Costunolide Induces Differentiation of Human Leukemia HL-60 Cells

  • Costunolide has been reported to be a cytotoxic and chemopreventive agent. This work investigated the mechanism of the anti proliferative effect of costunolide and determined that it induced differentiation of the human leukemia cell line HL-60. Costunolide exhibited a potent antiproliferative activity against HL-60 cells. It was also found to be a potent inducer of differentiation in human leukemia derived HL-60 cells through the examination of differentiation markers, as assessed by the reduction of nitroblue tetrazolium, the increase in esterase activities and phagocytic activity, morphology change and the expression of CD14 and CD66b surface antigens. These results, accompanied by a decline in the expression of c-myc protein, suggest that costunolide induces differentiation of human leukemia cells to granulocytes and monocytes/macrophages lineage.

Costunolide와 Dehydrocostuslactone을 이용한 목향의 함량분석 (Quantitative Analysis of Aucklandia Lappa Using Costunolide and Dehydrocostuslactone)

  • Aucklandia lappa Decne (Compositae) has been used for treatment of abdominal pain, vomiting, diarrhea, chronic inflammation, and antibacterial effect. The quality of these herbs has been affected by many factors such as collection time, place, temperature, cultivation environment and manufacturing process. We used costunolide and dehydrocostuslactone as marker compounds for quality evaluation of rhizome of Aucklandia lappa. 66 samples of Aucklandia lappa were collected from those habitats in Korea and China. The developed HPLC-DAD method was applied to investigate for quality control of Aucklandia lappa samples. The average contents of the costunolide and dehydrocostuslactone were 2.3895% and 0.9258%, respectively. The principal component analysis (PCA) exhibited that classification of Aucklandia lappa according to origin not separated. Results of this study may be satisfactory applied to determination of content criteria of Aucklandia lappa.

산국 꽃의 Germacranolides (Germacranolides from Flowers of Chrysanthemum boreale Makino)

  • Two sesquiterpene lactones were isolated from the flowers of Chrysanthemum boreale Makino by the silica gel column chromatography and recrystallization. On the basis of spectrometric studies including $^1H-NMR,\;^{13}C-NMR,\;DEPT,\;^1H-^1H\;COSY,\;{13}C-^1H\;COSY$, IR and Mass, compounds 1 and 2 were identified as germacranolide, tulipinolide and costunolide, respectively. And they showed antibacterial activity against Vibrio parahaemolyticus, Bacillus subtilis, Bacillus cereus and Staphylococcus aureus. This is the first report that Chrysanthemum boreale contained tulipinolide and costunolide.

In vitro Biological Activity of Germacranolide sesquiterpene lactones

  • Bioactivity-directed isolation has led to the isolation of (-)-ent-costunolide (1) as the major active compound from Hepatostolonophora paucistipula. This compound (1) was determined by spectroscopic data interpretation. This sesquiterpene lactone (1) inhibited the growth of the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (4 mm inhibition zone at $15{\mu}g$/disc), cytotoxic activity to murine leukaemia cell lines ATCC CCL 46 P 388D1 ($IC_{50}$ 687 ng/ml, at $0.075{\mu}g$/disk), BSC monkey kidney cell lines (100% of well at $15{\mu}g$/disk) and antiviral activity to Herpes simplex virus (0.25 mg/ml, 100% of well at $7.5{\mu}g$/disk) and Polio virus (0.125 mg/ml, 100% of well at $3.75{\mu}g$/disk). These results suggest that (-)-ent-costunolide (1) has potential antimicrobial and cytotoxic agents.

  • 함박꽃나무 수피 및 목부의 EtOH 조추출물로부터 prep. TLC, silica gel 및 Sephadex LH-20 칼럼 크로마토그라피를 이용하여 4종의 화합물을 단리하였다. 이들 화합물들은 NMR, MS 등의 기기분석에 의해 sesquiterpene lactone 구조의 costunolide (I)를 비롯하여, 리그난 배당체 화합물인 syringin (II)과 1, 2-dihydroxyxanthone (III) 및 vanillic acid (IV)으로 각각 동정하였다.

  • From the leaves of Magnolia sieboldii a new aporphine-type alkaloid named magnoporphine was isolated. The structure of magnoporphine was all assigned by $^1H-^1H$COSY, $^1H-^{13}C$ COSY and $^1H-^{13}C$ long range NMR. In addition, costunolide, syringin, syringenin $4-O-{\beta}-cellobioside$ and echinacoside was isolated.

  • Costunolide. a sesquiterpene lactone is isolated from Magnolia Sieboldi. It is known to possess antitumour and anti-inflammatory activities. This compound is synthesized from Ihe easily available decalin dione using the ring cleavage approach to construct the ten-membered ring system. The two keys points in this work are the chiral inductionon the allyl alcohol moiety using Sharpless epoxidation reaction and opening of the eopxide with an organocuprate reagent which leads to a $\alpha$-exomethylene lactone. 목향과 토목향의 성분함량과 패턴비교 연구 (Quantitative and Pattern Analysis of Aucklandia lappa Decne and Inula helenium L.)

  • Aucklandia lappa Decne (Compositae) has been used for treatment of vomiting, diarrhea, abdominal pain and indigestion and Inula helenium L. (Compositae) for sweat and urination. In spite of difference species and efficacy, we have been used Aucklandia lappa Dence together with Inula helenium L. To distinguish Aucklandia lappa Dence from Inula helenium L., we have compared the contents of dehydrocostuslactone and costunolide by HPLC. The average contents of costunolide of Aucklandia lappa Dence was 0.84% $({\pm}\;0.29)$ but was not detected in Inula helenium L. The contents of dehydrocostuslactone from Aucklandia lappa Dence and Inula helenium L. were about 1.70% $({\pm}0.37)$ and 1.82% $({\pm}0.59)$, respectively.
