Inhibitory Effects of the Roots of Cudrania tricuspidata Bureau on Osteoclast Differentiation (꾸지뽕나무 뿌리 추출물의 파골세포 분화 억제 효과)

  • Cudrania tricuspidata Bureau (Moraceae) is a traditional oriental medicine that has been widely used as anti-oxidant, anti-inflammatory and immunomodulatory in Korea. This study was performed that the 70% ethanol extract of the roots of C. tricuspidata (CTE) suppressed receptor activator of NF-${\kappa}B$ ligand (RANKL)-induced osteoclastogenesis, actin ring formation in RAW 264.7 cell lines. CTE significantly inhibited the JNK/mitogen-activated protein kinase (MAPK) signaling pathway without affecting ERK and p38 signaling in RANKL-stimulated RAW 264.7 cells. Also, CTE inhibited RANKL-induced expression of c-Fos, an upstream activator of NFATc1. Consequently, CTE suppresses osteoclast differentiation by inhibiting RANKL induced MAPK signaling pathways and disrupts the actin rings in mature osteoclasts. Thus, CTE can be used for the development of osteoporosis treatment drug with a natural material.

Analysis by GC of 1-Deoxynojirimycin in Leaves and Teas of Cudrania tricuspidata Bureau (꾸지뽕잎 및 꾸지뽕 잎차에 함유된 1-deoxynojirimycin의 GC 분석)

  • 1-Deoxynojirimycin(1-DNJ) was hardly detected by general UV detector. Therefore, in this study, we analyzed 1-DNJ, a effective functional material in which was contained the leaves of Cudrania tricuspidata and its tea by gas-chromatography. Synthesized a TMS derivative by attaching trimethylsilyl group to 1-DNJ, analyzed this by GC, and could detect a good peak. The leaves of Cudrania tricuspidata contains $1154.83{\pm}56.67$ ug/g.d.w of 1-DNJ and tea of Cudrania tricuspidata leaves does $8.01{\pm}0.61$ ug/g.d.w. This means the contents of 1-DNJ was nearly identical to each other. The larger size of the leaves had the more contents of 1-DNJ and the middle region of collection was the highest than any other collective regions of leaves.

Isolation and Quantitative Analysis of Cudratricusxanthone A and Cudraxanthone D from Roots of Cudrania tricuspidata Bureau (꾸지뽕나무 뿌리로부터 Cudratricusxanthone A와 Cudraxanthone D의 분리 및 함량 분석)

  • Cudrania tricuspidata Bureau (Moraceae) has been used for physical weakness, impotence and insomnia in traditional Korean medicine. In this study, cudratricusxanthone A and cudraxanthone D were isolated from roots of C. tricuspidata and quantification were achieved by using high performance liquid chromatography (HPLC) method with diode array detector. The isolated compound was identified by NMR analysis and HPLC method was validated by linearity, precision and specificity test. The results showed that calibration curves of two compounds indicated great linearity with a correlation coefficient ($R^2$) of 0.9976 and 0.9995. The limits of detection (LOD) for cudratricusxanthone A and cudraxanthone D were 1.15, $0.11{\mu}g/ml$. Intra-day precision of cudratricusxanthone A and cudraxanthone D were 0.32~1.99%, 0.09~3.32% and inter-day precision were 0.59~2.40%, 0.28~1.55% RSD (%) values, respectively. The specificity was confirmed by chromatograph and quantitative contents of cudratricusxanthone A and cudraxanthone D were $0.46{\pm}0.02%$ and $1.53{\pm}0.06%$. Therefore, this study can be used as a basic research data for the quantitative analysis of derived compounds from roots of C. tricuspidata.

Effects of Bedsoil and Growth Regulator on Cutting Propagation of Cudrania tricuspidata Bureau (상토 및 생장조절제 처리가 꾸지뽕나무의 삽목번식에 미치는 영향)

  • This Study was carried out to select bed soil and growth regulator which are effective for the macro propagation by cutting of Cudrania tricuspidata Bureau. The rooting of Cudraniae tricuspidata showed good result when the bed soil was coarse soil, rooted hardwood cutting 56.9%, and softwood cutting 62.3%. The rooting of Cudraniae tricuspidata showed good result when treated by growth regulator IBA 100 ppm, rooted hardwood cutting 57.9%, and softwood cutting 89.2%.

Antimicrobial Activity of Methanol Extracts from Cudrania tricuspidata Bureau according to the Parts Harvested and Time (수확부위 및 시기에 따른 꾸지뽕나무 메탄올 추출물의 항균성)

  • To evaluate the availability of Cudrania tricuspidata Bureau as a natural source of antimicrobials, the antimicrobial activity of methanol extracts of harvested parts was investigated using the paper disc diffusion method. The extracts from leaves and root bark had broad antimicrobial activity against various bacteria, including Bacillus subtilis, Bacillus cereus, Staphylococcus aureus, Streptococcus mutans, Listeria monocytogenes, Enterobacter aerogenes, Enterococcus faecalis, Salmonella choleraesuis subsp. choleraesuis, Vibrio vulnificus, and Pseudomonas aeruginosa and inhibited Bacillus cereus, Staphylococcus aureus, and Listeria monocytogenes, agents of food poisoning especially well. The extract from ripe fruit had a very high antimicrobial activity against Staphylococcus aureus with a 20.2 mm of clear zone at 50 mg/mL sample concentration. These results indicated that Cudrania tricuspidata could be used as new source for developing natural antimicrobial agents.

Molecular markers based on chloroplast and nuclear ribosomal DNA regions which distinguish Korean-specific ecotypes of the medicinal plant Cudrania tricuspidata Bureau

  • Cudrania tricuspidata Bureau is a widely-used, medicinal, perennial and woody plant. Obtaining information about the genetic diversity of plant populations is highly important with regard toconservation and germplasm utilization. Although C. tricuspidata is an important medicinal plant species registered in South Korea, no molecular markers are currently available to distinguish Korean-specific ecotypes from other ecotypes from different countries. In this study, we developed single nucleotide polymorphism (SNP) markers derived from the chloroplast and nuclear genomic sequences, which serve to to identify distinct Korean-specific ecotypes of C. tricuspidata via amplification refractory mutation system (ARMS)-PCR and high resolution melting (HRM) curve analyses. We performed molecular authentication of twelve C. tricuspidata ecotypes from different regions using DNA sequences in the maturaseK (MatK) chloroplast intergenic region and nuclear ribosomal DNA internal transcribed spacer (ITS) regions. The SNP markers developed in this study are useful for rapidly identifying specific C. tricuspidata ecotypes from different regions.

For conservation and germplasm utilization of the plant, it is imperative to obtaining information regarding the genetic diversity of the plant populations.   • Phytochemical investigation of the MeOH extract of the root barks of Cudrania tricuspidata Bureau (Moraceae), as guided by hepatoprotective activity in vitro, furnished four isoprenylated xanthones, cudratricusxanthone A (1), cudraxanthone L (2), cudratricusxanthone E (3), and macluraxanthone B (4). All of these compounds showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in human liver-derived Hep G2 cells. Compounds 1, 2, and 4 also exhibited the significant hepatoprotective effect on nitrofurantoin-induced cytotoxicity in human liver-derived Hep G2 cells.

Determination of Flavonoid by HPLC and Biological Activities from the Leaves of Cudrania tricuspidata Bureau (꾸지뽕나무 잎의 생리활성 및 HPLC에 의한 성분의 정량)

  • The methanol extract of the leaves of Cudrania tricuspidata and ethyl acetate fraction from the methanol extract showed inhibition for trypsin activity and the growth inhibition of Staphylococcus aureus. The content of kaempferol 7-O-$\beta$-D-glucopyranoside isolated from this plant was determined by HPLC and it was about 0.31% for the methanol extract.

  • Four flavonoids have been isolated from the leaves and stems of Cudrania tricuspidata Bureau (Moraceae) in Korea. They were kaempferol and kaempferol $7-O-{\beta}-D-glucopyranoside$ from the leaves, and kaempferide $7-O-{\beta}-D-glucopyranoside$ and naringenin $7-O-{\beta}-D-glucopyranoside$ from the stem respectively. The structures were established by spectroscopic and chemical methods.
