Flower Rot of Cotton Rose (Hibiscus mutabilis) Caused by Choanephora cucurbitarum (Choanephora cucurbitarum 에 의한 부용 꽃썩음병)

  • In July 2001, rotting and shivering flowers of cotton rose (Hibiscus mutabitis) were fecund in the flower beds along the roadsides in Jinju area. The disease first started as water-soaking, dark-green lesions on the petals, and then whole flower was rotted rapidly, Whitish mycelia and monosporous sporangiophore with monosporous sporangiola were formed abundantly on the lesions. Colony appeared as white to pale yellowish brown mycelia on potato dextrose agar medium (PDA). Monosporous sporangiophore was long slender and branched at the apex, each branch bearing a head of sporangiospores. Sporangium was subglobose in shape and was 42.6-114.2$\mu$m in size. Monosporous sporangiola were elliptic, fusiform or ovoid, and brown in color and 12.3~21.6 $\times$8.3~11.6$\mu$m Um in size. Sporangiospores were elliptic, fusiform or ovoid in shape, dark brown or brown in color and 16.3~23.8$\times$8.2~13.6$\mu$m in size, and they had three or more appendages at bipolar end. Zygospores were mostly globose, dark black colored and sized was 46.2-78.4$\mu$min diameter, The fungus grew on PDA between at 15 to 4$0^{\circ}C$, and the optimum temperature was 3$0^{\circ}C$. This is the first report on the flower rot of cotton rose caused by C. cucurbitarum in Korea.

Agrobacterium Mediated Transformation of Rehmannia glutinosa L. with Glutathione S-Transferase Gene (Gh-5)

  • Using Agrobacterium-me야ated transformation method the auxin-regulated cotton GST (Gh-5) constructs were used to transform Rehmannia glutinosa L. The PCR analysis was conducted to verify transgenicity. Based on the PCR analysis, there was verified that the 988 bp DNA band had showed in transgenic plant genomes in PCR anaJysis using Gh5-1 and Gh5-2 primers. The effects of cocultivation with Agrobacterium tumefaciens, regeneration and selection conditions on the transformation efficiency of Chinese foxglove (Rehmannia glutinosa L.) were investigated. Factors such as cocultivation period, use of acetosyringone, postcultivation in darkness, and different kanamycin concentrations for selection were assessed. In vitro regeneration, the number of leaves, shoot lengths and numbers on MS medium were superior to on B5 and WPM medium, and the shoot formation rate was highest level of 95% in cultured base part containing leaf stalk. Addition of acetosyringone at concentration of $200{\mu}M$ to cocultivation medium and 3-day of cocultivation improved transformation frequencies. Exposure of explants to darkness for 4 weeks on selection medium resulted in further increased the regeneration frequency of transgenic shoots. In PCR analysis, the amplified fragments of Gh5 gene were detected (988 bp), and GST-expressing transgenic R. glutinosa L. plants had approximately three-fold higher activity in leaf extracts compared with control plant.

  • The objectives of this investigation were to produce artificially an antitumor constituent by submerged culture of the mycelium of Coriolus versicolor (Fr.) Quel., to characterize the influence of various modifications of the nutrient and culture conditions with respect to the pro­duction, to determine chemical composition of the antitumor constituent, and to examine effects of the constituent on the immune response of mice. Submerged agitation of the mycelium in flasks containing a nutrient solution showed its adequate growth. Especially the mycelial growth in the medium containing glucose and yeast extract was abundant. The addition of cotton seed flour or ginseng waste to the medium increased the yield of mycelial growth and the production of the antitumor constituent. The replacement of glucose with starch also yielded the adequate growth. The antitumor constituent extracted from the mycelium and isolated from the culture filtrate was a protein-bound polysaccharide. The analyses of this constituent by GLC and amino acid autoanalysis showed that it contained four monosaccharides and fifteen amino acids. The protein-free polysaccharide of the constituent was also found to exert greater antitumor activity against sarcoma-180 in mice than the entire constituent. The antitumor constituent was found to potentiate the immune response of mice against sheep red blood cell. The protein-bound polysaccharide exerted more favorable influence on the immunity than the protein-free moiety.

Effect of Sonication and vir Genes on Transient Gene Expression in Agrobacterium-Mediated Transformation (Agrobacterium을 이용한 형질전환에서 sonication과 vir 유전자들의 효과)

  • Sonication tremendously improves the efficiency of Agrobacterium infection by introducing small and uniform fissures and channels throughout the targeted tissue. Using shoot tips of cotton as explants, the effect of sonication treatment and virulence genes in Agrobacterium tumefaciens on transformation efficiency was investigated. The pat gene which encodes resistance to the herbicide, glufosinate, was used as a selectable marker. Transformation efficiency was evaluated on th basis of survival rates of cocultivated shoot tips on selection medium containing 2.5 mg/l gulfosinate-ammonium(ppt) adn 25. mg/l Clavamax. Sonication from 5 to 15 second has a positive effect on shoop tip survival. However, whil virE as well as virG or vir GN54D showed an enhancement in transformation efficiency, virE,. virG resulted in the most significant enhancement. Overall, the combination of additional virG/virE gene and sonication treatment resulted in the most significant increase in transformation efficiency.

Nutritive Values of Major Feed Ingredient in Tropics - Review -

  • Majority of livestocks are kept in the tropics. Demand for animal products (meat and milk) is continuously increasing and is related to human population growth. Consequently, potential feeds should be continuously identified particularly on their nutritive values. Crop residues and agricultural by-products are the main feed sources for feeding livestock in the tropics. Their nutritive values ranging from low to medium quality level although Some agricultural by-products such as cotton seed meal and coconut meal are rich in nitrogaen contents. From literatures, nutritive values in these ingredients are mainly based on their chemical composition and to some extent based on limited number of in vivo studies. However, optimum of inclusion in the diet is suggested. Development of tree legumes should be thoroughly considered since they grow well in most tropical regions. In order to improve nutritive value of tropical feeds, biological treatments should be considered. Effect of secondary compounds decreasing efficiency of nutrient utilization in the rumen, to some extent could be reduced by introducing probiotics.

  • This study was carried to a textile conservation process included washing effect fiber analysis such as fiber identification fabric density and thickness color fading and of extracted soils. the following results were obtained. 1. AS a result of investigating to fabric surfaces by S.E.M all of cleaning methods wet cleaning-solvent cleaning in charge system were effective to remove soils from fabrics. 2. The buried fabrics were made of silk few of them were cotton ramie and hemp. 3. According to fabric density and thickness used fabrics were almost medium weight fabrics. 4. Low values of L, a, b indicated that the colors of these fabrics were faded to yellow and brown. 5. The soil components were hydrocarbon-alkane group alkyl alcohol and ketone group.

The Actual Condition and Problems of Current Shrouds (현행수의의 실태 및 문제점)

  • The purpose of this study is to analyze the actual condition and problems of current ready-made shrouds. Current shrouds were formed after the 20th century and therefore are quite apart from its traditional precedents of Chosun Dynasty. In this respect, several problems can be pointed out. 1. The use of natural colored hemp as a primary textile of current shrouds is far from our tradition. Silk, ramie, and cotton have to be used together with hemp. Also, more variety of colors can be used other than natural white. 2. The size of ready-made shrouds has to be categorized at least into three: large, medium and small 3. The pattern and sowing method of Dopo(도포) and Wonsam(원삼) should be corrected in line with those of the traditional Dopo(도포) and Wonsam(원삼). 4. The design and items of current ready-made shrouds have to break away from the standardized convention and should seek more of a variety that our rich tradition provide. 5. The rational index should be suggested so that consumers can compared the quality and price of ready-made shrouds available at their hands.

  • Following the prior studs regarding that 1.0g of cotton fabric cound be dyed uniformly with a reactive dye in the solvent mixture of 2㎖ of water and 23㎖ of dichloromethane, silk fabric was dyed with C. I. Reactive Blue 203 in the water/dichloromethane two-phase immiscible solvent media. In order to minimize dye loss due to its hydrolysis, the reactive dyeing was carried out in dichloromethane containing a small amount of water. With only 3㎖ of water in 22㎖ of dichloromethane, 1.0g of silk fabric could be dyed perfectly. The uptake ratio was increased greatly, compared with that of normal reactive dyeing in a water medium. It would seem that the one of hydrophobic solvents, dichloromethane, can assist the even dyeing as it disperses a small amount of dye-dissolved water phase and conveys this water phase to the fabric entirely and uniformly.

Antibiotic Susceptibility and Distribution of Vibrio parahaemolyticus Isolated from the Seafood (어패류에서 분리한 장염비브리오균의 분포 및 항균제 감수성)

  • Vibrio parahaemolyticus was investigated for analysis of distribution and antibiotic susceptibility at the markets, Seoul in 2004. The average isolation rate was 26.8%(306/1,143) and the highest rate with 64.9% was obtained in August. The isolation rates from December to April was 0.0%. It is more effective to inoculate into the enrichment medium, alkaline pepton water by swabbing a sample with a cotton ball rather than pouring mixed sample. The isolation rates were from 24.4% to 37.1% in domestic samples and from 9.8% to 26.8% in imported ones. All of the isolates were resistent to ampicillin and ticarcillin and most of them were sensitive to chloramphenicol, tetracycline, nalidixic acid, amoxicillin, ceftriaxone, streptomycin, kanamycin, trimethoprim/sulfamethoxazol, ampicillin/sulbactam, ciprofloxacin.
