The Antibacterial Component from Cinnamomi Cortex against a Cariogenic Bacterium Streptococcus matans OMZ 176

  • The methanol extract of Cinnamoni Cortex showed antibacterial action against cariogenic bacterium, Streptococcus mutans OMZ 176. The active principle of the extract was identified to be trans-cinnamaldehyde, which was bactericidal in the minimal inhibitory concentration (MIC) of $100\;\mu$g/ml against the strain. From the results of antibacterial activity of cinnamaldehyde and its derivatives, the acrolein group in the cinnamaldehyde was elucidated to be an essential element for the activity.

  • Four compounds were isolated from the dichloromethane fraction of Cinnamomi Cortex through bioassay-guided isolation. Their structures were identified as coumarin (1), 3,3-dimethoxy-1-propenyl benzene (2), cinnamic acid (3) and o-methoxy cinnamaldehyde (4) on the basis of spectroscopic data. All four compounds showed inhibitory activities in vitro against monoamine oxidase (MAO) prepared by mouse brain. The $IC_{50}$ values were $41.4\;{\mu}M\;(1),\;110.6\;{\mu}M\;(2),\;252.5\;{\mu}M\;(3)\;and\;83.1\;{\mu}M$ (4), respectively.

Comparative Evaluation of Korean Medicine Well-matched with Pork through an Inhibition of Pancreatic Lipase (저육(猪肉)과 잘 어울리는 췌장라이페이즈 억제능을 가진 한약재의 비교평가)

  • Objectives : The aim of this study is to search Korean Medicine with the superior inhibitory effect of pancreatic lipase including a possibility as a well-matched sauce material with pork. Methods : We chose 5 samples which have the potent inhibitory effect of pancreatic lipase enzyme among 9 samples. 5 samples were Glycyrrhizae rhizoma, Cinnamomi cortex, Ganoderma lucidum, Syzygium aromaticum, and Schisandra chinensis. Animals were divided into eight groups (n=7). The experimental groups except for normal group were fed 60% high-fat diet. 5 samples were orally administered at a dose of 200 mg/kg body weight and orlistat were orally administrated at a dose of 60 mg/kg body weight for 7 days. Biochemical anaylses of 5 samples were executed based on lipid parameters such as triglyceride (TG) and total cholesterol (TC). Results : Korean Medicines with $IC_{50}$ of below $1mg/m{\ell}$ were Glycyrrhizae rhizoma, Cinnamomi cortex, Ganoderma lucidum, Syzygium aromaticum, and Schisandra chinensis. Body weight change of all drug-treated groups except Glycyrrhizae rhizoma reduced significantly during the experimental period. Orlistat treatment reduced significantly both TG and TC in serum. The significant emission of TG in fece showed in Cinnamomi cortex treatment, whereas Ganoderma lucidum and Syzygium aromaticum showed a tendency to increase without a significance. Besides, Ganoderma lucidum and Schisandra chinensis emitted significantly TC to fece. Conclusions : In conclusion, Cinnamomi cortex may exert anti-obesity effect by directly inhibiting pancreatic lipase, which would prevent the absorption of lipid from the small intestine. Accordingly, Cinnamomi cortex may suggest a high possibility as a well-matched sauce material with pork.

Synergistic Growth Inhibition of Herbal Plant Extract Combinations against Candida albicans

  • Many skin diseases are caused by microbial infections. Representative pathogenic fungus and bacterium that cause skin diseases are Candida albicans and Staphylococcus aureus, respectively. Malassezia pachydermatis is a fungus that causes animal skin diseases. In this study, we propose a method for removing pathogenic microorganisms from the skin using relatively safe edible herbal extracts. Herbal extracts were screened for skin health through the removal of pathogenic microorganisms, and combinations for effective utilization of the screened extracts were identified. In this study, among methanol extracts of 240 edible plants, C. albicans, S. aureus, and M. pachydermatis were killed by extracts of 10 plants: Acori Gramineri Rhizoma, Angelicae Tenuissimae Radix, Cinnamomi Cortex, Cinnamomi Ramulus, Impatientis Semen, Magnoliae Cortex, Moutan Cortex Radicis, Phellodendri Cortex, Scutellariae Radix, and Syzygii Flos. By evaluating the synergistic antifungal activities against C. albicans using all 45 possible combinations of these 10 extracts, five new synergistic antifungal combinations, Acori Gramineri Rhizoma with Magnoliae Cortex extracts, Acori Gramineri Rhizoma with Phellodendri Cortex extracts, Angelicae Tenuissimae Radix with Magnoliae Cortex extracts, Magnoliae Cortex with Phellodendri Cortex extracts, and Phellodendri Cortex with Syzygii Flos extracts, were identified. By utilizing the selected extracts and five combinations with synergistic antifungal effects, this work provides materials and methods to develop new and safe methods for treating candidiasis using natural products.

Synergistic Antifungal Activity of Magnoliae Cortex and Syzyii Flos against Candida albicans

  • Candida albicans is a dermal fungus of the human body that is known to cause oral candidiasis, vaginal candidiasis, and bloodstream infections in immunocompromised people or in certain environmental conditions. As cases of strains resistant to antifungal agents in C. albicans have been reported, studies using plant materials as safe antifungal agents are being actively conducted. In this study, a total of 17 edible plant extracts showed antifungal activity against C. albicans as a result of evaluating a 280-plant extract library using paper disk diffusion method. Among them, the four extracts with the strongest antifungal activity (Cinnamomi Cortex, Cinnamomi Ramulus, Magnoliae Cortex, and Syzygii Flos) were selected and evaluated for synergistic antifungal activity against C. albicans. The combination of Magnoliae Cortex and Syzygii Flos showed a synergistic activity. The antifungal activity was evaluated based on the concentrations of magnolol and eugenol, the respective components of Magnoliae Cortex and Syzygii Flos. Magnolol and eugenol showed synergistic antifungal activities at the concentration ratio of 1:25 - 1:61. The antifungal activity of these two compounds contributes 28 to 48% to the synergistic antifungal activity of the combination of Magnoliae Cortex and Syzygii Flos extract. In this study, we propose that a combination of Magnoliae Cortex and Syzygii Flos can effectively inhibit the growth of C. albicans and that magnolol and eugenol are the responsible inhibitory compounds.

Effect of Co-administration of Aconiti Lateralis Preparata Radix and Cinnamomi Cortex on Osteoclast Differentiation (부자와 육계 병용투여 시 파골세포 분화 억제에 미치는 영향)

  • Objectives : Aconiti Lateralis Preparata Radix (Aconitum Carmichaeli, AC) and Cinnamomi Cortex (Cinnamomi Cortex, CC) have been treated to elderly for kidney yang enhancement in Korean traditional medicine. In this study, the effects of water extract of AC and CC on RANKL (Receptor Activator for Nuclear Factor ${\kappa}B$ Ligand)-induced osteoclast differentiation were evaluated in culture system. Methods : MTT assay was used to evaluate the potential cytotoxicity of AC and CC extracts in bone macrophage marrows (BMMs) stimulated with M-CSF. TRAP (tartrate-resistant acid phosphatase) staining and TRAP activity were performed to know the inhibitory effect on osteoclast differentiation. The protein expression levels of nuclear factors such as activated T cell(NFAT)c1, c-Fos, MAPKs and ${\beta}$-actin in cell lysates treated with AC and CC extracts were analysed by western blotting. Results : AC, CC extracts and their co-administration inhibited significantly RANKL-induced osteoclast differentiation in BMMs in a dose dependent manner without toxicity. Each AC and CC extracts inhibited the phosphorylation of p38. Also, AC and CC extracts, respectively, inhibited the protein expression of c-Fos and NFATc1 more than Co-administration of AC and CC even if all treatments did. It was observed that RANKL-induced degradation of I-${\kappa}B$ is significantly suppressed by all treatments. Conclusions : Taken together, It was concluded that AC and CC have beneficial effect on osteoporosis by inhibition of osteoclast differentiation. Thus, Atractylodis AC and CC could be a treatment option for osteoporosis.

Purification and properties of an antifungal component, AF-001, from Cinnamomi Cortex (계피로부터 항진균물질 AF-001의 분리.정제 및 특성)

  • Ether extract of Cinnamomi Cortex showing antifungal activity was purified and characterized. The active component from the extract was identified to be trans-cinnamaldehyde, which was effective in inhibiting the growth of the representative fungi of dermatomycosis with minimum inhibitory concentration of $39{\sim}78\;{\mu}g/ml$. The antifungal spectrum of trans-cinnamaldehyde was broader than that of commercial antifungal agent, Ketoconazole.

Pancytopenia Improves with Compromised Liver Function in A Hepatocellular Carcinoma Patient Treated by Modified Sipjeondaebo-tang (십전대보탕가감방을 투여한 간세포암종 환자에서 범혈구감소증 호전 및 간수치 상승 증례)

  • Objectives : To observe the therapeutic effects of an oriental herbal prescription in a hepatocellular carcinoma patient with splenomegaly and pancytopenia. Methods : Modified Sipjeondaebo-tang was prescribed three times a day to a hepatocellular carcinoma patient with splenomegaly and pancytopenia. Laboratory tests were carried out regularly to observe the therapeutic effects of the oriental herbal prescription for pancytopenia. Results : When treated with modified Sipjeondaebo-tang including Cinnamomi Cortex, the levels of white blood cells, red blood cells, hemoglobin, and hematocrit increased. However, aspartate transaminase, alanine transaminase, alkaline phosphatase, gamma-glutamic transpeptidase, total bilirubin, direct bilirubin levels also increased. Conclusions : Administering modified Sipjeondaebo-tang with Cinnamomi Cortex showed effect in improving pancytopenia but an increase in liver enzyme levels was also observed.

Effects of Cinnamomi Cortex on Hypercholesterolemia Induced by Estradiol Valerate in Rats (육계(肉桂)가 Estradiol valerate에 의하여 유발된 고콜레스테롤 혈증에 미치는 영향)

  • This study was designed to investigate the effects of Cinnamomi Cortex extract (CC) on hypercholesterolemia induced by Estradiol valerate (EV) in female rats. We investigated the effects of CC on Changes in body weights for 5 weeks. In addition, we examined the effects on fasting blood glucose (FBS) and tested oral glucose tolerance test after oral administration of CC for 5 weeks. We also investigated the effects on levels of total cholesterol, HDL-cholesterol and LDL-cholesterol in serum. Treatment with CC restored weight gain rates suppressed by EV significantly. CC did not affect serum glucose level and glucose tolerance. Levels of total cholesterol and triglyceride were lowered by administration of CC compared to EV treated control respectively. These result suggest that CC can lower serum cholesterol level then prevent weight loss induced by EV. We also suggest the possibility as anti-hyperlipidemic agent of CC.

Study on Herb Pair of Zanthoxyli Fructus(Pericarpium Zanthoxyli) (촉초(蜀椒)(천초(川椒)) 약대(藥對)에 관한 연구)

  • Objectives : The purpose of this study is to find out herb-pair prototype composed of Zanthoxyli Fructus, to examine compatibility regulation between them and to classify them into seven features of compatibility. Methods : To search usages of Zanthoxyli Fructus compatibility, we have investigated the classics of herb and prescription such as "五十二病方"(Fifty-two kinds of Diseases), "黃帝內經"(The Medical Classic of Yellow Emperor), "傷寒論"(Treatise on Febrile Caused by Cold), "金匱要略"(Synopsis of Prescriptions of The Golden), "附廣肘后方"(Handbook of Prescriptions for Emergencies), "本草綱目"(Compendium of Materia Medica), "得配本草"(Deukbae Materia Medica). Results : In these books Zanthoxyli Fructus was paired with below herbs; 桂(Cinnamomi Cortex), 薑(Zingiberis Rhizoma), 吳茱萸(Evodiae Fructus), 細辛(Asari Radix), 茴香(Foeniculi Fructus), 肉荳蔲(Cistanches Herba), 釜下土(Terba Flava Usta), 益智仁(Alpinae Oxyphyllae Fructus), 飴(Saccharum Granorum), 葱白(Allii Radix), 梨(Pyrus), 牙皂(Gleditsia Fructus), 杏仁(Armeniacae Amarum Semen), 蕎麥粉(Fagopyrum esculentum Moench), 胡粉(Minium), 白朮(Atractylodis Rhizoma White), 蒼朮(Atractylodis Rhizoma), 麻子仁(Cannabis Fructus), 大麥蘗(Hordei Fructus Germiniatus), 麴(Massa Medicata Fermentata), 生地黃(Rehmanniae Radix), 茯苓(Poria), 牡蛎(Ostreae Concha), 白礬(Alumen), 雄黃(Realgar), 烏頭(Aconiti Radix), 附子(Aconiti Lateralis Preparata Radix). Mutual assistance compatibility with : Armeniacae Amarum Semen, Fagopyrum esculentum Moench, Minium, Atractylodis Rhizoma White, Atractylodis Rhizoma, Cannabis Fructus, Hordei Fructus Germiniatus, Massa Medicata Fermentata, Rehmanniae Radix, Poria, Ostreae Concha, Alumen. 3. Mutual restraint compatibility with: Realgar, Aconiti Radix, Aconiti Lateralis Preparata Radix) 4. Mutual detoxication compatibility with: 桑汁(Juice of Mori Folium), 桑根汁(Juice of Morus Cortex), 大蒜(Allii Bulbus), 薺苨(Remotiflori Radix), 桂汁(Juice of Cinnamomi Cortex).
