십자화과 채소의 가공 및 저장 중 Glucosinolate의 분석과 그 함량변화 (Analysis of Glucosinolates and the Change of Contents during Processing and Storage in Cruciferous Vegetables)

  • Glucosinolate는 배추, 양배추, 무 및 케일에서는 각각 indole glucosinolate를 포함한 6, 5, 5 및 2종류의 glucosinolate가 분리 및 동정되었고, 겨자에서는 4종류의 glucosinolate가 분리 및 동정 되었으나, indole glucosinolate는 분리되지 않았다. 또한 배추에서는 glucosinolate함량이 속, 줄기 및 잎부분의 순으로 높게 나타났다. 총 glucosinolate의 함량은 시료별로는 건조량으로 환산하여 배추, 양배추, 무, 케일 및 겨자의 순으로 나타났으며, 가공상태에 따라서는 신선한 상태, 자연건조, 가열 건조 및 조리한 상태의 순으로 나타났다. 저장에 따른 총 glucosinolate의 함량은 저장기간이 경과함에 따라 조금씩 감소하였다.

배추 절임조건에 따른 Myrosinase 활성 및 Total Glucosinolates 함량 변화 (Changes in Myrosinase Activity and Total Glucosinolate Levels in Korean Chinese Cabbages by Salting Conditions)

  • 배추의 절임조건에 따른 수분함량, 염도 및 pH 변화를 측정하였다. 절임에 사용한 소금의 농도가 높고 절임 시간이 증가할수록 배추의 수분 함량은 감소하는 것으로 나타났다. 이는 삼투압에 의하여 조직액의 용출 및 소금의 침투로 나타나는 현상으로 절임배추 특유의 조직 감과 관련이 있는 것으로 판단된다. 염분 농도가 비교적적은 6%에서는 삼투압의 변화가 비교적 적게 일어나 비교적 일정한 염분 농도를 유지하는 반면에, 소금의 농도를 증가시키면 절임과정 중에 배추의 삼투압에 변화가 일어나 배추에서 유출된 수분으로 초기에 사용한 소금의 염도보다 감소하였다. 절임 전 신선한 배추의 pH는 약 알칼리(pH 7.8~8.2)에서 절임시간이 경과할수록 점차로 낮아지는 것으로 나타났다. 이는 절임 과정 중에 배추에 존재하는 유기산이 용출 및 초기 발효에 의해 pH가 감소했을 것으로 판단된다. 절임시간이 경과할수록, 절임에 사용한 소금의 농도가 증가할수록 myrosinase 활성이 점차로 감소하였다. 배추의 절임시간이 길어지고 사용한 소금의 농도가 증가할수록 pH가 감소하는 것은 myrosinase 의 최적 활성을 나타내는 pH 범위를 벗어나게 되어 효소활성이 급격히 감소하는 것으로 판단된다. 절임에 사용한 소금의 농도와 절임시간이 증가함에 따라 total glucosinolates의 함량도 더 많이 감소함을 확인하였다. 이는 배추와 소금물이 접촉하면서 배추의 세포막이 파괴되고 세포 내에 있는 myrosinase의 작용에 의해 glucosinolates가 가수분해 되어 다양한 분해산물을 형성하는 것으로 판단된다. 이러한 변화는 절임공정에서 물리적 절단, 삼투압 현상으로 인한 조직의 손상, pH 변화 등에 의하여 glucosinolates가 가수분해 과정을 거쳐 분해산물을 생산하는 것으로 판단된다.

고랭지 여름 무에서 시들음병 발생 현황과 병원균의 병원학적 특성 연구 (Status of Fusarium Wilt Incidence on Summer Radish and Etiological Characteristics of the Causal Fungus in Korea)

  • 2018년과 2019년에 강원도 여름 재배 무 포장에서 시들음병 발생이 조사되었다. 시들음병은 7월 상순 발생했고, 7월 하순과 8월 뜨거운 여름에 급속히 확산되었으며, 심한 경우, 일부 포장에서는 80%까지 발생하였다. 유묘기에 병든 무는 생육이 불량하고 뿌리 내부조직이 갈변되었다. 생육중기 동안 시간이 지나면서 병든 잎은 황화되고 뿌리표면은 흰색에서 흑색으로 변색되고, 도관조직은 갈변하였다. 병든 식물체로부터 23개 Fusarium균이 분리되었고, EF-1α와 IGS 염기서열 분석에 의해 Fusarium oxysporum f. sp. raphani로 동정되었다. 무와 다른 작물에 대한 인공접종에 의해 병원성이 검정되었다. 11개 품종 간에 차이는 있었으나 모든 균주들은 무에서 병원성이었다. 그러나 로켓, 방울양배추, 스토크, 순무를 포함하는 배추과 작물을 제외하고는 병원성이 없었다. 병원성 검정결과는 무 포장에서 시들음병을 방제하기 위해서는 배추과 채소 이외의 작물로 돌려짓기하는 것이 필요하다는 것을 나타낸다.

Genetic Diversity and Phylogenetic Relationships between Chinese Cabbages [B. campestris (syn. rapa) L.] and Cabbages (B. oleracea L.) in Korea

  • Members of the genus Brassica, which are known as oil crops or cruciferous vegetables, are widely cultivated in Canada, Australia, Asian and Europe. Because Brassica species have high yields, are well adapted to their environments, and are self-incompatible, the germplasm is abundant. Previous studies have reported abundant genetic diversity even within Brassica subspecies. In Korea, fresh cabbage leaves are eaten with roast meat, and to meet the current popular demand, new varieties are being increasingly bred. To determine the genetic diversity and relationships among the cabbage vegetables in Korea, we evaluated the genetic variation of 18 accessions based on 5S and 18S ribosomal RNA (rRNA) gene sequences. We detected many variable nucleotide sites, especially in the 5S rRNA gene sequences. Because the length of the 18S rRNA gene might influence the dissimilarity rate statistics, we used both the 5S and 18S sequences to analyze the phylogenetic relationships. S7 (B. oleracea) showed the most distant phylogenetic relationship with the other Brassica species. Interestingly, B2 (B. oleracea), B15, and B18 (B. campestris) have three different types of leaf profiles, and were divided into one group, and the other Brassica species formed another group. Statistical analysis of interspecies and intraspecies genetic distances revealed that B. campestris L. showed higher genetic diversity than B. oleracea L. This work provides additional data that facilitates the evaluation of the genetic variation and relationships among Brassica species. The results could be used in functional plant breeding programs to improve Brassica crops.

Changes in Isothiocyanate Levels in Korean Chinese Cabbage Leaves during Kimchi Storage

  • Glucosinolates are hydrolyzed by the enzyme myrosinase and are mainly found in cruciferous vegetables such as Chinese cabbage (Brassica campestris L. ssp. pekinensis). lsothiocyanates (ITCs) are glucosinolate degradation products with reported anticarcinogenic properties. Korean Chinese cabbage in the form of 'kimchi' is a staple part of the Korean diet. In this study, we examined the effects of storage temperature and duration on glucosinolate, ITC, soluble sugar, and organic acid levels in kimchi. Changes in pH and the impact of various parts of the Korean Chinese cabbage being used during the preparation of the dish were also assessed. Extracted ITC levels, analyzed via gas chromatography (GC) and GC/mass spectrometry (GC/MS), were higher in the midrib parts than in the cabbage leaves after storage at both 4 and $20^{\circ}C$. During storage, organic acid levels increased while soluble sugars were depleted. The pH initially increased (after 1 day at $20^{\circ}C$, and 1 week at $4^{\circ}C$), but subsequently decreased over time at both temperatures. Glucosinolate and ITC levels increased in the beginning of storage but then generally fell during further storage. Our data suggest that acidity-related reduction in myrosinase activity during storage may decrease glucosinolate and ITC levels. The changes in these levels depended on the storage conditions and the Korean Chinese cabbage parts used for the kimchi preparation.

Current Status of Plasmodiophora brassicae Researches in Korea

  • Clubroot disease is caused by the soil-born obligate plant pathogen Plasmodiophora brassicae. This pathogen can infect all cruciferous vegetables and oil crops, including Brassica rapa, B. oleracea, B. napus, and other Brassica species. Clubroot disease is now considered to be a major problem in Chinese cabbage production in China, Korea, and Japan. We collected several hundreds of P. brassicae infected galls from Korea, and isolated the single spore from the collection. For establishment of novel isolation, and mass-propagation methods for singe spore isolates of P. brassicae pathogen, we developed new filtration method using both cellulose nitrate filter and syringe filter. Accurate detection of P. brassicae pathogen in the field was done by using real-time PCR in the potential infested soil. When we tested the different pathogenicity on commercial Chinese cabbage varieties, P. brassicae from collected galls showed various morphological patterns about clubroot symptom on roots. To date, 8 CR loci have been identified in the B. rapa genome using the quantitative trait loci (QTL) mapping approach, with different resistant sources and isolates. We are trying to develop the molecular marker systems for detect all 8 CR resistant genes. Especially for the study on the interaction between pathogens and CR loci which are not well understood until now, genome wide association studies are doing using the sequenced inbred lines of Chinese cabbage to detect the novel CR genes.

Diet and Colorectal Cancer Risk in Asia - a Systematic Review

  • Diet is one of the major factors that can exert a majorly influence on colorectal cancer risk. This systematic review aimed to find correlations between various diet types, food or nutrients and colorectal cancer risk among Asian populations. Search limitations include dAsian populations residing in Asia, being published from the year 2008 till present, and written in the English language. A total of 16 articles were included in this systematic review. We found that red meats, processed meats, preserved foods, saturated/animal fats, cholesterol, high sugar foods, spicy foods, tubers or refined carbohydrates have been found by most studies to have a positive association with colorectal cancer risk. Inversely, calcium/dairy foods, vitamin D, general vegetable/fruit/fiber consumption, cruciferous vegetables, soy bean/soy products, selenium, vitamins C,E and B12, lycophene, alpha-carotene, beta-carotene, folic acid and many other vitamins and minerals play a protective role against colorectal cancer risk. Associations of fish and seafood consumption with colorectal cancer risk are still inconclusive due to many varying findings, and require further more detailed studies to pinpoint the actual correlation. There is either a positive or no association for total meat consumption or white meats, however their influence is not as strong as with red and processed meats.

배추 무사마귀병의 발생상황과 병원균(Plasmodiophora brassicae)의 병원성 및 배추품종의 병저항성 (Incidence, Pathogenicity of Clubroot Fungus(Plasmodiophora brassicae) and Varietal Resistance in Chinese Cabbage)

  • To obtain a basic information of breeding for resistance to clubroot in Chinese cabbage, disease incidence, pathogenicity, and varietal response to the pathogen were studied. Incidence of clubroot was observed at 3 districts in Gyeonggi-Do, 2 districts in Kangwon-Do, and 1 district each in Gyeongnam, Geongbuk and Jeonbuk, respectively. Disease infection rate and diseased ara were most severe in northern part of Gyeonggi-Do. The isolates of clubroot collected from 8 different districts were not different in their virulence one another in view of their infection rate and disease severity in Chinese cabbage. The clubroot fungus had a wide host range for the cruciferous vegetables. Disease severity was high in rape, turnip and mustard, moderate in Chinese cabbage and broccoli, and low in kale and cauliflower. All of Korean hybrids of Chinese cabbage tested were highly susceptible to clubroot, but Japanese varieties were resistant to the highly pathogenic isolate (EJ-93) which was isolated from the Chinese cabbage in Korea. The hybrid(F1) between clubroot resistant line(930WG) and the susceptible line(332MS) showed completely resistant reaction, which indicated that clubroot resistance was governed by a dominant gene.
