답잡초를 대상으로 벼 흰빛잎아름병 기주절위구명 (Paddy Weeds Serving as the Possible Reservoirs for Rice Bacterial Leaf Blight)

  • 답잡초 돌피 외 19종을 대상으로 병원균군, 접종방법 및 잡율의 생육시기를 달리하여 벼흰빛잎마름병의 기주범국 구명시험에서 얻어진 결과를 요약하면 다음과 같다. 1. 나도겨풀, 강아지풀, 독새풀 등은 밀양 2003에 준한 이병성을 나타냈으며, 그 밖에 바랭이, 왕바램이 알방동산이, 너도방동산이, 바람하늘지기 등에서 이병성을 보였으나 광엽잡초에는 전혀 이병되지 않았다. 2. 생육초기에 이병성을 크게 보인 강아지풀, 나도겨풀, 독새풀은 생육전기간을 통해서 이병성이 컸으며, 균군간에는 제일군이 병원성이 다소 큰 것 같았다. 3. 접종방법별로는 유상접종인 핀 및 가위 접종이 무상접종의 분무접종보다 이병성이 켰다.

호평벼와 남평벼의 혼합재배에 의한 도열병 방제와 레이스 다양성의 변화 (Rice Blast Control and Race Diversity by Mixed-Planting of Two Cultivars ('Hopyeongbyeo'/'Nampyeongbyeo') with Different Susceptibility to Magnaporthe oryzae)

  • 도열병에 대한 감수성 정토가 다른 두 품종의 혼합재배에 의한 무농약 도열병 방제효과를 조사하였다. 재배 생력화를 위해 도열병 저항성을 제외한 품질을 비롯한 벼의 유전적 형질이 유사한 품종조합을 선발한 결과 '호평벼'와 '남평벼'의 조합을 1:1로 종자 혼합을 통한 혼합재배를 실시하였을 때 50.4%의 이삭도열병 방제효과가 있었다(2007). 단일재배시 최대 67.4%까지 우점을 보이던 KJ-101레이스가 혼합재배구에서 최소 29.4%로 감소하였고 그 외의 기타 레이스들이 차지하는 비율은 5.4%에서 9.0%로 증가하였으며 11개의 새로운 레이스가 분리되었다. 단독 혹은 혼합재배에 관계없이 분리된 레이스의 수는 유사하였으나 혼합재배 시 새로운 레이스가 차지하는 비율이 12.5%에서 62.5%로 증가하였고 레이스 간 밀도의 차이는 23.6%에서 8.8%로 감소하였다. 이러한 병원균 레이스 다양성의 변화와 레이스 밀도의 균형이 혼합재배에 의한 도열병 발생의 감소와 깊은 관련이 있을 것으로 추정되며 이와 더불어 혼합재배 효과에 대한 좀 더 명확한 설명을 위해 병원력을 비롯한 레이스의 유전적 형질에 관한 연구가 추후 보완되어져야 할 것이다.

들깻잎 재배단지에서 분리한 Staphylococcus aureus의 독소 유전자와 항생제 감수성 분석 (Profiles of Toxin genes and Antibiotic Susceptibility of Staphylococcus aureus Isolated from Perilla Leaf Cultivation Area)

  • 본 연구는 들깻잎과 들깻잎 생산환경을 대상으로 31개의 시료를 채취하여 S. aureus 를 분리 하였다. 분리된 S. aureus 31주의 toxicity를 평가하고자 독소유전자와 항생제 내성을 검색하였다. 그 결과 분리된 균주에서 4개의 서로 다른 독소유전자 패턴은 확인하였으며 sea와 sed유전자를 동시에 보유하는 균주는 4균주 (12.9%)였고, sed 유전자를 보유하는 균주는 9균주 (29.0%)였으며 see 유전자를 보유하는 균주는 1균주 (3.2%)였다. 한편 seb와 sec 유전자를 보유하는 균주는 없었다. 항생제 내성평가결과, 12제의 항생제(penicillin, ampicillin, oxacillin, amoxicillin-clavulanic acid, cefazolin, cephalothin, imipenem, gentamicin, tetracycline, ofloxacin, norfloxacin, and erythromycin)에 내성을 보인 균주는 7균주 (22.6%)였다. 또한 분리된 균주의 2균주 (2.6%)는 5제의 항생제(penicillin, ampicillin, amoxicillin-clavulanic acid, gentamycin, and telithromycin)에 내성을 보였고 MRSA (Methicilline Resistant Staphylococcus aureus)는 포장지, 장갑, 들깻잎에서 발견되었다. 따라서 본 연구결과는 들깻잎에 오염된 S. aureus에 의하여 독소형 식중독이 발생할 가능성을 시사하며 들깻잎과 생산 환경에서 항생제 저항성 S. aureus가 검출되어 의약계뿐만 아니라 농업현장에서도 항생제내성균주 출현을 예방하는 대책이 요구된다.

사탕무씨스트선충의 기주범위 검정 (Host Range Screening of the Sugar Beet Nematode, Heterodera schachtii Schmidt)

  • 사탕무씨스트선충(Heterodera schachtii)의 피해가 2011년에 배추 주산지인 강원도 지역에서 확인되었다. 이 선충은 암컷이 씨스트를 형성하는 특성을 가지므로 농약에 의한 효과적인 방제가 어렵다. 따라서 사탕무시스트선충에 대한 비기주 작물을 선정하여 감염지역의 배추를 대체할 작물을 추천하고자 총 17과(科) 276품종의 식물에 대한 사탕무씨스트선충의 저항성을 검정하였다. 사탕무씨스트선충 접종 후 씨스트 발생 정도에 따라 감수성(susceptible), 중감수성(moderately susceptible), 저항성/비기주(resistant/immune)로 구분하였다. 검정 결과 감수성 106품종, 중감수성 40품종, 저항성/비기주 130품종으로 구분되었다. 씨스트가 전혀 형성되지 않은 작물은 가지, 토마토, 상추, 들깨, 당근, 셀러리, 수박, 참외, 오이, 호박, 부추, 양파, 파, 도라지, 더덕, 잔대, 콩 등이었다. 가지과, 국화과, 명아주과, 화본과 작물은 작물 및 품종에 따라 약감수성 또는 저항성/비기주 식물이 혼재하였다. 본 시험에서 사탕무씨스트선충이 전혀 발생하지 않아 저항성/비기주식물로 밝혀진 130품종은 사탕무씨스트선충 감염지의 배추를 대체할 작목을 추천하는데 활용할 수 있을 것으로 생각된다.

Interactions Between Leafminer Damage and Leaf Necrosis Caused by Alternaria alternata on Potato in the Sultanate of Oman

  • Four field experiments were carried out from 1998 to 1999 and from 1999 to 2000 growing seasons of potato to investigate the relationship between leafminers and Alternaria alternata. The experiments established differential susceptibility among potato varieties to alternaria leaf necrosis, relationship between leafminer infestation and the level of necrosis, use of insecticides to reduce leafminer and leaf necrosis damages, and use of polyester fleece to eliminate leafminer and eventually reduce leaf necrosis. Results of the study indicate that control of leafminer is of primary importance because this will automatically lead to control of the damage caused by A. alternata.

Breeding of 'Jinmani' Cultivar of Gomchwi with Disease Resistance, High Quality and Yield

  • Gomchwi using stuffed leaves is many cultivated for 'Gondalbi(Ligularia stenocephala)' among Gomchwi species. 'Gondalbi' species like to be cultivation on farm because of low the incense and the bitter taste, and high yield. But 'Gondalbi' caused to curtail yield that susceptibility of powdery mildew disease and shriveling and death of plant on summer season. To solve this problems, we crossed a Gomchwi and 'Handeari-gomchwi' to have resistance of powdery mildew disease and high yield. A new Gomchwi cultivar 'Jinmani' was bred by crossing between Gomchwi (Ligularia fischeri (Ledeb.) Turcz.) and Handaeri-gomchwi (Ligularia fischeri var. spiciformis Nakai). The selection and investigation of growth and yield characteristics were conducted from 2007 to 2020 in field and greenhouse of Highland Agriculture Research Institute, NICS, Rural Development Administration. The color of petiole ear was purple. trichome of petiole and leaf back non-existed, and luster of leaf back existed. Density of leaf vein was 4 degree among 1-5 degree in a newly developed cultivar 'Jinmani'. Plant height, leaf length, leaf width and petiole length were 55.7, 21.8, 22.2, and 33.9 cm, respectively in the 2nd year of growth characteristics. Plant size was similar with that of 'Gommany'. Bolting and flowering time were Aug. 5 and Sept. 5, respectively, and Bolting and flowering time of 'Gommany' showed similar to Aug. 8 and Sept. 1, respectively. 'Jinmani' showed higher number of leaves (202 ea.) per plant compared to 'Gommany' (159 ea.). Furthermore, yield was 67.9% higher in 'Jinmani' (2,569 g/plant) than in 'Gommany' (1,530 g/plant). 'Jinmani' showed lower leaf thickness (0.66mm) than 'Gommany' (0.69 mm), and consequently showed more hardness in leaf characteristics (25.1 kg/2) compared to 'Gommany' (24.3kg/cm2). 'Jinmani' showed similar strong resistance compared to 'Gommany' in the susceptibility of powdery mildew disease.

Relationship of Transformation Efficiency and Metabolites Induced in Korean Soybean Cotyledons Treated with Sonication

  • The interaction between Agrobacterium and soybean has been studied at the transcriptome level but not at the metabolic level. However, it is necessary to investigate the difference in metabolites between susceptible and non-susceptible cultivars for high efficiency transformation. We investigated the difference in metabolites from sonicated soybean cotyledons of Korean cultivars and Bert cultivar. To identify difference in metabolites, sonicated extracts were analysed by Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR/MS). The soybean cultivars were classified by susceptibility using green fluorescent protein expression. We found a difference in metabolites between the high susceptible and low susceptible cultivars. The FT-ICR/MS experimental m/z data of different metabolites were compared with theoretical m/z in KNApSAcK database. The candidate list was made using KNApSAcK and focused on phenolic compounds. These candidate metabolites are speculated to influence factors in the interaction. This list of candidates may be useful to investigate the interaction between Agrobacterium and plants to increase transformation efficiency.

Natural Photodynamic Activity of 5-Aminolevulinic Acid Produced by E. coli Overexpressing ALA Synthase from Bradyrhizobium japonicum

  • The present study was conducted to determine plant growth and physiological responses of corn, barnyardgrass, and soybean to ALA (5-aminolevulinic acid). ALA effect on early seedling growth of test plants was greatly concentration dependant, suggesting that it inhibits at higher concentrations. No significant difference in herbicidal activity of two types of ALA on plant height and weight of test plants was observed. Barnyardgrass was the most sensitive to ALA and followed by corn and soybean, indicating that both crop plants were less affected by ALA concentration as well as different growth stages than barnyardgrass. Greatly reduced chlorophyll contents from leaves of three plant species were observed with increasing of ALA concentration. Compared with untreated controls, higher amounts of three tetrapyrroles were detected from three crop plants, indicating more accumulation in ALA-treated plants. The differential selectivity among plant species would be explained with the differences in tetrapyrrole accumulating capabilities, the susceptibility of various greening groups of plant species to the accumulation of various tetrapyrroles, and their metabolism in various plant tissues. The results indicate that negative biological potential of ALA exhibited differently on plant species, and that the photodynamic herbicidal activity against susceptible plants highly correlated with the extent of tetrapyrrole accumulation by the species.

Comparison of Gene Expression Changes in Three Wheat Varieties with Different Susceptibilities to Heat Stress Using RNA-Seq Analysis

  • Wheat is highly susceptible to heat stress, which significantly reduces grain yield. In this study, we used RNA-seq technology to analyze the transcript expression at three different time-points after heat treatment in three cultivars differing in their susceptibility to heat stress: Jopum, Keumkang, and Olgeuru. A total of 11,751, 8850, and 14,711; 10,959,7946, and 14,205; and 22,895,13,060, and 19,408 differentially-expressed genes (log2 fold-change > 1 and FDR (padj) < 0.05) were identified in Jopum, Keumkang, and Olgeuru in the control vs. 6-h, in the control vs. 12-h, and in the 6-h vs. 12-h heat treatment, respectively. Functional enrichment analysis showed that the biological processes for DEGs, such as the cellular response to heat and oxidative stress-and including the removal of superoxide radicals and the positive regulation of superoxide dismutase activity-were significantly enriched among the three comparisons in all three cultivars. Furthermore, we investigated the differential expression patterns of reactive oxygen species (ROS)-scavenging enzymes, heat shock proteins, and heat-stress transcription factors using qRT-PCR to confirm the differences in gene expression among the three varieties under heat stress. This study contributes to a better understanding of the wheat heat-stress response at the early growth stage and the varietal differences in heat tolerancea.

Resistance to Anthracnose Caused by Colletotrichum acutatum in Chili Pepper(Capsicum annuum L.)

  • Pepper fruit anthracnose, caused by Colletotrichum acutatum, results in serious yield loss and affects crop quality in many Asian countries, making it a disease of economic consequence. A source resistant to C. acutatum was identified by the AVRDC within the line Capsicum chinense Jacq. PBC932. The resistant breeding line C. annuum AR is the $BC_3F_6$ generation derived from C. chinense Jacq. PBC932. The inheritance of resistance to C. acutatum was analyzed in segregating populations derived from the two crosses HN 11$\times$AR and Daepoong-cho$\times$AR. Detached mature green fruits were inoculated using microinjection method. The disease response was evaluated as the disease incidence at 7 DAI. The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_R$ populations derived from the two crosses fit significantly to a 1:3 Mendelian model. This indicates that the resistance of AR to C. acutatum is controlled by a single recessive gene.
