Secretory Production of Human Leptin in Bacillus subtilis

  • Human leptin is identified as a 16kDa (146 amino acids) protein secreted from adipocytes which influences body weight homeostasis. In order to produce active leptin, the human obese gene coding for leptin was expressed in Bacillus subtilis WB600 strain which is deficient in six extracellular proteases. The recombinant leptin was produced in a culture supernatant, and in a culture supernatant, it was contained as high as 48% for total proteins. After simple purification steps, which consisted of ammonium sulfate precipitation and anion-exchange column chromatography, 2.3 mg of leptin with a purity greater than 95% was obtained from the 0.51 culture with the recovery yield of 38.3%. The purified leptin showed the correct folding structure with one disulfied bond.

Defining 'Islamic' Urbanity Through A Trans-Regional Frame

  • The word 'urbanity' literally means 'quality or state of being urban' where the criterion of urban economic and civic culture is assumed despite the general celebration of cultural uniqueness of urban centers. The narratives celebrating the uniqueness of urban centers since the ancient past till recent times could not get rid of the broad categorization of the urban models depending on their contextual networks of trade, mobility and culture. This paper attempts to explore whether the urban cultures in South Asia even preceding a global phenomenon like colonialism were actually reflecting an idea of urbanity where the urban culture, including planning and architecture reflected a trans-national model. This paper particularly concentrates on the medieval period when a pattern of urbanity took shape in this subcontinent under the influence of Islam, which could be explained by its particular idea of urban model, cultural exchange and vibrant trade networks.

  • The New Culture Movement marked the dawning of a turbulent era in China. By the time of May Fourth Movement, the cultural contradiction and confliction grew into an extensive and enduring social movement extending deep and far-reaching influence on the Chinese society. The huge difference and contradiction existing between the Beijing and Shanghai cultures contributed to the outbreak of this social movement. To be specific, there was an active human and information exchange involving the two leading Chinese cities and at the same time the old and new cultures, thoughts and moralities constantly acted upon each other.

Content Analysis of Previous Research to Examine Categorization and Methodology of Korean Food History Studies (선행연구 내용 분석을 통한 한국 음식사(飮食史) 연구의 범주 및 방법론 고찰)

  • This study summarizes the research methods, subjects, research processes, and achievements of major researchers by analyzing previous research results on Korean food history. The goal of the current study aimed to seek the methodology and direction of 'food history research'. Literature data from 1945 to 2022 were examined. Results of excavation research on ancient food literature were divided into the following stages: the 1980s, when the foundation for 'cataloging' was laid, and the 2000s, when 'digitization' was achieved. Achievements of each period were collected, and the achievements and limitations were analyzed. Next, the research results were classified into 'Food technology history', 'Recipe change history', and 'Food culture and dietary history'. We observed that around the 2000s, anthropology and folklore research perspectives were reflected, and the research on 'history of diet' reached a turning point. Our results indicate the possibility that food history can develop as a special historical area. This could be achieved by establishing an exchange system with other disciplines and creating a systematic curriculum.

  • This study suggests the popular culture theory as a basic framework to find out the characteristics of contemporary fashion show meeting the popular culture, and describes that the show is the popular culture of this generation. The culture is the way of our life unifying the world at common area of human being. The popular culture is to subdivide it into the public culture that shares aesthetic taste, and to borrow each other and develop it mutually because of the collapse of dichotomous boundary with high quality culture, and to represent the people’s thought and behaviour expanding their areas. The author has examined the characteristics of popular culture of modern fashion show by four collections, that is, commercialism, mass media, political relationship and interaction, etc: Firstly, at the commercialism, the fashion show combines it with other genre organically to do mass production and sales and produces cultural consumption goods supplying a lot of sight-seeing. Secondly, at the mass media, the image of contemporary fashion show has been spread out with mass media such as fashion magazines, cable TV and Internet beyond time and space to produce advertisement effects and makes new fashion and bring democratic culture. Thirdly, at political relationship, the fashion in its peak can make a big stream of wealth, so that it plays important roles under governmental regulations in this era and governments make efforts to support and develop it. Lastly, at interaction, the fashion show shares media functions to let users participate in the program and to exchange information as a feedback to influence each other.

Relationship between TQM Performance and Organizational Culture of Dietitians in Institutional Foodservice (단체급식소 영양사의 종합적 품질경영(TQM) 수행과 조직문화와의 관계 규명에 관한 연구)

  • This study was designed to investigate the correlation between perceived TQM performance and organizational culture of dietitians. The objective of the study is to help the management of foodservice by providing a direction which will elevate perceived TQM performance of dietitians and presenting plans which will ensure effective dietitians. Questionnaires handed out to 308 dietitians worked in institutional foodservice operation including elementary, middle and high schools, hospitals, business and industries. In terms of TQM importance and performance, the more important dietitians perceived, the higher their performance level rose. Data form the IPA, external and internal customer satisfaction, executive ability, communication system and technology, information technology and application ability, food process, strategy, and leadership were required further improvement by dietitians. Of present organizational culture model, human relations model and open systems model were more likely to be adopted by dietitians in middle and high schools. According to the type of foodservice management, the open systems model was more preferred by dietitians from self-operated operations rather than respondents from contracted operations. Canonical correlation analysis between TQM performance and organizational culture showed canonical correlation to be higher (canonical correlations coefficient: .66). In conclusion, TQM performance-organizational culture showed higher canonical correlation. In the organizational culture, foodservice operation is capable of improving the aims for the rational goal model and the open systems model. The results showed that TQM performance and organizational culture had significant relationship, especially positive organizational culture emphasizing on internal process and rational goal model would have influence on TQM performance of dietitians. Foodservice operation, however, should recognize importance of open and development culture to improve dietitians' TQM performance. To apply open system foodservice organization should encourage dietitian and foodservice employees to challenge and compete for the works, Moreover, organizational effort such as information exchange program and support system should be established.

  • Regarding the ancient Sino-Kroea sea communication as the main theme, the propagation of the China-culture in Koran Peninsula and he influence of Chinese ancient shipbuilding technique for Korea are described in the paper. Frequent sea trade resulted in the flourishing of Chinese and Korean harbours on both side of Yellow Sea. Sino-Koran sea communication is of long-standing. Korean Peninsula (KP) is just as the large arm extended to the great sea from the North-east China. It is the Chinese mainland plate-lump's extended part in the Yellow Sea. In the ancient time, the land communication between China and Korea was often not unimpeded, the contact between China and KP, such as the South-Korea, depended on the sea communication mainly, Although the friendly relationship between China and Korea rose one after another in the various historical period, but considering from the great background of people's historical development, just as the modern people benefited fro the Europe-American culture, the people of ancient time also benefited from the China-culture. All China, KP and Japan obtained the development by propagating the ancient China-culture. The seagoing ships which carried out the Sino-Korea sea communication and trade, as the friendly envoys, finished the mission linked u and developed the Sino-Korea culture contact.

  • If we look closely to the power shift of the powerful countries historically. during the 16th century Spain's power came from gold, colony trade, the mercenary force, close relation with the thrown, 17th century Netherlands owes their power to trade, capital market, the marines, 18th century France owes it to population, agriculture, public administration, and the army. Also, England had their industry, political unity, finance and trust, the marines, liberalistic principles, a geographical merit of being an island which can be defended well and the 20th century America has their cosmopolitan culture, supernational communication, the capacity of the economy, science technology, military strength, alliances, liberalism international formation. But in the 21st century culture and art will prevail over the information age where technology and knowledge was the key, and it is predicted that this will be the source of power for a strong country. Rolfe Yesson, the head of The Copenhagen Research Center for future studies said, "Information age has ended and in the future Dream Society will arrive which focuses on making distinctions by delivering dreams and emotions to consumers". As cyberspace gradually substitutes reality, cyberspace has become more than an information search engine and has become a place where people fulfill their desires and exchange culture.   • The Purpose of this research is to examine and contrast the cultural characteristics of the street fashion in new generations of Korea and Japan. We also intended to help clothing companies understand the current trend in this new generation. As the method for street fashion analysis. various literature, internet information and new media materials were utilized. Key observations obtained are as following First, they pursue a complex image. New generations seem to admix images from different areas to create a synthetic image, rather than adopting a single image; Second, practicality is an important factor. They Pursue a sense of their own unique trend: Third, distinction. The difference in society values between Koreans and Japanese is incorporated. The Korean new generation considers the traditional moral principles as important social values. In contrast the Japanese new generation is greatly influenced by the open sexuality culture ; Fourth. public ownership and sharing of the culture. There is a gradual deviation from a dominating unidirectional influence from one country on another. Now the two countries are influencing each other ; Fifth, sense. There are numerous fashion items and generalized total coordination. New generations express more active and creative fashion sense in those fashion items.

Analysis of S-glutathionylated proteins during adipocyte differentiation using eosin-glutathione and glutaredoxin 1

  • Protein S-glutathionylation is a reversible post-translational modification on cysteine residues forming a mixed disulfide with glutathione. S-glutathionylation, not only protects proteins from oxidation but also regulates the functions of proteins involved in various cellular signaling pathways. In this study, we developed a method for the detection of S-glutathionylated proteins (ProSSG) using eosin-glutathione (E-GSH) and mouse glutaredoxin 1 (mGrx1). ProSSG was efficiently and specifically labeled with E-GSH to form ProSSG-E via thiol-disulfide exchange. ProSSG-E was readily luminescent allowing the detection of ProSSG with semi-quantitative determination. In addition, a deglutathionylation enzyme mGrx1 specifically released E-GSH from ProSSG-E, which increased fluorescence allowing a sensitive determination of ProSSG levels. Application of the method to the adipocyte differentiation of 3T3-L1 cells showed specific detection of ProSSG and its increase upon differentiation induction, which was consistent with the result obtained by conventional immunoblot analysis, but with greater specificity and sensitivity.
