경남 고성만 굴양식장의 위생학적 특성 (Sanitary characteristics of Seawater and Oyster (Crassostrea gigas) in Goseong Bay, Korea)

  • 고성만의 지속적이고 안정적인 굴을 생산하기 위한 방안을 모색하고자 2008년 7월부터 2009년 4월까지 위생학적인 조사를 실시하여, 현재 굴양식장의 환경 수준을 평가했다. 해수의 위생세균은 고성읍에 가까운 만의 북측에서 상대적으로 높게 출현하였으나, 미국 NSSP 기준치를 충족하는 청정한 해역 수준에 있었다. 양식 굴에서도 위생세균과 중금속 함량도 각각 미국 NSSP 및 식품공전에서 제시하는 기준치 이내로 나타나 위생학적으로 안전한 위치에 있는 것으로 나타났다. 이상의 결과로 볼 때, 경남 고성만의 굴양식장 해역은 여름철 큰 강우 시기를 제외하고는 해수와 양식굴의 분변계대장균군의 기준치를 충족하였고, 고성만 주변 유역의 오염부하가 늘어날 큰 위협요인은 없는 것으로 판단되지만, 해역의 안전성확보를 위해서는 지속적인 모니터링이 필요할 것으로 생각된다.

  • We explored the efficiency of natural antibacterial agents used to enhance the self-life and sensory properties of shucked and packed Pacific oyster Crassostrea gigas, which are in high demanded. First, we screened natural resources exhibiting antibacterial activity against food spoilage and pathogenic bacteria. Of theses, ignited oyster shell powder (IOS) and the natural food preservative, lactic acid bacteria fermented powder (LBF), were selected for further study considering the efficacy, mass production, and cost. The addition of 0.1% IOS (W/V) and 0.5% LBF (W/V) to shucked and packed oyster optimally extended the shelf-life without affecting the sensory evaluation. The results obtained in this study will provide a clue to enhance self-life in raw oyster products.

개체동결 굴(Crassostrea gigas) 엑스분을 이용한 굴 풍미계 과립조미료의 제조 및 품질특성 (Processings and Quality Characteristics of the Oyster Crassostrea gigas Granular Flavor Seasoning from IQF Oyster Extract)

  • The pacific oyster Crassostrea gigas has a desirable taste and flavor that differs from those of other fish and shellfish. In order to develop a high value-added product from individually quick-frozen oyster extract (IQFOE), we prepared an oyster granular flavor seasoning (OGS) from IQFOE and characterized its qualities. The OGS was prepared by granular molding and fluidized bed drying with inosine monophosphate (IMP, 0.1%), yeast extract powder (1.4%), tangle extract powder (0.6%), monosodium glutamate (MSG, 5.0%), microcrystalline cellulose (0.6%), lactose (27.5%), salt (33.0%), spray-dried IQFOE (22.5%) as a powdered materials, and IQFOE ($Brix\;25^{\circ}$, 7.0%), soy sauce (0.4%) and water (1.7%) as a liquid materials. The moisture, crude protein, pH and salinity of the OGS were 3.4%, 12.5%, 6.50 and 32.0%, respectively. Especially, the OGS revealed very higher amino-N content (1,856.0 mg/100 g) than that (1,291.2-1,610.2 mg/100 g) of other commercial flavor seasonings. In taste-active compounds, free amino acid contents was 1,359.0 mg/100 g, and major ones were glutamic acid, taurine, hydroxyproline, glycine, lysine, phosphoserine, proline in order. And OGS showed good organoleptic qualities for taste, odor and general preference compared with commercial flavor seasonings on a local market.

가막만 양식 굴, Crassostrea gigas의 산란에 따른 체조성과 생존율 변화 (Variation of Body Composition and Survival Rate According to Spawning of Pacific Oyster, (Crassostrea gigas) in Gamak Bay)

  • In 2007, 43.5% mortality of the cultured oyster population occurred in Gamak Bay. Mortality rapidly increase in September and peak in October. To prevent future mass-mortality event, we investigated spawning and variation of oyster body composition. The main spawning period of culture oyster occurred from August to September. Condition index and body composition (protein and glycogen) appeared to be influenced by the spawning activity. Condition index and glycogen content in September were lowest (13.5% and 5.6 mg/g, respectively). However, protein, lipid and glycogen contents did not rapidly recover after the spawning activity. The data indicates that mass-mortality of cultured oysters in Gamak Bay may be due to deteriorated health, spawning activity, stress of the high water temperature and decreasing food resources.

The Effects of PAHs (Polynuclear Aromatic Hydrocarbons) Exposure on Fertilization and Larval development of the Pacific Oyster, Crassostrea gigas

  • To evaluate the effect of PAHs on fertilization and larval development of the Pacific oyster, Crassostrea gigas via in vivo exposure for 30 days. Conditioned C. gigas adult brood stock was taken from oyster farm in May 2003 and raised in flow through tank at 20-22$^{\circ}C$ with feeding equivalently mixed marine microalgae: Chaetoceros simplex, C. gracilis, Isochrysis galbana and Tetrasemis tetrathele. The oysters were exposed to PAHs cocktail at 200ppb (10 species) until they are fully riped with an untreated group. Motility of sperm, Fertilization and The percent of D-shaped larvae was measured under microscope (Olympus BX-50). Fertilization capability using dry sperm and eggs from both non- and PAHs treated brood stocks was significantly reduced at the treated group (P<0.01). In addition to the origin effect, it was obvious that fertilization of eggs from both group were adversely suppressed in consistent with PAHs level (P<0.01). The percent of D-shaped larvae in treated group was significantly reduced comparing to non-treated. The percent was adversely correlated with PAHs level, which was more significantly reduced in treated group. Therefore, it was obvious that PAHs exposure on the Pacific oyster has adverse affects on the sperm as well as larvae in both maternal and acute source.

Regulation of adductor muscle growth by the IGF-1/AKT pathway in the triploid Pacific oyster, Crassostrea gigas

  • We investigated the insulin-like growth factor 1 (IGF-1)/AKT signaling pathway involved in muscle formation, growth, and movement in the adductor muscle of triploid Pacific oyster, Crassostrea gigas. Large and small triploid oysters (LTs and STs) cultured under identical conditions were screened, and the signaling pathways of individuals with superior growth were compared and analyzed. mRNA and protein expression levels of actin, troponin, tropomyosin, and myosin, proteins important in muscle formation, were higher in LTs compared with STs. Expression levels of IGF-1, IGF binding protein (IGFBP), and IGFBP complex acid-labile subunit were also higher in LTs compared with STs. Phosphorylation of the IGF receptor as well as that of AKT was high in LTs. In addition, the expression of phosphomammalian target of rapamycin and phospho-glycogen synthase kinase $3{\beta}$ was increased and the expression of Forkhead box O3 was decreased in LTs. Therefore, we suggested that the IGF-1/AKT signaling pathway affects the formation, growth, and movement of the adductor muscle in triploid oysters.

원유의 WSF (water soluble fraction)가 참굴의 초기생활사에 미치는 영향 (Effect of Crude Oil (WSF) on Early Life Stage of the Pacific Oyster, Crassostrea gigas)

  • The observed the toxic effect of WSF (Water Soluble Fraction) on development time, development rate, attachment rate, survival rate and growth of the larva during the early stage of the Pacific oyster, Crassostrea gigas. The time required early juvenile from the fertilized eggs exposed to crude oil (WSF) was about 13 hours in control group. But that of the crude oil dose group takes 12 hours, with shorter compared to control group. Development and attachment rate of the oyster significantly higher in 0.4 and 0.8 mg/L WSF compared to control group (P<0.05) but significantly lower more than 1.6 mg/L WSF (P<0.05). Survival rate of the oyster larva significantly higher in 0.4 and 0.8 mg/L WSF compared to control group (P<0.05) but significantly lower more than 2.4 mg/L WSF (P<0.05). Survival rate of spot showed no significantly difference compared to control group (P>0.05). Shell growth of the oyster were significantly lower more than 1.6 mg/L WSF compared control group (P<0.05). The effect concentration of crude oil showed that more 0.4 mg/L in development and attachment rate and more 1.6 mg/L in growth. Also, chronic exposure of crude oil even effect also in low concentration.

굴, Crassostrea gigas의 대사율에 미치는 수온 및 개체크기의 영향 (Effect of Temperature and Body Size on Oxygen Consumption and Ammonia Excretion of Oyster, Crassostrea gigas)

  • 수온 및 염분에 따른 굴의 일반적인 대사경향을 알아보기 위하여 수온과 개체크기에 따른 산소소비율과 암모니아 질소 배설률을 측정하였으며, O:N원자비를 산출하였다. 산소소비율과 암모니아 질소배설률은 개체의 크기가 작을수록 높았으며, 수온증가에 따라 증가하였다. O:N 원자비는 일반해수에서 8-40의 범위에 있었으며, 수온 $25^{\circ}C$에서 O:N 원자비는 8로 감소하였는데, 이는 산란기에 주요 대사기질로서 단백질을 이용하며, 단백질의 요구가 큰 것으로 추정된다. 이 결과는 굴양식장의 지속적인 관리 및 적정 수용력 산정을 위한 기초자료로 활용이 가능하다.

  • 온도, pH및 염도 stress가 굴(Crassostrea gigas)의 malate dehydrogenase isozyme에 미치는 영향을 전기영동법으로 분석하였다. 대조군의 MDH isozyme은 2개의 band가 양극 쪽으로 이동되어 분리되었다. 각 실험 온도와 pH를 12시간 노출시킨 실험군에서는 1개의 band만 양극 쪽에서 분리되었고, 24시간과 48시간 노출한 실험군에서는 2개의 band가 모두 양극 쪽에서 분리되었다. 염분 농도가 5ppt 와 3ppt에서 12시간 노출시켰을 때 각각 2개의 band가, 10ppt와 40ppt에서는 각각 3개의 band가 분리되었고, 24시간과 48시간 노출 실험군에서는 2개의 band가 양극 쪽에서 분리되었다. 모든 isozyme pattern에서 각 band의 활성도는 실험 조건에 따라 각각의 특이성이 나타났다. 결과적으로 MDH isozyme의 변화는 굴이 환경 stress에 대한 생화학적 반응의 결과로 간주된다.

2011년 11월 고성만 굴(Crassostrea gigas) 양식장 수질환경 모니터링을 통한 이산화탄소 수지 평가 (Evaluation of Simple CO2 Budget with Environmental Monitoring at an Oyster Crassostrea gigas Farm in Goseong Bay, South Coast of Korea in November 2011)

  • Real-time monitoring for environmental factors (temperature, salinity, chlorophyll-a, etc.) and fugacity of carbon dioxide ($fCO_2$) was conducted at an oyster Crassostrea gigas farm in Goseong Bay, south coast of Korea during 2-4th of November, 2011. Surface temperature and salinity were ranged from $17.9-18.7^{\circ}C$ and 32.7-33.8, respectively, with daily and inter-daily variations due to tidal currents. Surface $fCO_2$ showed a range of $390-510{\mu}atm$ and was higher than air $CO_2$ during the study period. Surface temperature, salinity and $fCO_2$ are showed significant correlations with chl.-a and nutrients, respectively. It means when chl.-a value is high in surface water of the oyster farm, active biological production consume $CO_2$ and nutrients from environments and produce oxygen, suggesting a tight feedback between biological processes and environmental reaction. Thus, factors affecting the surface $fCO_2$ were evaluated using a simple mass balance. Temperature and biological productions by phytoplankton are the main factors for $CO_2$ drawdown from afternoon to early night, while biological respiration increases seawater $CO_2$ at night. Air-sea exchange fraction acts as a $CO_2$ decreasing gear during the study period and is much effective when the wind speed is higher than $2-3m\;s^{-1}$. Future studies about organic carbon and biological production/respiration are required for evaluating the roles of oyster farms on carbon sink and coastal carbon cycle.
