Introduction of a speedy growing trait into Pleurotus eryngii by backcrossing (큰느타리(새송이)버섯에서 역교배를 통한 속성형질의 도입)

  • Two strains which have good traits in quality and speedy growing were selected based on previous report(Ryu, 2007) to breed a new strain carrying the traits in it. KNR2322 carrying high quality and KNR2503 carrying a speedy growing trait were cultivated to harvest monokaryons. The monokaryotic mycelia were characterized in growth rate, color, and stability. Selfing of monokaryons from KNR2322 and crossing between KNR2322- and KNR2503-derived monokaryons were performed. Two parental lines, $KNR2322-4{\times}37-6{\times}12(B)$ showing thremmatological values in weight and quality, 68.5g and 6.5 and $KNR2322-30{\times}KNR2503-60(A)$ was harvested in 13.5 days after scraping old medium were selected. Crossing between A and B resulted in two high quality and speedy growing strain, A8B8 and A8B10. Their days for harvest after scraping, weight, and quality were 14.0 days, 88.8g and 92.7g, 7.0 and 7.1. Originality tests were conducted with genomic DNAs of A8B8 and Keuneutari 3ho(commercial strain) by DNA fingerprinting a confrontation culture. The results showed polymorphism and a inhibition line between them. It suggested that the new strain have originality from the commercial strain. A8B8 have been registered in Korea Seed and Variety Service with as "Saesongi 1ho".

Nutrition Practice to Alleviate the Adverse Effects of Stress on Laying Performance, Metabolic Profile and Egg Quality in Peak Producing Hens: II. The Probiotic Supplementation

  • In this experiment, the effects of cage density (CD) and probiotic supplementation (PS) on laying performance, metabolic profile, and egg quality in peak-producing hens were evaluated. After blocking according to the cage location, Lohman layers (n = 180, 46 wks of age) were allocated randomly to two levels of CD (540 vs. 360 $cm^2$/hen) and three levels of PS (0, 0.15, and 0.30%). Probiotic contained Enterococcus faecium culture (10${\times}$10$^9$ cfu/g). Egg production (EP) and feed consumption (FC) were measured daily; egg weight (EW) was measured bi-weekly; BW was measured before and after the experiment; and blood samples were obtained at the end of the experiment. The data were analyzed using two-way ANOVA. Increasing CD decreased FC (125.0 vs. 120.8 g/d, p<0.0001) and FCR (1.93 vs. 1.87, p<0.0001) and did not alter EP, EW, and BW. Increasing level of PS linearly decreased FC (p<0.02) and FCR (p<0.006). Averages were 123.9, 123.2, and 121.6 g/d for FC and 1.91, 1.92, and 1.86 for FCR in hens supplemented with 0, 0.15, and 0.30% probiotic, respectively. Hens placed in high-density cages had greater serum corticosterone concentration than hens placed in normal-density cages (12.8 vs. 11.3 $\mu$g/dL, p<0.04); CD did not affect concentrations of other metabolites. Increasing level of PS linearly increased serum glucose, albumin, and creatine concentrations and quadratically increased total protein, globulin, Ca, and P concentrations. Average concentrations (mg/dL) were 260, 297, and 305 for glucose; 6.28, 8.09, and 7.58 for total protein; 1.98, 2.48, and 2.38 for albumin; 4.30, 5.62, and 5.19 for globulin; 0.40, 0.52, and 0.54 for creatine; 16.0, 16.5, and 16.3 for Ca; and 6.27, 8.14, and 7.17 for P in hens supplemented with 0, 0.15, and 0.30% probiotic, respectively. There was no effect of CD on egg quality. Increasing level of PS linearly improved yolk color (YC) and quadratically increased albumen index (AI) and Haugh unit (HU). The mean values were 9.67, 9.75, and 10.58 for YC; 8.94, 6.93, and 8.72% for AI; and 85.6, 74.9, and 82.9 for HU for hens supplemented with 0, 0.15, and 0.30% probiotic, respectively. There was also CD by PS effect on FC, EP, and serum glucose, total protein, albumin, globulin, creatine, Ca and P concentrations. In conclusion, increased CD partially depressed laying performance and caused stress. Probiotic supplementation improved laying performance and metabolic profile. It also partially alleviated the adverse effects of stress resulting from increased caging density.

Evaluation of adenosine triphosphate testing for on-farm cleanliness monitoring compared to microbiological testing in an empty pig farrowing unit

  • Careful cleaning and disinfection of pigpens is essential to prevent disease spread and avoid the resultant economic losses. Hygiene in pigpens is generally evaluated by visual monitoring supplemented with bacteriological monitoring, which includes counting the total aerobic bacteria (TAB) and/or fecal indicator bacteria (FIB). However, these methods present drawbacks such as time and labor requirements. As adenosine triphosphate (ATP) is ubiquitous in all living organisms including microorganisms, this study aimed to directly compare the results of microbial assessment and ATP quantification, and to suggest possible detailed application methods of the ATP test for hygiene evaluation in pigpens of a farrowing unit. Before and after standard cleaning procedures, samples were collected from the floor corner, floor center, and feeding trough of four pigpens at different time points. No FIB were detected and both the TAB and ATP levels were significantly decreased in the floor center area after cleaning. FIB were continuously detected after cleaning and disinfection of the floor corners, and there was no significant ATP level reduction. The feeding trough did not show any significant difference in these values before and after cleaning, indicating insufficient cleaning of this area. The levels of TAB and ATP after cleaning were significantly correlated and the average ATP value was significantly lower in the absence of FIB than in their presence. In the absence of standard references, a more thorough hygiene management could be achieved evenly by supplementing cleaning or disinfection based on the lowest ATP results obtained at the cleanest test site, which in the present study was the floor center. Overall, these results indicate that the on-farm ATP test can be used to determine the cleanliness status, in addition to visual inspection, as an alternative to laboratory culture-based testing for the presence of microorganisms.

  • Recently, as a culture that values work-life balance has become more important in society as a whole, interest in hobbies and leisure activities that will enrich life is increasing. This phenomenon is due to an increase in platforms where people can enjoy and learn hobbies online. Among them, craft-related content is growing rapidly, and as offline education has been brought online, learning immersion has been reduced online. Therefore, in this study, we want to identify current problems in online and offline craft learning and seek ways to improve UI design to solve them. To this end, we conducted a literature survey on the online learning platform, online craft learning platform and UI design, and based on this, conducted case investigation and analysis to derive the third stage of the learning process of the online craft learning platform. Subsequent surveys showed that as a result, the online craft learning platform was able to find improvements in terms of differentiating the content exploration process, visualizing the craft progress stage, ease of working with video tools, and providing smooth feedback, and in addition to the need for a device for active learning. Through the results of this study, we hope to expand the craft content market by making it easier for users to enjoy the contents provided on the online craft learning platform.

A Study on the Importance and Valuation of Public Functions in Private Botanical Gardens (사립식물원 공익적 기능의 중요도 및 가치평가 연구)

  • The role of the botanical garden in securing biodiversity, responding to climate change, and sustainable development in modern cities is becoming more important. Private botanical gardens that lead the domestic botanical culture are declining due to complex reasons such as an increase of tourist destinations, lack of policy support, lack of introduction of advanced management strategies, and similar leisure activities to national and public botanical gardens. The Private Botanical Garden not only has a large number of plant species with high conservation value, but also provides various public utilities as a cultural and educational space and government efforts are needed to activate the operation of this amenity. The purpose of this study is to evaluate the value of public functions provided by private botanical gardens using the Contingent Valuation Method (CVM). In this way, the government aims to provide a basis for policy and institutional support for private botanical gardens. The main results are as follows. First, public utility functions provided by private botanical gardens were recognized as 'preservation' (23.4%), 'recreation and tourism' (17.5%) and 'research' (16.6%) in order of analytic hierarchy process (AHP). Second, 'heritage value' (33.0%) and 'existence value' (32.5%) were recognized as significant among the values provided by private botanical gardens. Third, the willingness to pay (WTP) to preserve the public functions of the private botanical garden was 12,234 won. Based on this, the economic value of all private botanical gardens in the whole country was estimated, resulting in about 233.8 billion won. There is a need to revise laws and regulations related to financial support for the revitalization and quality improvement of private botanical gardens. It is also necessary to establish a cooperative operating system between national, public and private botanical gardens.

Optimal Fermentation Conditions of Hyaluronidase Inhibition Activity on Asparagus cochinchinensis Merrill by Weissella cibaria

  • This study was conducted to evaluate the hyaluronidase (HAase) inhibition activity of Asparagus cochinchinesis (AC) extracts following fermentation by Weissella cibaria through response surface methodology. To optimize the HAase inhibition activity, a central composite design was introduced based on four variables: the concentration of AC extract ($X_1$: 1-5%), amount of starter culture ($X_2$: 1-5%), pH ($X_3$: 4-8), and fermentation time ($X_4$: 0-10 days). The experimental data were fitted to quadratic regression equations, the accuracy of the equations was analyzed by ANOVA, and the regression coefficients for the surface quadratic model of HAase inhibition activity in the fermented AC extract were estimated by the F test and the corresponding p values. The HAase inhibition activity indicated that fermentation time was most significant among the parameters within the conditions tested. To validate the model, two different conditions among those generated by the Design Expert program were selected. Under both conditions, predicted and experimental data agreed well. Moreover, the content of protodioscin (a well-known compound related to anti-inflammation activity) was elevated after fermentation of the AC extract at the optimized fermentation condition.

  • Public data has been transformed from provider-oriented information disclosure to a form of personalized information sharing centered on individual citizens since government 3.0. As a result, the government is implementing policies and projects to maximize the value of public data and increase reuse. This study analyzes the issues related to public data in the news and seeks the status of government agencies and government projects by issue. We conducted semantic analysis on domestic online news and public agency bidding information including public data and conducted the work of linking major key words derived with social and economic values inherent in public data. As a result, major issues related to public data were divided into broader access to public data, growth of new technology, cooperation and conflict among stakeholders, and utilization of the private sector, which were closely related to transparency, efficiency, participation, and innovation mechanisms. Also major agencies of four issues include the Ministry of Strategy and Finance and Seoul, Ministry of Culture, Sports and Tourism and Gyeonggi-do, Ministry of Trade, Industry and Energy and Incheon, and Ministry of Land, Infrastructure and Transport and Gyeongsangbuk-do. Most of the issues are being led by the government.

  • This study analysed the components and characters of welfare rights through discussions of the concepts and types of universal rights, and discussions of human rights and citizenship rights. Welfare rights is claims rights which requires somewhat, and it is positive rights. And it is generally passive rights, but it contains collective participation rights which is active rights. The result of total discussions of rights, human rights, and citizenship rights led us to know the components and characters of welfare rights. Welfare rights contains social rights, economic rights, and cultural environmental rights. Social rights are composed of the right of social security, social welfare service, health, education, and residency. Economic rights are composed of the right of labor, intervension of labor market, job security, and capital control. cultural environmental rights are composed of the right of culture and environment. And welfare rights has several characters. First, it is natural rights which is bestowed on the citizens or people in modern civil societies. it is samely characterised as liberties and political rights. second, it has the same values like other rights such as lberties and political rights. Or it is more important, because it is necessary for other rights. Third, it is not the objective being which is constant, but it is changed, formed and constructed as total rights with human rights and citizenship rights. Fourth, it is truely rights, but is simultaniously accompanied by obligations. But the obligations is unconditional like as other rights. Endly, levels of welfare on the welfare rights must be modicum rather than minimum. The meaning of modicum level is uncertain, but it aims to the entire participation of peoples as citizen and social integration. And it has to aim to the prevention of heridity and continuity of inequality.

Effect of Feeding Frequency of Extruded Pellet on Growth and Body Composition of Juvenile Flounder, Paralichthys olivaceus During the Winter Season (겨울철에 부상사료 공급횟수가 넙치 치어의 성장과 체성분에 미치는 효과)

  • A feeding trial of two feeding rate (satiation and $80\%$ satiation) $\times$ four feeding frequency (three meals a day, two meals a day, one meal a day and one meal every two days) factorial design with three replications was carried out to investigate the optimum feeding frequency and rate for growth of flounder during the winter season. Juvenile flounder averaging 45 g were fed a extruded pellet for 60 days. Weight gain was affected by both feeding frequency (P<0.001) and feeding rate (P<0.01). The best weight gain obtained from fish fed two meals a day with satiation was not different from that of fish fed three meals a day with satiation and $80\%$ satiation. Feed efficiency and protein efficiency ratio of fish fed the diet three times daily were the highest among groups, but not different to other groups of fish except for one meal every two days with satiation and $80\%$ satiation feeding. Daily feed intake was affected by both feeding frequency and feeding rate (P<0.001), these values of fish fed two and three meals a day with satiation were the highest among the groups. Moisture, crude protein, crude lipid and ash contents of fish were not affected by both feeding frequency and feeding rate (P>0.05). The results of this study indicate that the optimum feeding frequency for the growth of juvenile flounder grown from 45 g to 53 g is two meals a day with satiation feeding during the winter season.

Quality Changes of Vegetables by different Cooking Methods (조리방법에 따른 채소류의 품질 변화)

  • The quality changes of vegetables including cucumbers, young pumpkins, carrots, radishes, onions by different cooking methods in $90^{\circ}C$ water, 0.5% NaCl solution, 1.0% NaCl solution and $400^{\circ}C$ superheated steam were investigated. The changes in pH of cooked vegetables were increased. Hunter color system values for L(lightness), a(redness) and b(yellowness) of the vegetables cooked in $400^{\circ}C$ supetbeated steam for 30 seconds were less compared with those in $90^{\circ}C$ water, 0.5% NaCl solution and 1.0% NaCl solution. The microbial growth was inhibited very effectively when the vegetables cooked in $400^{\circ}C$ superheated steam for 30 seconds. After cooked in $90^{\circ}C$ water, 0.5% NaCl solution, 1.0% NaCl solution, $400^{\circ}C$ superheated steam, vitamin C content decreased by different ratio depending on cooking methods and vegetable varieties. Based on the results of the quality changes of cooked vegetables, the superheated steam cooking method showed a positive effect on sterilization and maintaining pH, color, hardness of vegetable tissues, vitamin C content. The data obtained from this study could be utilized as basic information for the optimum conditions of superheated steam cooking to develope HMR(Home Meal Replacement).
