Biological Control of n Severe Viral Strain Using a Benign Viral Satellite RNA Associated with Cucumber mosaic virus

  • Two strains of Cucumber mosaic virus (CMV) isolated in Kuwait were confirmed their infectivity based on symptomatology and host range on different cultivars of tomato (Lycopersicon esculentum), tobacco(Nicotiana tabacum L.) and squash (Cucurbita pepo). The pattern of symptoms differed for the two CMV strains in tomato and tobacco, showing severe stunting and mosaic symptoms with one strain designated KU2, and almost symptomless with the other strain designated KU1. A satellite RNA 5 (sat-RNA) was found to be associated with the KU1 strain and was characterized as a benign viral satellite RNA. Using reverse transcription and polymerase chain reaction (RT-PCR) with sat-RNA specific primers, an amplified PCR product of about 160bp was determined and analyzed by gel electrophoresis. This naturally occurring benign viral satellite RNA was successfully used as a biological control agent to protect tomato plants against the severe KU2 strain. Tomato plants grown in plant-growth chambers, were preinoculated with KU1 containing the benign viral satellite and then challenge inoculated with the severe KU2 strain at different time intervals. All plants challenged three weeks after preinoculation showed nearly complete protection from subsequent infection by the severe strain. This biological control technology using plant viruses was found protective and could be successfully established sooner after the preinoculation.

Pathogenicity of Didymella bryoniae on the Seedlings of Cucurbits (오이류 유묘에 대한 덩굴마름병균의 병원성)

  • The objective of the study is to determine differences between cucurbits in the pathogenicity of Didymella bryoniae isolated from the naturally infected seeds of cucumber and pumpkin. Primary seedling infection of cucumber(Cucumis sativus L.), oriental melon(Cucumis melo var. makuwa Makino), pumpkin(Cucurbita pepo L.) and watermelon (Citrullus vulgaris Shrad.) occurred on the radicle, hypocotyl and cotyledons and symptoms on each crop were very similar. Infection of the radicle generally caused pre-emergence rot, while infection on the hypocotyl and cotyledons provided further inoculum for infection of the first true leaves and the stem. In cross inoculation tests, all isolates of D. bryoniae could infect cucumber, oriental melon, pumpkin and watermelon at different growth stages and there were not much differences in pathogenicity or susceptibility between isolates of the pathogen and crops tested. The susceptibility of cucumber and pumpkin was markedly influenced by prevailing humid conditions.

Establishment of a Simple and Rapid Gene Delivery System for Cucurbits by Using Engineered Zucchini Yellow Mosaic Virus

  • The infectious full-length cDNA clone of zucchini yellow mosaic virus (ZYMV) isolate PA (pZYMV-PA), which was isolated from pumpkin, was constructed by utilizing viral transcription and processing signals to produce infectious in vivo transcripts. Simple rub-inoculation of plasmid DNAs of pZYMV-PA was successful to cause infection of zucchini plants (Cucurbita pepo L.). We further engineered this infectious cDNA clone of ZYMV as a viral vector for systemic expression of heterologous proteins in cucurbits. We successfully expressed two reporter genes including gfp and bar in zucchini plants by simple rub-inoculation of plasmid DNAs of the ZYMV-based expression constructs. Our method of the ZYMV-based viral vector in association with the simple rub-inoculation provides an easy and rapid approach for introduction and evaluation of heterologous genes in cucurbits.

  • 308 isolates of Ampelomyces quisqualis were isolated from powdery mildew fungi of 73 plant species in Korea from 1994 to 2004. Among them, the new mycohosts and new plant hosts of A. quisqualis were found in 13 species of powdery mildew fungi in 38 species of plant. The new hosts of A. quisqualis were Erysiphe heraclei on Heracleum moellenderfii; E. hommae on Elsholtzia splendins; E. glycines on Glycine max; E. lespedezae on Lespedeza biclor; E. pileae on Pilea mongolica; E. pisi on Phaseolus radiatus; E. polygoni on Rumex aquatica and Rumex crispus; Golovinomyce artemisiae on Artemisia princeps var. orientalis; G. cichoracearum on Rudbeckia laciniata var. hortensis; G. rubiae on Rubia akane; Oidium sp. on Valeriana fauriei, Lactuca indica var. laciniata, Carpesium triste var. manshuricum, Aster ageratoides var. turczaninow, Lufa cylindrica, Ixeris demtats Nakai, Phlox paniculata, Bidens bipinnata and Pathenocissus tricuspidata; Microsphaera alphitoides on Quercus aliema, M. pseudolonicerae on Cocculus trilobos; Podosphaera sp. on Ligustrum obtusifolium; Sphaerotheca aphanisi on Fragaria ananassa; S. balsaminae on Impatiens textori; S. fusca on Cucurbita pepo, Cucurbita maxima, Ligularia fischeri, Solanum melongena, Lagenaria leucantha, Cucumis melo var. makuwa, Acalypha australis, Cosmos bipinnatus and Aster scaber; Uncinuliella simulans on Rosa muliflora and Uncinula australiana on Lagerstroemia indica.

Characterization of the Lsi1 Homologs in Cucurbita moschata and C. ficifolia for Breeding of Stock Cultivars Used for Bloomless Cucumber Production

  • Bloomless cucumber fruits are commercially produced by grafting onto the pumpkin stocks (Cucurbita moschata) to restricted silicon ($SiO_2$) absorption. Inhibition of silicon absorption in bloomless stocks is conferred by a mutant allele of the CmLsi1 homologous to Lsi1 in rice. In this study, we characterized the Lsi1 homologs in pumpkin (C. moschata) and its cold-tolerant wild relative C. ficifolia ('Heukjong') in order to develop a DNA marker for selecting a bloomless trait and to establish the molecular basis for breeding bloomless stock cultivars of C. ficifolia. A Cleaved amplified polymorphic sequence (CAPS) marker (CM1-CAPS) was designed based on a non-sysnonymous single nucleotide polymorphism (SNP, C>T) of the CmLsi1 mutant-type allele, and its applicability for Marker-assisted selection (MAS) was confirmed by evaluating three bloom and five bloomless pumpkin stock cultivars. Quantitative RT-PCR of the CmLsi1 for these stock cultivers implied that expression level of the CmLsi1 gene does not appear to be associated with the bloom/bloomless trait and may differ depending on plant species and tissues. A full length cDNA of the Lsi1 homolog [named CfLsi1($B^+$)] of 'Heukjong' (C. ficifolia), was cloned and sequence comparison between CmLsi1($B^+$) and CfLsi1($B^+$) revealed that there exists total 24 SNPs, of which three were non-synonymous. Phylogenetic analysis of CfLsi1($B^+$) and Lsi1 homologs further revealed that CfLsi1($B^+$) is closesly related to Nodulin 26-like intrinsic proteins (NIPs) and most similar to CpNIP1 of C. pepo than C. moschata.

Inflammation inhibitory effect of water extract from pumpkin's tendril (호박 덩굴손 물 추출물의 염증제어 활성)

  • Pumpkin has long been used as traditional health materials in oriental medicine, pharmacy, medicine, and food industries in many countries. In this study, the water extract and two active components from tendril of young C. moschata Duch. were investigated on inflammation inhibitory activity. The water extract of young C. moschata Duch. showed high cell viability over 95% and it decreased the production of interlukin 6 (IL-6) and tumor necrosis factor (TNF-${\alpha}$) in the capacity of mouse bone marrow-derived macrophages (BMDM) upon lipopolysaccharide (LPS) stimulation. Also, isolated fraction (B4) suppressed the secretion of IL-6 and TNF-${\alpha}$. Among the domestically cultivated pumpkins, B4 fraction contained in the tendril part of them and it comprised of the order of tendril from Cucurbita pepo var. cylindrica, old of C. moschata Duch., and young of C. moschata Duch. These results suggest that water extracts of C. moschata Duch. and purified active compound, rutin, show anti-inflammation activity by suppression of the secretion of IL-6 and TNF-${\alpha}$. It can be applicable as pharmaceutical materials.

Historical Classification and Evaluation Character of Pumpkin and Squash Genetic Resources (Cucurbita spp.) Reserved in National Agrobiodiversity Center (농업유전자원센터 호박속 자원의 구분 및 특성)

  • 호박(Cucurbita spp.)은 박과 호박속 식물로 원산지는 열대 남아메리카로 추정되며 1년생 혹은 다년생 초본으로서 우리나라를 포함한 전 세계에서 널리 분포하고 있다. 호박은 비타민A와 비타민C가 풍부하고 이뇨작용 효과가 탁월해 피부를 좋게 하고 부기를 빼주는 것으로 보고되고 있다. 본 연구에서는 농업유전자원센터에서 보유한 호박속 자원의 내력과 형태적인 특성을 그룹화하여 자원 활용을 위한 기초자료를 제공하고자 하였다. 농업유전자원센터에서 보유한 호박속 등록자원은 총 5종(C.pepo, C. moschata, C.maxima, C.argyrosperma, C .ficifolia) 1,640자원으로 주요 원산지는 한국 589자원(35.9%), 미국 122자원(7.4%), 러시아 79자원(4.8%), 멕시코 49자원(4.2%), 터키 56자원(3.4%)이었으며 국내 원산의 경우 강원도에서 수집된 자원이 66자원으로 가장 많았다. 전체 1,640 자원 중 재래종은 733자원(44.7%)을 차지하였으며 다음으로는 육성품종이 133자원(8%)으로 많았다. 생장형이 조사 된 301자원 대부분이 덩굴성이며 준왜성 16자원, 왜성 14자원이 있었다. 호박은 과실 모양과 색, 잎 모양, 줄기 모양 등 생육 특성이 매우 다양하게 조사 되었으며 과경 등 몇 가지 특징은 완벽하지 않지만 종(species)을 구분하는 간이적인 지표로 사용되는 것으로 조사되었다. 흰가루병 저항성 자원은 조사 된 1,170 자원 중 우리나라 재래종인 참호박(IT104575)을 포함한 7자원이 조사되었다. 293개 자원의 과실당도는 2.2~15.4brix까지 다양했으며 당도 12brix 이상 자원은 IT200650을 포함하여 6자원이 조사되었다. 본 연구에서 조사된 호박속 자원의 내력 및 생육 특성은 육종소재의 기초 정보로서 이용되고 있으며 자원 내력정보의 보완, 형질과 기능성 물질 분석 등 추가적인 연구를 통해 연구소재의 기초 정보로서 활용의 가치가 더 커질 것으로 보인다.

Nucleotide Sequence of Coat Protein Gene of Kyuri Green Mottle Mosaic Virus Isolated from Zucchini

  • The coat protein (CP) gene of kyuri green mottle mosaic virus zucchini strain (KGMMV-Z) isolated from zucchini (Cucurbita pepo) in Chonfu, Korea in 1999 was sequenced by the reverse transcription and polymerase chain reaction with degenerate and generate primers originated from tobamoviruses. The degenerate primers were very effective in amplification of KGMMV-Z CP region. The KGMMV-Z CP gene consisted of 486 nucleotides and had the same nucleotide length compared with those of cucurbit-infecting tobamoviruses. KGMMV-Z CP gene shared 43.8, 44.2, and 44.4% nucleotide sequence similarity with the CP gene of cucumber green mottle mosaic virus watermelon strain (CGMMZ-W), CGMMV-KW1, and CGMMV-SH, respectively, whereas three CGMMV strains among themselves showed 98.6-99.6% nucleotide similarity. The deduced amino acids of KGMMV-Z CP gene were 161 amino acid residues with the molecular weight of 17,181 daltons. The first 24 codons of KGMMV-Z CP gene corresponded to the sequences of the N-terminal amino acid of the viral capsid protein. The amino acid sequences of KGMMV-Z CP had 45.3% similarity compared with those of three CGMMV strains. However, the amino acid sequences of CGMMV strains were identical. These results showed that two cucurbit-infecting tobamovirus members, KGMMV-Z and CGMMV were genetically distantly related.

First Report of Cucumber mosaic virus Isolated from Wild Vigna angularis var. nipponensis in Korea

  • A viral disease causing severe mosaic, necrotic, and yellow symptoms on Vigna angularis var. nipponensis was prevalent around Suwon area in Korea. The causal virus was characterized as Cucumber mosaic virus (CMV) on the basis of biological and nucleotide sequence properties of RNAs 1, 2 and 3 and named as CMV-wVa. CMV-wVa isolate caused mosaic symptoms on indicator plants, Nicotiana tabacum cv. Xanthi-nc, Petunia hybrida, and Cucumis sativus. Strikingly, CMV-wVa induced severe mosaic and malformation on Cucurbita pepo, and Solanum lycopersicum. Moreover, it caused necrotic or mosaic symptoms on V. angularis and V. radiate of Fabaceae. Symptoms of necrotic local or pin point were observed on inoculated leaves of V. unguiculata, Vicia fava, Pisum sativum and Phaseolus vulgaris. However, CMV-wVa isolate failed to infect in Glycine max cvs. 'Sorok', 'Sodam' and 'Somyeong'. To assess genetic variation between CMV-wVa and the other known CMV isolates, phylogenetic analysis using 16 complete nucleotide sequences of CMV RNA1, RNA2, and RNA3 including CMV-wVa was performed. CMV-wVa was more closely related to CMV isolates belonging to CMV subgroup I showing about 85.1-100% nucleotide sequences identity to those of subgroup I isolates. This is the first report of CMV as the causal virus infecting wild Vigna angularis var. nipponensis in Korea.

Occurrence of Papaya ringspot virus Infecting Cucurbit Crops in Korea (박과작물에 발생하는 파파야원형반점바이러스의 발생 보고)

  • A flexuous rod-shaped virus was isolated from Cucurbita pepo leaves showing as green mosaic and puckering symptoms at Anseong, Korea. Based on the biological analysis, electron microscopy, and reverse transcription-polymerase chain reaction (RT-PCR), the virus isolate was identified as Papaya ringspot virus type watermelon (PRSV-W). From biological analysis, the host range of PRSV-W was limited to the families Cucurbitaceae and Chenopodiaceae. Most susceptible cucurbit species, such as Cucumis lanatus, Cucumis sativus, Cucurbita pepo, and Citrullus lanatus, showed symptoms of green mosaic, malformation, puckering, and narrow laminae by infection with PRSV-W. The local lesion were showed on the inoculated leaves of both Chenopodium amaranticolor and C. quinoa. Field survey of PRSV, Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV), three major viruses infecting cucurbit, was done during 2001 to 2003 on 173 commercial cucurbit cultivating fields distributed over the three regions of Gyeonggi, Gyeongbuk and Jeonnam Provinces where cucurbits are grown in different environmental conditions and cropping patterns. Typical viral symptoms were observed from 107 cultivating fields, and all three kinds of potyviruses were detected from 206 samples out of the 235 samples using RT-PCR. Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV) are the most widely distributed viruses in outdoor and retarding-culture fields, at an infection rating of 48 and 33 percents, respectively. PRSV was detected from 12 percent of 235 samples. The nucleotide and amino acid sequences of coat proteins (CP) of eight PRSV isolates, collected from several areas including Anseong, were determined and sequenced heterogeneity among the isolates was performed. The CP gene of PRSV showed 88.6~97.3 percent homology in nucleotide sequences and 95.1~99.3 percent homology in amino acid sequences with other PRSV isolates worldwide. The phylogenetic analysis indicated that the Korean PRSV isolates belong to the southern-east Asian cluster.
