$Fasciola$ $gigantica$ Fatty Acid Binding Protein (FABP) as a Prophylactic Agent against $Schistosoma$ $mansoni$ Infection in CD1 Mice

  • Although schistosomicidal drugs and other control measures exist, the advent of an efficacious vaccine remains the most potentially powerful means for controlling this disease. In this study, native fatty acid binding protein (FABP) from $Fasciola$ $gigantica$ was purified from the adult worm's crude extract by saturation with ammonium sulphate followed by separation on DEAE-Sephadex A-50 anion exchange chromatography and gel filtration using Sephacryl HR-100, respectively. CD1 mice were immunized with the purified, native $F.$ $gigantica$ FABP in Freund's adjuvant and challenged subcutaneously with 120 $Schistosoma$ $mansoni$ cercariae. Immunization of CD1 mice with $F.$ $gigantica$ FABP has induced heterologous protection against $S.$ $mansoni$, evidenced by the significant reduction in mean worm burden (72.3%), liver and intestinal egg counts (81.3% and 80.8%, respectively), and hepatic granuloma counts (42%). Also, it elicited mixed $IgG_1/IgG_{2b}$ immune responses with predominant $IgG_1$ isotype, suggesting that native $F.$ $gigantica$ FABP is mediated by a mixed Th1/Th2 response. However, it failed to induce any significant differences in the oogram pattern or in the mean granuloma diameter. This indicated that native $F.$ $gigantica$ FABP could be a promising vaccine candidate against $S.$ $mansoni$ infection.

Anti-metastatic and Anti-angiogenic Activities of Ekong-san and Its Metabolites by Human Intestinal Bacteria (이공산의 혈관신생 및 암전이 억제효과에 관한 연구)

  • Ekong-san(EKS) was expected to have inhibitory effects on angiogenesis, considering the fact that its constituents such as Ginseng Radix, Glycyrrhizae Radix and Citri Pericarpium were reported to inhibit angiogenesis. Moreover, recently several metabolites transformed by the human intestinal microflora were reported to enhance effectiveness compared to their crude drugs. Based on these data, this study was designed to confirm whether the EKS metabolites (EKS-M) can significantly exert the anti-angiogenic and anti-metastatic activites. Hence, with EKS and EKS-M, viability assay, proliferation assay, in vitro tube formation assay, gelatin zymogram assay, in vitro invasion assay were carried out. EKS showed less toxicity in ECV304 and HT1080 cells than EKS-M. EKS-M inhibited the proliferation of HT1080 cells by 30% at 200㎍/㎖ and 42% at 400 ㎍/㎖ respectively. Also, EKS-M degraded the tube network at 200㎍/㎖. EKS and EKS-M inhibited the expression of MMP-9 at 200 and 400㎍/㎖ in HT1080 cells. EKS reduced the invasive activity of HT1080 cells through matrigel coated transfilter atthe concentration of 200㎍/㎖ more effectively than EKS-M. These data suggest that EKS and EKS-M has anti-angiogenic and anti-metastatic activities.

Quality Evaluation of the Cinnamon Essential Oils Based on Gas Chromatographic Analysis and Cytotoxicity (가스 크로마토그래피 분석과 세포독성에 의한 계피 정유의 품질평가)

  • To evaluate the quality of the crude drugs using three kinds of Cinnamomum Cortex (CC), Vietnamese CC (VCC, the stem bark of Cinnamomum obtusifolium), periderm-peeled Chinese CC (PPCC, periderm-peeled stem bark of C. cassia), Chinese CC (CCC, stem bark of C. cassia) and a Cinnamomi Ramulus (CR, the twig of C. cassia), the four essential oils were prepared by steam distillation method. Cinnamaldehdye (CAN) and an unknown substance tentatively named hydroxy-cinnamaldehdye(HCNA) were detected in the four essential oils by gas chromatography-mass spectrometry, the contents of which are significantly different one another. Vietnamese CC had the highest content of HCNA whereas CR had the highest CAN content and the lowest HCNA. Vietnamese CC exhibited the greatest cytotoxic activity against the cancer cell lines, A549, HepG-2, HL-60, P-388, U-937, and KB and CR the lowest cytotoxicity. Contents of CAN and HCNA in CCC and PPCC are positioned between VCC and CR. These results suggest that measurement of HCNA and cytotoxicity may determine the quality of CC and CR.

Effects of Water and Methanol Extracts of Cricket (Gryllus bimaculatus) on Alcohol Metabolism (귀뚜라미의 물 및 메탄올 추출물이 알코올 대사에 미치는 효과)

  • The cricket has been used in East Asia as crude drugs for treating fever and hypertension, and is presently reared as a pharmaceutical insect in China and a food for animals. For the purpose of evaluating protective extracts against alcohol-induced toxicity, the extracts of the cricket (Gryllus bimaculatus) were examined in animal models acutely administered alcohol by the cricket in ICR-mice. Water and methanol extracts from the cricket, were found to cause a significant decrease (37%) in the blood ethanol concentration as well as enhancement of liver mitochondrial alcohol dehydogenase (ADH) and acetaldehyde dehydogenase (ALDH) activitieson on a single intraperitoneal administration in mice. Futhermore methanol extract was demonstrated to exhibit more potent enhancing activity on ethanol metabolism than water extract. These results suggest that water/alcohol extract of G. bimaculatus may be used as a food for reducing the toxicity of alcohol.

Studies on the Efficacy of the Combined Preparation of Crude Drugs (VII) -Effect of 'Saengkankunbi-Tang' on the Liver Tissue Recovery- (생약복합제제(生藥複合製劑)의 약효연구(藥效硏究) 제7보(第7報) -생간건비탕(生肝健脾湯)의 간조직부활(肝組織復活)에 대(對)한 작용(作用)-)

  • To investigate the effect of 'Saengkankunbi-Tang' on the liver tissue recovery in about 70% cutted rat liver, the observation on the photographing by the Gamma-camera and the electromicroscopic section against the rabbit liver induced by $CCl_4$, these studies were conducted. The results were summarized as follows; Observed on the liver tissue recovery, function regeneration and body weight increase in about 70% cutted rat liver, the recovery of liver tissue and body weight were increased with the passage of the time and also they were showed to the high increase rate in proportion to administration amount. The glucose content in the serum was found in increasing tide as compared with the control group. The alkaline phosphate activity in the serum showed the significant difference in the 10th day and 13th day as compared with the control group. The ammonia content in the blood showed the significant difference only in the 10th day. Observed on the photographing by the Gamma-camera against the rabbit liver induced by $CCl_4$, sample groups were shown to be significant recovery as compared with the control group. Observed on the electromicroscopic section, sample groups showed the control action of the nuclear destruction against the rabbit liver induced by $CCl_4$ and also they showed the control action of the expansion of glycogen and granular endothelial reticulum in cytoplasm. According to the above finding, it is presume that 'Saengkankunbi-Tang' can be applicable to the extensive treatment of liver disease.

Studies on the Processing of Crude Drugs(VII) -On the Constituents and Biological Activities of Gardeniae Fructus by Processing- (한약의 수치에 관한 연구(제 7보) -치자의 수치에 의한 성분변화 및 생리활성-)

  • We have conducted to characterize the physico-chemical change and pharmacological transformation of traditional herbal medicines by means of processing. Processed Gardeniae Fructus was prepared by heating of fruit of Gardenia jasminoides(GF) for $30{\sim}50\;minute$ in the roster designed for herb processing. The contents of drying loss, water extract, diluted ethanol extract, ether extract and geniposide in non-processed GF and processed GF were examined. The contents of drying loss, water extract and geniposide in processed GF showed a decrease as compared with those of non-processed GF, however the contents of dilute ethanol and ether extract showed a increase as compared with those of non-processed GF. The rate of decrease/increase of those index were in proportion to heating time. And, biological activities of methanol extract of non-processed GF and processed GF were investigated. DPPH scavenging effects and inhibitory effect of xanthine oxidate and hemolysis of processed GF exhibited more effective than those of non-processed GF in vitro. Accelerating effect of large intestinal transport and purgative action of non- processed GF were discriminated by processing of GF. Methanol extracts of non-pro- cessed GF and processed GF showed the protective effects against the hepatotoxicity induced by ${\alpha}-naphthylisothiocyanate$ in rats. These results suggested that the transformation of biological activities of GF by means of processing may be due to the physico-chemical change of the constituents in GF by heating.

Studies on the Crude Drugs of Atractylodis Species (출류(朮類) 대조 생약에 관한 연구)

  • Atrartylodis Rhizoma(蒼朮)'s origin plants are Atratylodes lancea and A. Chinensis in Chinese, Japanese and Korean pharmacopoeia. A. Japonica is only indigenous in Korea, it is actually used as Atractylodis Rhizoma in Korean market. A. lancea is used in Hunan province, China and A. Chinensis is used in Hubei province, China. It is impossible to distinguish with species differency as macro- and micro-morphology. We tried to distinguish with species differency by HPLC and GC-Mass spectra. Atractylone(mw. 216) which is a marker compound in Atractylodis Rhizoma Alba(白朮) was detected in A. japonica. Atractylodin (mw.182) was detected in A. lancea and two eudesmadien derivatives (mw. 204) were detected in A. chinensis. HPLC chromatogram showed the same patterns. As a result, we propose that A. japonica will be added as Atractylodis Rhizoma (蒼朮)'s origin plant in Korean Pharmacopoeia. Atractylodis Rhizoma Alba(白朮)'s origin plants are A. macrocephala in China, and A. Japonica and A. ovata in Korea and Japan. In GC-Mass analysis, all samples showed same patterns and the main compound was atractylone.

Studies on Domestic Production of Imported Botanical Drugs-On Domesticational Cultivation of Coptis japonica- (수입 생약(生藥)의 국산화(國産化)에 관한 연구-황연(黃蓮)의 순화재배(馴化栽培)에 대하여-)

  • A study was carried out to investigate the possibility of acclimation of Coptis japonica Makino in Korea. Coptis japonica could be grown within 4 years as a medicinal crude drug if it is cultivated in the area of mountain facing north(300m above sea level) with 70% light interception and appropriate soil texture. It is to be noted that the propagation of the plant is more important than the yield of berberine because the berberine contents do not change in different conditions. The results were as follows; 1. The germination rates of the seeds by covering with the pine tree leaves were the highest and averaged 63%. 2. The germination rates of dry seeds were not affected in spite of low temperature and $GA_3$ treatment. 3. The germination rates of the seeds stored in moistured sand were averaged 85%, but in dry state, they were not germinated. 4. The number of leaves and fresh weight of Coptis japonica from the area of mountain facing north were increased significantly. 5. The growth and fresh weight of Coptis japonica were reduced in clay. 6. The best growth was obtained under 70% light interception. It is possible to intercrop between evergreen trees. 7. The foliage dressing did not affect the growth. 8. The number of leaves and fresh weight were increased from the 3rd year, and the average weight of rhizomes was 5.7g in 4th year. 9. The berberine contents of the rhizomes of Coptis japonica under various treatment conditions ranged from 9 to 10%. Coptidis Rhizoma from this study and from Korean market showed much higher berberine contents than those from Japanese market.

Inductions of Caspase-, MAPK- and ROS-dependent Apoptosis and Chemotherapeutic Effects Caused by an Ethanol Extract of Scutellaria barbata D. Don in Human Gastric Adenocarcinoma Cells

  • Objectives: The crude extracts of Scutellaria barbata D. Don (SB) have traditionally demonstrated inhibitory effects on numerous human cancers both in vitro and in vivo. Gastric cancer is one of the most common types of cancer on world. The authors investigated the effects of an ethanol extract of Scutellaria barbata D. Don (ESB) on the growth and survival of MKN-45 cells (a human gastric adenocarcinoma cell line). Methods: The MKN-45 cells were treated with different concentrations of ESB, and cell death was examined using an MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. Analyses of sub-G1 peaks, caspase-3 and -9 activities, and mitochondrial membrane depolarizations were conducted to determine the anti-cancer effects of SB on MKN-45 cells. Also, intracellular reactive oxygen species (ROS) generation was investigated. Results: ESB inhibited the growth of MKN-45 cells, caused cell cycle arrest, and increased the sub-G1 population. In addition, ESB markedly increased mitochondrial membrane depolarization and the activities of caspase-3 and -9. ESB exerted anti-proliferative effects on MKN-45 cells by modulating the mitogen-activated protein kinase (MAPK) signaling pathway and by increasing the generation of ROS. Furthermore, combinations of anti-cancer drugs plus ESB suppressed cell growth more than treatments with an agent or ESB, and this was especially true for cisplatin, etoposide, and doxorubicin. Conclusion: ESB has a dose-dependent cytotoxic effect on MKN-45 cells and this is closely associated with the induction of apoptosis. ESB-induced apoptosis is mediated by mitochondria-, caspase- and MAPK dependent pathways. In addition, ESB enhances ROS generation and increases the chemosensitivity of MKN-45 cells. These results suggest that treatment with ESB can inhibit the proliferation and promote the apoptosis of human gastric adenocarcinoma cells by modulating the caspase-, MAPK- and ROS-dependent pathway.

Effect of Chinae Rhizoma on the Motility of Isolated Rabbit Duodenum (토복령알코홀추출물이 토끼 적출 십이지장관운동에 미치는 영향)

  • Chinae Rhizoma is dried crude roots and rhizomes obtained from Smilax china Linne which belongs to Smilax Genus of Liliaceae Familia, and it has been in wife use as a folk remedy for a long time. But very little systemic studies on its pharmacological actions have been made until now. Furthermore, the effective components except saponin of Chinae Rhizoma have been not known yet. It came to the attention of the authors that Chinae Rhizoma are used as antidiarrhetics in Chinese herb medicine, therefore they attempted to observe the effects of Chinae Rhizoma Alcoholic Extracts (Chinae Rhizoma Ethanol Extract=CREE, Chinae Rhizoma Methanol Extract=CRME) from Smilax china Linne on motility of the isolated rabbit duodenum in this experiment. The results of the study were as follow: 1. The movement of the isolated rabbit duodenum was slowed down and its tonus was relaxed in proportion to the concentration of Chinae Rhizoma Alcoholic Extracts (CREE & CRME). 2. With a concentration of CREE $10^{-4}g/ml$, the contractile effects of acetylcholine $10^{-6}g/ml$, pilocarpine $10^{-5}g/ml$ and physostigmine $10^{-5}g/ml$ were remarkably blocked in the movement of isolated rabbit duodenum. 3. The concentration of the isolated rabbit duodenum induced by barium chloride $10^{-4}g/ml$ was significantly inhibited by CREE $10^{-4}g/ml$. 4. With the administrations of histamine $10^{-5}g/ml$, serotonin $10^{-6}g/ml$ and prostaglandin $A_2\;10^{-6}g/ml$ respectively, contractions were observed in the rabbit duodenum. But, treatment of CREE $10^{-4}g/ml$ inhibited markedly the tonus and motility of the duodenum pretreated with the above drugs. 5. The effects of CREE and CRME were very much alike in this experiment. From the above results, it is concluded that the mechanism of action of Chinae Rhizoma on motility of isolated rabbit duodenum was partially due to cholinergic blocking effect.
