  • In March 2010, grafted cucumber cultivated in the greenhouse showed a severe rot on crown resulting yellowing and wilting of the leaves. The symptoms of naturally infected plants showed dark brown, watersoaked lesions at the base of the stem. The fungus produced mass of white mycelium and yellow to orange spores in necrotic lesions on dead and dying plants. Fungus was isolated from rotted tissues of the crown and root. On the basis of morphological characteristics, ITS sequence and pathogenicity tests, the isolate was identified as Fusarium solani f. sp. cucurbitae. This is the first report of the crown and foot rot of grafted cucumber caused by F. solani f. sp. cucurbitae in Korea.

  • Colonization of Pseudomonas chlororaphis O6, a nonpathogenic rhizobacterium, on the roots induced systemic resistance in cucumber plants against tai-get leaf spot, a foliar disease caused by Corynespora cassiicola. A cDNA library was constructed using mRNA extracted from the cucumber leaves 12 h after inoculation with C. cassiicola, which roots had been previously treated with O6. To identify the genes involved in the O6-mediated induced systemic resistance (ISR), we employed a subtractive hybridization method using mRNAs extracted from C cassiicola-inoculated cucumber leaves with and without previous O6 treatment on the plant roots. Differential screening of the cDNA library led to the isolation of 5 distinct genesencoding a GTP-binding protein, a putative senescence-associated protein, a galactinol synthase, a hypersensitive-induced reaction protein, and a putative aquaporin. Expressions of these genes are not induced by O6 colonization alone. Before challenge inoculation, no increase in the gene transcriptions could be detected in previously O6-treated and untreated plants but, upon subsequent inoculation with the pathogenic fungus, transcription levels in O6-treated plants rose significantly faster and stronger than in untreated plants. Therefore, the O6-mediated ISR may be associated with an enhanced capacity for the rapid and effective activation of cellular defense responses which becomes apparent only after challenge inoculation on the distal, untreated plant parts, as suggested by Conrath et al. (2002). • This study was conducted to investigate the effect of U. atrum treatment on control of Botrytis leaf blight of lily and Botrytis gray mold of cucumber, and to evaluate the U. atrum as the biological control agent of Botrytis diseases. The antagonistic isolates CNU 9037 and CNU 9054 isolated from tomato leaves were identified as Ulocladium atrum Preuss based on morphological characteristics. This is the first record of U. atrum in Korea. In bioassays on dead leaves of tomato and cucumber, treatment of U. atrum colonized the dead leaves and suppressed sporulation of Botrytis as compared with the untreated control. The suppression of spoulation of Botrytis on dead leaf segments by U. atrum was higher when U. atrum was treated before Botrytis was treated. The effect of treatments with conidial suspension of U. atrum on leaf blight of lily and gray mold of cucumber caused by Botrytis elliptica and B. cinerea, respectively, was investigated under greenhouse conditions. Spraying U. atrum ($1{\times}10^6$ conidia per ml) at intervals of 1 week for three times resulted in a significant reduction of natural infections of lily leaves caused by B. elliptica. Protective value of U. atrum treatment was higher than that of the fungicide (procymidone) treatment. Spraying U. atrum also resulted in a significant reduction of cucumber gray mold caused by B. cinerea. Our results show that U. atrum has a potential for biological control against diseases caused by Botrytis spp, in lily and cucumber.

  • Root-knot nematodes represent a significant problem in cucumber, causing reduction in yield and quality. To develop screening methods for resistance of cucumber to root-knot nematode Meloidogyne incognita, development of root-knot nematode of four cucumber cultivars ('Dragonsamchuk', 'Asiastrike', 'Nebakja' and 'Hanelbakdadaki') according to several conditions such as inoculum concentration, plant growth stage and transplanting period was investigated by the number of galls and egg masses produced in each seedling 45 days after inoculation. There was no difference in galls and egg masses according to the tested condition except for inoculum concentration. Reproduction of the nematode on all the tested cultivars according to inoculum concentration increased in a dose-dependent manner. On the basis of the result, the optimum conditions for root-knot development on the cultivars is to transplant period of 1 week, inoculum concentration of 5,000 eggs/plant and plant growth stage of 3-week-old in a greenhouse ($25{\pm}5^{\circ}C$). In addition, under optimum conditions, resistance of 45 commercial cucumber cultivars was evaluated. One rootstock cultivar, Union was moderately resistant to the root-knot nematode. However, no significant difference was in the resistance of the others cultivar. According to the result, we suggest an efficient screening method for new resistant cucumber to the root-knot nematode, M. incognita.

  • Damping-off, caused by Rhizoctonia solani and Pythium ultimum, is a very important plant disease and affects seeds and seedlings of many plant species worldwide. To investigate control efficacy of Serenade formulation (1.34%, SC) against Rhizoctonia and Pythium damping-off diseases, seeds of chili pepper and cucumber were sown in soils inoculated with R. solani and P. ultimum and Serenade formulation was applied by soil-drenching with 9-, 19-, and 39-fold dilutions. Control values of Serenade formulation on Rhizoctonia damping-off of chili pepper was 58% and 29% for 9- and 19-fold dilutions, respectively. In the case of cucumber Rhizoctonia damping-off, Serenade treatments showed similar control efficacy with damping-off of chili pepper. On the other hand, control efficacy of Serenade formulation on Pythium damping-off of cucumber was less than control effects on Rhizoctonia damping-off. Only Pythium damping-off of chili pepper treated with 9-fold dilution Serenade was statistically different with untreated control. This result suggest that Serenade formulation could be effectively used for controlling Rhizoctonia and Pythium damping-off diseases.

  • A dctA gene encoding a protein with identity to a C4-dicarboxylate/H+ was cloned from a beneficial biocontrol bacterium, P. chororaphis O6. Expression of the dctA was induced in minimal medium by several organic acids and was repressed by glucose. Highest expression was observed in early-log cells grown on fumarate and succinate with decline as cells approached late-log phase. The dctA transcript accumulated weakly when cells were grown on malate but strong expression was observed with benzoate. Expression of the dctA transcript was repressed in early-log cells upon addition of glucose to fumarate, but was detected as the cell culture aged. A dctA-deficient mutant of O6, constructed by marker exchange mutagenesis, did not grow on minimal medium containing succinate, benzoate, or fumarate, and growth on malate was delayed. The dctA mutant and wild type grew equally on glucose. The dctA mutant on cucumber roots in sterilized potting soil was colonized at levels comparable to those of the wild type, but induction level of disease resistance by the mutant against target leaf spot disease was decreased. These results may indicate that the dctA is essential for utilization of certain organic acids and its expression is controlled by the availability of sugars. In addition, the dctA is not essenitial for cucumber root colonization, but important for induction of disease resistance.

  • Biological control activity of Acremonium strictum BCP, a mycoparasite on Botrytis cinerea, was examined against six plant diseases such as rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew in growth chambers. The spore suspension of strain BCP showed strong control activities against five plant diseases except against wheat leaf rust. On the other hand, the culture filtrate of A. strictum BCP was effective in controlling only cucumber gray mold and barley powdery mildew. Further in vivo biocontrol activities of A. strictum BCP against tomato gray mold were investigated under greenhouse conditions. Control efficacy of the fungus on tomato gray mold increased in a concentration-dependent manner. Treatment of more than $1{\times}10^6$ spores/ml significantly controlled the disease both in tomato seedlings and in adult plants. The high disease control activity was obtained from protective application of the strain BCP, whereas the curative application did not control the disease. Foliar infections of B. cinerea were controlled with $1{\times}10^8$ spores/ml of A. strictum BCP applied up to 7 days before inoculation. In a commercial greenhouse, application of A. strictum BCP exhibited the similar control efficacy with fungicide procymidone (recommended rate, $500{\mu}g/ml$) against strawberry gray mold. These results indicate that A. strictum BCP could be developed as a biofungicide for Botrytis diseases under greenhouse conditions.

  • In this study, antifungal activity of essential oils of Cymbopogon citratus and Ocimum basilicum and two fungicides Mancozeb and Metalaxyl-Mancozeb in six different concentrations were investigated for controlling three species of Phytophthora, including P. capsici, P. drechsleri and P. melonis on pepper, cucumber and melon under in vitro and greenhouse conditions, respectively. Under the in vitro condition, the median effective concen- tration ($EC_{50}$) values (ppm) of plant essential oils and fungicides were measured. In greenhouse, soil infested with Phytophthora species was treated by adding 50 ml of essential oils and fungicides (100 ppm). Disease severity was determined after 28 days. Among two tested plant essential oils, C. citratus had the lowest $EC_{50}$ values for inhibition of the mycelial growth of P. capsici (31.473), P. melonis (33.097) and P. drechsleri (69.112), respectively. The mean $EC_{50}$ values for Metalaxyl-Mancozeb on these pathogens were 20.87, 20.06 and 17.70, respectively. Chemical analysis of plant essential oils by GC-MS showed that, among 42 compounds identified from C. citratus, two compounds ${\beta}$-geranial (${\alpha}$-citral) (39.16%) and z-citral (30.95%) were the most abundant. Under the greenhouse condition, Metalaxyl-Mancozeb caused the greatest reduction in disease severity, 84.2%, 86.8% and 92.1% on melon, cucumber, and pepper, respectively. The C. citratus essential oil reduced disease severity from 47.4% to 60.5% compared to the untreated control ($p{\leq}0.05$). Essential oils of O. basilicum had the lowest effects on the pathogens under in vitro and greenhouse conditions. These results show that essential oils may contribute to the development of new antifungal agents to protect the crops from Phytophthora diseases.
