The Effects of Biofilm Care on Subgingival Bacterial Motility and Halitosis

  • Background: Oral diseases are caused by various systemic and local factors, the most closely related being the biofilm. However, the challenges involved in removing an established biofilm necessitate professional care for its removal. This study aimed to evaluate and compare the effects of professional self and professional biofilm care in healthy patients to prevent the development of periodontal diseases. Methods: Thirty-seven patients who visited the dental clinic between September 2018 and February 2019 were included in this study. Self-biofilm care was performed by routine tooth brushing and professional biofilm care was provided using the toothpick method (TPM) or the oral prophylaxis (OP) method using a rubber cup. Subgingival bacterial motility and halitosis (levels of hydrogen sulfide, $H_2S$; methyl mercaptan, $CH_3SH$; and di-methyl sulfide, $(CH_3)_2S$) were measured before, immediately after, and 5 hours after the preventive treatment in the three groups. Repeated measures analysis of variance test was performed to determine significant differences among the groups. Results: TPM was effective immediately after the prevention treatment, whereas OP was more effective after 5 hours (proximal surfaces, F=16.353, p<0.001; smooth surfaces, F=66.575, p<0.001). The three components responsible for halitosis were effectively reduced by professional biofilm care immediately after the preventive treatment; however, self-biofilm care was more effective after 5 hours ($H_2S$, F=3.564, p=0.011; $CH_3SH$, F=6.657, p<0.001; $(CH_3)_2S$, F=21.135, p<0.001). Conclusion: To prevent oral diseases, it is critical to monitor the biofilm. The dental hygienist should check the oral hygiene status and the ability of the patient to administer oral care. Professional biofilm care should be provided by assessing and treating each surface of the tooth. We hope to strengthen our professional in biofilm care through continuous clinical research.

  • 대학생은 청소년기에서 성인기로 진입하는 발달단계상의 과도기로, 아직 학생의 신분이기 때문에 경제적인 측면에서 부모에게 의존을 하는 등 주체적으로 결정할 수 있는 범위가 제한되어 있다. 하지만 스스로 공부해야 하고 직업 탐색을 통해 나아갈 길을 모색해야 하는 등의 자기 결정권과 책임 또한 높아지는 시기이다. 따라서 중 고등학생 시기와 마찬가지로 자아정체감의 혼란을 겪을 수 있다. 이때 전통적 가치를 배우고 현 사회를 조망할 수 있는 주제 분야의 책을 함께 읽고, 여러 사람들과 이야기를 나누며 인식의 범위를 확장할 수 있다면 성숙을 기할 수 있을 것이다. 이에 본 연구에서는 전국 문헌정보학과 대학생 독서회의 사례를 바탕으로, 대학생 대상의 독서회를 조직하고 운영하는데 필요한 방안들을 제안하고자 한다.

중년기 기혼남녀의 가족생활 스트레스, 가족가치관이 결혼안정성에 미치는 영향 (The Effects of Family Life Stress and Family Values on Marital Stability among Middle-Aged Couples)

  • The purpose of this study was to explore the effects of family life stress and family values on marital stability among middle-aged couples. The subjects included a total of 302 men and women aged 45-64 who had been married for more than 20 years and were living in Seoul. The collected data were statistically analyzed with the SPSS 21.0 Package. First, Cronbach's alpha was calculated to measure the reliability of the scale that assessed the variables of the research model. Then, mean and standard deviation were calculated to determine the degrees of family life stress, family values, and marital stability among the subjects. Finally, a hierarchical multiple regression analysis was performed to identify factors that affected the subjects' marital stability. The results revealed that the level of family life stress in middle-aged married men and women was slightly higher than the mean value. Of all the sub-scales of family life stress, the subjects were found to experience the most stress from factors related to financial pressure, followed by family relationships, a sense of loss within a family, and work-family compatibility. Therefore, financial problems were one of the critical stressors for middle-aged married couples. Also, for this demographic, the levels of family values and marital stability were higher than the medians. The factors affecting marital stability in middle-aged couples were identified to be family relationship stress, financial stress, subjective economic status, family values, stress related to work-familycompatibility,religion,andage.Thedegreeofmaritalstabilitywashigherinmiddle-agedcouplesa stheirstresslevelsfromfamilyrelationshipsandfinancialproblemswerelower.Moreover,thehigherthes ubjectiveeconomicstatusandthemoretraditionalthefamilyvalues,thehigherthedegreeofmaritalstabili ty.Finally,marriedcoupleswhowerereligioushadalowerlevelofstressfromwork-family compatibility, and the younger the couples, the higher the degree of marital stability.

Links between accelerated replicative cellular senescence and down-regulation of SPHK1 transcription

  • We have identified a mechanism to diminish the proliferative capacity of cells during cell expansion using human adipose-derived stromal cells (hAD-SCs) as a model of replicative senescence. hAD-SCs of high-passage numbers exhibited a reduced proliferative capacity with accelerated cellular senescence. Levels of key bioactive sphingolipids were significantly increased in these senescent hAD-SCs. Notably, the transcription of sphingosine kinase 1 (SPHK1) was down-regulated in hAD-SCs at high-passage numbers. SPHK1 knockdown as well as inhibition of its enzymatic activity impeded the proliferation of hAD-SCs, with concomitant induction of cellular senescence and accumulation of sphingolipids, as seen in high-passage cells. SPHK1 knockdown-accelerated cellular senescence was attenuated by co-treatment with sphingosine-1-phosphate and an inhibitor of ceramide synthesis, fumonisin $B_1$, but not by treatment with either one alone. Together, these results suggest that transcriptional down-regulation of SPHK1 is a critical inducer of altered sphingolipid profiles and enhances replicative senescence during multiple rounds of cell division.

Induction of pro-inflammatory cytokines by 29-kDa FN-f via cGAS/STING pathway

  • The cGAS-STING pathway plays an important role in pathogen-induced activation of the innate immune response. The 29-kDa amino-terminal fibronectin fragment (29-kDa FN-f) found predominantly in the synovial fluid of osteoarthritis (OA) patients increases the expression of catabolic factors via the toll-like receptor-2 (TLR-2) signaling pathway. In this study, we investigated whether 29-kDa FN-f induces inflammatory responses via the cyclic GMP-AMP synthase (cGAS)/stimulator of interferon gene (STING) pathway in human primary chondrocytes. The levels of cGAS and STING were elevated in OA cartilage compared with normal cartilage. Long-term treatment of chondrocytes with 29-kDa FN-f activated the cGAS/STING pathway together with the increased level of gamma-H2AX, a marker of DNA breaks. In addition, the expression of pro-inflammatory cytokines, including granulocyte-macrophage colony-stimulating factor (GM-CSF/CSF-2), granulocyte colony-stimulating factor (G-CSF/CSF-3), and type I interferon ($IFN-{\alpha}$), was increased more than 100-fold in 29-kDa FN-f-treated chondrocytes. However, knockdown of cGAS and STING suppressed 29-kDa FN-f-induced expression of GM-CSF, G-CSF, and $IFN-{\alpha}$ together with the decreased activation of TANK-binding kinase 1 (TBK1), interferon regulatory factor 3 (IRF3), and inhibitor protein ${\kappa}B{\alpha}$ ($I{\kappa}B{\alpha}$). Furthermore, NOD2 or TLR-2 knockdown suppressed the expression of GM-CSF, G-CSF, and $IFN-{\alpha}$ as well as decreased the activation of the cGAS/STING pathway in 29-kDa FN-f-treated chondrocytes. These data demonstrate that the cGAS/STING/TBK1/IRF3 pathway plays a critical role in 29-kDa FN-f-induced expression of pro-inflammatory cytokines.

Englerin A-sensing charged residues for transient receptor potential canonical 5 channel activation

  • The transient receptor potential canonical (TRPC) 5 channel, known as a nonselective cation channel, has a crucial role in calcium influx. TRPC5 has been reported to be activated by muscarinic receptor activation and extracellular pH change and inhibited by the protein kinase C pathway. Recent studies have also suggested that TRPC5 is extracellularly activated by englerin A (EA), but the mechanism remains unclear. The purpose of this study is to identify the EA-interaction sites in TRPC5 and thereby clarify the mechanism of TRPC5 activation. TRPC5 channels are over-expressed in human embryonic kidney (HEK293) cells. TRPC5 mutants were generated by site-directed mutagenesis. The whole-cell patch-clamp configuration was used to record TRPC5 currents. Western analysis was also performed to observe the expression of TRPC5 mutants. To identify the EA-interaction site in TRPC5, we first generated pore mutants. When screening the mutants with EA, we observed the EA-induced current increases of TRPC5 abolished in K554N, H594N, and E598Q mutants. The current increases of other mutants were reduced in different levels. We also examined the functional intactness of the mutants that had no effect by EA with TRPC5 agonists, such as carbachol or $GTP{\gamma}S$. Our results suggest that the three residues, Lys-554, His-594, and Glu-598, in TRPC5 might be responsible for direct interaction with EA, inducing the channel activation. We also suggest that although other pore residues are not critical, they could partly contribute to the EA-induced channel activation.

Sulfatase 1 and sulfatase 2 as novel regulators of macrophage antigen presentation and phagocytosis

  • Background: Sulfation of heparan sulfate proteoglycans (HSPGs) is critical for the binding and signaling of ligands that mediate inflammation. Extracellular 6-O-endosulfatases regulate posttranslational sulfation levels and patterns of HSPGs. In this study, extracellular 6-O-endosulfatases, sulfatase (Sulf)-1 and Sulf-2, were evaluated for their expression and function in inflammatory cells and tissues. Methods: Harvested human peripheral blood mononuclear cells were treated with phytohemagglutinin and lipopolysaccharide, and murine peritoneal macrophages were stimulated with interleukin (IL)-1β for the evaluation of Sulf-1 and Sulf-2 expression. Sulf expression in inflammatory cells was examined in the human rheumatoid arthritis (RA) synovium by immunofluorescence staining. The antigen presentation and phagocytic activities of macrophages were compared according to the expression state of Sulfs. Sulfs-knockdown macrophages and Sulfs-overexpressing macrophages were generated using small interfering RNAs and pcDNA3.1 plasmids for Sulf-1 and Sulf-2, respectively. Results: Lymphocytes and monocytes showed weak Sulf expression, which remained unaffected by IL-1β. However, peritoneal macrophages showed increased expression of Sulfs upon stimulation with IL-1β. In human RA synovium, two-colored double immunofluorescent staining of Sulfs and CD68 revealed active upregulation of Sulfs in macrophages of inflamed tissues, but not in lymphocytes of lymphoid follicles. Macrophages are professional antigen-presenting cells. The antigen presentation and phagocytic activities of macrophages were dependent on the level of Sulf expression, suppressed in Sulfs-knockdown macrophages, and enhanced in Sulfs-overexpressing macrophages. Conclusion: The results demonstrate that upregulation of Sulfs in macrophages occurs in response to inflammation, and Sulfs actively regulate the antigen presentation and phagocytic activities of macrophages as novel immune regulators.

딸기 중 침투성 및 비침투성 농약에 따른 잔류특성 연구 (Comparison of Residue Patterns for Systemic and Non-systemic Pesticides in Strawberry)

  • BACKGROUND: The healthy food trend has encouraged the consumption of natural products, including berries. This trend is expected to increase the strawberry consumption. There has been a concern about the exposure of pesticides approved for use on strawberry. In this study, the dissipation patterns of systemic and non-systemic pesticides were evaluated in strawberry under plastic-covered greenhouse conditions. METHODS AND RESULTS: Cyflumetofen and dimethomorph were applied on strawberry in the critical GAP (Good Agricultural Practices). Strawberries were harvested at 0, 1, 2, 3, 5, 7 and 10 days after final application of the pesticides. The analyses of the residual pesticides were performed by HPLC-DAD with C18 column. The limits of quantitation (LOQ) of cyflumetofen and dimethomorph were 0.04 and 0.02 mg/kg, respectively. The recovery of cyflumetofen and dimethomorph were 88.1 ~ 103.3% and 79.0 ~ 110.2% for the spiked two levels (LOQ and 10LOQ), respectively. The biological half-lives of cyflumetofen and dimethomorph werer 7.5 and 8.9 days, respectively. The dissipation rates in strawberry were calculated by the statistics method at a 95% confidence level. The distribution showed that pesticides with low log Pow were indicated by the decreased dissipation rate and pesticides with similar log Pow and low solubility also showed the decreased dissipation rate. CONCLUSION: The residues of cyflumetofen and dimethomorph in strawberry at time 0 after the final application were below the established MRL in Korea. The dissipation behavior of systemic and non-systemic pesticides in strawberry is affected by their log Pow and water solubility values.

Ground Tracking Support Condition Effect on Orbit Determination for Korea Pathfinder Lunar Orbiter (KPLO) in Lunar Orbit

  • The ground tracking support is a critical factor for the navigation performance of spacecraft orbiting around the Moon. Because of the tracking limit of antennas, only a small number of facilities can support lunar missions. Therefore, case studies for various ground tracking support conditions are needed for lunar missions on the stage of preliminary mission analysis. This study analyzes the ground supporting condition effect on orbit determination (OD) of Korea Pathfinder Lunar Orbiter (KPLO) in the lunar orbit. For the assumption of ground support conditions, daily tracking frequency, cut-off angle for low elevation, tracking measurement accuracy, and tracking failure situations were considered. Two antennas of deep space network (DSN) and Korea Deep Space Antenna (KDSA) are utilized for various tracking conditions configuration. For the investigation of the daily tracking frequency effect, three cases (full support, DSN 4 pass/day and KDSA 4 pass/day, and DSN 2 pass/day and KDSA 2 pass/day) are prepared. For the elevation cut-off angle effect, two situations, which are 5 deg and 10 deg, are assumed. Three cases (0%, 30%, and 50% of degradation) were considered for the tracking measurement accuracy effect. Three cases such as no missing, 1-day KDSA missing, and 2-day KDSA missing are assumed for tracking failure effect. For OD, a sequential estimation algorithm was used, and for the OD performance evaluation, position uncertainty, position differences between true and estimated orbits, and orbit overlap precision according to various ground supporting conditions were investigated. Orbit prediction accuracy variations due to ground tracking conditions were also demonstrated. This study provides a guideline for selecting ground tracking support levels and preparing a backup plan for the KPLO lunar mission phase.

정보보호 전문인력의 경력에 따른 지식 및 기술 수요 특성 (Considering Information Security Professionals' Career to Analyze Knowledge and Skills Requirements)

  • 정보보호에 대한 인식이 제고되면서 정보보호 인력의 효율적인 양성 및 관리에 대한 다양한 연구가 수행되었다. 효율적인 인력양성을 위해서는 정보보호 인력이 직무수행을 위해 갖추어야 할 지식 및 기술 요구사항을 파악하여 교육프로그램의 개발 및 운영에 반영하는 것이 필요하다. 본 연구에서는 문헌고찰 및 델파이 방법을 통하여 정보보호 분야의 71개 요소 지식 및 기술을 도출하고 정보보호 전문인력을 대상으로 요소 지식 및 기술에 대한 필요정도 및 숙련정도를 조사하여, 정보보호 전문인력의 업무 경력에 따라 상대적으로 중요한 지식 및 기술을 도출하였다. 본 연구의 결과는 정보보호 전문인력의 경력 개발 단계별로 신규 교육 및 재교육이 필요한 요소 지식 및 기술을 제시함으로써, 산업체 및 교육기관에서 정보보호 교육 프로그램을 개발하고 운영할 때 활용될 수 있을 것으로 기대된다.
