Distribution and Taxonomy of Entomopathogenic Fungal Species from Korea (한국에서 채집된 동충하초의 분포와 분류)

  • Thirty three entomopathogenic species belonging to twelve genus were collected throughout 16 collecting sites from 1990 to 1996. Among those collected species, 14 unrecorded species such as Cordyceps bifusispora, C. martialis, C. oxycephala, C. paludosa, C. pentatomi, C. rosea, C. ryogamiensis, Shimizuomyces paradoxa, Akanthomyces aculeatus, Polycephalomyces ramosus, Tilachlidiopsis nigra were added to Korean entomopathogenic species through this study. In nature, occurrence of Cordyceps nutans, C. sphecocephala and Paecilomyces tenuipes were from early June to late September. On the other hand, C. militaris, C. kyushuensis and C. pruinosa were mainly found from mid July to mid August when relative humidity are increased. Nine species of the genus Cordyceps including C. bifusispora and four deuteromycetous species were isolated. As a result of cultural test using six Cordyceps species, anamorph of C. militaris, C. kyushuensis were proved as Verticillium sp. C. pruinosa as Acremonium sp., C. sphecocephala as Hymenostilbe sp. and C. scarabaeicola as Beauveria sp., respectively.

Isolation and Identification of Entomopathogenic Fungi Collected from Mountains and Islands in Korea (한국의 산과 섬 지역에서 채집한 곤충기생 진균의 분리 및 동정)

  • Dead insects infected with fungi were collected from mountains and islands in several locations in Korea from 2003 to 2007. Morphological characteristics of the fungal fruiting bodies produced on the insect samples were examined. A total of 542 entomopathogenic fungal isolates were obtained from the insect samples and identified based on their morphological and cultural characteristics. Three species of Beauveria, eight species of Cordyceps and four species of Isaria were mainly isolated from the insect samples. The other entomopathogenic fungi isolated from the insect samples were Metarhizium anisopliae, Nomuraea rileyi, Paecilomyces sp. and Verticillium sp.

Pharmacological Activities of Paecilomyces japonica, A New Type Cordyceps sp. (눈꽃 동충하초의 약물활성)

  • Cordyceps is reputed for its broad biological activities and as a tonic for replenishing vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of a new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on hyperglycemia induced by streptozotocin(STZ) and by epinephrine in rats and in mice as well as on immunological functions in mice were investigated. The 70% methanol extract of the fungus, when administered orally at 100 and 300 mg/kg in STZ-induced hyperglycemic rats, caused a significant decrease in blood glucose level 3hr after sample treatments. The methanol extract, when administered p.o. at the same dose levels in epinephrine-induced hyperglycemic mice, also caused a significant decrease in serum glucose levels as well as a significant reversal of the liver glycogen contents suggesting its hypoglycemic activity might be due to glycogen breakdown in the liver. Treatment of normoglycaemic mice with the methanol extract of the fungus exhibited a significant glucose tolerance up to 3hr after oral glucose load(2.0 g/kg). The methanol extract also showed immuno-stimulating activity as measured by carbon clearance in mice and a significant antifatigue effect as measured by weight loaded forced swimming performance in mice.

Report on the Genus Cordyceps and Its Anamorphs Collected in Korea (한국자생 동충하초의 채집 및 자원보고)

  • Morphological characteristics of Cordyceps species and its allies collected in Korea were clarified. Through the survey conducted from June 1999 to October 2002 in 19 mountains in Korea, 667 samples of entomogenous fungi were collected. Cordyceps and its allies of 17 species of 5 genera were identified as Cordyceps gracilioides, C. japonica, C. longissima, C. martialis, C. militaris, C. myrmecophila, C. nutans, C. pruinosa, C. sphecocephala, C. tricentri, Hirsutella nutans, Paecilomyces cicadae, P. farinosus, P. tenuipes, Paecilomyces sp., Shimizuomyces paradoxa, Tilachlidiopsis nigra. The fungi with insect hosts have been collected mainly in the place of shade or mosses near brooks and streams that had high humidity. Overall the frequenct of fungal infection in natural ecosystem was relatively low as few as 10 collections per each species. However, many species were found in terms of the few number of colleciton sites with seasonal limitations. Occurrence of the fungi in Jeju island remote from inland of the Korean peninsula were diverse in their species due to the varied weather of vertical distribution following the altitude. Three most common species were C. nutans, P. tenuipes and C. militaris, mainly found early in August when the relative humidity and temperature were high, of which C. nutans occupied the highest frequency consisting of 65% in total collections. Neither variation in ascomata arrangement in stromata nor developement of secondary spores was recognizable, while the number, shape and colour of stromata varied with insect hosts and weather conditions.

  • A cordycipitoid fungus infecting Hepialidae sp. in Nepal was supposed to be identical to Cordyceps liangshanensis, originally described from southwestern China, and thus, transferred to the genus Metacordyceps or Papiliomyces in previous studies. However, our multi-gene (nrSSU-nrLSU-tef-1α-rpb1-rpb2) phylogenetic and morphological studies based on the type specimen and additional collections of C. liangshanensis revealed that the fungus belongs to the genus Ophiocordyceps (Ophiocordycipitaceae). Therefore, a new combination O. liangshanensis was made, and a detailed description of this species was provided.

Genetic Diversity of Paecilomyces japonica and Cordypces militaris Strains by URP-PCR Fingerprinting (URP-PCR핵산지문에 의한 눈꽃동충하초 (Paecilomyces japonica.)와 번데기동충하초(Cordypces militaris) 유전적 다양성분석)

  • This study was carried out to identify the genetic characteristics among isolates of Paecilomyces spp.and Cordyceps spp. by URP-PCR analysis. Twenty URP (universal rice primer) primers of 20 mer which were designed from repetitive sequence of rice, were used for producing PCR DNA fingerprints of the mushrooms. Of them, 5 URP primers, URP2F, URP2R, URP9F, URP4R, and URP17R amplified genomic DNA of the mushrooms with polymorphic PCR patterns. On isolates of Cordyceps militaris, primers URP1F, URP2R, URP6R and URP17R produced PCR polymorphic bands of 4 types. Isolates of Cordypces sp. that are isolated from different area of Korea were identical to isolate of C. militaris, while other species of Cordypces were different to the PCR profiles. However, the URP primers did not identify the polymorphism of PCR profile on isolates of P. japonica.

Genomic Structure of the Cu,Zn Superoxide Dismutase (SOD1) Gene of Paecillomyces tenuipes and Paecilomyces sp.

  • We describe here the complete nucleotide sequence and the exon-intron structure of the Cu,Zn superoxide dismutase (SOD1) gene of Paecilomyces tenuipes and Paecilomyces sp. The SOD1 gene of P. tenuipes spans 966 bp, and consisted of three introns and four exons coding for 154 amino acid residues. Three unambiguous introns in P. tenuipes separate exons of 13, 332, 97, and 20 bp, all exhibiting exon sizes identical to Cordyceps militaris SOD1 gene. The SOD1 gene of Paecilomyces sp. contains 946 bp and consisted of four introns and five exons coding for 154 amino acid residues. Five exons of Paecilomyces sp. SOD1 are composed of 13, 180, 152, 97, and 20 bp. Interestingly, this result showed that the total length of exons 2 (180 bp) and 3 (152 bp) of Paecilomyces sp. SOD1 is same to exon 2 length (332 bp) of C. militaris SOD1 and P. tenuipes SOD1. The deduced amino acid sequence of the P. tenuipes SOD1 showed $95\%$ identity to C. militaris SOD1 and $78\%$ to Paecilomyces sp. SOD1. Phylogenetic analysis confirmed that the C. militaris SOD1, P. tenuipes SOD1 and Paecilomyces sp. SOD1 are placed together within the ascomycetes group of fungal clade.

Changes in Nutritional Composition of Gryllus bimaculatus Fermented by Bacillus sp. and Mycelium of Basidiomycetes (고초균 및 버섯 균사체를 이용하여 발효한 쌍별귀뚜라미 발효물의 단백질 및 무기질 성분 변화)

  • This study examined the changes in the protein and mineral composition of Gryllus bimaculatus fermented with Bacillus substilis and the mycelia of Basidiomycetes. Normal Gryllus bimaculatus (S) and experimental group data obtained after an inoculation of Bacillus substilis (SC) (KACC 19623), Pleurotus eryngii (SP) and Cordyceps millitaris (SC) were compared. The crude protein content of the Gryllus bimaculatus (control) was 75.48%, but it decreased to 64.55, 54.32, and 63.53% after fermentation with SB, SP and SC, respectively (p<0.05). An analysis of the organic elements showed that the contents of the carbon and nitrogen sources were also reduced after fermentation, and the most significant decrease was observed after fermentation with SP. In SDS-PAGE, a 120 kDa and a 48 kDa protein of Gryllus bimaculatus were found. On the other hand, protein bands faded after fermentation with SP and SC, respectively. Moreover, no visible band was observed after fermentation with SB. According to amino acid analysis, the total free amino acid content increased 3.84 and 1.74 times after fermentation with SB and SP, respectively, compared to the corresponding baseline data. In contrast, it decreased by 0.52 times after fermentation with SC. Among the essential amino acids found in crickets fermented with SB, the valine and isoleucine content was 3.57 and 2.64 times higher, respectively, than the recommended daily amount of essential amino acids.

Isolation of Strain for the Preparation of the Fermented Antler and Its Physiological Activities (발효녹용의 균주 선별 및 발효녹용의 생리활성)

  • This study was conducted to isolate strain for the preparation of fermented antler (Cervus cornu parvum) and evaluate its physiological activities. The growth degrees of twenty-one samples from Bacillus sp., Lactobacillius sp. and mushroom strain on antler extract agar were evaluated in this study, and Bacillus subtilis KH-15, SCB-3, Cordyceps militaris, Phellinus linteus, Inonotus obliquus 26136, and Inonotus obliquus 26147 were selected. The fermented antler extract by C. militaris had relatively higher contents of total sugar (1619.3 ${\mu}g$/mL), uronic acid (302.0 ${\mu}g$/mL), sulfated-glycosaminoglycan (S-GAGs) (119.9 ${\mu}g$/mL) and sialic acid (21.6 ${\mu}g$/mL) than any other extracts. The anti-complementary activities of all fermented antler extracts were higher than non-fermented antler extract, and among these samples, fermented antler extract by C. militaris showed the highest anti-complementary activity (inhibition of 50% total complement hemolysis, $ITCH_{50}$; 50.1% at 1,000 ${\mu}g$/mL). The ability of fermented antler extract by B. subtilis KH-15 to scavenge 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical ($IC_{50}$; 4.97 mg/mL) was significantly the highest (p<0.05), whereas the extract from I. obliquus exerted significantly (p<0.05) high 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity ($IC_{50}$; 16.98 mg/mL) among all samples. The results of this study suggest that physiological effects including immuno-modulating and antioxidant activities of the antler may be increased through fermentation process.
