The hyphal tip laccase of Coprinus congregatus which is a membrane-associated enzyme and shows diffdrdnt banding patterns of PAGE analysis when compared with the enzyme of liquid culture (Choi et al. 1987) has been successfully secreted to culture medium in liquid shake culture by lowering the pH of medium to 4.0. When the fungus is cultivated in YpSs(pH 4.0) liquid, only the hyphal tip laccase is found in the medium after 6 hr incubation and there is no liquid-type enzyme when examined by PAGE analysis.

Coprinus congregatus 의 염색체 DNA를 주형으로 사용하고 균류 laccase에서 잘 보존된 copper binding region의 염기서열을 primer 로 사용하여 PCR을 수행한 결과 144 bp 길이의 laccase 유전자 일부를 cloning 하였다. 이의 염기서열을 분석한 결과 다른 균류의 lacacse 와 60-69% 정도의상동성을 보였으며 추정된 아미노산 서열은 68-75%의 상동성을 보였다.

Coprinus congregatus를 pH 4.2로 낮춘 YpSs 액체배지에 접종할 경우 세포막연관효소인 laccase가 배양 1일만에 상등액으로 대량 생성분비된다. 분비된 효소는 매우 빠르게 효소력의 감소를 보이며 전기영동상의 이동변화가 있다. 이와 같이 변화된 전기영동상을 보이는 효소단백질을 분광광도계로 분석할 경우 정제된 효소단백질과는 다른 분광 스펙트럼을 보인다.

새로운 고체배지 응고제인 Pluronic Polyol F127을 한천대신 사용하여 C. congregatus를 배양하였고 균사 끝(hyphaltip)에 있는 phenoloxidase들의 세포내에서의 존재위치를 확인하였다. 고체배지의 균사에서 원형질체를 얻었고 concanavalin A를 처리하여 세포막을 분리하였다. 세포막 분획에 phenoloxidase들이 존재하였고 이는 이 효소가 light receptor complex의 한 부분임을 시사한다.

Coprinus congregatus에서 laccase를 과량생성하는 변이주를 대상으로 phosphinothricin 저항성을 선택표지로 사용하여 형질전환을 수행하였다. 항생물질에 대한 저항성을 부여하는 plasmid DNA(pBARGEM 7-1)를 제한효소로 가수분해하여 원형질체에 형질전환을 수행한 결과 plasmid DNA $1{\mu}g$ 당 약 500여개의 형질전환체를 얻었다. 도입된 plasmid는 형질전환체의 chromosomal DNA에 삽입되어 있음을 Southern blot으로 확인하였다.

C. congregatus를 산성 액체배지(pH4.2)에 배앙할 경우 세포막연관 laccase가 배양 초기에 대량 합성분비되었으며 분비된 효소의 작용에 의하여 melanin색소가 중성배지에 비하여 빨리 생성되었고 배양 24시간 후에 배지의 산도는 pH5.2로 증가되었다. 이 효소의 합성은 transcription inhibitor인 5-fluorouracil에 의하여 크게 억제되었다.

Coprinus congregatus has a membrane-associated laccase which is not secreted into culture media. A mutant monokaryon obtained, by U. V. irradiation followed by protoplast generation and regeneration method, was successfully isolated. When the mutant was grown on a agar plate or in a liquid medium, it secreted laccase while the wild type did not under the same growth conditions. The laccase of the mutant was compared with that of wild type did not under the same growth conditions. The laccase of the mutant was compared with that of wild type of native PAGE analysis, and showed identical mobility.
