한국산 곡류에서의 곰팡이독소 오염현황 및 동시분석 (Occurrence of Mycotoxins in Korean Grains and Their Simultaneous Analysis)

  • 11종(아플라톡신 $B_1$, $B_2$, $G_1$, $G_2$, 오크라톡신 A, 푸모니신 $B_1$, $B_2$, 제랄레논, 데옥시니발레놀, T-2 toxin과 HT-2 toxin) 곰팡이독소 동시분석법을 정립하였다. 1/6 면역친화컬럼을 이용하여 정제하였으며, LC/MS/MS로 분석하였다. 각각 독소별 정성한계는 S/N=3에서 0.1-3.0 ${\mu}g/kg$이었으며, 정량한계는 S/N=10에서 0.3-10.0 ${\mu}g/kg$이었다. 회수율(recovery)은 70.45-111.11%이었으며, 상대표준편차(RSD)는 0.10-14.45%로 나타났다. 동 분석법을 이용하여 대표적인 한국산 농산물인 백미, 보리, 옥수수에 대한 곰팡이독소 오염실태를 조사하였다. 푸모니신, 데옥시니발레놀, 제랄레논이 백미와 보리에서 각각 평균 29.8, 6.4, 36.2%와 2.3, 55.8, 34.9%의 평균 오염율을 보였으며, 다른 독소들은 나타나지 않았다. 옥수수에서는 1개의 시료에서 푸모니신($B_1+B_2$)이 100.90 ${\mu}g/kg$ 검출되었다. 하지만, 곰팡이독소 오염 수준은 현재 우리나라 각 곰팡이독소 허용 최대기준치 이내로 나타났다. 한편, 벼의 가공단계, 재배지역, 벼 품종 및 품질 등급에 따른 곰팡이독소 오염현황을 비교하였다. 벼에서 현미, 백미로 가공될수록 곰팡이독소 오염도는 감소하였다. 동일 품종의 경우에 상대적으로 기온이 높은, 낮은 위도의 지역에서 재배된 벼가 곰팡이독소가 많이 오염된 것으로 나타났으며, 벼의 품질 등급이 낮아질수록 높은 곰팡이독소 오염도를 나타내었다. 이러한 결과를 종합적으로 살펴보았을 때 국내산 주요 농산물의 곰팡이독소 오염수준이나 위해도가 기준치 이내이며, 위험한 수준은 아닌 것으로 판단된다. 그러나, 곰팡이독소는 코덱스 등 국제사회에서 중요하게 다루어지고 있고, 현재까지 알려지지 않은 위험성이나 신규 독소들에 대한 연구가 활발히 진행되고 있는 만큼 우리나라도 충분한 연구를 통하여 우리 식품에 대한 안전성 논란에 사전 대비토록 하여야 할 것이다.

목질계 농부산물을 이용한 고체발효에서 발효조건 최적화를 통한 구연산 생산 증대 (Optimization of Fermentation Conditions for the Production of Citric Acid by Aspergillus niger NRRL 567 Grown on Agricultural by Products)

  • 본 실험에서는 농부산물인 밀짚을 고체배지로 사용하여 Aspergillus niger NRRL 567에서 구연산 생산 시, 발효조건과 첨가제가 구연산 생산에 미치는 영향을 단일변수(one-factor-at-a-time) 최적화를 이용하여 주요 인자의 순차적 최적화를 수행하였다. 발효 72시간에서 온도, 수분함량, 입자크기, pH와 첨가제 농도를 최적화했을 때, 각각 $30^{\circ}C$, 70%, 0.5~1.0 mm, pH 5.5와 4% 메탄올 첨가조건에서 최대 구연산 생산인 206.0 g/kg 건조중량 (DM)을 확인할 수 있었다. 이는 최적화 이전 구연산 최고 생산인 74.5 g/kg DM 대비 177% 증가한 결과이다. 최적화 실험에서 도출된 조건을 밀짚, 옥수수대와 피트모스(peat moss)에 적용하여 고체발효를 수행하였을 때, 발효 120시간에서 각각 231.8, 213.8, 240.2 g/kg DM 구연산 생산을 확보하였다. 본 실험 결과는 밀짚과 옥수수대 등의 목질계 농부산물을 이용한 구연산 생산 시, 고체발효법이 기존의 액체발효법의 대체가 가능함을 시사하였다.

MODIS 위성 영상 기반의 일차생산성 알고리즘 입력 기상 자료의 신뢰도 평가: 미국 Corn Belt 지역을 중심으로 (The Evaluation of Meteorological Inputs retrieved from MODIS for Estimation of Gross Primary Productivity in the US Corn Belt Region)

  • 다양한 공간규모에 대해서 식생의 생산성을 추정하기 위해 대기와 생태계내의 탄소순환 과정을 모니터링 하는 것은 탄소순환과정과 원격탐사기법을 결합함으로써 기능하다. 그 대표적인 예로서 미국 항공우주국(NASA)에서 개발한 지구관측시스템(Earth Observing System, EOS)은 총 일차생산성 (Gross Primary Productivity, GPP), 순 일차생산성 (Net Primary Productivity, NPP)을 전 지구에 대해 1km의 공간해상도로 제공한다. 그러나 MODIS의 생산성 추정에 기상입력자료로 이용되는 The NASA Data Assimilation Office (DAO)자료는 $1{\circ}\;{\times}\;1.25{\circ}$의 거친 해상도를 가지기 때문에 지역규모에서 지표기상의 상세한 공간적인 이질성을 반영하기 어렵다. 본 연구에서는 MODIS의 Aqua와 Terra 위성의 영상만을 이용하여 생산성을 추정하기 위한 입력자료로 사용되는 일 단위 일사량(MJ $day^{-1}$), 열 최저기온($^{\circ}C$, 낮 시간 평균 대기수증기압 포차(Yapor Pressure Deficit, Pa)을 5 km 해상도로 각각 추정하였다. 각각의 입력자료의 평가를 위해 미국 중서부 Corn Belt 지역 내에 위치하고 있는 7 개소의 지상기반의 관측자료를 수집하였다. MODIS 위성영상을 이용하여 추정한 기상입력자료와 관측자료를 비교한 결과, 일 최저기온은 ME와 상관계수가 각각 0.83에서 0.98, $-0.9^{\circ}C$ 에서 $+5.2^{\circ}C$ 의 범위로 양호한 상관성을 보였다. VPD는 약간 산개하는 경향을 보인다(ME = -183.8 Pa ~ +382.1 Pa; R = 0.51 ~ 0.92). 반면에, 일 단위 일사량은 MODIS가 약간 과대평가하는 경향을 보였지만 대부분의 지역에서 관측자료와 유의한 일치도를 보였다(ME = -0.4 MJ $day^{-1}$ +7.9 MJ $day^{-1}$; R = 0 67 ~ 0.97). 본 연구에서는 맑은 날에 대해서 MODIS 위성영상으로부터 추정된 기상입력자료가 MODIS가 제공하는 기상입력자료보다 상세한 지표 기상의 이질성을 반영한 자료를 생산할 수 있음을 확인하였다.

DEVELOPMENT AND TESTING OF MEDIUM CAPACITY GRAIN FLOUR SEPARATOR

  • A power operated 90.5 hp electric motor) grain flour separator was designed and developed for separation of grain (wheat, corn, chickpea and soybean) flour into various fractions based on the size of the particles of the product. The separator agitating mechanism, feed control, cylindrical separator unit and an eccentric mechanism. The machine was tested for wheat ( variety ; Sujata) flour separation into four fractions, viz ; semolina, Gr-I and II, flour (coarse) and white (fine) flour. Wheat samples (6.8% m.c., db) were first pearled by CIAE pearler for 15.8% bran removal . The pearled wheat grains were then milled for semolina by a burre mill. The product and machine characteristics were determined at different capacities varying from 24 kg/h to 143 kg/h. It was found that 76 kg/h capacity gave reasonably best results in terms of purity and recovery of semolina vis-a-vis the market product. The energy requirement of the machine at no-load was found to be 230 W and at load c nditions, it varied between 36.3-6.4 KJ per kg of fead seperation. The macine could be used by small flour millers small/medium size traders and retailers and other processors for making available various flour products of different particle size in the market for ready use of the consumers.

Mechanical Tenacity Analysis of Moisture Barrier Bags for Semiconductor Packages

  • We have been using Moisture Barrier Bags for dry packing of semiconductor packages to prevent moisture from absorbing during shipping. Moisture barrier bag material is required to be waterproof, vapor proof and offer superior ESD (Electro-static discharge) and EMI shielding. Also, the bag should be formed easily to the shape of products for vacuum packing while providing excellent puncture resistance and offer very low gas & moisture permeation. There are some problems like pinholes and punctured bags after sealing and before the surface mount process. This failure may easily result in package pop corn crack during board mounting. The bags should be developed to meet the requirements of excellent electrical and physical properties by means of optimization of their raw material composition and their thickness. This study investigates the performance of moisture barrier bags by characterization of their mechanical endurance, tensile strength and through thermal analysis. By this study, we arrived at a robust material composition (polyester/Aluminate) for better packing.

수질제어 및 부식억제제에 의한 상수도관의 내부부식 제어 (Internal Corrosion Control of Drinking Water Pipes by pH and Alkalinity Control and Corrosion Inhibitor)

  • The internal corrosion of water distribution systems is the main cause for the problem of the public health threat as well as water leakage in the damaged pipeline, red water, and odor and taste of the tap water. This study was examined the effect of chemicals used for pH and alkalinity control and corrosion inhibitors for producing the optimal corrosion control method. Corrosion study at different pH and alkalinity indicated that these control using alkaline chemicals was effective in corrosion rate, Fe release reduction, but examined to be increased in turbidity and corrosion-by-products(TTHMs) problems. The turbidity was slightly increased, requiring caution in controlling corrosion with $Ca(OH)_2$. At pH 9.0, TTHMs concentration is increased two times corn pared with non-control of pH. Using the pipe which had experienced 28 years of exposure, iron release was decreased with the corrosion inhibitor. Consequently, pH, Alkalinity control method using alkaline chemicals must be complemented by corrosion inhibitor application for efficient corrosion control.

Xylanase Production by Mixed Culture Using Crude Hemicellulose from Rice Straw Black Liquor and Peat Moss as an Inert Support

  • Black liquor (BL) is a by-product of rice straw pulping process. It is a low costs raw material for production value-adding proteins and enzymes, which has been paid more and more attention to reduce its environmental pollution. Mixed cultures of micelial fungi, Trichoderma reesei Northern Regional Research Laboratory (NRRL)11236, Trichoderma reesei NRRL 6165 and Aspergillus niger strains NRC 5A, NRC 7A, and NRC 9A were evaluated for their ability to produce xylanase using crude hemicellulose (CHC) prepared from BL and peat moss as an inert support under solid state fermentation (SSF). The most potent strains, A. niger NRC 9A (818.26 U/g CHC) and T. reesei NRRL 6165 ($100.9{\pm}57.14$ U/g CHC), were used in a mixed culture to enhance xylanase production by co-culturing under SSF. In the mixed culture, xylanase production ($1070.52{\pm}12.57$ U/g CHC) was nearly1.3 and 10.6-fold increases over the activities attained in their monocultures, A. niger NRC 9A and T. reesei NRRL 6165, respectively. Optimization of the culture parameters of the mixed culture SSF process, concentration of ammonium sulfate and corn steep liquor, CHC/peat moss ratio, inoculum size and ratios of the two strains, initial pH value, initial moisture content and incubation time, exhibited a significant increase ($2414.98{\pm}84.02$ U/g CHC) in xylanase production than before optimization.

LC/ESI-MS를 이용한 Acrylamide의 정성확인 및 정량분석 (Quantitative Analysis and Qualification of Acrylamide Using LC/ESI-MS)

  • Acrylamide, difficult to analyze by GC and GC/MS due to the polarity and low volatility, was analyzed by LC/ESI/MS in the study. Acrylamide its(molecular weight 71amu) showed m/z=72 $(M+H)^+$ and high peak intensity at 22V in SIR mode. The mass spectrum ratios of acrylamide for qualitative identification had m/z=72 in precursor ion and m/z=55 in products ion, respectively. Those ratios at 30V in SIR mode ranged from 1: 1.4 to 1:1.17 despite various acrylamide concentrations. The ion intensity ratios of acrylamide $(m/z=72,\; [M+H]^+)$ to acrylamide isotopes $(m/z=73,\;[M+H]^+)$ ranged from 100 : 3.57 to 100 : 3.92. The results verified theoretical mass spectrum ratio that was 100:3.82. The linearity of standard calibration curve was y : 520.584x + 1815.26 with $r^2=0.99.$ In quality assurance and quality control, the recovery rate ranged from 81.64 percent to 90.97 percent and relative standard deviation was less than $10\%$ with 5 repeated injections at individual standard calibration solutions. The method was applied to analyze acrylamide in food at grocery stores. Snacks made of potatoes showed the highest acrylamide concentration followed by products made of French fries, wheat, and corn.

어육(魚肉) 연제품(煉製品)의 보수력(保水力)에 관(關)한 연구(硏究) (A Study on Water Holding Capacity of Fish Meat Paste Products)

  • It is well known that water holding capacity plays an important role in processing such meat products as frankfurter-type sausage and fish meat paste products as kamaboko and fish sausage. Consumer qualities of meat products, such as appearance, flavor, as well as drip and shrinkage on cooking, depend greatly on the degree of water binding. In this paper, the water holding capacities of fish paste and salt added paste of white corvenia, Argyrosomus argentatus and file fish, Novodon modestus were measured by centrifuging and press method before and after cooking. And the effects of the addition of phosphates and starch to enhance water binding and stabilize gel formation were also discussed. In addition, the experimental conditions which are suitable to determine the water binding of fish meat paste product were suggested. The results were expressed in percent of water absorbed by the filter paper when pressed or released by pressor or centrifuge to the weight of sample. From the results. a proper condition to measure the water holding capacity of fish meat paste was that 3.0 g of sample which was previously added with 10 percent water was centrifuged at 13,400 G or 12,000 rpm for 15 minutes for the centriguging method and for press method, 0.3 g sample with 10 percent of water added was extracted by an oil pressor at $30\;kg/cm^2$ for 1 minute. Water holding capacity of fresh paste of white corvenia was relatively higher than that of file fish and the difference between species of fish was greater than the difference between measurments by two methods. Sodium chloride had a great effect on enhancing the water holding capacity of fish meat paste giving better effect when 3.0 percent of salt was added. Phosphates used except calcium phosphate revealed a certain enhancement in water binding, yielding best effect at 0.3 percent addition, and metaphosphate seemed to be more effective in order. The addition of corn starch, however, appeared to be not so effective for enhancement of water binding in fresh-salt-added fish meat paste but in cooked fish paste which might be attributed to absorption of water by starch grain and swelling during the heating and consequently enforced gel strength of cooked fish paste. And the water holding capacity of cooked fish paste was proportionally related to its gel strength.

  • A power operated (0.5 hp electric motor) grain flour separator was designed and developed for separation of grain (wheat, corn, chickpea and soybean) flour it no various fractions based on the size of the particles of the product. The separator is made of mild steel and consists of a hopper, power driven agitating mechanism, feed control , cylindrical separator unit and an eccentric mechanism. The machine was tested for wheat (variety : Subjata) flour separation into four fraction, viz : semolina ; Gr-I and II, flour (coarse) and white (fine) flour. Wheat samples (6.8% m.c., db) were first pearled by CIAE pearler for 15.8% bran removal . The product and machine characteristics were determined at different capacities varying from 24 kg/h to 143 kg/h. It was found that 76 kg/h capacity gave reasonably best results in terms of purity and recovery of semolina vis-a-vis the market product.
