충북지역의 천연 자원을 활용한 화장품 산업의 발전 전망 (Prospects for development of cosmetic industry using natural products in Chungbuk)

  • 유전 자원에 대한 접근 및 이용에 따른 이익의 공정한 공유를 표방하는 나고야 의정서가 발효되면서, 최근 국내 천연자원을 이용한 바이오 소재 및 제품 개발 연구에 많은 지원이 이뤄지고 있다. 국내 화장품 연구 및 개발 동향은 기존의 합성 화합물 중심에서 벗어나 안전성이 확보된 천연 바이오 소재 중심으로 변화하고 있다. 또한 기능성화장품의 범위가 기존에 크게 미백, 주름개선, 자외선 차단 3가지에서 염모, 탈모 완화, 여드름 완화, 아토피성 피부의 보습 등으로 세분화 및 추가됨으로써 화장품 연구 개발의 범위가 확대되었다. 이러한 기능성 화장품 범위 확대, 화장품 소재에 대한 안전성 규제 강화, 동물실험 금지 등 화장품 연구 환경 변화로 인해 다양한 천연물 기반의 생물학적 활성을 갖는 유효 성분들이 기능성 화장품 소재로 연구되는 사례가 점차 증가하고 있다. 충북은 바이오산업을 지역 특화산업으로 선정하여 바이오산업 육성을 위한 중부권의 오송생명과학단지/오창과학산업단지와 북부권 제천 바이오벨리 조성, 충주기업도시 등을 통해 바이오 제약 및 화장품 기업 유치 와 바이오 산업클러스터를 완성해 가고 있다. 이와 더불어 오송생명과학단지 내에 위치한 오송화장품임상연구지원센터는 충북 지역 중소 화장품 기업들의 글로벌 시장 진출 및 동남아 수출 지원을 위해 임상시험, 효능평가, 해외 인증, 해외시장 박람회 등의 지원을 중점적으로 수행하고 있다. 이러한 하드웨어 인프라 조성과 함께 충북의 천연 소재를 이용한 다양한 국책 연구 개발 사업을 추진하여 충북 특화자원인 황기, 오미자 뿐 아니라 두충, 익지인, 소목 등을 이용한 화장품 소재 개발 연구를 수행하고 있다. 충북지역 기업체 및 연구기관을 중심으로 천연물의 지표 및 유효 물질 원료 확보와 이를 활용한 기능성 화장품 제품화가 본격적으로 추진되고 있어 향후 충북 천연물 산업은 화장품 산업을 중심으로 더욱 성장해 나갈 것으로 기대된다.

생약재 추출물로부터 주름 개선 소재의 발굴 및 이를 이용한 화장품의 안정성 시험 (Screening of Anti-wrinkle Resource from Herbal Medicinal Extracts and Stability Test of Its Cosmetic Products)

  • Various herbal medicinal extracts were examined for the development of cosmetic products with anti-oxidative and anti-wrinkle activity. First, total polyphenol contents and DPPH radical scavenging activities of herbal medicinal extracts were measured. Most herbal samples, except for DW extracts of Portulaca oleracea, Caesalpinia sappan, Taraxacum platycarpum, Carthamus tinctorius, and 70% EtOH extracts of Taraxacum platycarpum and Carthamus tinctorius, showed DPPH radical scavenging activity over 80% at a concentration of $400{\mu}g/m{\ell}$. SOD-like antioxidant activity of DW extracts of Syzygium aromaticum, DW extracts of Eriobtrya japonica and 70% EtOH extracts of Sophora japonica was measured as 40%, 35% and 80%, respectively at a dry matter concentration of $50{\mu}g/m{\ell}$. In elastase inhibition assay, DW extracts of Lycium chinense ($50{\mu}g/m{\ell}$) and 70% EtOH extracts of Areca catechu ($50{\mu}g/m{\ell}$) showed 50% and 40% of inhibition, respectively. At a concentration of $1.250{\mu}g/m{\ell}$, DW extracts of Lycium chinense and 70% EtOH extracts of Areca catechu showed 10% and 30% of collagenase inhibition, respectively. Skin and lotion samples were prepared using the two herbal extracts of high anti-wrinkle activity: Lycium chinense extract and Areca catechu extract. The storage stability of skin and lotion containing each of the selected herbal extracts was evaluated. pH and viscosity were used as stability indicators for the stability test under different storage temperatures and freeze-thaw cycle conditions. The skin and lotion containing each of DW extract of Lycium chinense and 70% EtOH extract of Areca catechu was showed high pH and viscosity stability. The skin and lotion containing DW extracts of Lycium chinense showed relatively higher stability than the skin and lotion containing 70% EtOH extract of Areca catechu, at cycle chamber and freeze-thaw conditions. In summary, these results indicated that cosmetics containing DW extract of Lycium chinense were relatively stable, and this herbal extract could be used as a stable functional cosmetic material.

관중(Dryopteris crassirhizoma Nakai) 추출물의 Propionibacterium acnes에 대한 항균 효과 (Antimicrobial Effect of Ethanol Extract of Dryopteris crassirhizoma Nakai on Propionibacterium acnes)

  • 본 연구는 예로부터 민간요법 및 한방에 유용한 약재로 사용되어지고 있는 관중(Dryopteris crassirhizoma Nakai)을 이용하여 Propionibacterium acnes에 대한 항균활성을 평가하고, 이를 토대로 여드름 억제 및 치료에 관련된 화장품 소재를 개발하는데 그 목적이 있다. P. acnes에 대한 관중 crude 추출물 및 헥산분획물의 minimum inhibitory concentration (MIC) 및 paper disk diffusion assay로 항균 효과를 측정하였으며, 추출물에 대한 온도 및 pH 변화에 따른 안정성을 평가하였다 추출물을 이용한 P. acnes에 대한 MIC 값은 관중 crude 추출물은 0.008 mg/mL, 헥산추출물은 0.001 mg/mL로 측정되었으며, 이는 대조군으로 사용한 항생제인 triclosan의 MIC 값(0.004 mg/mL)과 비교했을 때 시료가 관중 crude 추출물 및 헥산 분획물임을 고려하면 매우 항균활성이 높은 것으로 평가되었다. 추출물에 대한 paper disk diffusion assay를 수행한 결과, 대조군으로 사용한 triclosan과 대비, 높은 활성을 나타내는 것으로 측정되었으며, HaCaT 세포에 대한 독성은 $20{\mu}g/mL$ 이하의 농도에서는 나타나지 않았다. 관중추출물의 온도, pH 안정성을 평가한 결과, 25, 70, 80, 90, 100, $121^{\circ}C$ 등 다양한 온도 조건과 $pH 2{\sim}pH 11$의 변화에 있어서도 그 효능이 변하지 않고 안정된 상태로 유지되는 것으로 관찰되었다.

선학초 추출물의 항여드름균 효능 연구 (Studies on the Anti-acne Effect of Agrimonia pilosa Ledeb.)

  • 본 연구는 우리나라의 야산이나 길가, 들판 등에 흔히 자라는 여러해살이 풀이며, 오래 전부터 민간요법 및 한방에 유용 한 약재로 사용되어 지고 있는 선학초(Agrimonia pilosa Ledeb.)를 이용하여 여드름 유발균에 대한 항균활성을 평가하고, 이를 토대로 항여드름 관련 화장품 소재를 개발하는데 그 목적이 있다. 선학초를 methanol로 추출한 후, 이 추출물을 여드름 유발균주인 P. acne를 이용하여 minimum inhibitory concentration (MIC) 및 paper disk diffusion assay로 항균 효과를 측정하였으며, 추출물에 대한 온도 및 pH 변화에 따른 안정성을 평가하였다. 추출물을 이용하여 P. acne에 대한 MIC 값은 0.005 mg/mL로 측정되었으며, 이는 대조군으로 사용한 항생제인 triclosan의 MIC 값(0.004 mg/mL)과 비교했을 때 시료가 선학초 전초에 대한 crude extract임을 고려하면 매우 항균활성이 높은 것으로 평가되었다. 추출물에 대한 paper disk diffusion assay를 수행한 결과, 대조군으로 사용한 triclosan 처럼 높은 활성을 나타내는 것으로 측정되었으며, Staphylococcus aureus를 비롯한 여러 종의 호기성 균주에도 선학초추출물은 항균활성이 우수한 것으로 평가되었다. 선학초 추출물의 온도, pH 안정성을 평가한 결과, 70, 80, 90, 100, $121^{\circ}C$ 등 다양한 온도 조건과 $pH 2{\sim}pH 11$의 변화에 있어서도 그 효능이 변하지 않고 안정된 상태로 유지되는 것으로 관찰되었다.

염증성 피부질환 개선 기능 소재로서 둥근전복(Haliotis discus) 유래 항균펩타이드 Ab4-7의 항염증 효과 (Anti-inflammatory Effects of Haliotis discus-derived Antibacterial Peptide (AMP) Ab4-7 as a Functional material for Improving Inflammatory Skin Diseases)

  • 서해안에 서식하는 해양생물인 전복은 항산화 및 항염증 효과가 있으며, 항생제 개발 및 화장품 원료 등 다양한 산업에 활용될 수 있는 잠재력을 가진 자원이다. 본 연구에서 우리는 서해안의 다양한 해양 생물 중에서 전복을 선택하였으며, 전복에서 분리한 항균펩타이드(AMP)를 이용하여 항균펩타이드 유도체인 Ab4-7를 규명하고 생리활성을 연구하였다. Ab4-7의 항염증 효능을 확인하기 위해 염증이 유도된 RAW 264.7에 Ab4-7을 처리한 결과, 염증성 사이토카인, TLR4, TNF-α, IL-1β, COX-2, iNOS 등을 억제하고 항산화와 관련된 유전인자인 HO-1의 mRNA 발현을 증가시켰다. Ab4-7의 강력한 항산화 및 항염증 효과를 바탕으로, 염증이 유도된 RAW 264.7 세포에서 Ab4-7이 extrocellular matrix metalloproteinase (ECM) 분해에 관여하여 다양한 염증성 피부질환을 유도하는 유전자인 matrix metalloproteinase (MMPs)를 조절하는 효과를 RT-PCR을 통해 확인하였다. Ab4-7은 강력한 항염증 및 항산화 효과를 가지고 있으며, 항염증 및 항산화 효과를 통한 MMPs 관련 유전인자 조절을 통해 다양한 염증성 피부질환 치료제의 기능성 재료로 활용될 수 있다고 사료된다.

The Strategy for the Development of Bio-Resources Utilizing Sericultural Products and Insects

  • Experiments related to the field of sericulture started in the years 1900, in Korea. The sericultural experimental station in Korea was first organized among agricultural fields in Korea, indicating that sericulture in Korea was regarded as an important field of agriculture. Sericulture has been devoted to a great deal for the improvement of Korean economy during the past 100 years even under the coarse social circumstances caused particularly by the Korean War, However, the traditional Korean sericulture, aimed to produce silk yarn, was weakened, because of several reasons such as diminishment in silk consumption, increased labor charge in Korea, and so on. After this difficulty time, the Korean sericulture was revolutionized by shifting into functional sericulture from 1995, and the Korean sericulture now plays an important role for the improvement of human health. Mulberry tree, silkworm, and silk have a boundless potential to be developed as resources. We expect the know-how obtained through silkworm research would expand to the other insect research too. Thus, an area of entomological industry is hoped to prosper owing to insect research as well as sericulture. Mulberry tree is known to possess many bio-active substances, so it can be utilized as a resource for substitute medicine and a raw material for the functional food. In addition, an invention of genetically engineered mulberry variety, which will produce more bioactive substances, is expected. Silkworm is one of the most extensively studied insect organisms on the genome so far, Thus, silkworm is expected to be an "insect bio-factory", enabling mass-production of useful proteins by transformation, in which useful foreign genes are assimilated into silkworm. Silk can be transformed into several phases, because it possesses useful functional groups, which are sensitive to chemical reaction. Also, because silk fibrin itself is protein, it has a superior applicability as tissue membrane. Due to this usefulness, many researchers are now working on the silk as food, cosmetic, medical resource, and bioengineering resource, and even an expanded application is expected using silk in the future. Until now, the researches on insects were largely focused on the prevention of the damage caused by pest, instead of a beneficial aspect. However, insects are thought to be the fourth natural resource in the world, possessing unlimited potential as world resources in the near future. Therefore, our entomological research effort should be focused on the subject with potential for industrialization. Such subject includes selecting the insect species useful for environmental evaluation, construction of environment-friendly agricultural ecosystem, pollen mediation, pet, and advanced bio-resources.

해양심층수 관련 국내 특허출원 동향 (Current Status of Applied Korean Patents Regarding the Deep Sea Water)

  • Deep sea water exists at depths of over 200m under the sea. As no sunlight reaches it, photosynthesis does not take place within it, and it contains no organic matter. In addition, its temperature is maintained at a stable low level throughout the year, so it does not get mixed with the sea water on the surface. It contains a large amount of nutritious salts, whose cleanness is maintained. It is a marine resource that has matured for a long period of time. Research into deep sea water, which started in the 1970s, has been made around the whole world, including the USA and Japan. In Korea, research has been active in this area since 2000. As there has been a good amount of research into industrial applications for deep sea water, since 1993, patents for the relevant technologies have been applied. This paper intends to provide a resource to researchers of deep sea water, by summarizing of all domestic deep sea water-related patents applied with Korean Intellectual Property Office from 1993 to 2008. This research was conducted using a computer and KIPRIS Database owned by the Korea Institute of Patent Information. 'Deep sea water' was used as the search keyword. A total of 222 Korean patents relating to deep sea water have been registered on the basis of IPC. Of these, 126 patents relate to the manufacturing and the treatment of foods, foodstuffs, or non-alcoholic beverages(A23L), while 50 patents relate to the production for medical, dental, or cosmetic purposes(A61K). 38 patents relate to water purification, treatment of wastewater, sewage and sludge (C02F), while 8 patents relate to fishery and farming(A01K). In summary, it was found that studies for the practical use of deep sea water have been conducted in relation to the manufacturing and the treatment of foods, foodstuffs, beverages, and cosmetics.

Phytochemical Screening, Isolation, Characterization of Bioactive and Biological Activity of Bungkang, (Syzygium polyanthum) Root-bark Essential Oil

  • Bungkang (Syzygium polyanthum) is a medium to tall plant which produces medicinal root-bark, the plant is normally found along inland river bank and produces small white flowers and fruits. Essential oils are among the most interesting components of the plant extracts consisting mostly of monoterpenoid or sesquiterpenoids. They are used as therapeutic agents in ethno, conventional, and complementary alternative medicines. Investigation and evaluation of the essential oil of Syzygium polyanthum as well as the antibacterial, antioxidant and antifungal activity was ascertained. The experiment was performed. 100 chemical constituents were obtained and two pure compound was isolated as Eugenol (1) and Farnesol (2). Significant growth inhibition of Staphylococcus aureus, (ATCCⓒ25923) Klebsiellia pneumonia (ATCCⓒ19155), Salmonella typhi (ATCCⓒ14028) and Escherichia coli (ATCC©25922) and the fungal strains Aspergillus flavin, Aspergillus niger, Candida, tropicalis, and Fusarium oxysporium was observed from the essential oil at concentration of 500 ㎍/mL. Antioxidant potential was observed to be strong of 18.42 ㎍/mL when compared to the control of 15.23 ㎍/mL. The result indicated that the oil obtained from root-bark of Syzygium polyanthum can be considered as an agent for antioxidant, antibacterial and antifungal in pharmaceutical food and cosmetic industries trails.

Molecular characterization and biological changes caused by Agrobacterium-mediated infiltration of PgTRX1

  • In order to test the functionality of Panax ginseng thioredoxin 1 (PgTRX1) isolated from fermented wild ginseng roots, a transient effect on physiological activity were performed over a short time frame using the Agrobacterium infiltration technique. The PgTRX1 gene isolated from fermented wild ginseng was confirmed to have a size of 579 bp, and the expression of PgTRX1 was the highest in the sample after 6 h of fermentation. As a result of constructing this gene and confirming the infiltration reaction mediated by Agrobacterium in tobacco leaves, it was found that the expression of the NbHSR203j gene was also induced as PgTRX1 expression increased. As a result of measuring the biological activity of the infiltration samples, the total phenol content increased by 35.45±1.84 to 49.01±1.84 ㎍ GAE/mL compared to the control, and the total flavonoid amount of 9.52±0.41 to 9.82±0.25 ㎍ QE/mL was slightly high. From these results, Agrobacterium-mediated PgTRX1 appears to be related to the hypersensitive response induction mechanism of plants and the production of secondary metabolites such as phenolic substances.

Inhibitory effects of Kirengeshoma koreana Nakai on Melanogenesis in B16F10 melanoma cells

  • Kirengeshoma koreana Nakai (K. koreana)was Saxifragaceae and rare plants in Korea, which is classified as an Critically Endangered (CR) species in Korea. Therefore, most of the studies on it were ecological and taxonomic, and there are no studies on biological activity. In this study, we evaluated the whitening activity of K. koreana extract (KKE). Melanogenesis Inhibitory effects were demonstrated by western-bot and RT-PCR for the effects of KKE on MITF, tyrosinase, TRP-1 and TRP-2 in IBMX-treated B16F10 melanoma cells. IBMX were reported as melanin synthesis enhancers. It could increase intracellular melanin synthesis by activation of the microphthalmia-associated transcription factor (MITF) signaling pathway. KKE showed no cytotoxicity at B16F10. In addition, KKE effectively inhibited the protein and mRNA levels of MITF, tyrosinase, TRP-1 and TRP-2. In conclusion, KKE inhibited melanin synthesis by inhibiting the expression of MITF and its downstream pathways tyrosinase, TRP-1 and TRP-2. Therefore, it was confirmed that K. koreana is a valuable resource for functional cosmetic and biomaterials.
