Forward-Mode $Na^+-Ca^{2+}$ Exchange during Depolarization in the Rat Ventricular Myocytes with High EGTA

  • During depolarization, extrusion of $Ca^{2+}$ from sarcoplasmic reticulum through forward-mode $Na^+-Ca^{2+}$ exchange was studied in the rat ventricular myocytes patch-clamped in whole-cell configuration. In order to confine the $Ca^{2+}$ responses in a micro-domain by limiting the $Ca^{2+}$ diffusion time, rat ventricular myocytes were dialyzed with high (14 mM) EGTA. $K^+$ current was suppressed by substituting KCl with 105 mM CsCl and 20 mM TEA in the pipette filling solution and by omitting KCl in the external Tyrode solution. $Cl^-$ current was suppressed by adding 0.1 mM DIDS in the external Tyrode solution. During stimulation roughly mimicking action potential, the initial outward current was converted into inward current, $47{\pm}1%$ of which was suppressed by 0.1 mM $CdCl_2.$ 10 mM caffeine increased the remaining inward current after $CdCl_2$ in a cAMP-dependent manner. This caffeine-induced inward current was blocked by $1\;{\mu}M$ ryanodine, $10\;{\mu}M$ thapsigargin, 5 mM $NiCl_2,$ or by $Na^+\;and\;Ca^{2+}$ omission, but not by $0.1\;{\mu}M$ isoproterenol. The $I{\sim}V$ relationship of the caffeine-induced current elicited inward current from -45 mV to +3 mV with the peak at -25 mV. Taken together, it is concluded that, during activation of the rat ventricular myocyte, forward-mode $Na^+-Ca^{2+}$ exchange extrudes a fraction of $Ca^{2+}$ released from sarcoplasmic reticulum mainly by voltage-sensitive release mechanism in a micro-domain in the t-tubule, which is functionally separable from global $Ca^{2+}{_i}$ by EGTA.

Response Characteristics of Forced Vibration Model with Sinusoidal Exciting Force (정현파로 가진한 강제진동 해석과 응답특성)

  • The characteristics of forced vibration with excited sinusoidal force was introduced. Also, numerical analyses and FRF in frequency domain were performed in detail. In this regard, the responses of displacement, velocity and acceleration were investigated in a forced vibration model. The FRF characteristics in real and imaginary part around natural frequency are also discussed. This response approach of forced vibration in time domain is used for the identification and monitoring of sinusoidal forced vibration. For acquiring a displacement, velocity and acceleration, a numerical technique of Runge-Kutta-Gill method was performed. For the FRF(frequency response function), These responses are used. Also, the FRF can represent the intrinsic characteristics of the forced vibration. These performed results and analysis are successful in each damped condition for the forced vibration model. After numerical analysis of the different mass, damping and stiffness, the forced vibration response characteristics with sinusoidal force was discriminated considering its amplitude and frequency simultaneously.

  • The purpose of this study is to investigate the relationship between facial image efficacy and job satisfaction in a sample of 167 workers in SMEs in Seoul and Gyeonggi province. and to examine the effect of SME workers' face image efficacy on job satisfaction. The results of the study are as follows. First, there was a statistically significant correlation between management ability and job satisfaction, which is a sub - area of facial image efficacy. This means that the higher the management ability, the higher the degree of job satisfaction. There was a statistically significant correlation between perception attitude and job satisfaction, which is a sub - area of face image efficacy. Second, management ability, which is a sub - area of face image efficacy of SME workers, has a statistically significant effect on job satisfaction. The sub - domain of face image efficacy has no effect on job satisfaction, Expression confidence, which is a sub-domain of image efficacy, has a statistically significant effect on job satisfaction.

Crystal Structure of DsbA from Corynebacterium diphtheriae and Its Functional Implications for CueP in Gram-Positive Bacteria

  • In Gram-negative bacteria in the periplasmic space, the dimeric thioredoxin-fold protein DsbC isomerizes and reduces incorrect disulfide bonds of unfolded proteins, while the monomeric thioredoxin-fold protein DsbA introduces disulfide bonds in folding proteins. In the Gram-negative bacteria Salmonella enterica serovar Typhimurium, the reduced form of CueP scavenges the production of hydroxyl radicals in the copper-mediated Fenton reaction, and DsbC is responsible for keeping CueP in the reduced, active form. Some DsbA proteins fulfill the functions of DsbCs, which are not present in Gram-positive bacteria. In this study, we identified a DsbA homologous protein (CdDsbA) in the Corynebacterium diphtheriae genome and determined its crystal structure in the reduced condition at $1.5{\AA}$ resolution. CdDsbA consists of a monomeric thioredoxin-like fold with an inserted helical domain and unique N-terminal extended region. We confirmed that CdDsbA has disulfide bond somerase/reductase activity, and we present evidence that the N-terminal extended region is not required for this activity and folding of the core DsbA-like domain. Furthermore, we found that CdDsbA could reduce CueP from C. diphtheriae.

Computation of 2-D mixed-mode stress intensity factors by Petrov-Galerkin natural element method

  • The mixed-mode stress intensity factors of 2-D angled cracks are evaluated by Petrov-Galerkin natural element (PG-NE) method in which Voronoi polygon-based Laplace interpolation functions and CS-FE basis functions are used for the trial and test functions respectively. The interaction integral is implemented in a frame of PG-NE method in which the weighting function defined over a crack-tip integral domain is interpolated by Laplace interpolation functions. Two Cartesian coordinate systems are employed and the displacement, strains and stresses which are solved in the grid-oriented coordinate system are transformed to the other coordinate system aligned to the angled crack. The present method is validated through the numerical experiments with the angled edge and center cracks, and the numerical accuracy is examined with respect to the grid density, crack length and angle. Also, the stress intensity factors obtained by the present method are compared with other numerical methods and the exact solution. It is observed from the numerical results that the present method successfully and accurately evaluates the mixed-mode stress intensity factors of 2-D angled cracks for various crack lengths and crack angles.

Photonic sensors for micro-damage detection: A proof of concept using numerical simulation

  • Damage detection has been proven to be a challenging task in structural health monitoring (SHM) due to the fact that damage cannot be measured. The difficulty associated with damage detection is related to electing a feature that is sensitive to damage occurrence and evolution. This difficulty increases as the damage size decreases limiting the ability to detect damage occurrence at the micron and submicron length scale. Damage detection at this length scale is of interest for sensitive structures such as aircrafts and nuclear facilities. In this paper a new photonic sensor based on photonic crystal (PhC) technology that can be synthesized at the nanoscale is introduced. PhCs are synthetic materials that are capable of controlling light propagation by creating a photonic bandgap where light is forbidden to propagate. The interesting feature of PhC is that its photonic signature is strongly tied to its microstructure periodicity. This study demonstrates that when a PhC sensor adhered to polymer substrate experiences micron or submicron damage, it will experience changes in its microstructural periodicity thereby creating a photonic signature that can be related to damage severity. This concept is validated here using a three-dimensional integrated numerical simulation.

  • 컴퓨터를 활용한 지식과 정보의 고도 처리를 위해 온톨로지를 구축하고 활용하려는 요구가 강해지고 있다. 이런 요구에 부흥하여 온톨로지의 구축 방법에 대한 많은 연구가 이루어지고 있다 그러나 현재까지의 온톨로지 구축 이론과 온톨로지 개발 도구가 실제 구현 목적과 사례에 따라 제한적인 부분에서 개발되어 사용되고 있다. 그러므로 구축하고자 하는 분야의 온톨로지에 맞는 적절한 구축 프로세스와 도구가 필요하다. 분야 온톨로지(domain ontology)를 구축함에 있어 특정 분야의 전문가 또는 비전문가가 언어자원으로부터 지식을 개념화(conceptualization), 형식화(formalization)하여 온톨로지를 구축할 수 있도록 분야 관련 언어자원에 근거하여 온톨로지를 구축할 수 있는 온톨로지 구축 프로세스(OntoProcessl))를 제안한다. 그리고, 다수의 온톨로지 구축자가 동시에 같은 분야의 온톨로지를 구축할 경우, 개념화 과정에서 서로 다른 구축자가 동일한 개념을 중복 정의하거나, 형식화 과정에서 형식언어 이해 부족으로 인한 구축 능률 저하 문제가 발생할 수 있다. 이를 위해 메타 온톨로지(meta ontology)를 이용하여 다중 온톨로지를 구축할 때 발생하는 문제를 해결하는 다중 온톨로지 구축 프로세스를 제안한다. 현재 이 프로세스에 근거하여 온톨로지 구축 시스템(OntoCS2))을 개발하였고 국가 IT 온톨로지 인프라 기술 개발 프로젝트에서 IT분야 온톨로지의 개발에 활용되고 있다.

A study on the possibility of using dual task performance as a screening test for driving ability of elderly drivers (노인운전자 운전능력 선별검사로서 이중과제수행의 활용 가능성 연구)

  • This study was conducted to identify the relationship between the dual task performance and driving behavior of the elderly driver in order to identify whether the dual task can be utilized as a test to screen the driving ability of the elderly driver. We surveyed general information and driving-related information for 32 normal elderly drivers, and evaluated Y-DuCog(Yonsei-Dual task Cognitive screening) and the K-DBQ (Korean-Driving Behavior Questionnaire). As a result of the study, the performance of the dual task using the pegboard task and the animal name speaking showed a significant correlation with the score of the violation domain of K-DBQ. With this study, it was possible to confirm the possibility as a test for discrimination of driving ability.

  • The response of an anti-Compton phoswich detector to gamma rays was investigated using Monte-Carlo method, and the pulses from different crystal cases, including gamma deposition only in the LaBr3(Ce) or CsI(Tl) crystal and coincidence in both crystals, were analyzed. A novel pulse discrimination method for gamma deposition events based on wavelet transform analysis, called SSD (Scale Shape Discrimination), was developed in this study. Compared to the traditional PSD (Pulse Shape Discrimination) method, SSD has the advantage of transforming one-dimensional pulses in the time-domain into two-dimensional time-frequency spectra, providing the more useful features for pulse discrimination. The performances of the Compton suppression and Full-energy peak loss using PSD and SSD methods was studied. The results show that the Compton suppression factor IPSD = 5.12 and ISSD = 5.32, and FEP loss factor PLPSD = 0.0554 and PLSSD = 0.0587. Meanwhile, the influences of the cutoff values for pulse discrimination on the results of I and PL with different method were analyzed.

  • In this research, there is a purpose to develop an informatics education curriculum of the general level that the preliminary teacher at elementary school prepares for information education. First of all, in order to achieve the purpose, we compare and analyze the standard CS 2013 of the educational curriculum of elementary school teacher training university, University-Level Program cooperation deepening course, higher education course, and prepared content and level. Secondly, subjects were structured in consideration of standard J07-GE analysis of higher education curriculum and cooperation with secondary education course. Third, subject names were determined by examination by experts, taking into consideration the scope of subjects, content system composition, etc. Computer Science II, Computer Science II, Data Management in the domain to understand the basic principles of Computer Science I, Computing System, in order to approach expert opinions, analysis results, problems arising in information society from the viewpoint of computer science We proposed data management and analysis to grasp the patterns and relationships involved. In this research, not only improving ability to solve problems based on the basic capacity strengthening of teachers but also presenting subjects of general level, considering continuity star of high school information subjects it makes sense.
