Inhibitory Effect of Coprinus comatus Ethanol Extract on the Liver damage in Benzo(a)pyrene-treated Mice (먹물버섯 에탄올추출물이 Benzo(a) pyrene 투여에 의한 마우스의 간 손상에 미치는 영향)

  • This study was carried out to investigate the inhibiton effects of Coprinus comatus ethanol extract of edible mushroom on liver damage in benzo(a)pyrene (B(a)P) treated mice. The activities of serum aminotransferase, cytochrome P 450 and hepatic content of lipid peroxide after B(a)P treatment were increased than those of control, but those levels were significantly decreased by the treatment of Coprinus comatus ethanol extract. Whereas, the hepatic glutathione content and glutathione S transferase activity were decreased by B(a)P treatment than those of control, but those were increased by the treatment of Coprinus comatus ethanol extract. Also the activities of superoxide dismutase, catalase and glutathione peroxidase after B(a)P treatment were markedly increased than those of control, but those levels were decreased by the treatment of Coprinus comatus ethanol extract. These results suggest that Coprinus comatus ethanol extract have a protective effect on liver damage by benzo(a)pyrene through the mechanisms of decreasing lipid peroxide and activities of free radical generating enzymes.

Antimutagenic Effect of the fruiting Body and the Mycelia Extracts of Coprinus comatus (먹물버섯 자실체 및 균사체 추출물의 돌연변이 억제효과)

  • The inhibitory effect of Coprinus comatus on the mutagenicity in Salmonella assay system and SOS chromotest were studied. In Ames test, the ethanol and water extracts and the cultured mycelia fractions of Coprinus comatus did not show any mutagenicity, but the Coprinus comatus ethanol extracts showed inhibitory effects of 8 0∼90% on the mutagenicity induced by indirect mutagen of benzo(a)pyrene(B(a)P) and aflatoxin B1(AFB1) in Salmonella typhimurium TA98 and TA100. The antimutagenic effect increased with increasing concentration of the ethanol extract toward N methyl N' nitro N nitrosoguanidine(MNNG). However, the water extracts inhibited about 40∼50% against direct and indirect mutagen. The cultured mycelial filtrate of Coprinus comatus, the fractionⅡ, showed antimutagenic effect of 90% on MNNG and 25∼50% on B(a)P and AFB1. In SOS chromotest, the ethanol extracts of Coprinus comatus showed antimutagenic effect of 65∼81% on SOS function induced by 4 NQO, and the cultured mycelia fractionⅡ showed low inhibitory effect of 20∼50%.

  • Coprinus congregatus를 pH 4.2로 낮춘 YpSs 액체배지에 접종할 경우 세포막연관효소인 laccase가 배양 1일만에 상등액으로 대량 생성분비된다. 분비된 효소는 매우 빠르게 효소력의 감소를 보이며 전기영동상의 이동변화가 있다. 이와 같이 변화된 전기영동상을 보이는 효소단백질을 분광광도계로 분석할 경우 정제된 효소단백질과는 다른 분광 스펙트럼을 보인다.

  • The hyphal tip laccase of Coprinus congregatus which is a membrane-associated enzyme and shows diffdrdnt banding patterns of PAGE analysis when compared with the enzyme of liquid culture (Choi et al. 1987) has been successfully secreted to culture medium in liquid shake culture by lowering the pH of medium to 4.0. When the fungus is cultivated in YpSs(pH 4.0) liquid, only the hyphal tip laccase is found in the medium after 6 hr incubation and there is no liquid-type enzyme when examined by PAGE analysis.

Genetic Relationships of Coprinus spp. on the Basis of Sequences in ITS II Region (먹물버섯류(Coprinus spp.)의 ITS II 영역 염기서열에 의한 유연관계 분석)

  • The internal transcribed spacer II regions (ITS II) of the ribosomal DNA gene repeat from Coprinus spp. were amplified using polymerase chain reaction (PCR) and sequenced. Sequences from 11 species including Coprinus comatus, C. atramentarius, C. micaceus, C. lagopus, C. cinereus, C. rhizophorus, C. flocculosus, C. radians, and C. echinosporus were compared. The spacer regions of them were $253{\sim}275$ nucleotide in length and partially contained 5.8S and 25S. The reciprocal homologies of each ITS II sequence among these strains were in the range of $50.6{\sim}100%$. According to the analysis of ITS II sequences, Coprinus spp. were classified into three clusters. Cluster I consisted of Coprinus lagopus, C. cinereus, C. echinosporus, C. rhizophorus, C. niveus, and C. atramentarius. Cluster II comprised C. micaceus, C. flocculosus, C. radians, and C. disseminatus. On the other hand C. comatus is in Cluster III even though this species is belonging to the section Coprinus in morphological aspect. These results suggest that Coprinus comatus, which was considered as a type species of the genus Coprinus in morphological classification, gives a doubt of monophyletic evolution and is assumed to be paraphyletic or polyphyletic.

Cluster I consisted of Coprinus lagopus, C. cinereus, C. echinosporus, C. rhizophorus, and C. atramentarius. Cluster II comprised C. micaceus, C. radians, C. disseminatus, Psathyrella candolleana, and P. spadiceogrisea. On the other hand C. comatus is in Cluster III with Stropharia rugosoannulata even though this species is belonging to the section Coprinus in morphological aspect. These results suggest that taxonomic position of Psathyrella would better be inculded in genus Coprinus. Coprinus comatus, the type species of Coprinus, gives a doubt on monophyletic evolution and is assumed to be paraphyletic or polyphyletic.

  • Degenerate primers corresponding to the sequences of the copper-binding regions in the fungal laccases were used to isolatc laccase gene specific fragment by PCR in Coprinus congregahts. A 144 bp DNA hagrnent was cloned and was identified to have 60-69 % homology with other fungal laccase genes. The predicted amino acid sequcnces showed 68-75% homology with other fungal laccase proteins.

  • Coprinus cinereus resistant strain against validamycin was successfully isolated. Germination rate of oidiospores, branching pattern, and localization of actin protein of the resistant strain were compared with normal strain. The resistant strain showed better germiantion rate of oidia (about 20 fold), more frequent branching, and even actin localization on validamycin plate where actin content was severely reduced in case of normal strain.

  • Transformation of Coprinus congregatus with a linearized plasmid has been successfully carried out using phosphinothricin resistance gene as a dominant selectable marker. The transforming frequency was about 500 transformants per microgram of DNA using the protoplast-$CaCl_2$ method. The transforming vector pBARGEM 7-1 which had the phosphinothricin resistance gene was detected in the restriction enzyme fragments of chromosomal DNA from a transformant by Southern hybridization.

  • An inky cap, Coprinus comatus synthesizes and secretes a laccase in the liquid yeast extract peptone dextrose medium. We have successfully purified the enzyme through the ion-exchange chromatography and the preparative gel electrophoresis. The estimated molecular weight was 67 kDa by the SDS-PAGE analysis. Optimum pH was pH 4.3 and optimum temperature was $25^{\circ}C$. The Km value was 0.45 mM and the Vmax was 0.0132 OD/min/unit for o-tolidine. Purified laccase showed 49.3% decolorizing activity against remazol brilliant blue R (RBBR) and 41.6% decolorizing activity against Poly R-478 after 12 h incubation.
