This study was conducted to investigate quality characteristics of white pan bread with Cudrania tricuspidata leaf powder having functional components. Mixograph, pH, TPA, volume, specific volume, moisture, CrumbScan, color value, sensory evaluation and shelf-life test were performed on the white pan bread with Cudrania tricuspidata leaf powder. The obtained results are as follows. Mixogram analysis results showed that the dough with Cudrania tricuspidata leaf powder was suitable for baking quality. The more Cudrania tricuspidata leaf powder was added in the dough, the higher dough pH was and the lower pH values after fermentation and of the final productwere. TPA analysis results showed that higher amounts of Cudrania tricuspidata leaf powder resulted in increased hardness, chewiness, and gumminess of the white pan bread. Volume and specific volume of white pan bread were decreased with increased amounts of Cudrania tricuspidata leaf powder. CrumbScan images showed that fineness of grain, crust thickness, and volume of the bread were significantly different depending on the amount of Cudrania tricuspidata leaf powder. While color value analysis showed that increasing the amount of Cudrania tricuspidata leaf powder resulted in low L and a values. The results of the acceptance test showed that CLP3 had better scores in the appearance and texture characteristics than the control, while taste and overall acceptance were higher in the CLP2 compared with the control. According to the results of the shelf-life test, the white pan bread with 1~2% of Cudrania tricuspidata leaf powder had a significantly lower hardness when compared with the control. It was judged that a lower amount of Cudrania tricuspidata leaf powder could help in maintaining the softness of bread crumbs. Moisture content in the white pan bread decreased with the increased amounts of Cudrania tricuspidata leaf powder, but there were no significant differences among specimens.
The baking performance of Cudrania tricuspidata leaf powder as a value-added food ingredient was investigated in a model system of sponge cakes. Cudrania tricuspidata leaf powder was incorporated into cake batter at 5 levels (0, 5, 10, 15, and 20%, w/w) by adding equivalent amount based on total weight of wheat flour. The specific volume, height, moisture content, volume index, and symmetry index of sponge cakes decreased significantly with the increase in Cudrania tricuspidata leaf powder content (p < 0.05). Sponge cakes became darker and firmer with increase in Cudrania tricuspidata leaf powder content (p < 0.05), as indicated by decrease in the $L^*$ and increase in the firmness measured by the texture measuring instrument. Total polyphenol content also increased gradually as the Cudrania tricuspidata leaf powder content increased. Finally, the consumer acceptance test indicated that addition of Cudrania tricuspidata leaf powder up to 10% in the formulation of sponge cakes did not significantly influence the consumers' acceptability with respect to taste and flavor. In addition, the overall acceptability was not different from each other for control and 5% sample (p > 0.05).
In this study, we examined antioxidative effects and the anti-adipogenesis effect of different parts of Cudrania tricuspidata (C), and Morus alba (M). Total polyphenol contents were highest in M-root ($34.56{\pm}0.045mg\;GAE/g$), and there was no significant difference, between C-root and M-leaf. Total flavonoid contents of C-root were highest ($23.07{\pm}0.004mg\;QE/g$). To examine antioxidant activities of C and M extracts, DPPH and ABTS radical scavenging activity, and FRAP assay, was used. Results show that antioxidant activities of C and M extracts increased, in a dose-dependent manner. Adipocytes are generated by preadipocyte differentiation, during adipogenesis. Matured adipocytes accumulate in abnormal and cause obesity. We investigated effects of leaf and root extracts of C and M, on lipid accumulation, in 3T3-L1 adipocytes. Changes in cell morphology, and degrees of lipid accumulation in adipocytes, were evaluated by Oil Red O staining. Root extracts of C and M, reduced lipid content in a dose-dependent manner. Therefore, root extracts of C and M, may be good candidates for managing obesity.
Water extracts of root, leaf, branch and fruit of Cudrania tricuspidata were tested to use as natural antiwrinkle and whitening cosmetic ingredient. Root extract indicated 86.5 mg/g polyphenol concentration and 55.5 mg/g flavonoid concentration. From MTT test, root, branch and fruit extract did not show cell toxicity up to $1,000{\mu}g/mL$, however, leaf extract displayed significant cell toxicity in concentrations higher than $200{\mu}g/mL$. Cudrania tricuspidata root extracts displayed excellent 81% tyrosinase inhibition (whitening effect), 58% elastase inhibition (antiwrinkle effect) at $500{\mu}g/mL$ and showed minor UVB absorbance. When solution and lotion formulation were made including Cudrania tricuspidata root extract, they are stable at $25^{\circ}C$ for 21 days, but showed significant deviation in viscosity and particle size at $47^{\circ}C$. From the study, Cudrania tricuspidata root extract demonstrated good potential for natural whitening and antiwrinkle cosmetic agent.
This study was conducted to classify the variation in leaf and fruit morphology among natural grown 16 Cudrania tricuspidata stands. As results of the analysis in C. tricuspidata stands total 134 Species were found and as results of the analysis in leaf morphology, it was grouped as Group IV. By the analysis of variance in length, width, and weight of fruits were differed in 15 C. tricuspidata stands. The length, width, and weight of fruits were higher in Jinan and Sunchang, than Gochang, Gimje, and Goheung. By the cluster analysis of 3 fruit morphology were classified as 3 Groups. According to the results of this study C. tricuspidata leaves and fruit morphological characteristics for each group were compared by analysis there were differences between the morphological characteristics and the characteristics of the fruit did not match.

This study was conducted to suggest the Cudrania tricuspidata leaf and Achyranthes japonica Nakai Complex (CAC) possibility of use as a functional natural material for improving bone disease. Cudrania tricuspidata leaf and Achyranthes japonica Nakai were mixed in the same amount, extracted with hot water, and then powdered and used in the study. After, the cytotoxicity of CAC for osteoblasts (MG63 cell), osteoclasts (differentiated RAW264.7 cell), and macrophages (RAW264.7 cell) were evaluated by MTT assay, and ALP assay and TRAP assay were performed to confirm the differentiation capacity of osteoblasts and osteoclasts, respectively. In addition, the anti-inflammatory effect in macrophages was evaluated by ELISA, qRT-PCR, and western blot assay. CAC did not proliferated osteoblasts and osteoclasts, but increased ALP activity against osteoblasts differentiation and decreased TRAP activity against osteoclasts differentiation. CAC did not proliferated macrophages but decreased nitric oxide production. Also, decreased NOS2, IL1B, IL6, PTGS2, and TNFA gene expression, and JNK and p38 protein phosphorylation in a concentration-dependent manner, but ERK protein phosphorylation was not changed. As a result, CAC increased the differentiation and activation of osteoblasts, inhibited the differentiation and activation of osteoclasts, and regulated the expression of inflammatory cytokines in macrophages. Therefore, it is thought that CAC can be used as a functional natural material that prevents bone disease and has an anti-inflammatory effect.

Objectives : Cudrania tricuspidata (C. tricuspidata) is well-known traditional herbal remedy and its root, leaf and fruit were used for treatment of inflammation, tumor and painkilling. However, effect of C. tricuspidata fruit on tight junction is still unknown. The aim of this research was to determine the effect of C. tricuspidata fruit extract on human keratinocyte HaCaT cells. Methods : The antioxidant effects of water extract of C. tricuspidata (WECT) and ethanol extract of C. tricuspidata (EECT) were analyzed by using an ABTS assay. To confirm the cytotoxicity of WECT and EECT, MTS assay was performed. The mRNA expression levels of tight junction related genes were analyzed using quantitative RT-PCR analysis. Furthermore, dispase assay was used to investigate the alteration of cell-cell adhesion strength of EECT treated HaCaT cells. Results : WECT and EECT showed strong antioxidant activity. No obvious cytotoxicity was observed in both WECT and EECT until $2.0mg/m{\ell}$ concentration. The mRNA expression level of Claudin 6 were significantly increased by EECT treatment, whereas the WECT did not affect the expression of Claudin 6. Furthermore, EECT treatment enhances cell-cell adhesion strength. Conclusions : In this study, we investigated the physiological activities of the extracts of Cudrania tricuspidata fruit extracts on human keratinocytes by two different extraction methods. EECT might have an anti-aging activity on the skin by reducing oxidative stress. Moreover, it may be a useful ingredient in atopic dermatitis and skin-moisturizing, given its effects of altering Claudin 6 gene expression and enhancing cell-cell adhesion strength.

This study examined the immunomodulatory effects of milk fermented with based on the synbiotic interaction between Cudrania tricuspidata leaf extract (CT) and Lactobacillus gasseri 505. Raw 264.7 macrophages were treated with milk (Control), milk supplemented with 0.2% CT (CT-milk), or CT-milk fermented with L. gasseri 505 (FCT). FCT-treated cells showed higher NO production than the other groups. ELISA revealed that FCT produced the highest enhancement of the production of pro-inflammatory cytokines such as $TNF-{\alpha}$, $IFN-{\gamma}$, $IL-1{\beta}$, IL-6, IL-2, IL-12, and IL-10. Furthermore, FCT also increased the gene expression of pro-inflammatory mediators such as iNOS and COX-2 in a concentration-dependent manner. Collectively, these results indicate that the synbiotic interaction between CT and L. gasseri 505 might enhance their immunomodulatory effects of fermented milk.

This study investigates the effects of diets with mulberry leaf and Cudrania tricuspidata leaf supplements on blood glucose-related biomarkers in streptozotocin-induced diabetic rats. Male diabetic Sprague-Dawley rats were divided into groups and fed several diets. Group A was fed a diet with a 10% supplement of mulberry leaves. Group B was fed a diet with a 10% supplement of mulberry leaves and an additional 10% supplement of Cudrania tricuspidata leaves. Group C was fed a diet with a 10% supplement of mulberry leaves and an additional 30% supplement of Cudrania tricuspidata leaves. Group D was fed a diet with a 10% supplement of Cudrania tricuspidata leaves. Group E was the DM control. All diets were based on AIN-93G diet and they all lasted 6 weeks. The blood glucose levels of groups B and C stayed similar to initial levels (reference blood glucose) for the experimental period, which was significantly lower than levels in groups A, D, and E at the 6th week (p<0.05). And the area under the blood glucose curve (AUC) was also significantly lower in groups B and C than in groups D and E (p<0.05). The fasting blood glucose level after the experiment was significantly lower in groups B and C (p<0.05), and the C-peptide content of group C was as high as 50 pmol/L on average. The HbA1c content was also significantly lower in groups A and B than in group E (p<0.05). The serum TG, AST, and ALT levels were significantly decreased in groups A, B, and D than in group E (p<0.05). From the findings, it is shown that a diet of 1:1 mulberry and Cudrania tricuspidata leaf supplements could improve blood glucose-related biomarkers.
