  • Samyang Genex succeeded in commercialization of anticancer agent-paclitaxel by plant cell culture technology. The core technology of Samyang Genex relating paclitaxel production includes cell line development, cell line preservation, cell culture, scale-up technology, and purification technology. On the basis of the research, Samyang Genex built the factory operated by CGMP (current good manufacturing practice). The $paclitaxel-Cenexol^{TH}-is$ commercially available in Korea, and it will be launched to world market including USA after approval of US FDA.

  • The centrosome is a subcellular organelle from which a cilium assembles. Since centrosomes function as spindle poles during mitosis, they have to be present as a pair in a cell. How the correct number of centrosomes is maintained in a cell has been a major issue in the fields of cell cycle and cancer biology. Centrioles, the core of centrosomes, assemble and segregate in close connection to the cell cycle. Abnormalities in centriole numbers are attributed to decoupling from cell cycle regulation. Interestingly, supernumerary centrioles are commonly observed in cancer cells. In this review, we discuss how supernumerary centrioles are generated in diverse cellular conditions. We also discuss how the cells cope with supernumerary centrioles during the cell cycle.

  • A new method of calculating sensitivity coefficients of core characteristics relative to infinite-dilution cross sections has been developed. Conventional sensitivity coefficients are evaluated for the changes of effective cross sections which are dependent on individual models of core and cell. Therefore a correction has been derived to the conventional sensitivity coefficients based on the perturbation theory. The accuracy of the present method has been verified by comparing numerical results of sensitivity coefficients with a reference Monte-Carlo method.

태양전지용 CdSe 나노입자의 합성 (Preparation and Characterization of CdSe nanoparticle for Solar Cell application)

  • CdSe nanoparticles were prepared by chemical solution methods using $CdCl_2{\cdot}4H_2O$ (or $Cd(NO_3)_ 2{\cdot}4H_2O$) and $Na_2SeSO_3$. The characteristics of CdSe nanoparticles were controlled by the react ion time, reaction temperature and reaction method as well as the surfactants. Cetyltrimethyl ammonium bromide(CTAB) was used as a capping agent to control the chemical reactions in aqueous solution. Polyvinylalcohol(PVA) was used as a templet in sono-chemical method. CdSe nanoparticles synthesized in aqueous solution showed homogeneous size distribution with relatively stable surface. CdSe nanoparticles synthesized in non-aqueous solution containing diethanolamine(DEA) showed the structure transformation from cubic to hexagonal as the reduction temperature increased from 80 to $160^{\circ}C$. Core shell CdSe was synthesized by sono-chemical method. Characteristics of CdSe nanoparticles were analyzed using transmission electron microscopy(TEM), x-ray photoelectron spectroscopy(XPS), x-ray diffraction(XRD), UV-Vis absorption spectra, fourier transform infrared spectroscopy(FT-IR) and photoluminescence spectra spectroscopy(PL). This paper presents simple routes to prepare CdSe nanoparticles for solar cell applications.

  • 고분자전해질 연료전지의 성능은 매우 복잡한 물리 현상들에 의해 변화하게 된다. 반응면적이 25 $cm^2$인 5-pass, 4-turn 사행성 유동유로의 립 부분에 보조유동유로를 가지는 형상에 대하여 물관리 측면에서의 연료전지 성능을 수치해석을 통해 비교하였다. 보조유동유로를 추가함에 따라 촉매층 공급 대류의 유동 방향이 변경되어 유로 내부의 물 배출 특성을 향상시키는 결과를 나타내었다. 또한 입구에서의 공급기체를 보조유동유로로 분산시킴에 따라 입구에서의 전류 밀도는 낮아지며 보조유동유로로 이동하는 공급기체들은 주 유동유로의 내에서의 체류시간보다 길어져서 전체적인 전류밀도 분포가 균일해지는 것을 확인하였다.

  • 포유동물 세포내에서 간염 바이러스의 내면항원의 발현과 전위내면 항원(precore) 부위의 역할을 규명하기 위하여 고등동물세포 발현용 벡터에 전위내면항원 부위를 갖거나 또는 갖지 않는 내면항원 유전자를 클로닝 하여 COS 세포내에서의 발현을 조사하였다. 전위내면항원 부위를 포함한 내면항원 유전자를 갖는 플라스미드로감염시킨 COS 세포는 항원들이 세포추출물과 배양액에서 검출되었다. 분비된 항원의 증가율은 감염후 2일과 3일 사이가 가장 높았고, 부분결실된 제조합 플라스미드 중 내면항원의 ATG codon에서 180bp 떨어진 것이 가장 발현이 잘 되었다. 전위내면항원을 갖지 않거나 하나의 염기가 첨가되어 변형된 전위내면항원을 갖는 제조합 플라스미드의 경우 항원들이 세포 추출물에서만 검출되었다. 이러한 사실은 전이내면항원 부위가 HBe 항원의 분비에 관여 한다는 경우 항원들이 세포 그러나 대장균이나 효모 세포의 경우는 전위내면항원의 존재와 상관없이 항상 세포추출물에서만 존재하는 것으로 보아 이들 세포의 경우에서는 전위내면항원 부위가 HBe 항원의 분비에 영향을 줄 수 없음을 의미한다.

Dual TORCs driven and B56 orchestrated signaling network guides eukaryotic cell migration

  • Different types of eukaryotic cells may adopt seemingly distinct modes of directional cell migration. However, several core aspects are regarded common whether the movement is either ameoboidal or mesenchymal. The region of cells facing the attractive signal is often termed leading edge where lamellipodial structures dominates and the other end of the cell called rear end is often mediating cytoskeletal F-actin contraction involving Myosin-II. Dynamic remodeling of cell-to-matrix adhesion involving integrin is also evident in many types of migrating cells. All these three aspects of cell migration are significantly affected by signaling networks of TorC2, TorC1, and PP2A/B56. Here we review the current views of the mechanistic understanding of these regulatory signaling networks and how these networks affect eukaryotic cell migration.

  • Sandwich sheet with inner structure is expected to find many applications because of high stiffness to mass ratio. In order to simulate forming of sandwich sheet with pyramid core, an effective simulation method is required. Compared to the expensive model using solid elements, cost effective model using simplified elements such as shells and beams is developed. By comparing two models in terms of the cost and accuracy for unit cell deformation, a developed model shows some advantages over the model using solid elements. Evolution of two kind of forming limits, face buckling and core buckling are successfully expressed by developed model. Developed model is also applied in the simulation of square cup drawing and L-type bending. The corresponding experiments are carried out. Deformation shape and wrinkling behavior are compared and discussed. It is found that simulation results using a developed model are in good agreement with experiments.

In vitro Anticancer Activity of Paclitaxel Incorporated in Low-melting Solid Lipid Nanoparticles

  • Triglyceride solid lipid with medium chain fatty acid, tricaprin (TC), was used as a core matrix of lipid nanoparticles (LN) to solubilize water-insoluble paclitaxel and enhance the stability of nanoparticles by immobilization of incorporated drug in the solid core during storage at low temperature. In the present study, TC-LN containing paclitaxel was prepared by hot melt homogenization method using TC as a core lipid and phospholipids as stabilizers. The particle size of TC-LN containing paclitaxel was less than 200 nm and its zeta potential was around -40 mV. Calorimetric analysis showed TC core could be solidified by freezing and thawing in the manufacturing process in which the hot dispersion should be prepared at elevated temperature and subsequently cooled to obtain solid lipid nanoparticles. The melting transition of TC core was observed at $27.5^{\circ}C$, which was lower than melting point of TC bulk. The particle size of TC-LN remained unchanged when kept at $4^{\circ}C$. Paclitaxel containing TC-LN showed comparable anticancer activity to the Cremophore ELbased paclitaxel formulation against human ovarian (OVCAR-3) and breast (MCF-7) cancer cell lines. Thus, lipid nanoparticles with medium chain solid lipid may have a potential as alternative delivery system for parenteral administration of paclitaxel.
