Differentiation of the Vascular System in Mature Embryo and Seedling of Prunus davidiana FR. (산복사나무(Prunus davidiana FR.)의 성숙배와 유식물에 있어서 유관속계의 분화)

  • To examine the differentiation and transition of vascular system in Prunus davidiana FR., the mature embyro and developing seedlings were embedded in paraplast and treated by clearing method. In mature embryo, the procambium was connected with the epicotyl-hypocotyl-radicle axis and cotyledons, whereas protophloem and protoxylem were restricted primarily to the mid-vein and two lateral veins of the cotyledonary base. With the onset of germination, protophloem and protoxylem were differentiated both acropetally and basipetally from the cotyledonary base. The first and second leaf traces appeared in the cotyledonary node, and then differentiated bidirectionally toward the epicotyl and the root tip. The 3rd to 6th leaf traces were connected with the cotyledonary traces in hypocotyl. At the part of the root tip, the xylem was a diarch. As the first and second leaf traces were superimposed at the middle part of the root, the diarch xylem was changed to a tetrarch. As the cotyledonary traces were diverged below the root base, the tetrarch xylem was changed to an octarch. It was suggested that the vascular system of the epicotyl might be superimposed on that of the cotyledon-hypocotyl-root during the formation of the primary vascular system of Prunus davidiana.

Origin of the Vascular Combium in the Developing Hypocotyl of Glycine max Seedling (대두 유식물의 배축에 있어서 유관속형성층의 기원)

  • The pattern of elongation in the developing hypocotyl of Glycine max shows that the elongation generally proceeds from base to the cotyledonary node in acropetal diredtion, although earlier elongation takes place through the entire hypocotyl. Because the differentiation of the vascular cambium in the hypocotyl advances also acropetally, it can be seen that the acropetal wave of hypocotyl elongation is associated with the acropetal differentiation of the cambium in the hypocotyl elongation is associated with the acropetal differentiation of the cambium in the hypocotyl elongation is associated with the acropetal differentiation of the cambium in the hypocotyl. The elongation of procambial cells occurs not only during active elongation but also after cessation of elongation of the hypocotyl. In tangential view, the procambium of the hypocotyl in early stage has homogeneous structure composed of short cells. Subsequently, these procambial cells elongate actively and then become elongated long cells. These long cells eventually become fusiform initials, while some of elongated long cells are transversely divided and then converted into ray initials. The characteristics of the vascular cambium are entirely acquired some time after hypocotyl elongation is completed, and the transitin from procambium to vascular cambium in the hypocotyl is a rather gradual process.

Vascular Transition and Hypocotyl Elongation in Soybean(Glycin max) Seedlings (대두(Glycine max) 유식물에서 하배축의 신장과 유관속전이)

  • The relationship between elongation of hypocotyl and its vascular transition was studied with seedling of Glycine max. the hypocotyl elongation proceeded acropetally from the base of hypocotyl toward the cotyledonary node. The vascular transition did not occur in the basal region of the hypocotyl, which did not nearly elongate, with exarch radial vascular bundles. However, the vascular transition was almost completed at the middle part of the hypocoty, more or less elongated, with endarch collateral vascular bundles. Such bundles also appeared in the uppermost region of the hypocotyl, in which the elongation was the most striking. These results suggested that the vascular transition was related to the hypocotyl elongation and that the transition of primary vascular system in Glycine max seedling was established by rather rapid process. Our observations of the serial sections from root to hypocotyl revealed that the vascular system through the root-hypocotyl-cotyledon was a unit, to which one of the epicotyl that did not participate in transition was superimposed.

  • To study the capacity of callus and shoot formation on seedling stage in soybean, excised hypocotyl, epicotyl, shoot tip, cotyledonary node and primary leaf were cultured on artificial media (MS and B$_{5}$ medium) supplemented with several hormones. Regeneration of shoots was fairly successful from shoot tip and cotyledonary node tissues in soybean. These shoots could be rooted in vitro through tissue culture technique and transplanted normally into soil. Hypocotyl and epicotyl tissues formed only callus, of which growth and appearance were different according to the kinds of media and additives. A small number of shoots were formed from primary leaf tissues, but they did not develop further.r.

  • Cotyledonary and hypocotyl explants of melon seedlings were cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) and benzyladenine (B.A).Up to 22% of cotyledonary explants and 7%, of hypocotyl explants, respectively: Produced somatic embryos through intervening two types of calli: bright yellow compact (BYC) callus and pale-yellow compact (PYC) callus. BYC callus was capable of producing somatic embryos at initial culture, but it became necrotic as subrulhues proceeded. In contrast UC callus was incapable of producing somatic embryos during initial culture (first 6 weeks), but it became bright-yellow friable (BYF) callus with forming a few globular embryos after 2 months of subculture, indicating that the callus turned embryogenic. The embryogenic capacity of BYF maintained for over one year when the callus was sucultured at 4-week interval. Upon transfer onto MS basal medium the callus gave rise to numerous somatic embryos and subsequently converted to plantlets. Plantlets were transplanted to potting soil and grown to maturity in the phyotron.

  • To establish high frequency shoot formation from two cultivars (Cheongyangjaerae and Myungan) of boxthorn (Lycium chinense Mill.), hypocotyl segments with cotyledons from seedlings were used as explants. High frequency adventitious shoot formation (more than 80%) were obtained from hypocotyl segments with cotyledon on MS medium supplemented with 1.0 mg/L zeatin, when precultured for 3 weeks under dark conditions followed by transfer to light conditions. But there was no shoot induction in the explants cultured without preculture under dark conditions. Roots were induced from the shoots when transferred to rooting medium supplemented with 1.0 mg/L IAA for 4 weeks. Regenerated plantlets were grown to normal mature plants in soil.

Structural Characteristics of Vascular Tissue in Carrot Seedlings with Anomalous Cotyledon Developed from Somatic Embryos (당근의 체세포배로부터 발생한 이상자엽 유식물의 유관속 조직의 구조적 특성)

  • The somatic embryos of Daucus carota L. cv. Hongshim developed in MS basal medium far 4 weeks had varied number of cotyledons. Palisade and spongy parenchyma of cotyledon were not clearly discriminated in all seedlings developed from the embryos. No independent existence of collateral vascular bundle was observed in all seedlings with various types of cotyledon ; instead, vascular bundles were either interconnected or partially connected with one another. Most of the cotyledonary bases on hypocotyl showed short cylinder structure which encircle plumule. The vascular tissues of cotyledonary bases and nodes of seedlings with jar-shaped or 1 cotyledon were connected in ring forms, showing the pattern of ectophloic shiphonostele, and similar ring form structure was also found in the vascular arrangement of 5 cotyledon seedlings. The vascular bundles of seedlings with 2, 3 and 4 cotyledons in many cases had independently arranged within the cotyledonary bases and nodes, showing the pattern of eustele. In hypocotyl, tetrarch or hexarch xylems prevailed in seedlings with jar-shaped cotyledon or 1 and 5 cotyledon; tetrarch xylems prevailed in 2 cotyledon seedlings; and triarch xylems prevailed in 3 cotyledon seedlings. In most of seedlings, cortex vascular bundles were dispensed in the region from cotyledonary node to hypocotyl, but double vascular bundles were also observed occasionally. In roots, diarch xylems were observed in most of seedlings with 2 cotyledons, triarch xylems in half of seedlings with 3 cotyledons, and diarch xylems in most of the remaining seedlings with the occasional occurrences of tetrarch xylems.

Procambium differentiation and shoot apical meristem development in somatic embryos of soybean (Glycine max L.) (대두 체세포배에서 전형성층 분화와 경단분열조직의 발달)

  • Immature embryos of Glycine max L. was cultured on Murashige and Skoog's (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D). After 6 to 8 weeks of culture, immature embryos produced somatic embryos. Of somatic embryos, two cotyledonary embryo (14%), one cotyledonary embryo (37%), fused cotyledonary embryo (43%), and stunted globular embryos (6%) were observed. The procambial strand of cotyledons originated from circular procambial tissues of lower hypocotyl. The circular procambial tissues were independently divided into one or two procambial strand at the edge of cotyledonary-node, and then connected to each cotyledon to form somatic embryos with one or two cotyledons. When cotyledon was a fused type, the circular procambial strand in lower hypocotyl was continuously connected to the cotyledon. Also, somatic embryos with two cotyledons developed a functional shoot apex with the tunica-corpus structure. In contrast, somatic embryos with one or fused cotyledon formed an abnormal shoot apex without the tunica-corpus structure or with non-dome shape in the inter-cotyledonary area. These results indicated that the variation of cotyledon in somatic embryos is closely related to procambial differentiation and shoot apical meristem development.

High Frequency Organogenesis and Plant Regeneration in Tissue Cultures of Lettuce Seedling Explants (상추 유식물체 절편의 조직배양에 의한 고빈도 기관발생과 식물체 재분화)

  • To induce adventitious buds, hypocotyl and cotyledonary explants from 7 to 10 day-old seedlings of lettuce (Lactuca sativa L.: two Japanese cultivars of crisphead lettuce and four Korean cultivars of leaf lettuce) were cultured or Murashige and Skoog (MS) and Schenk and Hildebrandt (SH) media supplemented with BA and NAA in the light for five weeks. Cotyledonary explants produced adventitious shoots at greater frequencies than hypocotyl explants. MS medium was more favorable to adventitious shoot formation than HS medium. Combination of 0.5 mg/L BA and 0.1 to 1 mg/L HPh in MS medium led to the greatest frequency (86%) in adventitious shoot formation. Creator than 95% of shoots excised from explants were rooted when cultured on MS basal medium.

Optimization of Regeneration Condition Under Agrobacterium-mediated Transformation in in vitro Cultured Korean Soybean

  • Soybean is the important crop in Asian countries as protein source, oil production and animal feed. Improving soybean using genetic transformation is the principal tool in nowadays. Developing herbicide resistant transgenic soybean plants through Agrobacterium-mediated transformation has been worked in many previous studied. However, the transformation efficiency is still low. Many attempts try to find the optimum media condition for plant regeneration after infection. After transformation, the plant regeneration is very important condition to promote growth of transgenic plant. In this study, we optimized a regeneration condition for two Korean soybean cultivar, Dawonkong and Pungsannamulkong using cotyledon, cotyledonary nodes and hypocotyl as explant. The results showed that shoot regeneration of cotyledonary nodes on B5 medium containing 2 mg/L 6-benzylaminopurine showed the highest percentage of regeneration in Dawonkong (75.8%) while Pungsannamulkong presented high number of shoots 2.12 shoots per explant. For transformation condition, co-cultivation in 7 days showed a high number of GUS positive expression. Most of explants can survived under media including 5 mg/L of glufocinate which refers phosphinotricin for 2-week selection. Washing with 400 mg/L of cefotaxime in several times and selection in plant regeneration media with 400 mg/L of cefotaxime can prevent bacteria growth, effectively.
