The purpose of this study was to investigate the cultivation site of Cornus officinalis (C. officinalis) by analyzing the contents of loganin and morroniside according to the cultivation environment of C. officinalis. The loganin and morroniside contents were analyzed by ultra performance liquid chromatography. Mg content and pH in soil data were found to have the greatest effect on logainin content. In soil data, Mg, Na content, pH, salinity, silt ratio and clay ratio were found to have the greatest effect on morroniside content. Therefore, Mg content, pH, and clay ratio were found to have a high effect on the contents of loganin and morroniside, which are indicators of C. officinalis. In conclusion, Mg content, pH, and clay ratio in soil may have a high effect on the contents of loganin and morroniside, which are indicators of C. officinalis.

Objectives : The aim of the present study is to evaluate the wound healing-enhancing and anti-inflammatory effects of Pinus densiflora, Cornus officinalis, Zingiber officinale, Ganoderma japonicum and Scutellaria baikalensison human keratinocyte, HaCaT cells. Methods : We adopted in vitro wound healing assay to measure the proliferation-and migration-enhancing effects in HaCaT cells. The expressions of cytokine genes were measured in HaCaT cells using real-time PCR analysis. Results : The extracts of Pinus densiflora, Cornus officinalis, Zingiber officinale, Ganoderma japonicum and Scutellaria baikalensis enhanced the proliferation and migration of HaCaTcells. The expression of keratinocyte growth factor receptor(FGFR2-IIIb) gene was also induced. The extracts inhibited iNOS, IL-$1{\beta}$ and TNF-$\alpha$ gene expression. Conclusions : The extract of Pinus densiflora, Cornus officinalis, Zingiber officinale, Ganoderma japonicum and Scutellaria baikalensis has wound healing-enhancing effects and anti-inflammatory effects.

The purpose of this study is to predict the effects of macroscopic and integrative therapies by finding active ingredients, potential targets of Astragalus membranaceus (Am) and Cornus officinalis (Co) for diabetic nephropathy. We have constructed network pharmacology-based systematic and network methodology by system biology, chemical structure, chemogenomics. We found several active ingredients of Astragalus membranaceus (Am) and Cornus officinalis (Co) that were speculated to bind to specific receptors which had been known to have a role in the progression of diabetic nephropathy. Four components of Am and eleven components of Co could bind to iNOS; two ingredients of Am and six ingredients of Co could docking to cGB-PDE; one component of Am and nine components of Co could bind to ACE; three ingredients of Co with neprilysin; three components of Co with ET-1 receptor; four ingredients of Am and fourteen ingredients of Co with mineralocorticoid receptor; one component of Am and seven components of Co with interstitial collagenase; one ingredient of Am and ten ingredients of Co with membrane primary amine oxidase; one component of Am and four components of Co with JAK2; two ingredients of Am and one ingredient of Co with MAPK 12; one component of Am and five components of Co could docking to TGF-beta receptor type-1. From this work we could speculate that the possible mechanisms of Am and Co for diabetic nephropathy are anti-inflammatory, antioxidant and antihypertensive effects.
Objectives : In this animal study, we performed the single oral dose toxicity test of Standardized Cornus officinalis Sieb. et Zucc. and Psoralea corylifolia L. 30% ethanol extract (SCP) in Sprague-Dawley (SD) rats owing to aims for verifying approximate lethal dose (ALD). Methods : According to OECD guidelines for the testing of chemicals section 4 health effects test No. 420 acute oral toxicity study - fixed dose procedure (17 December 2001), single oral dose toxicity test was performed. Animals were divided into two groups: Group 1, vehicle-treated rats (Control); Group 2, SCP 5000 mg/kg treated rats. SCP is composed of two medicinal herbs: Cornus officinalis Sieb. et Zucc. (650 g) and Psoralea corylifolia L. (350 g) in 30% ethanol. SCP was once orally administered to female and male SD rats at dose levels of 5000 mg/kg. Animals were monitored on the mortality, clinical signs, body weight changes and necropsy findings for 14 days. Results : After single oral treatment of SCP, we could not find any mortality up to 5000 mg/kg. Compared with the control group, there were also no significant differences in clinical sign, weight change, weight gain and gross abnormalities in SCP 5000 mg/kg-treated group. Conclusions : Taken together, these results suggest that the ALD of SCP in both female and male SD rats were considered as over 5000 mg/kg. Results from this study provide scientific evidence for the safety of SCP.
Hydrogen peroxide is one of major chemicals mediating antitumor and antimicrobial activities of macrophages. We searched natural products enhancing hydrogen peroxide generation from murine macrophage-like cell line J774. Among 21 methanol extracts of Korean medicinal plants, the extract from Cornus officinalis was the most effective. The active component from the fractions was searched by activity guided fractionation, and identified as ursolic acid by spectral data.
Cornus officinalis plants that grow in several locations in Korea have been found to be infected with leaf spot disease. Symptoms include necrotic lesions, which are angular, irregularly shaped, vein-limited, and dark brown, on both sides of the leaves. The causal agent of the disease was identified to be Pseudocercospora cornicola based on the morphological characteristics of the fungus and molecular phylogenetic analysis of the obtained multi-locus DNA sequence data. This is the first report investigating P. cornicola found on C. officinalis in Korea.

Twenty male soccer players volunteered as the experimental subjects. The subjects were randomly assigned to the "Winter training with Cornus officinalis extract intake group"(WCI; n=10) and "Winter training group"(WT; n=10). The variables of blood fatigue substance, muscle damage and liver function where measured in all the subjects before the start of the Cornus officinalis intake after at the end of the 4 weeks winter training. Winter training program was proceeded with 6 days a week for 4 weeks. Exercise intensity was measured using the HRR(70~95%). Changes in the heart rate were also measured with the Polar. WCI group were ingested 80 mL of Cornus officinalis extract after every breakfast and dinner for 4 weeks. The test data were analyzed by paired t-test, and independent t-test(%diff), and the alpha level of p<.05 was set for all tests of significance. Lactate in WCI group had significantly decreased, and the changes between the groups were significantly lower in the WCI group than in the WT group. Ammonia in WCI group had significantly increased. CK in WCI group had significantly decreased. LDH in both group had significantly decreased. AST in WCI group had significantly decreased, and the changes between the groups were significantly lower in the WCI group than in the WT group. ALT in WCI group had significantly decreased. In conclusion, the results of this study show that Cornus officinalis extract seem to have a positive influence on blood fatigue substance, muscle damage and liver function. Therefore Cornus officinalis extract is considered to be able to contribute an ergogenic aids.

An 80% EtOH extract and the solvent fractions of the seeds of Cornus officinalis were evaluated for their inhibitory activities against advanced glycation end products (AGEs) formation and AGEs-induced protein cross-linking in vitro. In vitro assay for AGEs-bovine serum albumin (BSA) formation showed that the 80% EtOH extract, n-hexane, EtOAc, n-BuOH and water fractions significantly inhibited AGEs formation with observed $IC_{50}$ values of 1.13, 17.64, 1.52, 1.24 and $3.27{\mu}g/ml$, respectively. In indirect AGEs-ELISA assay, the 800% EtOH extract, EtOAc and n-BuOH fractions exhibited more potent inhibitory activity on AGEs-BSA formation than aminoguanidine, a well know AGEs inhibitor. Furthermore, the 80% EtOH extract and all the solvent fractions inhibited concentration-dependently AGE-BSA cross-linking to collagen. The 80% EtOH extract, EtOAc, n-BuOH and water fractions also had a breaking activity against preformed AGE-BSA cross-linking concentration dependently. Thus these results suggest that the 80% EtOH extract and fractions of the seeds of C. officinalis could be an inhibitor as well as breaker of AGE-BSA cross-linking.
Ten compounds, (+)-pinoresinol (1), (-)-balanophonin (2), gallicin (3), vanillin (4), 4-hydroxybenzaldehyde (5), coniferaldehyde (6), betulinic acid (7), ursolic acid (8), 5-hydroxymethyl furfural (9), and malic acid (10), were isolated from a EtOAc-soluble fraction of the seeds of Cornus officinalis. The structures of these compounds were elucidated by spectroscopic methods as well as by comparison with reported values. Compounds 1, 2, and 4-7 were isolated from this species for the first time. All the isolates (1-10) were subjected to an in vitro bioassay to evaluate their inhibitory activity against advanced glycation end products (AGEs) formation. Among these, compounds 2 and 3 showed the significant inhibitory activity on AGEs formation with $IC_{50}$ values of 27.81 and 18.04${\mu}M$, respectively.
