Genetic Relationship of Cordyceps spp. Based on Internal Transcribed Spacer Sequences of Ribosomal DNA (Ribosomal DNA의 ITS 염기서열에 의한 동충하초속균의 유연관계)

  • The genetic relationships among six Cordyceps spp. were investigated based on internal transcribed spacer sequences of ribosomal DNA .A portion of the these genes was amplified by PCR. Approximately 590 base pairs were successfully amplified, cloned, sequenced, compared. The nucleotide sequence of the six amplified fragments were aligned by the clustal W program. As a result, Cordyceps militaris shared 87, 96, 98, 90 and 97% sequences homology with Paecilomyces japonica, Paecilomyces sp. J300, Paeciomyces farinosa. Paecilomyces sp. J500 and Cordyceps sinensis, respectively. Paecilomyces japonica also shared 87, 88, 92 and 87% sequence similarity with Paecilomyces sp. J300, Paecilomyces farinosa, Paecilomyces sp. J500 and Cordyceps sinensis, repectively.

Excellent Strains Selection from Wild Cordyceps spp.(II) (야생동충하초로부터 우량균주선발(II))

  • Many Cordyceps spp. were collected for excellent strains selection at near Wanju-kun from May to October, 2000. They were identified. As the resulting, They were 2 genera and 4 species. 6 strains were got through 4 species culture. 6 strains are Cordyceps militaris f. albino, C, gracilaides, C. militaris and Isaria japoica. Among them, 4 species used for induction of artifitial fruiting body. 3 strains of them were selected for excellent strains. excellent strains are Cordyceps militaris and Isaria japoica.

  • Hepatoprotective effects of polysaccharides extracted from liquid cultured mycelia were screened by measuring the glutamic pyruvate transaminase activity of the primary cultured rat hepatocytes intoxicated with carbon tetrachloride. Sixty among 75 isolates of higher fungi showed to be hepatoprotective effect, and these were 13 of Ganoderma lucidum, 5 of Lentinus edodes, 1 of Pleurotus ostereatus, 4 of Coriolus versicolor, 2 of Lyophyllum spp., 7 of Grifora frondosa, 3 of Agaricus spp., 14 of Schizophyllum commune and 11 of Cordyceps spp.. Especially, 10 isolates, Ganoderma lucidum IY003 and IY009, Lentinus edodes IY103, Lyophyllum sp. IY402, Agaricus sp. IY701 and IY703, Schizophyllum commune IY804, IY810 and IY818, Cordyceps sp. IY902, were indicated below 80% of glutamic pyruvate transaminase activity.

  • This study was carried out to identify the genetic characteristics among isolates of Paecilomyces spp.and Cordyceps spp. by URP-PCR analysis. Twenty URP (universal rice primer) primers of 20 mer which were designed from repetitive sequence of rice, were used for producing PCR DNA fingerprints of the mushrooms. Of them, 5 URP primers, URP2F, URP2R, URP9F, URP4R, and URP17R amplified genomic DNA of the mushrooms with polymorphic PCR patterns. On isolates of Cordyceps militaris, primers URP1F, URP2R, URP6R and URP17R produced PCR polymorphic bands of 4 types. Isolates of Cordypces sp. that are isolated from different area of Korea were identical to isolate of C. militaris, while other species of Cordypces were different to the PCR profiles. However, the URP primers did not identify the polymorphism of PCR profile on isolates of P. japonica.

Antibacterial Activities of Cordyceps spp., Mugwort and Pine Needle Extracts (동충하초, 쑥 및 솔잎 추출물의 항균작용)

  • The purpose of this study was to investigate antibacterial activity of extracts of Cordyceps spp.(Paecilomyces japonica and Cordyceps militaris), mugwort and pine needle. Fruiting body and mycelium of Cordyceps spp., mugwort and pine needle were extracted with water and 70% ethanol. Antibacterial activities of each extracts against 3 kinds of Gram positive (Bacillus subtilis, Listeria monocytogenes and Staphylococcus aureus) and 3 kinds of Gram negative pathogenic bacteria(Escherichia coli O157 : H7, Shigella sonnei and Salmonella typhimurium) were tested. The yields of water and ethanol extracts of fruiting body (39∼58%) were 2.4 ∼4.4 times higher than mycellium(9∼24%) in Cordyceps sup., while those of mugwort and pine needle were less than 9%. Ethanol extract of P. japonica mycelium(JFE) had antibacterial to S. monocytogenes at 1% level and ethanol extract of C. militaris fruiting body (MFE) had antibacterial to S. aureus at 3% level. Ethanol extract of mugwort was antibacterial against L monocytogenes and S. aureus at 1% level. Water extracts of Cordyceps spp.(P. japonica and C. militaris) and mugwort had no antibacterial activity against tested bacterial strains. Water extract of pine needle had antibacterial activity against all bacterial strains except E. coli and ethanol extract had antibacterial activity against all tested bacterial strains at 1% level. Pine needle extracts had the most wide antibacterial spectrum against bacterial strains used for this experiment. Growth inhibiting activities of pine needle extracts were higher in ethanol extract than water extract for most of tested bacteria in tryptic soy broth.

  • Many Cordyceps rpp were collected for excellent strains selection at Mts. Moak, Younsuk and Daea-ree herbarium near Wanju-kun from mid May to October,1999. They were identified as the resulting 2 genera and 5 species, 10 strains were got through 5 species culture. Among them 4 species used for induction of artifitial fruiting body and 6 strains of them were selected to excellent strains: Cordyceps militaris (99-0007), Isaria japonica (99-0001, 99-0002, 99-0003), Cordyceps soborifera (99-0008), Cordyceps kanzashiana(99-0009).

Identification and Antibacterial Activity of Volatile Flavor Components of Cordyceps Militaris

  • Flavor characteristics of raw Cordyceps militaris significatntly different from those of dried one. In the case of raw Cordyceps militaris , major flavor components were composed of 5 alcohols, 3 ketones, 4 phenols, 9 alkanes , and 3 alkenes. The major alcohol was 1-octen-3-ol(22.56%, 1147.3% ng/ml), which contributed to the characteristic green flavor. Ketones (3-ocatone, inparticular )were present in the highest concentration in raw Cordyceps militaris . In contrast, major flavor components of dried Cordyceps militaris were composed of 4 alcohols, 4 ketones, 3 furans, 4 pyrizines, 2 dithiazines, 5 phenols , 8alkenes , 17 alkanes, and 8 fatty acids. Dried Cordyceps militaris had unique sweet aroma of sesame as wella s a milky flavor. Green or fruit flavor were rarely detected . In alkanes , 10 cosanes, component fo wax were present. Typical flavor components of alkanes such as $\beta$-caryophyllen and Δ-cadinene were also detected. Fatty acids of dried Cordyceps militaris ranged from myristic acid (14 :0) to linoleic acid (18 ; 2). The sweet aroma of dried Cordyceps militaris was mostly due to pryazines, dithaiazines, and furans. Two dithaizines were identified and characteristics of these flavor components was a roasted bacon flavor. Strong antibacterial acitivity was observed toward Vibrio spp. such as V. vulnificus, V.cholerae, V. parahaemlyticus. Relatively high antibacterial acitivity was shown toward Bacillus subtilis , B,cereus, Staphyllococcus aureus, and Corynebacterium xerosis.

Classification of Isolates of Ganoderma lucidum, Schizophyllum commune and Cordyceps spp. by Electrophoretic Patterns of Isozymes (Isozyme patterns 차이에 의한 영지, 치마버섯 및 동충하초의 계통분류)

  • To classify fungal species employed for pharmacological effects, mycelial proteins of six isolates of Ganoderma lucidum, five isolates of Schizophyllum commune and five isolates of Cordyceps spp. were separated on polyacrylamide gel to compare them by esterase, acid phosphatase, leucine aminopeptidase and peroxidase patterns. Similarity of isozyme patterns among the isolates of G. lucidum IY003, IY004, IY005 and IY008 was indicated over 70%, but that among the isolates of G. lucidum IY009, IY010 and others was indicated from 48% to 9%. Highest similarity of isozymes of S. commune was observed to be between IY803 and IY805, and similarity between these two isolates was 57%. Similarity among other isolates was shown to be from 40% to 56%. Isozyme patterns of Cordyceps spp. were comparatively different, even though they were originated from the same kind of insect as their isolate. Similarity between Cordyceps spp. IY901 and IY904, which was isolated from moths, was 67% and that of IY905 and IY909, which was originated from the larvae, was 42%. Similarity among other isolates was shown to be from 12% to 67%.
