Inhibition of C-terminal O-Methyltransferase by a Rat Liver Cytosolic Peptide

  • The activity of SD-framesylcysteine O-methyltransferase was assayed by incubating the enzyrne with a synthetic in vitro substrate, [N-acetyl-S-trans, trns-famesyl-L-cysteine (AFC)], together with S-adenosyl-L-[emthyl-$_{14}$C)ester(AFCME)], was then analyzed either directly on HPLC or by converting the AFC[$methyl^{14}C$]ME to [$methyl^{14}C$] aclcohol by basehydrolysis. Employing these two analytical methods, it was established that a peptide purifed from rat liver cytosol fraction [Int. J. Biochem., 25, 1157 919930] strongly inhibited the above enzyme activity with $IC_{50}\; of\; 7.1\times 10^{-8}$ M. Also, the S-famesylcysteine O-methyltransferase from several human colon cancer cells was equally inhibited by the peptide.

Comparison betwee Bovine Hide and Pigskin Gelatins and Preparation of Gelatin Hydrolysates

  • Bovine hide and pig skin gelatins were prepared and their molecular weight profiles were examined by SDS-PAGE. The major molecular weights of bovine hide gelatin were 220 kDa, 140kDa, and 130kDa and the weights of pigskin gelatin were 210 kDa, 135kDa and 120kDa. Also , as a typical parameter of rheological property of the gelatin , viscosities of gelatin were measured under various conditions. Gelatin hydrolysates were prepared using typical commerical proteases and their angiotensin converting enzyme inhibitory activities were examined.

  • An angiotensin converting enzyme (ACE) inhibitory peptide was isolated and purified from the hydrolysates of duck meat protein. Duck meat protein was hydrolyzed using trypsin at 37$^{\circ}C$ for 2 hrs. An ACE inhibitory peptide was purified using membrane filtration, anion exchange chromatography, gel permeation chromatography, fast protein liquid chromatography, normal phase HPLC. The purified inhibitory peptide was identified to be a tetrapeptide, Glu-Asp-Leu-Glu having $IC_{50}$/ value of 85.9 $\mu$M.

  • The messenger RNA-based vaccine for the coronavirus disease 2019 (COVID-19) may induce glomerulonephritis, including immunoglobulin A nephropathy (IgAN). New-onset IgAN triggered by vaccination against COVID-19 has been reported rarely, especially in children. Herein, we report a pediatric case of newly diagnosed IgAN after administration of the Pfizer vaccine for COVID-19. A 12-year-old girl was referred to our hospital for evaluation of gross hematuria after inoculation with the second dose of Pfizer's COVID-19 vaccine; she had no adverse effects after the first dose. At the time of admission, she showed heavy proteinuria and persistent hematuria. Kidney biopsy revealed an IgAN, and she was treated with an oral steroid and an angiotensin-converting enzyme inhibitor. Four months after discharge, the proteinuria and hematuria resolved completely.

  • A 22-year-old male patient was diagnosed with autosomal dominant polycystic kidney disease (ADPKD). He received conservative treatment with an angiotensin-converting enzyme inhibitor. Two years later, oral therapy, consisting of 60 mg tolvaptan per day, was initiated. Compared with height-adjusted total kidney volume, the rate of kidney growth reduced significantly from 7.33% to 0.66% annually, since commencement of the tolvaptan therapy. The liver enzyme profile and serum sodium level and osmolality were constantly within normal ranges. In Korea, this is the first reported case of a patient with ADPKD who received tolvaptan treatment for more than 1 year. This case demonstrates that long-term tolvaptan treatment appears to be safe, well tolerated, and effective for ADPKD.

Isolation of Polyphenol from Green Tea by HPLC and Its Physiological Activities (HPLC에 의한 녹차의 polyphenol 화합물의 분리 및 polyphenol의 생리활성)

  • Polyphenols were isolated from Korean green tea using Sephadex LH-20 and HPLC. The isolated polyphenols were procyanidin B-4, procyanidin B-2-3,3'-digallate, prodelphinidin C-2-3,3'-di-O-digallate, (+)-catechin-3-O-rhamnose, procyandin B-5, procyanidin B-7-3-0-gallate, gallate, epiafzelechin-$(4{\beta}{\rightarrow}8)$-epiafzelechin, procyanidin B-3-3-O-rhamnose, afzelechin-$(4{\alpha}{\rightarrow}8)$-catechin, prodelphinidin B-5-3,3'-di-O-digallate and (+)-taxifolin-3-O-D-xyloside. The inhibitory effects of prodelphinidin C-2-3,3'-di-O-gallate and procyanidin B-2-3,3'-digallate $(at\;100{\mu}M)$ on angiotensin.converting enzyme were 68.8 and 54.6%, respectively, while the inhibitory effects of prodelphinidin C-2-3,3'-di-O-gallated and procyanidin B-2-3,3'-digallate $(at\;100{\mu}m)$ on xanthine oxidase were 54.5 and 38.2%, respectively. Lastly, the inhibitory activities of prodelphinidin C-2-3,3'-di-O-gallate $(at\;100{\mu}m)$ on tyrosinase was 42.1%.

Inhibitory Activity on Angiotensin Converting Enzyme (ACE) of Korean Medicinal Herbs (한국산 약용식물의 안지오텐신 전환효소에 대한 저해활성)

  • The study was conducted to develop new materials of functional foods from natural resources. Fortyeight medicinal plants were screened on inhibitory activity of angiotensin-converting enzyme (ACE), a protein elevating blood pressure. The water extracts of Armoracia aerial part, Cynanchum radix, Euonymus branch, Phyllostachys stem, Trichosanthes seed, and methanol extract of Silybum aerial part were showed strong inhibitory activity (above 70%) on ACE at the final concentration of $4,000\;{\mu}g\;ml^{-1}$. Among the plant extracts, water extracts of Phyllostachys taeniam and Trichosanthes seed were especially showed strong inhibitory activity (above 25%) at $500\;{\mu}g\;ml^{-1}$. These two plants are expected good candidate for development into anti-hypertensive materials.

Effects of the Biological Function of Yogurt Added with Lycium chinence Miller Extract (구기자(Lycium chinence Miller)추출액을 첨가한 요구르트의 생리활성 효과)

  • This experiment was carried out to investigate the biological function of Lycium chinence Miller yogurt. Antioxidant activity was higher in methanol extract yogurt than in water extract yogurt. The antioxidant activity was shown at 83.9% in Lycii folium extract yogurt, 47.0% in Lycii fructus extract yogurt and 54.0% in Lycii cortex extract yogurt. Angiotensin converting enzyme inhibitor activity was shown highly both in water extract and methanol extracts from the Lycii folium. The $\alpha$-glucosidase inhibitor activity was shown at the 4.0% concentration of Lycii folium extract yogurt, Lycii fructus extract yogurt and Lycii cortex extract. In an orally administrated rat, normal yogurt, Lycii fructus extract yogurt and Lycii cortex extract yogurt have no effect on blood cholesterol content. IgG production in blood is more increased in Lycii cortex extract yogurt than in normal yogurt and Lycii fructus extract yogurt.

Physiological Functionality of Various Extracts from Danmemil and Legumes (단메밀과 콩 추출물들의 생리 기능성)

  • Physiological functionalities of various extracts from Danmemil and legumes were determined and its optimal extraction conditions were also investigated. Angiotensin I-converting enzyme (ACE) inhibitory activity and tyrosinase inhibitory activity of Danmemil were higher in water extracts (53%, 58%) than those of ethanol extracts. However, its electron-donating ability was the highest in ethanol extracts (72%). ACE inhibitory activity and electron-donating ability of Black bean No. 1 and Taekwangkong(one of bean) were higher in water extracts than those of ethanol extracts, whereas SOD-like activity was the highest in ethanol extracts. ACE inhibitor and tyrosinase inhibitor of Danmemil were maximally extracted when it were treated with 20 times of distilled water at 35$^{\circ}C$ for 24 h and 36 h, respectively. Its electron donating compound was maximally extracted by treatment of 50$^{\circ}C$ for 18 h. ACE inhibitor of Black bean No. 1 was extracted maximally when it was treated with distilled water (1 :20) at 20$^{\circ}C$ for 24 h, whereas the other functional compounds were maximally extracted at 20$^{\circ}C$ for 18 h.

The Increases of Proenkephalin A mRNA Levels and the Secretion of $[Met^{5}]-Enkephalin$ Induced by Long-term Stimulation with Nicotine are Mediated by a Lipoxygenase Pathway in Bovine Adrenal Medullary Chromaffin Cells (소 부신수실 크롬친화성 세포에서 Nicotine의 장기간 자극으로 유발된 Proenkephalin A mRNA의 증가 및 $[Met^{5}]-enkephalin$의 분비 증가가 Lipoxygenase 경로에 의해 매개됨)

  • The effect of nicotine on the secretion of $[Met^{5}]-enkephalin$ (ME) in addition to proenkephalin A (proENK) mRNA levels and effects of indomethacin, nordihydroguaiaretic acid (NDGA), and captopril on nicotine-induced responses were studied in bovine adrenal medullary chromaffrin (BAMC) cells. Long-term exposure of BAMC cells to nicotine at a concentration of $10{\mu}M$ significantly increased proENK mRNA level and the secretion of ME into the medium. Treatment of BAMC cells with NDGA (a lipoxygenase inhibitor, $10{\mu}M$), indomethacin (a cycloooxygenase inhibitor) or captopril (an angiotensin converting enzyme inhibitor) alone did not affect ME secretion and proENK mRNA levels. The pretreatment of BAMC cells with NDGA inhibited the increased ME secretion and proENK mRNA level induced by nicotine. However, indomethacin and captopril did not affect nicotine-induced responses. Our results indicate that neuronal regulations of ME secretion and proENK mRNA level induced by nicotine in BAMC cells are in part mediated by a lipoxygenase-but not cyclooxygenase-and endogenous renin-angiotensin pathway.
