Studies on Combined Usage of Combined Preparation of Crude Drugs and Anti-neoplastic Drugs(II) -Alleviative effect of cis-Platin and Mitomycin C through Combined Usage of Gamidaebo-Tang- (생약복합제제(生藥複合製劑)와 항암제(抗癌劑)의 병용투여(倂用投與)에 관한 연구(硏究) (제2보)(第2報) -cis-Platin 및 Mitomycin C와 가미대보탕(加味大補湯)의 병용투여(倂用投與)에 의한 부작용(副作用) 경감(輕減) 효과-)

  • The water extract of Gamidaebo-Tang depressed the lethal toxicity of cis-platin$(45{\mu}M/kg,\;s.c.)$, and mitomycin C (6mg/kg, s.c.). Significant depression of renal toxicity(indicated by an increased blood urea nitrogen value and creatinine value) of cis-platin $(35{\mu}M/kg)\;s.c.)$, was observed in mice treated with Gamidaebo-Tang. In rats treated with mitomycin C and Gamidaebo-Tang, the increased LDH activity and reduced total protein and albumin contents by mitomycin C were significantly depressed. The number of WBC was significantly increased in rats treated with mitomycin C but in the rats treated with mitomycin C and Gamidaebo-Tang, the number of WBC was increased and the hematocrit value and the number of RBC and Hgb were significantly increased in the dose-dependent manner. Therefore, bone marrow toxicity of mitomycin C in the rats treated with Gamidaebo-Tang was depressed. In the end, alleviative effects of the side actions of cis-platin and mitomycin C were acknowledged by combined usage of Gamidaebo-Tang and these anti-neoplastic drugs.

Medicinal Components in Bupleurum Species (시호의 약리성분 특성)

  • This review deals briefly with the various medicinal components(mainly saikosaponins), their biological activities and the variation of their contents by different cultivation environment and plant parts in Bupleurum species. Bupleuri radix, a crude drug, is the root of Bupleurum falcatum L. (Korea, Japan), B. chinense(China), and their related species (Umbelliferae). There are over 120 species in Bupleurum genus throughout world, mainly Asian area, and over 5 species in Korea, investigated up to now. These plants contain many physiological active compounds and the principal components are saikosaponins. Major activities of this crude drug and saikosaponins are the anti-inflammatory and antihepatotoxic activities. Saikosaponins and their derivatives in Bupleurum spp. have been chemically studied, isolated and identified over 70 compounds in over 50 species. Other components, physiologically active ones, also have been investigated, which are the groups of lignan, flavonoid, essential oil, polyacetylene, polysaccharide, etc. Saikosaponins belong to the group of triterpenoid saponin chemotaxonomically and occur the accumulation and turnover in plant tissues through secondary metabolism, mevalonic acid pathway. The contents and kinds of saikosaponins and other components in Bupleurum spp. plants are various due to different species and growing environments, as the plant growth characters and yield are various. Most of medicinal plants as well as Bupleurum species are very useful as agricultural products and traditional medicines, and also are very valuable as genetic resources and natural products. So we need to collect, evaluate, preserve, and utilize various medicinal plants, and also to under-stand secondary metabolism and improve the breeding and cultivation techniques for the safe production of crude drugs with high quality and yielding.

Chemosensitizing Activity Against Human Leukemia Cell of Crude Extracts of Native Camellia(Camellia japonica) in Jeonnam

  • This study has been undertaken to increase availability of native camellia in Jeonnam as a medicinal resource and to isolate the effective components from them. Multidrug resistance(MDR) by tumor cells is a major obstacle to successful cancer chemotherapy. We report that the crude extracts of camellia flowers, leaves has a chemosensitizing effect that can reverse Pgp-mediated MDR by increasing the intracellular accumulation of drugs. The cytotoxic and chemosensitizing effects of MeOH extract from 12 spp. citrus fruits on the AML-2/D100 were determined using MTT assay. Chemosensitizing effects was screened in the presence of vincristine, a good substrate of Pgp. IC$\sub$50/ for extracts in AML-2/WT was found to be 65∼350$\mu\textrm{g}$/$m\ell$ whereas the range of its mean IC$\sub$50/ value in Pgp-overexpressing cells (AML-2/Dl00) in the presence of vincristine was 90∼400$\mu\textrm{g}$/$m\ell$. Of the extracts tested, mature leaf extract displayed the most potent chemosensitizing effect[IC$\sub$50/;100 $\mu\textrm{g}$/$m\ell$, CR;1.06, RF;2.97 in the presence of VCR]. This indicates that the toxicity (IC$\sub$50/;288.89$\mu\textrm{g}$/$m\ell$) of mature leaf extract is minimal at concentrations required for a complete reversal of the drug resistance. Also, this result indicates that crude extracts of camellia mature leaves would contain some principles which have chemosensitizing activity.

Comparison of the Anti-inflammatory Effects of the Extracts from Rubus coreanus and Rubus occidentalis (토종 복분자와 외래종 복분자 추출물의 항염증효과 비교)

  • The dried fruit of the Rubus coreanus, which is well-known in Korea and referred to as 'Bokbunja,' has been employed as a traditional medicine for centuries. This crude drug has been utilized in Korea for the management of impotence, spermatorrhea, enuresis, asthma, and allergic diseases. Our previous study demonstrated that the ethanol extracts of R. coreanus have anti-inflammatory effects. The principal objective of the present study was to conduct a comparison of the anti-inflammatory effects of the ethanol extracts of R. coreanus and R. occidentalis; here, we tested the unripe (URCE), half-ripened (HRCE), and ripened fruits (RCE) of R. coreanus, and the unripe (UROE), half-ripened (HROE), and ripened fruits (ROE) of R. occidentalis. We found that URCE, UROE, HRCE, and HROE reduced the production of nitric oxide and prostaglandin $E_{2}$ as well as pro-inflammatory cytokines in lipopolysaccharide-stimulated RAW264.7 murine macrophages. Interestingly, the R. coreanus extracts showed stronger inhibitory effects on the production of these inflammatory mediators than the R. occidentalis extracts.

  • HPLC is a useful method for identifying amygdalin which has been the subject of considering controversy for cancer remedy. The resolution of amygalin was satisfactory and the separation of amygdalin was accomplished using carbohydrate analysis column, mobile phase of acetonitrile: $H_2O$ (90:10), and UV detector. Quantitative analysis showed that amygdalin contents of Prunus tomentosa, P. serrulata, and P. ishidoyana (Amygdalaceae) were 2.18, 2.41, and 2.26%, respectively. It was suggested that the seed of Prunus species was worth useful resource of amygdalin.

  • The rhizomes of Salvia miltiorrhiza Bunge(Labiatae) has been used in Chinese traditional medicine for the treatment of coronary heart diseases and myocardial infarction. As a part of a research for standardization of crude oriental drugs, we have determined the content of tanshinone IIA in Salvia miltiorrhiza purchased from various regions of Korea. The HPLC method by which quantitative analysis was conducted, showed reproducible results and chromatographic isolation of tanshinone IIA was accomplished successively.

  • 'Pae Jang (敗醬)' is one of Chinese crude drugs used mainly as a edema, abdominal pain and hemoptysis, etc. With regard to the botanical origin of 'Pae Jang', it has been considered to be Patrinia scabiosaefolia of Valerianaceae, but there has never been studied pharmacognostically. To clarify the botanical origin of the 'Pae Jang', we studied on the anatomical characteristics of the roots of three species growing in Korea. Through our studies, the botanical origin of 'Pae Jang' from Korea was proved to be the under ground portions of Patrinia scabiosaefolia and Patrinia villosa.

  • Studies were undertaken to develop for the anti-inflammatory analgesic activities of 'Insam-Tang' extract, which composed of four crude drugs; Ginseng Radix, Glycyrrhizae Radix, Atractylodis Rhizoma, and Zingiberis Rhizoma, was known to be applied in edema, convulsion, excremental blood, and cooling pain etc, in oriental herb remedies. It was exhibited significant anti-inflammatory effect on carrageenin inflammation, and preventive and therapeutic effects to chronic inflammation on Freund' complete adjuvant in rats, Especially, therapeutic effect of extract 500mg/kg was observed more effective than that of phenylbutazone 50mg/kg. Also, it was shown significant effect on leakage of dye into the peritoneal cavity and analgesic effect on squirm symptom induced by acetic acid in mice.

  • This study was undertaken to investigate the effect of 'Jakyakgamcho-Tang' and parched 'Jakyakgamcho-Tang' on anti-inflammatory and antiulcerative actions, and motility of isolated intestine of mice. The results of this study were summarized as follows; Anti-inflammatory effect was showed markedly by the administration of water extract of 'Jakyakgamcho-Tang' and parched 'Jakyakgamcho-Tang'. Isolated intestine of mice was relaxed with the treatment of 'Jakyakgamcho-Tang' and parched 'Jakyakgamcho-Tang' and intestinal contraction induced by $BaCl_2$ and acetylcholine was also inhibited. The water extract of 'Jakyakgamcho-Tang' and parched 'Jakyakgamcho-Tang' were significantly inhibited Shay's ulceration in rats.

Studies on the Heavy Metal Contents in Cultivated Medicinal Plants and their Correlations with Soils (수종 재배 약용식물과 토양중 중금속 함량에 관한 연구)

  • Heavy metals may cause trouble(s) in human body. For consideration of the safety of crude drugs cultivated in Korea, we selected some medicinal plants and soils from cultivated site and estimated their heavy metal contents by inductively coupled argon plasma spectrometry (ICAP). Among heavy metals tested, the highest level of Cu and Zn were shown in plants cultivated in Kyung Ki; the highest level of Fe in those in Youngnam and the highest level of Mg and Mn in those in Youngdong. The heavy metal content s were as low as to be permeasible at least for the medicinal plants we selected. There were no significant correlations between heavy metal contents of soils and cultivated medicinal plants.
