Effects of Copper, Zinc and Cadmium on the Recovery Pattern of Aryl Sulfotransferase IV Activity in Rats fed 2-Acetylaminofluorene Diet

  • Purified rat liver aryl sulfotransferase IV (AST IV) was found to be inhibited in vitro by zinc, copper, cadmium and terbium. Among these four elements, zinc, copper and cadmium were all strongly inhibitory to the AST IV activity at very low concentrations (2.5 $\mu$M to 0.025 $\mu$M). In rat liver cytosol, zinc, copper and cadmium at 25 $\mu$M to 0.025 $\mu$M also decreased the AST IV activity to $50\%$ of the controls. In order to assess the possible effects of these metals on the AST IV activity recovery pattern in vivo, studies on the relationship between these minerals and dietary 2-acetylaminofluorene were conducted. Total of forty rats were fed one of five diets for 6 weeks: diet 1, Control diet plus 2-acetlyaminofluorene ($0.05\%$); diet 2, zinc-deficient diet plus 2-acetlyaminofluorene; diet 3, zinc-supplement diet plus 2-acetylaminofluorene; diet 4, copper-supplement diet plus 2-acetylaminofluorene; diet 5, cadmium-supplement diet plus 2-acetylaminofluorene. Half of the rats from each diet were changed to individual diet after 3 weeks of 2-acetylaminofluorene feeding. Placement of rats on the control diet following one cycle of 2-acetylaminofluorene feeding of 3 weeks without 2-acetylaminofluorene resulted in nearly full recovery of AST IV activity within 3 or 4 weeks. However, the rats fed diets that supplemented with zinc, copper or cadmium without 2-acetylaminofluorene showed a new pattern of lowered AST IV activity as early as the first cycle. Also, lowering in cytosolic AST IV contents was appeared in the livers from the rats, following one cycle of 2-acetylaminofluorene feeding of 3 weeks, fed one of the diets that supplemented with copper, cadmium or zinc without 2-acetylaminofluorene for ensuing 3 weeks.

  • To estimate the influence of plasma copper and zinc concentration on infertility, we measured copper and zinc levels in plasma of 117 infertile women and 14 infertile men by atomic absorption spectrophotometry from January, 1987 through June, 1987 and the results were as follows. 1. The mean plasma copper level was significantly lower in infertile women than in fertile control women while mean plasma zinc level significantly higher in infertile women than in fertile control women. 2. A significant difference in the mean plasma level between infertile women with tubal and endometrial factor and normal controls was noted. 3. The mean level of copper in plasma of azoospermic patients was significantly higher than in fertile control subjects. 4. In fertile control and infertile women, no diurnal and menstrual cyclic variations in plasma zinc and copper levels were present, respectively. Also, there were no significant correlation between plasma zinc and copper levels in all studied group of subjects.

Zinc and Copper Status of Middle- and Old-Aged Women in Type 2 Diabetes (중.노년기 제2형 당뇨병 여성의 아연 및 구리 영양상태)

  • The purpose of this study was to assess the zinc and copper status of type 2 diabetic women and to analyse the relation-ship among zinc and copper status, and diabetic control indices of diabetes. The mean age of diabetes was 57.9 years old. The mean duration of diabetes was 8.0 $\pm$ 6.5 years. The mean daily energy intake of diabetes was 1562 kcal. There were no significant differences of age, BMI, %body fat, nutritional intakes, total energy intake, and energy composition from carbohydrate, protein, and fat between diabetes (n = 50) and control group (n = 68). However, both zinc intake density (4.15 mg/1000 kcal) and zinc %RDA (62.0%) of diabetes were significantly lower than control group (p <0.05, P < 0.01, respectively). The plasma zinc level was not significantly different between diabetes and control group (90$\mu$g/dl, 91$\mu$g/dl respectively). The proportion of diabetic women whose plasma zinc levels were lower than 76 $\mu$g/dl(borderline zinc deficiency) was 18.8%. This was about 38% higher than control group. It has been suggested that insulin secretion might decrease in borderline plasma zinc and copper deficiency and increase in normal plasma zinc and copper status. The urinary zinc excretion was twice higher in diabetes than in control group (p < 0.001). The urinary zinc loss was positively correlated with the duration of diabetes (p < 0.05), hyperglycemia (p < 0.001) and insulin resistance (p < 0.05). These results lead us to conclude that normal blood glucose level controlled by diet therapy could improve the hyperzincuria in diabetic women.

Nutritional Status of Zinc and Copper in Type 2 Diabetic Patients after Short-term Zinc Supplementation (제 2형 당뇨병 환자에서 단기간 아연 보충에 따른 아연과 구리 영양상태)

  • This study was carried out to determine whether a short-term zinc supplementation could improve the zinc status without adverse changes in copper status among type 2 diabetic patients. Seventy-six diabetic subjects and 72 normal adults participated in this study. Subjects were randomly divided into supplemented and control groups. Forty-four diabetic patients and 34 normal subjects were supplemented with 50 mg zinc gluconate daily for 4 weeks. Dietary intakes of participants were measured for two non-consecutive days by 24-hour recall method. Nutritional status of zinc and copper were also evaluated by biochemical measurement of fasting plasma samples and spot urinary collection. At baseline, diabetic patients showed significantly lower levels of dietary zinc intake and higher urinary zinc excretion than the normal adult group(p<0.05, p<0.0001). Plasma level of zinc was not significantly different between diabetic and normal adults at baseline. However, plasma zinc level increased significantly in both diabetic patients and normal adults after zinc supplementation. The changes in plasma copper levels following zinc supplementation were not statistically significant in diabetic subjects as well as in normal adults. These results indicated that four weeks of zinc supplementation did not influence Cu status and that it may contribute to improving the zinc status. Therefore, we suggest that Zn supplementation for a short-term period may improve marginal zinc status of diabetic patients without interfering with their copper status

  • The zinc, copper and chromium contents in the hair in 103 healthy Korean and 126 Korean with schizophrenic(aged 20 to 59 years in Yanbian district)by atomic absorbtion spectrometer(Type WFZ-II). Research Period : from May 1985 to September 1986. The zinc, copper and chromium contents in the schizophrenic hair are significantly lower than those of healthy persons, respectively. The standard rates of discriminant from healthy persons and the patients by method of (Department of Pathophysiology, Yanbian Medical Colloge ; Yanbian Neuropsychosis Hospital, Yanji Jilin Prov, P.R. China) Fisher discrominant analysis are 72.9% and 84.4%, respectively. Study on Zinc and Copper intakes of Breast-fed Infants (모유 영양아의 아연과구리 섭취량에 관한 연구)

  • The purpose of this study is to investigate zinc and copper intakes of exclusively breast-fed infants. We measured consumed volume and the minerals concentrations of human milk from thirty three lactating women and their infants during the first 5 months of lactation , Zinc concentrations of human milk showed 0.394, 0.290, 0.191, 0.147 and 0.139mg/100g at 0.5, 1, 2, 3, 4 and 5 months of lactation respectively. Copper concentrations in the milk showed 61.04, 45.97, 37.15, 3.94, 26.22 and 26.21$\mu\textrm{g}$/100g respectively. Consumed volume of the milk of infants were 515, 650, 718, 731, 746 and 769g/day. There is significant difference between boys and girls in consumed volume of the milk at 2, 3 and 4 months of lactation. Zinc intakes of breast-fed infants declined(p<0.05) from 2.13mg/day at 0.5 months to 1.21mg/day at 3 months with no change thereafter significantly. Copper intakes of the infants declined(p<0.05) from 323.2$\mu\textrm{g}$/day at 0.5 months to 238.9$\mu\textrm{g}$/day at 3 months with no change thereafter. Form this survey, the reevaluation of zinc and copper intakes and recommended dietary allowance of those minerals during infancy are merited. (Korean J Nutrition 33(8) : 857-863, 2000)

  • In order to explain the positive catalytic action of copper compound for the rate of leaching of zinc sulfide minerals, the electrode and redox potentials of both synthetic and natural sulfides were measured at various conditions of temperatures and pressures. The potentials of Chalcopyrite and copper sulfide were considerably higher than that of zinc sulfide, whereas lead sulfide and Galena had slightly lower potentials than that of zinc sulfide. At elevated temperatures and pressures, the same tendency was obtained. By means of comparing the calculated and measured values of potentials for sulfides, it was suggested that the electrode potentials in acid solution were generated by oxidation of sulfur ion. As a result, it was concluded that the catalytic action of copper compound in the leaching of synthetic zinc sulfide should be arised from the galvanic action between sulfides keeping intimate contact one another in which copper sulfide worked as cathodic and zinc sulfide as anodic part analogous to the metal corrosion under galvanic action.

  • Research was conducted to determine the micromineral status of grazing cattle in three climatic regions of the province of South Sulawesi, Indonesia. Soil, forage, blood and liver samples were collected within the Western, Central and Eastern regions in February-March and August-September of 1987. Forages were deficient in copper, selenium and zinc. Serum copper was deficient in all regions but zinc was deficient more in the dry season. Liver copper was deficient in all regions and both seasons except in the central region in the rainy season. Based on the analyses, microminerals most likely deficient in both the rainy and dry seasons for each region were as follows: Western-copper, selenium and zinc; Central-manganese and copper; Eastern-selenium, copper and zinc. Some degree of molybdenum excess was found in the Eastern region.

  • Optimum condition for neutralization has been studied to remove the copper and zinc ions from spent sulfate solutions which resulted from the acid washing of the base of brass bulb. Chemical distribution of copper and zinc species and the variation of solubility of the two ions with solution pH were obtained by considering the complex formation reaction and mass balance. Removal percentage of zinc was more sensitive to solution pH than that of copper. This results from the fact that the solubility of zinc is higher than that of copper. The form of coagulant affected little the removal percentage of zinc, while addition of coagulant as the phase of solution resulted in higher removal percentage of copper than as the phase of solid.

  • The purpose of this study was to investigate the dietary habits, nutritional status, and serum copper and zinc concentrations of adolescent athletes. 50 high school adolescent athletes (29 male, 21 female) took part in physical education high school and 47 high school adolescent non-athletes (21 male, 26 female) in general high school. Questionnaires, anthropometric measurements, 24-hour dietary recalls and blood analysis were conducted. In the males, the mean height, weight and body fat percents of athletes and non-athletes were 174.0/172.9 cm, 67.4/68.3 kg, and 8.9/20.0% (p<0.001) respectively. In the females, the mean height, weight and body fat percents of athletes and non-athletes were 163.7/159.3 cm (p<0.01), 58.7/55.7 kg (p<0.05), and 18.6/30.1 % (p<0.001) respectively. In both male and female athletes, intakes of animal fat were significantly higher and intakes of vitamin C were significantly lower than those of non-athletes. There was no significantly difference in zinc and copper intakes between athlete and non-athlete groups. The average serum zinc level of male athletes was significantly lower than that of male non-athlete. The average serum copper levels of male and female athletes were significantly lower than those of male and female non-athletes. Based on these results, exercise may effect on zinc and copper utilization of adolescent. Further studies on zinc and copper nutrition of adolescent athletes were needed to understand more mineral nutrition and exercise.
