and thus, the gene was designated as alaA.

Objective : Recent studies have shown encouraging progress toward the use of autogenic and allogenic mesenchymal stem cells (MSCs) to arrest, or even lead to partial regeneration in, intervertebral disc (IVD) degeneration. However, this technology is still in its infancy, and further development is required. The aim of this study was to analyze whether rat adipose-derived mesenchymal stem cells (ADMSC) can differentiate towards IVD-like cells after treatment with transforming growth factor ${\beta}3$ (TGF-${\beta}3$) in vitro. We also performed quantitative analysis of gene expression for ADMSC only, ADMSCs treated with TGF-${\beta}3$, and co-cultured ADMSCs treated with TGF-${\beta}3$. Methods : ADMSCs were sub-cultured to homogeneity and used in fluorocytometry assays for CD11, CD45, and CD90/Thy1. ADMSCs were differentiated in spheroid culture towards the chondrogenic lineage by the presence of TGF-${\beta}3$, dexamethasone, and ascorbate. We also co-cultured pure ADMSCs and nucleus pulposus cells in 24-well plates, and performed immunohistochemical staining, western blotting, and RT-PCR for quantitative analysis of gene expression. Results : Results of fluorocytometry were positive for CD90/Thy1 and negative for CD11 and CD45. TGF-${\beta}3$-mediated induction of ADMSCs led to the expression of the differentiation markers of intervertebral disc-like cells, such as aggrecan, collagen II, and sox-9. Co-cultured ADMSCs treated with TGF-${\beta}3$ showed higher expression of differentiation markers and greater extracellular matrix production compared with ADMSCs treated with TGF-${\beta}3$ alone. Conclusion : ADMSC treated with TGF-${\beta}3$ may be an attractive source for regeneration therapy in degenerative IVD. These findings may also help elucidate the pathologic mechanism of MSC therapy in the degeneration of IVD in vivo.

Fresh Kills is the largest landfill in the world located in the west side of Staten Island, New York. The landfill served as a storage area for New York City's trash for more than 50 years. After years of civilian and political pressure, state and local legislation decided its closure of landfill operation in Fresh Kills in March 2001. Soon after, Department of City Planning announced a Fresh Kills international design com-petition: 'Landfill to Landscape'. The winning entry was promised to be outline for the redevelopment of the 2,200 acre site which the size of three times Central Park. Forty-eight teams representing more than 200 offices from around world submitted proposals, from which six finalists that mostly led by landscape architects were selected. In December 2001, a jury of architects, landscape architects and city officials unanimously selected Field Operations as the winner. The plan, named Lifescape, visualizes the gradual 20-year transformation of the whole Staten Island into a 'natural lifestyle island' recognizing that Staten Island is home to coastal wetlands that shelter one of the most diverse ecosystems in the New York metropolitan area. It suggested that an ecologically reconstituted Fresh Kills could become the center of integrated parks and greenways system on the island otherwise fragmented. The project will be one of the largest and most ambitious undertakings in the metropolis in years developing a complex web of habitats and parklands on top of mountain of trash. This study tries to achieve two goals: One is to provide general explanations on the project, Lifescape, breaking down to its background, geographical context, design concepts and phased development plan. Another is to introduce the unique and innovative design approaches by Field Operations that are different from a conventional landscape architectural attitude. Since this project was well published through many magazines and newspapers, main focus will be upon aspects that differentiate this project from usual landscape projects. Conceptually Lifescape brought provocative notions on nature/culture relationship and the role of urban park as an active agency rather than just a green rest area. Also this project introduced pioneering graphics like plan collage, diagrammatic plan, phasing diagram and photo montage as vehicles conveying information, imagination and provocation. Witnessing the influence of the project gradually in the field of academic and practice in the States, this study is intended to become a constructive reference to similar landscape projects dealing with large and complex urban context in conjunction with restructure of contemporary city.
A psychrotrophic bacterial strain, Pseudomonas fluorescens BM07, synthesized unsaturated fatty acids (UFA) from fructose in response to lowering of growth temperature, and incorporated them into both polyhydroxyalkanoic acid (PHA) and membrane lipid. The blocking of PHA synthesis by adding 5 mM 2-bromooctanoic acid to the growth medium, containing 70 mM fructose, was found to be a useful means to profile the composition of membrane lipid by gas chromatography. As the growth temperature changed from 35 to $50^{\circ}C$, the total content of two UFA, 3-hydroxy-cis-5dodecenoic acid ($C_{12:1}$) and 3-hydroxy-cis-7-tetradecenoic acid ($C_{14:1}$), in PHA increased from 31 to 44 $mol\%$. The growth at lower temperatures also led to an increase in the level of two major UFA, palmitoleic acid (C16:1 cis9) and cis-vaccenic acid (C18:1 cis11), in membrane lipid. A fraction of these membrane-lipid UFA was converted to their corresponding cyclopropane fatty acids (CFA). The CFA conversion was a function of culture time, exhibiting biphasic increase before and after entering the stationary phase. However, pH changes in growth media had no effect on the CFA conversion, which is contrary to the case of E. coli reported. The cells grown at $30^{\circ}C$ responded to a cold shock (lowering the medium temperature down to $10^{\circ}C$) by increasing the level of C16:1 cis9 and C 18: I cis II up to that of $10^{\circ}C$-grown control cells and concomitantly decreasing the relative level of cis-9,10methylenehexadecanoic acid (the CFA converted from C16:1 cis9) from 14 to 8 $mol\%$, whereas the 10-grown cells exhibited little change in the lipid composition when exposed to a warmer environment of $30^{\circ}C$ for 12 h. Based on this one- way response, we suggest that this psychrotrophic strain responds more efficiently and sensitively to a cold shock than to a hot shock. It is also suggested that BM07 strain is a good producer of two unsaturated 3-hydroxyacids, $C_{12:1}\;and\;C_{141:1}$.
Trichoderma reesei is the major filamentous fungus used to produce cellulase and there is huge interest in promoting its ability to produce higher titers of cellulase. Among the many factors affecting cellulase production in T. reesei, the mycelial phenotype is important but seldom studied. Herein, a close homolog of the Neurospora crassa COT1 kinase was discovered in T. reesei and designated TrCOT1, which is of 83.3% amino acid sequence identity. Functional disruption of Trcot1 in T. reesei by RNAi-mediated gene silencing resulted in retarded sporulation on potato dextrose agar and dwarfed colonies on minimal medium agar plates containing glucose, xylan, lactose, xylose, or glycerol as the sole carbon source. The representative mutant strain, SUS2/Trcot1i, also displayed reduced mycelia accumulation but hyperbranching in the MM glucose liquid medium, with hyphal growth unit length values decreased to 73.0 ㎛/tip compared to 239.8 ㎛/tip for the parent strain SUS2. The hyperbranching phenotype led to slightly but significantly increased cellulase secretion from 24 to 72 h in a batch culture. However, the cellulase production per unit of mycelial biomass was much more profoundly improved from 24 to 96 h.

This paper is to consider the new urbanization process of Daegu in the 1990s, especially with its economy, politics, social culture, environment and space. In the economic aspect, the industrial structure of Daegu has shifted from the manufacturing- centered economy to the service economy. But it has not yet overcome the linear frame of industrial structure still relying on the textile industry, though the degree of industrial specialization has increased in the part of the fabricated metal products and the precision and optical instrument. In the political situation that the implementation of the local self-government has made the relation between localities more competitive, the local government of Daegu has led a boom for boosting the regional economic growth, organizing private-public-research relations to improve the weakening regional investment and production function. In the social and cultural aspect of the new urbanization, the investment into the soft-ware facilities has been increased, and the urban festivals have been changed so as to revitalize the regional economy. In the environmental aspect, as the self-government system has launched, conflicts around values and interests of local governments have revealed frequently due to locations of infra-structures and of abhorrence facilities. Finally, seen from the spatial dimension of the city, the extension of residential areas and unregulated use of urban space have a result of inefficience of land-use, and this kind of unplanned outer expansion of the city has brought about with further separation of house and working place, and increasing distance of movements and the an urban spatial structure which requires more energy consumption.
