Adventitious Bud Induction and Plant Regeneration from Cotyledon Explants of Camellia japonica L. (동백 자엽절편으로부터 부정아 형성을 통한 식물체의 재분화)

  • Culture conditions for plant regeneration of Camellia japonica were achieved by organogenesis in explants of cotyledon. Seed cotyledons were cultured on MS medium containing various auxin, 2,4-D or NAA and cytokinins BA. The adventitious shoot buds were efficiently formed without embryogenesis on the basal region of cotyledon cultured on MS medium supplemented with $0.1\;{\mu}g/m{\ell}$ 2.4-D and $1\;{\mu}g/m{\ell}$ BA. Seed cotyledons could be used as a source of explants in experiments of genetic transformation of the genotypes evaluated for improving the efficiency of production of transgenic Camellia plants.

Analysis of the Change of Amino Acids by Cadmium and Polyamine-Treatment in Spring Radish Young Cotyledons and Roots (무우 유식물의 자엽과 뿌리에서 Cadmium 이온과 Polyamine 처리에 의한 아미노산 변화의 분석)

  • The change of amino acids was analysed with cotyledons and roots of young spring radish by the treatment of $Cd^{2+}$ ion and $Cd^{2+}$ ion plus PA. The concentration of Asp was decreased by the treatment of $Cd^{2+}$ ion, but Ala, Phe, Val, Ile, Typ, Lys and Arg was increased in the cotyledons. The concentration of Lys and Arg was decreased by the treatment of $Cd^{2+}$ ion plus PA at the same time. The concentration of basic amino acids, His, Lys and Arg was increased by the treatment of $Cd^{2+}$ ion, and decreased by the treatment of $Cd^{2+}$ ion and $Cd^{2+}$ ion plus PA in the roots. Only the concentration of Pro was increased by the treatment of $Cd^{2+}$ ion plus PA. This results showed that Pro was induced against stress of PA, and assumed that the change of other amino acids concentration may be relation to the metabolism against stress.

  • The concentration of free PAs in the cells was decreased gradually from $500{\mu}mole$ to $290{\mu}mole$ for PUT, from $153{\mu}mole$ to $79{\mu}mole$ for SPD, from $69{\mu}mole$ to $20{\mu}mole$ for SPM during the ABA treatment within 1 hour, and was slightly changed with the increasing or decreasing between $290{\mu}mole{\sim}220{\mu}mole$ in the constant content level of PUT during the continuously ABA treatment of 2 days in the young cotyledons. The concentration of free PUT was gradually decreased from $160{\mu}mole$ to $9{\mu}mole$ during the ABA treatment within 1 hour like as cotyledons, and decreased from $9{\mu}mole$ to $5{\mu}mole$ during the continuously ABA treatment 2 days in the young hypocotyls. PUT and other PA were existed $5{\mu}mole$ during the continuously ABA treatment of 2 days in the hypocotyls. It showed that during the ABA treatment was decreased all PA concentration in the cells and PAs were not concerned directly to stress, but might regulated physiological change against stress.

  • Cadmium was transported through the special carrier system into the cells of cotyledons, hypocotyls, and roots spring radish young plants. The transport of cadmium was inhibited by metabolic inhibitor, like DNP. The $K_m$ values for cadmium were 0.7 ppm for cotyledons, 1.72 ppm for hypocotyls, and 0.3 ppm for roots, and $V_{max}$ for cadmium was $40ppm{\cdot}h^{-1}{\cdot}g{\cdot}fresh\;weight^{-1}$ for cotyledons, $313ppm{\cdot}h^{-1}{\cdot}g{\cdot}fresh\;weight^{-1}$ for hypocotyls, and $606ppm{\cdot}h^{-1}{\cdot}g{\cdot}fresh\;weight^{-1}$ for roots. Cadmium cannot prove to be inducer for proline accumulation. Therefore, proline accumulation cannot be used as a marker to test the level of heavy metal pollution.

Effect of Blue, Red and Far-red Lights on Seeding Growth and Cotyledon Chlorophyll Content of Lagenaria siceraria Standl (광질처리에 따른 박 유묘의 생장 및 자엽의 엽록소 함량 변화)

  • Various types of the seeding of bottle gourd widely used as a rootstock of watermelon has been required to satisfy the farmers need. The study was done to determine the effect of light quality of blue, red and far-red lights treated with light emitting diodes on growth and morphology of bottle goured seeding and chlorophyll content of its cotyledons. The lights were treated in the growth chamber for 7 days to the seeding elapsed 8 days after sowing under natural condition, and 64 hole trays with commercial bedsoil. Plant height, length and diameter of hypocotyl, leaf area of cotyledon and first true leaf, its leaf length, number of true leaves, fraction and total dry weight were measured. Red light shortened and slenderized the hypocotyl, which lengthened by far-red light and thickened by blue light. Plant height was declined in order of far-red light treatment, blue and red lights mainly due to difference of hypocotyl length, Area and length of the first true leaf became smaller and shorter under far-red light than under the other lights. However, blue light increased leaf area of cotyledons. Two cultivars cv. Yongjadaemok and cv. Kunghap had different response to the light treatments in total seedling dry weight(dw); far-red and red light treatments showed the greatest and the least dw of hypocotyl, respectively, while blue and red lights did the greatest dw of the other organs. Among the ratio of each organ dw to total dw, those of hypocotyl and true leaves were different between the light treatments; the highest ratio of hypocotyl dw to total dw was observed in far-red light treatment but the lowest was in red light treatment. Those of the true leaves were the lowest in far-red light and similar response in blue or red light treatment. Chlorophyll content of cotyledons was decreased in order of red light treatment, blue and far-red lights, meaning that short period light treatment may influence photosynthesis of seeding and afterward its growth.

  • The contents of sucrose and reducing sugars of cotyledons and hypocotyls of Ricinus communis L were increased slightly during cold treatment at $4^{\circ}C$. The concentration of total amino acids was increased continuosly during cold treatment. But the contents of hydrophilic amino acids, Asp/Asn, Glu/Gln, Thr, Ser, Ala and cationic amino acids, Arg and Lys were varied dramatically with the cold treated time. The cold treatment induced 24, 52, 54, 55, 56 and 73.5kD of proteins in cotyledons and 55, 56 and 73.5kD of proteins in hypocotyls. 24, 42, 49 and 52kD of proteins in cotyledons and hypocotyls were boiling stable. They were not denatrated by boiling at $100^{\circ}C$.

Screening of Pumpkin (Cucurbita spp.) Germplasm for Resistance to Powdery Mildew at Various Stages of Seedlings Growth

  • Powdery mildew (Podosphaera xanthii) causes severe damage to pumpkin crops grown in open fields and plastic house. Initially, we used ten accessions of pumpkin species; Cucurbita pepo (3), C. maxima (2), C. moschata (2), and C. argyrosperma (3) to study the disease progress in different stages of seedling development. Commercial pumpkin varieties were used as susceptible host for inoculum management and young seedlings were inoculated by dusting powdery mildew spores on the cotyledons, 1-true-leaf and 2-true-leaf seedling stages. Based on initial study, we further evaluated 218 pumpkinaccessions for their resistance to powdery mildew in different seedling stages under plastic house. Area under disease progress curve (AUDPC) and relative AUDPC (rAUDPC) was higher in cotyledonary and 1-true-leaf than 2-true-leaf stage. Seedlings at cotyledons and 1-true-leaf seedling stage displayed more susceptibility to powdery mildew. Based on evaluation of 2-true-leaf stage, IT 110859 and IT 278459 from C. pepo and C. argyrosperma identified as resistant (<0.2). Of the 228 pumpkin accessions, 21 (9.2%) pumpkin accessions consisting of C. pepo (2), C. maxima (5), C. moschata (13), and C. argyrosperma (1) exhibited intermediate resistance (<0.4) to powdery mildew and these accessions are useful to growers for its rational management.
