자망어구 부이의 깃대가 부이줄 장력에 미치는 영향 (Effect of flagpole attached to buoy on tension of buoy rope of gillnet)

  • This study aims to reduce the force exerted to the buoy of the gillnet by wave and current. Five buoy models were selected for experiments and their rope tensions under wave and current action were compared. Five models were EL (ellipsoid), EL-H (ellipsoid-hole), SL (streamlined body), SP (sphere) and CL (cylinder, traditional type). In the first experiment, the Five models were tested without any attachment. In the second experiment, a flagpole was attached to each model. As a result, in the condition without flagpole, the tensions of four models with the exception of the CL were about a half of that of the CL. In the condition with flagpole, the tension of all models was twice larger than that without flagpole. Thus, a new model was suggested to improve the problem, which has a combined body that of a flagpole and a buoy Three new models of CL-L (long and thin cylinder), LF (leaf shape) and LF-F (leaf shape with fin) were designed. Also a cylinder type (CLD) with a flagpole as a control was included in the experiment. As a result, the LF-F had the smallest tension and a half tension of the CLD. Therefore, it is supposed that the flagpole and buoy combined model could reduce the tension on buoy rope and contribute to improve the gillnet loss problem.

Developing a Virus-Binding Bacterium Expressing Mx Protein on the Bacterial Surface to Prevent Grouper Nervous Necrosis Virus Infection

  • Grouper nervous necrosis virus (GNNV) infection causes mass grouper mortality, leading to substantial economic loss in Taiwan. Traditional methods of controlling GNNV infections involve the challenge of controlling disinfectant doses; low doses are ineffective, whereas high doses may cause environmental damage. Identifying potential methods to safely control GNNV infection to prevent viral outbreaks is essential. We engineered a virus-binding bacterium expressing a myxovirus resistance (Mx) protein on its surface for GNNV removal from phosphate-buffered saline (PBS), thus increasing the survival of grouper fin (GF-1) cells. We fused the grouper Mx protein (which recognizes and binds to the coat protein of GNNV) to the C-terminus of outer membrane lipoprotein A (lpp-Mx) and to the N-terminus of a bacterial autotransporter adhesin (Mx-AIDA); these constructs were expressed on the surfaces of Escherichia coli BL21 (BL21/lpp-Mx and BL21/Mx-AIDA). We examined bacterial surface expression capacity and GNNV binding activity through enzyme-linked immunosorbent assay; we also evaluated the GNNV removal efficacy of the bacteria and viral cytotoxicity after bacterial adsorption treatment. Although both constructs were successfully expressed, only BL21/lpp-Mx exhibited GNNV binding activity; BL21/lpp-Mx cells removed GNNV and protected GF-1 cells from GNNV infection more efficiently. Moreover, salinity affected the GNNV removal efficacy of BL21/lpp-Mx. Thus, our GNNV-binding bacterium is an efficient microparticle for removing GNNV from 10‰ brackish water and for preventing GNNV infection in groupers.

Morphological and molecular characteristics of Paralecithodendrium longiforme (Digenea: Lecithodendriidae) adults and cercariae from Chinese pipistrelle bats and viviparid snails in Thailand

  • This study aimed to describe the morphological and molecular characteristics of Paralecithodendrium longiforme (Digenea: Lecithodendriidae) adults and cercariae isolated in Thailand. Adult flukes were isolated from the Chinese pipistrelle bat (Hypsugo sp.), and cercariae were detected in the viviparid snail (Filopaludina martensi martensi) from Chiang Mai province. The morphological characteristics were observed and described using conventional methods, and the molecular characteristics with internal transcribed spacer 2 (ITS2) and 28S rDNA gene sequences. The adult flukes were fusiform, 0.84-0.98 mm in length, and 0.37-0.49 mm in width, and were distinguishable from other species by the presence of longitudinal uterine coils. The cercariae were nonvirgulate xiphidiocercariae, with the oral sucker bigger than the acetabulum, the tail without fin fold, a body size of 117.5-138.3×48.3-52.2 ㎛, and a tail size of 100.7-103.7×15.0-18.9 ㎛. Molecular studies revealed that the adults and cercariae shared 99.3% (ITS2) and 99.6% (28S rDNA) homology with each other. They were phylogenetically close to P. longiforme with an identity of 94.5% for ITS2 and 98.7% for 28S rDNA. This study provides new information on the natural definitive host and first intermediate host of P. longiforme in Thailand. The discovery of its cercarial stage in Filopaludina snails highlights the importance of monitoring the associated second intermediate host and prevention and control of this potentially zoonotic trematode.

생태 지표종을 이용한 해부학적 건강성 평가지수(HAI) 적용을 위한 예비실험 (Preliminary Studies on the Necropsy-Based Health Assessment Index (HAI) Using a Ecological Indicator Species)

  • 수환경의 생물학적 건강도는 다양한 생물상을 이용하여 평가가 이루어져 왔으며 특히, 어류는 수환경의 건강성 평가를 위한 가장 좋은 생물종 중의 하나이다. 해부학적 기초에 의한 건강성 평가방법을 시도한 이번 예비실험의 목적은 대전시 갑천에 위치한 하수종맡처리장의 방류수가 수환경의 건강성에 어떤 영향을 미치는지 평가해 보는 것이다. 이러한 영향을 분석하기 위해 어류의 흉선(Thymus), 지느러미 (Fins), 지라 (Spleen), 후장 (Hindgut),신장 (Kidney), 피부 (Skin), 간 (Liver), 눈 (Eyes), 아가미(Gill), 및 헛아가미 (Pseudobranch)의 해부학적인 10개 메트릭을 이용하였다. 각각 2개의 대조군(control)과 처리군 (Treatment)으로 실험용 수조를 분리하여, 처리군에는 하수종말처리장의 방류수를 사용하였고 대조군에는 자연수를 사용하였으며, 각각의 수조에 버들치 (Rhynchocypris oxycephalus)를 5개체 씩 투여하였다. 투여한 버들치는 해부학적 측면의 관찰을 실시하였고, 이후 처리군의 죽은 버들치와 대조군의 개체를 각각 해부하고 10개 기관부위를 분석하여 HAI 건강도지수를 산정하였다. 해부하적 분석결과에 따르면, 3개의 대조군의 HAI지수는 0으로 나타나 최적상태로 판명되었고, 4 대조군은 40 ${\sim}$ 150점으로서 모두 악화상태로 나타났다. 또한, 생물통합지수 분석, 종풍부도분석, 내성도 및 트로픽길드 분석에 의거한 다양한 건강도 분석은 HAI 분석결과를 지지하였다. 본 HAI 기법은 현재 국내에서는 적용 평가된바 없어, 기존의 유수생태계의 건강도 평가에 상호보완적지수로의 활용도 가능할 것으로 사료된다.

Effects of Nonylphenol and 2,2', 4,6,6'-pentachlorobiphenyl on in vitro Sex Steroid Production in Maturing Oocytes of the Yellowfin Goby, Acanthogobius flavimanus

  • Several studies have reported that nonylphenol (NP) and 2,2', 4,6,6'-pentachlorobiphenyl (PCB104) exhibit estrogenic activity. To investigate the estrogenic potency of NP and PCB104 during oocyte maturation, fully vitellogenic oocytes (0.76 mm diameter in average) of yellow fin goby, Acanthogobius flavimanus, were exposed in vitro to these chemicals at different concentrations (0.1, 1, 10, 100, and 1,000 ng/mL) with the exogenous precursor $17\alpha$-hydroxyprogesterone ($17{\alpha}OHP$) 50 ng/mL in the presence or absence of human chorionic gonadotropin (HCG). The production of testosterone (T), estradiol-$17\beta$ (E2), and $17\alpha,20\beta$-dihydroxy-4-pregnen-3-one ($17\alpha20{\beta}OHP$) in response to NP or PCB104 were measured by radioimmunoassay. Steroid levels were also expressed as E2/T and E2/$17\alpha20{\beta}OHP$ ratios. In the absence of HCG, no significant differences in either NP or PCB104 treatment groups were observed. In the presence of HCG, NP treatment did not show significant differences in the production of T, E2, and $17\alpha20{\beta}OHP$ at any concentrations tested, but E2/T ratios were decreased at concentrations of 0.1, 1, 10, and 1,000 ng/mL compared with the control group. PCB104 decreased E2 production at concentrations of 0.1, 10, and 1000 ng/mL, but did not show significant differences in the production of T and $17\alpha20{\beta}OHP$ at any concentration tested. While E2/T ratios were decreased at PCB104 concentrations of 0.1, 1, 10, and 1,000 ng/mL, E2/$17\alpha20{\beta}OHP$ ratios were also decreased at 0.1, 10, and 1,000 ng/mL compared with the control. Results indicate that both NP and PCB104 appeared to have antiestrogenic effects during this phase.

꽁치육의 고지혈증 유발 쥐에서의 항고지혈증 및 항동맥경화증 효능 (Effects of Saury Meat on Antihyperlipidemic and Antiarteriosclerosis Activities in Sprague-Dawley Rats)

  • 본 연구는 일상에서 우리가 쉽게 접할 수 있는 꽁치로 동물실험을 통해 항고지혈증 및 항동맥경화증의 효과를 검증 하였다. 실험동물에게 1% cholesterol과 0.5% Na-cholic acid를 첨가하여 인위적으로 고지혈증을 유발시킨 후 꽁치의 전체, 육, 내장, 머리+꼬리+뼈의 4종류로 나눈 분획을 경구투여 하여 항고지혈증 및 항동맥경화증의 효과를 살펴본 결과, 정상군에 비해 poloxamer-407을 투여하여 고지혈증을 유발한 흰쥐의 중성지방의 함량이 현저하게 감소하였다. 또한 poloxamer-407에 의한 고지혈증의 경감효과를 재확인할 목적으로 Triton WR-1339를 투여한 결과도 역시 중성지방의 함량이 현저하게 감소하였다. 이러한 결과를 기초로 4종류의 분획 중 다른 시료보다 육분획에서 대조군에 비해 지방조직, 혈청 지질량, 혈청 콜레스테롤 함량, 동맥경화지수, 간조직중의 지질 및 콜레스테롤의 함량, 분변 중의 지질의 함량 등이 현저하게 감소하였다. 혈중 지질과산화와 활성산소의 양은 현저하게 감소하지 않았지만, 활성산소를 제거하는 SOD의 양은 32.5%만큼 현저하게 증가한 것으로 보아 꽁치 육분획에서 항고지혈증 및 항동맥경화증의 효과가 탁월함을 알 수 있었다. 이상의 결과로부터 꽁치육의 항고지혈증 및 항동맥경화증의 효과를 확인할 수 있었다. 하지만 구체적으로 꽁치육의 어떠한 성분이 항고지혈증 및 항동맥경화증의 효과를 나타내는 지에 대해서는 추가 실험을 해볼 필요가 있다.

성견 3면 골내낭에서 Paste형 Calcium Sulfate 가 치주조직 치유에 미치는 영향 (THE EFFECTS OF PASTE TYPE CALCIUM SULFATE ON THE PERIODONTAL HEALING OF 3-WALL INTRABONY DEFECTS IN DOGS)

  • There are numerous kind of materials and techniques to regenerate the periodontal tissue which has been lost due to destructive periodontal disease, including bone graft material. Many bone graft materials have been reported and among these materials, synthetic material has been developed fin the long time because of its sufficient supply economically. Calcium sulfate which was evaluated as including much calcium, has been used in the clinical field. In the dental field calcium sulfate has been used as bone graft material and Kim reported that improved bone formation and more amount of new attachment after grafting calcium sulfate. but, because calcium sulfate has the problem that it generates the heat in setting period and resolves fast, we need to evaluate the effect of the improved calcium sulfate on periodontal tissue. The present study evaluates the effect of paste type calcium sulfate on the epithelial migration, alveolar bone regeneration, cementum formation and gingival connective tissue attachment in intrabony defect in dogs. Four millimeter deep and four millimeter wide 3-wall defects were surgically created in the mesial or distal aspects of premolars or molars. the test group received paste-type calcium sulfate with a flap procedure and the control group underwent flap procedure only. Histologic analysis after 8 weeks of healing revealed the following results : 1. The length of epithelial growth(the distance from CEJ to the apical end of JE) was 0.52${\pm}$0.26mm in the control and 0.56${\pm}$0.25mm in the test group. there was no statistically significant difference between the two groups. 2. The length of connective tissue adhesion was 1.74${\pm}$1.06mm in the control and 1.28${\pm}$0.57mm in the test group. there was no statistically significant difference between the two groups. 3. The length of new bone was 2.01${\pm}$0.95mm in the control and 2.62${\pm}$0.81mm in the test group. there was no statistically significant difference between the two groups. 4. The length of new cementum was 1.86${\pm}$0.80mm in the control and 2.77${\pm}$ 0.86mm in the test group. there was a statistically significant difference between the two groups.(P<0.01) These results suggest that the use of paste type calcium sulfate in 3-wall intrabony defects has significant effect on new cementum formation , but doesn't have any significant effect on the prevention of junctional epithelium migration and new bone formation. Finally, the paste type calcium sulfate that is used in this study is suggested to be the material that can have a significant effect on the periodontal healing, if its biocompatibility is improved.

사료내 Paprika와 동물플랑크톤 혼합첨가가 흰줄납줄개 Rhodeus ocellatus의 Carotenoid 축적 및 체색에 미치는 영향 (Effect of Paprika and Zooplankton Supplementation in the Diets on Carotenoid Deposition and Skin Pigmentation of the Rose Bitterling Rhodeus ocellatus)

  • 본 연구는 사료내 색소원으로 paprika와 동물플랑크톤 혼합첨가가 흰줄납줄개의 carotenoid 축적 및 색택에 미치는 영향을 조사하였다. 대조사료(Con)를 제외한 모든 실험사료에 paprika 분말 5%를 첨가하고, paprika 분말과 동물플랑크톤의 상호작용을 알아보기 위하여 paprika 분말만 첨가한 실험사료(P5), P5사료에 water flea Moina macrocopa 3% 첨가사료(P5M3), Artemia nauplius (Artemia sp.) 3% 첨가사료(P5A3), rotifer Brachionus Plicatilis 3% 첨가사료(P5R3)로 총 5개의 실험사료를 제조한 후, 2반복으로 10주간 사육실험 하였다. 실험종료 후, 모든 실험구간에서 생존율의 차이는 없었으며, 총 carotenoids 함량은 동물플랑크톤이 첨가된 실험 구에서 높은 다소 높은 값을 보였다. 특히, P5A3 실험구에 가장 높은 값을 나타냈다. 흰줄납줄개의 색택은 paprika와 동물플랑크톤 혼합 첨가한 실험구가 대조구 및 P5 실험구에 비해 붉은색이 더 선명하였다. 이상의 결과를 고려할 때, 흰줄납줄개의 배합사료내 paprika와 동물플랑크톤의 혼합첨가는 붉은 색 개전에 도움이 되었고, 특히 Artemia nauplius 또는 rotifer 첨가가 좋을 것으로 판단된다.

막걸리 분획물에 의한 암세포 성장 억제 및 Quinone Reductase 활성 증가 효과 (Effect of Growth Inhibition and Quinone Reductase Activity Stimulation of Makgeoly Fractions in Various Cancer Cells)

  • 막걸리의 암 예방효과를 알아보기 위하여 막걸리 농축물을 핵산, 메탄올, 부탄올 및 물로 순차적으로 분획하여 각 분획별로 암세포에 대한 성장 억제효과와 암 예방 지표인 QR활성 증가 효과를 측정하였다. 그 결과 메탄올 분획물의 경우 낮은 농도의 시료첨가에도 불구하고 괄목할 만한 높은 암세포 성장 억제효과를 나타내었으며 4종의 모든 암세포주 HepG2, B16-F10, HT29 및 MCF-7에서 농도 의존적인 암세포 성장 억제효과를 나타내었다. 또한 HepG2에서 측정한 QR활성 증가 효과에 있어서도 메탄올 분획물이 가장 높은 QR활성 증가 효과를 보여 암에 대한 예방효과가 기대된다. 따라서 앞으로 막걸리를 이용하여 항암관련 기능성식품을 개발할 수 있는 가능성이 보이며, 이를 위하여 특히 메탄올 분획물에 대한 집중적인 연구가 요구된다.

설비공학 분야의 최근 연구 동향: 2008년 학회지 논문에 대한 종합적 고찰 (Recent Progress in Air-Conditioning and Refrigeration Research: A Review of Papers Published in the Korean Journal of Air-Conditioning and Refrigeration Engineering in 2008)

  • This article reviews the papers published in the Korean Journal of Air-Conditioning and Refrigeration Engineering during 2008. It is intended to understand the status of current research in the areas of heating, cooling, ventilation, sanitation, and indoor environments of buildings and plant facilities. Conclusions are as follows. (1) Research trends in thermal and fluid engineering have been surveyed in the categories of general fluid flow, fluid machinery and piping, new and renewable energy, and fire. Well-developed CFD technologies were widely applied in developing facilities and their systems. New research topics include fire, fuel cell, and solar energy. Research was mainly focused on flow distribution and optimization in the fields of fluid machinery and piping. Topics related to the development of fans and compressors had been popular, but were no longer investigated widely. Research papers on micro heat exchangers using nanofluids and micro pumps were also not presented during this period. There were some studies on thermal reliability and performance in the fields of new and renewable energy. Numerical simulations of smoke ventilation and the spread of fire were the main topics in the field of fire. (2) Research works on heat transfer presented in 2008 have been reviewed in the categories of heat transfer characteristics, industrial heat exchangers, and ground heat exchangers. Research on heat transfer characteristics included thermal transport in cryogenic vessels, dish solar collectors, radiative thermal reflectors, variable conductance heat pipes, and flow condensation and evaporation of refrigerants. In the area of industrial heat exchangers, examined are research on micro-channel plate heat exchangers, liquid cooled cold plates, fin-tube heat exchangers, and frost behavior of heat exchanger fins. Measurements on ground thermal conductivity and on the thermal diffusion characteristics of ground heat exchangers were reported. (3) In the field of refrigeration, many studies were presented on simultaneous heating and cooling heat pump systems. Switching between various operation modes and optimizing the refrigerant charge were considered in this research. Studies of heat pump systems using unutilized energy sources such as sewage water and river water were reported. Evaporative cooling was studied both theoretically and experimentally as a potential alternative to the conventional methods. (4) Research papers on building facilities have been reviewed and divided into studies on heat and cold sources, air conditioning and air cleaning, ventilation, automatic control of heat sources with piping systems, and sound reduction in hydraulic turbine dynamo rooms. In particular, considered were efficient and effective uses of energy resulting in reduced environmental pollution and operating costs. (5) In the field of building environments, many studies focused on health and comfort. Ventilation. system performance was considered to be important in improving indoor air conditions. Due to high oil prices, various tests were planned to examine building energy consumption and to cut life cycle costs.
