BACKGROUND: Castor oil and wormseed extract are important active ingredients for biopesticide, and ricinoleic acid in castor oil and three monoterpenes (ascaridole, carvacrol and p-cymene) in wormseed extract are known bioactive substances. However, their stabilities had not been studied, even though the stability was the core property for estimation of shelf-life of biopesticide. Aimed to investigate the thermal stabilities of the bioactive substances in castor oil and wormseed extracts. METHODS AND RESULTS: The contents of ricinoleic acid and three monoterpenes (ascaridole, carvacrol and p-cymene) were analyzed by gas chromatography (GC). The thermal stabilities of the bioactive substance were measured at $0^{\circ}C$, $23^{\circ}C$, $30^{\circ}C$, $40^{\circ}C$, $45^{\circ}C$ and $54^{\circ}C$ for 84 d. The half-lives of ricinoleic acid in biopesticides was ranged from 28.9 d to 57.8 d at $30^{\circ}C$, and the stability of pure castor oil were located in the range ($t_{1/2}$=46.2d for Indian product and 27.7 d for Korean product) at the same temperature. The half-lives of the total monoterpenes in biopesticides were ranged from 3.9 d to 27.7 d at $30^{\circ}C$. Among the monoterpenes, the stability ascaridole and p-cymene were decreased in acidic condition. All the bioactive substances showed similar stability on the different thermal conditions. CONCLUSION:The half-lives of most bioactive substance from castor oil and wormseed extracts were less than 100 d. To increase the stability of bioactive substance in biopesticide, stabilizing additives like antioxidant and oxygen remover should be considered to extend of the shelf-life.
Protection agriculture is the essential choice for human to increase the efficiency of limited crop production area under harsh and changeable weather boundary conditions, extend growing season, maximize the crop yields, and then increase the sustainable income of the grower. The investment costs far greenhouses as well as labor and energy costs are much higher than for conventional plant production systems, so these can only be balanced by better crop yields, higher labor productivity, and higher energy efficiency. Recently, serious problems occurred such as insecticide eggs, sanitary pads with carcinogens, radon(Rn) emitting beds in domestic. It had not been establishing the inspection system to evaluate safety and human harmfulness before occurring real accidents. This research was conducted to test preliminary experimental inspection for consideration of safety insensibility. The influence of corrosion of metals and plants was studied through aerosol type spraying of agricultural pesticide products coming into the market. These products contain primary three insecticide ingredients(Flufenoxuron, Etoxazole, Fipronil) in recent accidents. Visual examination, SEM-EDS and optical microscope were used for the analysis for corrosion effect. Results show that a lot of ingredients contain in the crop protection products, and various type of corrosion exist in the surface of metals and plants. Therefore, it is necessary to provide health warning and accurate range of use for crop protection products containing insecticide ingredients.
A specific and rapid real-time PCR assay for detecting Ralstonia solanacearum in horticultural soil and plant tissues was developed in this study. The specific primers RSF/RSR were designed based on the upstream region of the UDP-3-O-acyl-GlcNAc deacetylase gene from R. solanacearum, and a PCR product of 159 bp was amplified specifically from 28 strains of R. solanacearum, which represent all genetically diverse AluI types and all 6 biovars, but not from any other nontarget species. The detection limit of $10^2\;CFU/g$ tomato stem and horticultural soil was achieved in this real-time PCR assay. The high sensitivity and specificity observed with field samples as well as with artificially infected samples suggested that this method might be a useful tool for detection and quantification of R. solanacearum in precise forecast and diagnosis.
Total 291 isolates of Ampelomyces quisqualis were obtained from 32 species of powdery mildew fungi and a selected isolate, Ampelomyces quisqualis 94013 (AQ94013) effectively hyperparasitized 6 species of Sphaerotheca and one species of Erysiphe which cause serious damage on many important crops in Korea. Moreover, AQ94013 showed antagonistic effects against 12 major fungal plant pathogens as well. Results indicated that the present isolate is not a host specific hyperparasite and has a broad spectrum of biocontrol potential. Providentially, AQ94013 revealed resistance to a number of agrochemicals so as to be applied with the chemicals reciprocally.(중략)
In replicated field trials, the efficacy of pentachloronitrobenzene (PCNB; quintozene) for control of damping-off of ginseng seedlings was found to be affected by timing of application and formulation. Application at the time of seeding and prior to placement of straw mulch was found to provide the moat consistent level of disease control. However, decline in plant stand during the four-year production cycle resulted in most treatments providing similar levels of plant populations at harvest. Soil residues of pentachloronitrobenzene were generally highest (1 $\mu\textrm{g}$ PCNB/g soil) in those treatments that exhibited the highest levels of disease control in the seedling year. Straw contained high levels of quintozene after application. Beet seed assays with artificially-infested soils indicated that current use rates provide an amount of product suitable for high levels of disease control.

Sustainable agriculture in Korea will gain important momentum by implementing The Direct Payment Scheme for Environmentally Friendly Agriculture in 1999. The farmers who want to participate in the program will be paid 530,000won/ha directly by the Government in return for engaging sustainable agriculture. The policy scheme which is a pilot program will be restricted to environmentally regulated areas to apply such as water supply protection zone, environmental protection districts and national parks. As a consequence of the policy, the supply of transitional organic agricultural products is expected to increase significantly. However the increased supply of low input agricultural products may have difficulties to find an appropriate marketing channel which is differentiated from the general agricultural product market. Development of an open market system for mass transaction of the low imput agricultural products is necessary. Also various government programs and projects which are designed for clean environment and regional community development must be integrated into the sustainable agriculture program to have synergy effects.
Melon necrotic spot virus (MNSV) was recently identified on watermelon (Citrullus vulgaris) in Korea, displaying as large necrotic spots and vein necrosis on the leaves and stems. The average occurrence of MNSV on watermelon was found to be 30-65% in Hapcheon and Andong City, respectively. Four isolates of the virus (MNSV-HW, MNSV-AW, MNSV-YW, and MNSV-SW) obtained from watermelon plants in different areas were non-pathogenic on ten general indicator plants, including Chenopodium quinoa, while they infected systemically six varieties of Cucurbitaceae. The virus particles purified by 10-40% sucrose density gradient centrifugation had a typical ultraviolet spectrum, with a minimum at 245 nm and a maximum at 260 nm. The morphology of the virus was spherical with a diameter of 28-30 nm. Virus particles were observed scattered throughout the cytoplasm of watermelon cells, but no crystals were detected. An ELISA was conducted using antiserum against MNSV-HW; the optimum concentrations of IgG and conjugated IgG for the assay were $1{\mu}l/ml$ and a 1:8,000-1:10,000 dilutions, respectively. Antiserum against MNSV-HW could capture specifically both MNSV-MN from melon and MNSV-HW from watermelon by IC/RT-PCR, and they were effectively detected with the same specific primer to produce product of 1,172 bp. The dsRNA of MNSV-HW had the same profile (4.5, 1.8, and 1.6 kb) as that of MNSV-MN from melon. The nucleotide sequence of the coat protein of MNSV-HW gave a different phylogenetic tree, having 17.2% difference in nucleotide sequence compared with MNSV isolates from melon.
소면적 재배작물 약효, 약해시험 그룹화를 위하여 소면적 재배작물 중 국내에 재배되는 엽채류 중 종류가 많고 재배면적이 비교적 큰 국화과 및 십자화과 작물인 상추, 잎브로콜리, 치커리, 배추, 열무, 유채, 쑥갓, 우엉, 앤디브, 겨자를 선정하여 십자화과 및 국화과에 공통으로 발생하는 병해충에 대한 포장시험 및 국내외 약효 시험성적을 비교검토 하였다. 국화과 및 십자화과 소면적 재배작물에 발생하는 흰가루병, 잿빛곰팡이병, 균핵병, 노균병, 무름병, 파밤나방, 도둑나방, 무테두리진딧물, 배추흰나비, 배추좀나방, 아메리카잎굴파리에 대한 약효시험 및 자료분석 결과에서도 기주 작물은 다르더라도 병해충이 동일종이면 방제효과가 유사한 경향으로 나타나는 것으로 보아 약효시험을 생략해도 될 가능성이 높아 국내 소면적 작물을 과별로 분류하고 동일 병원균을 대상으로 약효 외삽이 가능한 작물들을 분류한 후 작물잔류 상호적용 작물 그룹과의 연계를 검토하면 약효시험 및 잔류성 시험생략 후 약해시험만으로 간략히 적용하는 방안을 마련할 수 있을 것이다.
BACKGROUND: Yuza seed by-product has been produced in a large amount from the agricultural farms in the southern area of Korea. It has been mostly abandoned after commercial process for the production of juice, jam and tea. The study on the reuse of the yuza seed by-product has received much attention as a bio-resource material for the production of active compound in agriculture. METHODS AND RESULTS: Insecticidal rhamnolipid-producing Pseudomonas sp. EP-3 was grown in mineral salt media with the yuza seed by-product at 2, 20, 50 and 100 g/L. The growth of EP-3 was accompanied by a increase in insecticidal activity against green peach aphid. The highest insecticidal activity was observed when EP-3 was grown in the medium containing 50 g/L of the seed sample, producing approximately 996 mg/L of rhamnolipid at 96 h. Palmitic acid, stearic acid, oleic acid and linoleic acid were determined as the major fatty acids of the seed sample. The EP-3 cultures grown on the fatty acid mixture extracted from the seed sample showed a aphid mortality similar to that of cultures grown on the seed sample. The EP-3 cultures grown on 50 g/L of the seed sample showed aphid mortality more than 90% under greenhouse conditions. CONCLUSION: This study suggested that the yuza seed by-product may be used as a renewable material for microbial production of rhamnolipid against green peach aphid.
