옻나무 추출액의 급여가 육계의 생산성, 맹장내 균총 및 유해가스 발생량에 미치는 영향 (Effects of Feeing Extracts from Rhus trees on Performance, Microflora Population and Noxious Gas Emission in the Cecum of Broiler)

  • The effect of feeding extracts from rhus trees on performance of broiler were performed to investigate development of natural antibiotic in process of broilers production. A total of 320 broiler chicks at 7 day of age were fed the commercial diet and water, water containing 0 ppm (control), 300 ppm (T1), 600 ppm (T2) and 1000 ppm (T3) of extracts from rims trees for 7 weeks. The body weight gain and feed conversion ratio were improvement by feeding extracts from rims trees at 7 to 21 days of age (P<0.05), but amount of feed intake and mortality were not different among the four group. When 21 to 35 days of age, body weight gain, amount of feed intake, feed conversion ratio and mortality were almost same results when 7 to 21days of age, but it was not significantly different. When 35 to 49 days of age, body weight gain in T1 and T3 group was significantly improvement than control group (P<0.05), also feed conversion ratio was improve by feeding extracts from rhus trees. There was a decrease in the microflora population of both E coli. and salmonella in the cecum contents and feaces in broilers by feeding extracts from rhus trees. Emission of ammonia and VFA gas from excreta were significantly decreased at 5 to 10 days of storaged period (P<0.05). The digestibility of dry matter, crude fat, and crude ash of feed were tend to increased in broiler feeding extracts from rims trees. Digestibility of crude protein of feed in broiler three treatmental groups was significantly improved (P<0.05) as compared with those in control group. These results indicated that the feeding extracts from rhus trees of 300 to 1000 ppm of broiler production were improvement in th6 body weight gain, feed conversion ratio, and mortality due to increasing nutrients digestibility from feed, decreasing of microflora population of both E coll. and salmonella in the cecum contents and feaces, decreasing emission of ammonia and VFA gas from excreta.

국내 자생 고로쇠 (Acer mono Max.) 추출물의 항산화 활성 (Antioxidant Activity of the Extracts Derived from Korean Native Acer mono Max.)

  • Maple tree is a useful medical plant for obtaining bioactive materials such as pharmaceutics, cosmetics, food additive, etc., and there are 16 species of native maple trees in Korea. In this study, we evaluated the antioxidant activity of sap and crude extracts of Acer mono Max, a representative maple species. The crude extracts were obtained by solvent extraction (water, ethanol, and ethyl acetate) from its branches (bark and xylem). The phenolic contents and radical scavenging capacities of the extracts and the sap were evaluated in terms of half maximal effective concentration ($EC_{50}$) and kinetics by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The ethanol extracts showed the highest extraction yield, phenolic contents and antioxidant activity, and bark extracts showed better antioxidant activity than xylem extracts. The antioxidant activity of the sap was very low, but the $EC_{50}$ of ethanol and ethyl acetate extracts ranged from 68 to $79{\mu}g/mL$, similar to that ($60{\mu}g/mL$) of the control, butylated hydroxytoluene (BHT). The DPPH radical scavenging rate ($220{\sim}760{\mu}M/min$) and the second-order reaction rate constant ($6.48{\sim}7.04L/g{\cdot}min$) of these extracts were better than those of BHT ($55{\sim}370{\mu}M/min$ and $3.60L/g{\cdot}min$). These results suggest that A. mono Max. is one of the useful bioresources for obtaining antioxidant biologically active substances, and it is possible to obtain physiologically active substances from by-product of its pruning while minimizing the effect on the growth of the tree.

녹차, 오룡차 및 홍차 추출물의 생리활성과 항산화 효과 (Physiological and Antioxidant Activities of Green, Oolong and Black Tea Extracts)

  • This study investigated the physiological and antioxidant activities of green, oolong, and black tea extracts. The crude catechin extract yields of green, oolong, and black tea were 4.9%, 3.4%, and 2.5%, respectively. Total phenol contents of green, oolong, and black tea were 40.9%, 43.0%, and 38.5%, respectively. The order of the electron donating abilities of green, oolong and black tea were green tea>oolong tea>black tea extracts. The SOD-like activities of green, oolong and black tea extracts at 5,000 ppm were 21.2%, 17.5% and 13.9%, respectively. The nitrite-scavenging abilities of green, oolong and black tea extracts were higher than that of ascorbic acid (p<0.05). Antioxidant activities in soybean oil substrates at 500 ppm were in the order of green tea>oolong tea>black tea${\geq}$BHT (200 ppm). Therefore these results showed that the physiological and antioxidant activities of green tea were better than those of oolong and black tea.

탈지들깨박 Ethanol 추출물의 항산화 효과 (Studies on Antioxidant Activity of Ethanol Extracts from Defatted Perilla Flour)

  • The antioxidant activity of ethanol extracts from defatted perilla flour was investigated by measuring peroxide value of perilla oil during storage at $45^{\circ}C$. The antioxidant activity of ethanol extracts was also compared with BHA, BHT and tocopherol. Anti-oxidant activity of ethanol extracts was also examined in corn oil and lard. The ethanol extracts contents of defatted perilla flour and the original perilla seed were 7.69 and 4.56% respectively. The antioxidant activity of ethanol extracts was superior to that of 0.02% BHT, BHA and tocopherol in the perilla oil substrate, merely in concentration of one-twentieth as much as that contained in original perilla oil seeds. The fractions of non-polar solvent (hexane and chloroform) obtained from silicic acid column chromatography are less effective than that of polar solvent as an antioxidant. Antioxidant activity of partially purified ethanol fraction is slightly inferior to that of original crude ethanol extracts. Ethanol extracts were also effective in corn oil and lard almost same as in perilla oil. The total phenolic compound contents of crude ethanol extracts and partially purified ethanol fraction were 9.3, 6.4%, respectively.

  • 본 연구에서는 전통적으로 담석증, 신장치료 등의 한약제로 많이 사용하는 대왕을 여러 용매를 사용하여 얻은 대왕추출물 분액에 대한 세포독성을 여부를 MTT 정량법, NR 정량법, SRB 정량법을 이용하여 조사하였다. 1. 추출 용매 methylene chloride, ethyl acetate, butanol, water로부터 얻은 대왕추출물 모두 처리농도에 따라 세포에 미치는 영향이 증가하였다. 2. Butanol을 용매로 사용하여 얻은 대왕추출물 분액이 다른 3가지 용매로부터 얻은 대왕추출물보다 세포에 미치는 영향이 크게 나타났고 water를 용매로 사용하여 얻은 추출물이 A498 세포주에 미치는 영향이 가장 낮게 나타났다. 3. Butanol을 추출 용매로 하여 얻은 대왕추출물이 A498 세포주에 미치는 영향이 가장 컸는데 그 추출물에 대한 MTT$_{50}$, NR$_{50}$, SRB$_{50}$값은 각각 0.63mg/ml, 0.65mg/ml, 0.68mg/ml이었고, 가장 영향이 적은 water의 경우 MTT$_{50}$, NR$_{50}$, SRB$_{50}$값은 각각 0.84mg/ml, 0.82mg/ml, 0.80mg/ml이었다. 4. 정량방법 간의 대왕추출물에 대한 반응은 MTT 정량법이 가장 민감하게 나타났다.

Production of Thaxtomin A by Korean Isolates of Streptomyces turgidiscabies and Their Involvement in Pathogenicity

  • Crude extracts of pathogenic Streptomyces turgidiscabies isolates produced necrotic reaction on potato slices. Necrosis was first visible 24 hr after application and increased in severity over several days. However, necrosis was not observed when crude extracts of nonpathogenic strains were applied onto tuber slices. Presence of thaxtomin A from crude extracts of pathogenic S. turgidiscabies was identified by thin layer chromatography and high performance liquid chromatography. Nonpathgenic strains did not produce thaxtomin A in oatmeal broth. Inoculation of potato tuber slices with thaxtomin A partially purified from crude extract of a pathogenic S. turgidiscabies ST5 reproduce necrotic reaction suggesting that thaxtomin A is a pathogenicity determinant in S. turgidiscabies.

  • The effects of crude extracts from medicinal plants on biological activity were investigated. The crude ethanol extract of H. paucistipula inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (2 mm inhibition zone at 150 ㎍/disc) and the dermatophyte Trichophyton mentagrophytes ATCC 28185, (7 mm inhibition zone at 150 ㎍/disc), and toxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC/sub 50/ 2.48 ㎍/㎖ at 75 ㎍/disc). This crude ethanol extract of H. paucistipula is the strongest antimicrobial and cytotoxic activities against P388 murine leukaemia cells ATCC CCL 46 P388D1.

쑥추출물이 혈중 에탄올 농도와 간기능에 미치는 효과 (The Effects of Extracts from Mugwort on the Blood Ethanol Concentration and Liver Function)

  • This study was conducted to investigate the effects of mugwort extracts on the blood ethanol concentration and liver function in rats. Sprague-Dawley rats were used, the rats administered with 25% ethanol (5g/kg$.$B.W.) were devided into three groups (CON-E ; 25% ethanol administered to the CON-E) according to the administered ethanol concentration and the levels of administered mugwonts. Mugwont roots extracts were administered via the caudal vein. Ethanol concentration was measured at the time of 0, 1, 2 and 3hr by gas chromatography. GOT(Glutamic Oxaloacetic Transaminase) and GPT(Glutamic Pyruvic Transaminase) were measured at the time of 0 and 5hr. Components of each extracts were analyzed by using high performance liquid chromatography. Cell number, GOT and GPT were investigated by using rat hepatocyte culture. Megwort extracts were added at the levels of 1% or 2%. Hepatocyte culture were into five groups according to the addition levels. The results were summarized as follows ; 1. Catechin contents of 8∼10mg/100g and the contents of (-)-epigallocatechin was high in the water extracts. 2. Ethanol degradation efficiency declines in the following order : MDW-E>MOH-E>CON-E. 3. The numbers of rat hepatocytes declines in the following order : 2% MDW-L>1%MDW-L>1%MOH-L>CON-L>2%MOH-L. These results suggest that crude catechin of mugwort extracts may play important roles to degrade ethanol and recover liver function in rats.

감초 세근의 생리활성 탐색 (Biological Activities in roots of Glycyrrhiza uralensis Fisch)

  • 감초 세근의 추출물 및 glycyrrhizin과 그 효소분 해물의 생리활성 실험에서, 전체적으로 추출물의 효과가 단일 물질인 glycyrrhizin과 그 효소분해물보다 좋았으며 특히 에탄을 추출물의 효과가 더 좋았다. 여러 인간 암세포 성장저해 실험에서는 추출물 및 glycyrrhizin과 그 효소분해물은 모두 1.0 mg/ml의 농도에서 50%이상의 억제율을 보였으며, 특히 추출물들이 더 좋은 억제율을 나타내었다. 돌연변이 유발 억제실험에서는 물 : 에탄올(1 : 1 v/v) 추출물이 약 50% 억제 효과를 나타내었으며, 면역기능 실험에서 에탄을 추출물이 1.0 mg/ml의 농도에서 B세포는 1.3배, T세포는 1.2배의 촉진 활성을 보였다. 혈당강하실험에서는 물 추출물이 1.0 mg/ml의 농도에서 68%, 에탄을 추출물 65%, 물 : 에탄올(1 : 1 v/v) 추출물이 62%의 높은 활성을 보였으며 glycyrrhizin과 그 효소분해물은 40% 이상의 효과를 보였다. GST활성은 1.0 mg/ml의 농도에서 에탄을 추출물이 257%, 물 추출물 239%, 물 :에탄올(1 : 1 v/v) 추출물은 231%의 높은 생리 활성능을 보였으며 glycyrrhizin과 그 효소분해물도 200%이상의 좋은 효과를 보였다. 이상의 결과로 볼 때 추출열과 추출용매 등에 의해 감초에서 분해된 여러 가지 monomer들이 풍부하게 존재하는 crude 추출물들의 생리활성이 glycyrrhizin과 그 효소분해물보다 더 좋은 효과를 보였다는 것을 알 수가 있었으며 이러한 추출물에 대한 연구가 더 필요하다고 하겠다.
