  • Ultrstructural studies of germ cell differentiation and vitellogenesis in the oocytes of the female Rapana venosa in the brackish water area of Seomjin River were investigated by transmission electron microscope observations. In the early vitellogenic oocytes, the Golgi complex and mitochondria were involved in the formation of glycogen particle, lipid droplets, and yolk granules. In the late vitellogenic oocytes, the rough endoplasmic reticulum and multivesicular bodies were involved in the formation of proteid yolk granules in the cytoplasm. However, heterosynthetic vitellogenesis in this species were not observed in vitellogenic oocytes during oogenesis. A mature yolk granule was composed of three components: crystalline core, electron lucent cortex and the limiting membrane. As shown in some large gastropods, vitellogenesis in R. venosa occurred by way of endogenous autosynthesis without heterosythetic vitellogenesis (exogeneous endocytosis), which are found in the oocytes in bivalves. The mating period and spawning activity were related with the increases of seawater temperatures and salinities.

  • In this paper, a small-area 32-bit differential paired eFuse OTP memory for power ICs is designed. In case of smaller number of rows than that of columns for the OTP memory cell array, a scheme for the cell array reducing the number of SL driver circuits requiring their larger layout areas by routing the SL (source line) lines supplying programming currents for eFuse links in the row direction instead of the column direction as well as a core circuit is proposed. In addition, to solve a failure of being blown for non-blown eFuse links by the electro-migration phenomenon, a regulated voltage of V2V ($=2V{\pm}0.2V$) is used to a RWL (read word line) driver circuit and a BL (bit line) pull-up driver circuit. The layout size of the designed 32-bit eFuse OTP memory is $228.525{\mu}m{\times}105.435{\mu}m$, which is confirmed to be 20.7% smaller than that of the counterpart using the conventional cell array routing, namely $197.485{\mu}m{\times}153.715{\mu}m$.

  • Objective: The purpose of this study was to report the effect of traditional Korean medicine (TKM) in alleviating the side effects of lung cancer patient undergoing immunotherapy. Method: A 43-year-old man, who was diagnosed with non-small cell lung cancer, received pembrolizumab treatments. The patient was treated with acupuncture and herbal medicine (Geoeoyangpye-tang) to control various uncomfortable symptoms. The degree of pain was measured by the numeric rating scale (NRS). The European Organization for Research and Treatment of Cancer Quality of Life Questionnaire, Core 30 (EORTC QLQ-C30) and the EORTC 13-item lung cancer-specific module (EORTC LC-13 questionnaire) were used to assess the change in the quality of life. Results: After the TKM treatment, the flank pain and arthralgia based on the NRS were significantly improved. Various uncomfortable symptoms such as fatigue, dyspnea, insomnia, and loss of appetite were also significantly improved, based on the EORTC QLQ-C30 and EORTC QLQ-LC13. The size of the primary tumor was decreased during treatment. The disease status was stable radiologically after two months from discharge.

  • Recently, the prevalence of remotely managed patient care systems in medical institutions is increasing due to COVID-19. In particular, in the case of fluid monitoring, hospitals are considering introducing it as a system that can reduce patient safety and nurses' work. There are two products under development: a load cell method that measures weight and a method that detects drops of sap by infrared sensing. Although each product has differences in operation principle, sensor type, size, usage, and price, medical institutions are highly interested in the accuracy of the data obtained.In this study, two prototypes with different sensor methods were manufactured and the total amount of infusion per hour was measured to test the accuracy, which is the core of the infusion monitoring device. In addition, when there was an external movement, the change in the measured value of the sap was tested to evaluate the accuracy according to the measurement method. As a result of the experiment, there was a difference of less than 5% in the measurement value error of the two devices, and the load cell method showed a difference in the low-capacity measurement value and the infrared method in the high-capacity measurement value. As a result of this experiment, there was little difference in accuracy according to the sensor method of the infusion monitoring device, and it is considered that there is no problem in accuracy when used in a medical institution.

Evaluation of the Sealing Capacity of the Supercritical CO2 by the Measurement of Its Injection Pressure into the Tuff and the Mudstone in the Janggi Basin (초임계이산화탄소(scCO2) 주입압력 측정에 의한 장기분지 응회암과 이암의 scCO2 차폐능 평가)

  • The laboratory scale experiment was performed to evaluate the sealing capacity of the capping rock such as tuff and mudstone, measuring the intial supercritical $CO_2$ ($scCO_2$) injection pressure and the $scCO_2$-water-rock reaction for 90 days. The drilling cores sampled from 800 m in depth around the Janggi basin, Korea were used for the experiment. The mineralogical changes of mudstone and tuff were measured to evaluate the geochemical stability during the $scCO_2$-water-rock reaction at $CO_2$ storage condition (100 bar and $50^{\circ}C$). The rock core was fixed in the high pressurized stainless steel cell and was saturated with distilled water at 100 bar of pore water pressure. The effluent of the cell was connected to the large tank filled with 3 L of water and 2 L of $scCO_2$ at 100 bar, simulating the subsurface injection condition. The $scCO_2$ injection pressure, which was higher than 100 bar, was controlled at the influent port of the cell until the $scCO_2$ begin to penetrate into the rock and the initial injection pressure (> 100 bar) of $scCO_2$ into the rock was measured for each rock. The mineralogical compositions of mudstones after 90 days reaction were similar to those before the reaction, suggesting that the mudstone in the Janggi basin has remained relatively stable for the $scCO_2$ involved geochemical reaction. The initial $scCO_2$ injection pressure (${\Delta}P$) of a tuff in the Janggi basin was 15 bar and the continuous $scCO_2$ injection into the tuff core occurred at higher than 20 bar of injection pressure. For the mudstone in the Janggi basin, the initial $scCO_2$ injection pressure was higher than 150 bar (10 times higher than that of the tuff). From the results, the mudstone in Janggi basin was more suitable than the tuff to shield the $scCO_2$ leakage from the reservoir rock at subsurface.

  • The Earth outer core accomodates moderately considerable amount of lighter elements than pure iron itself. Hydrogen is one of the possible candidates of minor constituents in the outer core. It would be worth while to extend for the pressure effect on the solubility of hydrogen in the metal-hydrides including iron hydride. In view of hydrogen being one of the potential substitutes for petroleum, searching a more efficient way for storing hydrogen in the form of hydrides is of considerable value. For two purposes, $TiH_2$was selected among lot of hydrides for its characteristics under pressure and temperature. There have been two kinds of experiment carried out on $TiH_2$ under different experimental conditions. As one of these attempts, polycrystalline $TiH_2$ was loaded up to 15 GPa stepwise at the constant temperature 500${\circ}$ using a piston-cylinder diamond anvil cell equipped with a miniature furnace of an electric power supply. The X-ra diffraction technique was employed on the quenched samples after the simultaneous high pressure and temperature treatments. During these high pressure-temperature runs, and irreversible phase of $TiH_2$ has been observed at the pressures higher than 11.3 GPa, which would be assigned to the orthorhombic crystal system as one of the new phase(s) of $TiH_2$. Molar volume change on this phase transition is ∼10%.

Neurotoxicity of Synthetic Cannabinoids JWH-081 and JWH-210

  • Synthetic cannabinoids JWH-018 and JWH-250 in 'herbal incense' also called 'spice' were first introduced in many countries. Numerous synthetic cannabinoids with similar chemical structures emerged simultaneously and suddenly. Currently there are not sufficient data on their adverse effects including neurotoxicity. There are only anecdotal reports that suggest their toxicity. In the present study, we evaluated the neurotoxicity of two synthetic cannabinoids (JWH-081 and JWH-210) through observation of various behavioral changes and analysis of histopathological changes using experimental mice with various doses (0.1, 1, 5 mg/kg). In functional observation battery (FOB) test, animals treated with 5 mg/kg of JWH-081 or JWH-210 showed traction and tremor. Their locomotor activities and rotarod retention time were significantly (p<0.05) decreased. However, no significant change was observed in learning or memory function. In histopathological analysis, neural cells of the animals treated with the high dose (5 mg/kg) of JWH-081 or JWH-210 showed distorted nuclei and nucleus membranes in the core shell of nucleus accumbens, suggesting neurotoxicity. Our results suggest that JWH-081 and JWH-210 may be neurotoxic substances through changing neuronal cell damages, especially in the core shell part of nucleus accumbens. To confirm our findings, further studies are needed in the future.

Antioxidant and Anticancer Activities of Methanol Extracts Prepared from Different Parts of Jangseong Daebong Persimmon (Diospyros kaki cv. Hachiya) (장성 대봉감의 부위별 메탄올 추출물의 항산화 활성 및 항암 활성)

  • The antioxidant activity of methanol extracts from five different parts (flesh, peel, core, seed, calyx) of Jangseong Daebong persimmon (Diospyros kaki cv. Hachiya) were evaluated by determining total phenol content (TPC), DPPH radical scavenging activity (RSA), ABTS RSA, and reducing power (RP). The flesh extract gave the highest yield (92.93%) while the lowest yield was obtained from the seed (5.17%). The seed extract showed the highest total phenolic content ($76.47\pm0.009$ mg GAE/g extract), DPPH RSA ($IC_{50}=52.05\pm1.61\;{\mu}g/mL$), ABTS RSA ($IC_{50}=30.94\pm0.41\;{\mu}g/mL$) and RP ($IC_{50}=87.94\pm0.37\;{\mu}g/mL$). In addition, the calyx extract also showed high antioxidant activity. On the other hand, the core extract gave the lowest TPC and all antioxidant assays. In particular, HT-29 cells showed extensive cell death when treated with $500\;{\mu}g/mL$ of calyx extracts. Thus, these results suggest that methanolic extracts of Jangseong Daebong persimmon seed and calyx may serve as a potential source of natural antioxidant for food and nutraceutical application.

A Study of Structure of the Sucker of Common Freshwater Goby (Rhinogobius brunneus) and Triden Goby (Tridentiger brevispinis) (밀어 (Rhinogobius brunneus)와 민물검정망둑 (Tridentiger brevispinis)의 흡반 구조에 관한 연구)

  • The structures of sucker of two Cobiidae; Common freshwater goby and Triden goby were observed by light and electron microscopy. Scanning electron microscopy revealed the characteristic narrow ridges and grooves on the apical portion of sucker of Common freshwater goby, and hexagonal structures similar to a honeycomb representing the intercellular junctional area on the middle and basal portions. Some ridges were present on the epithelial surface on the middle and basal portions. The openings of several mucus-secreting cells were present between main epithelial cells. Light and transmission electron microscopy revealed the core of the fin; soft rays with a surrounding dense collagen fiber layer. Some loosely arranged fibers (collagen fiber) radiated toward the surface epithelium. The surface epithelium was cuboidal or columnar in shape. Scanning electron microscopy revealed the coiled irregular ridges and grooves, which was less developed and had sparser distribution than in Common freshwater goby, on the apical portion of sucker of Triden goby. The middle and basal portions had honeycomb structures as in Common freshwater goby. Fewer mucoussecreting cells were present. Light and transmission electron microscopy showed the core of soft rays, dense collagen fiber layer, however, the radiating fibers observed in the Common freshwater goby was rarely present. The sucker was thinner because the epithelium is squamous or polygonal in shape and rare presence of the radiating fibers.

Enzymatic Synthesis of Dithiolopyrrolone Antibiotics Using Cell-Free Extract of Saccharothrix algeriensis NRRL B-24137 and Biochemical Characterization of Two Pyrrothine N-Acyltransferases in This Extract

  • Saccharothrix algeriensis NRRL B-24137 produces naturally different dithiolopyrrolone derivatives. The enzymatic activity of pyrrothine N-acyltransferase was determined to be responsible for the transfer of an acyl group from acyl-CoA to pyrrothine core. This activity was also reported to be responsible for the diversity of the dithiolopyrrolone derivatives. Based on this fact, nine dithiolopyrrolone derivatives were produced in vitro via the crude extract of Sa. algeriensis. Three of them have never been obtained before by natural fermentation: acetoacetyl-pyrrothine, hydroxybutyryl-pyrrothine, and dimethyl thiolutin (holomycin). Two acyltransferase activities, acetyltransferase and benzoyltransferase catalyzing the incorporation of linear and cyclic acyl groups to the pyrrothine core, respectively, were biochemically characterized in this crude extract. The first one is responsible for formation of acetyl-pyrrothine and the second for benzoyl-pyrrothine. Both enzymes were sensitive to temperature changes: For example, the loss of acetyltransferase and benzoyltransferase activity was 53% and 80% respectively after pre-incubation of crude extract for 60 min at $20^{\circ}C$. The two enzymes were more active in neutral and basal media (pH 7-10) than in the acidic one (pH 3-6). The optimum temperature and pH of acetyltransferase were $40^{\circ}C$ and 7, with a $K_m$ value of $7.9{\mu}M$ and a $V_{max}$ of $0.63{\mu}M/min$ when acetyl-CoA was used as limited substrate. Benzoyltransferase had a temperature and a pH optimum at $55^{\circ}C$and 9, a $K_m$ value of $14.7{\mu}M$, and a $V_{max}$ of $0.67{\mu}M/min$ when benzoyl-CoA was used as limited substrate.
