Excessive Dietary Conjugated Linoleic Acid Affects Hepatic Lipid Content and Muscular Fatty Acid Composition in Young Chicks

  • The effects of dietary conjugated linoleic acid (CLA) on lipid concentrations and fatty acid composition of various tissues were studied in young chicks. From 7 days of age, a total of 160 chicks were divided into 4 groups, placed into 4 pens per group (10 birds per pen) and fed one of four experimental diets containing 6% tallow (TO 6%), 4% tallow plus 2% CLA (TO 4%-CLA 2%), 2% tallow plus 4% CLA (TO 2%-CLA 4%) or 6% CLA (CLA 6%) for 3 weeks. There were no significant differences in growth performances and the relative weights of various organs, but relative liver weight of chicks fed dietary CLA at 4 and 6% levels was significantly higher (p<0.05) than that of TO 6% group. The chemical compositions of leg muscle were not affected by CLA feeding. However, hepatic total lipid of chicks fed 6% CLA diet was significantly higher (p<0.05) than those of TO 6% and TO 4%-CLA 2% groups. The concentrations of various lipid fractions in serum were not affected by CLA feeding. With the increase in dietary CLA levels, cis 9-trans 11 CLA, trans 10-cis 12 CLA and total CLA of leg muscle increased linearly. The relative proportions of C18:1 $\omega$ -9 and C20:4 $\omega$-6 fatty acids in the leg muscles of chicks fed the CLA containing diets were significantly lower (p<0.05) than those of TO 6% group. These results indicate that the levels of CLA isomers were increased linearly in dose-dependent manner after feeding of synthetic CLA source. But it was also observed that excessive amount of dietary CLA resulted in the possible adversely effects, such as increase of liver weight, hepatic lipid accumulation and serum GOT level.

Effects of conjugated linoleic acid on the performance of laying hens, lipid composition of egg yolk, egg flavor, and serum components

  • Objective: This experiment investigated the effects of dietary supplementation with conjugated linoleic acid (CLA) on the serum components, laying hen productivity, lipid composition of egg yolk, egg flavor and egg quality. Methods: Healthy 28-week-old Hy-Line white laying hens (n = 480) were divided randomly into 4 groups, 6 replicates/group, 20 birds/replicate. The 30-day experimental diets included 0% (control), 0.4%, 0.8%, and 1.6% CLA. Some serum indices of the birds, and egg production, quality, fatty acid composition, egg quality were measured. Results: The dietary supplementation with 0.4%, 0.8%, and 1.6% CLA did not significantly affect the laying rate and feed intake, as well as calcium ion and phosphorus ion concentration in serum (p>0.05). However, the CLA had significantly increased the strength of eggshell, decreased the odor, flavor, and taste of egg yolk, deepened the color of egg yolk, increased saturated fatty acids and polyunsaturated fatty acids, and reduced the monounsaturated fatty acids (p<0.05). On the other hand, the dietary supplementation with 1.6% CLA had significant effects on feed/gain, and improved serum hormones. Dietary supplementation with 0.4% and 0.8% CLA can significantly enhance the activity of alkaline phosphates. Conclusion: CLA has no effect on production performance, but does enhance the lipid content of the egg yolk and the strength of the eggshell.

  • To examine the functional properties of conjugated linoleic acid (CLA) and ascidian tunic extracts in fish, we compared mackerel fed ascidian tunic extract and CLA (CA25) and a control group. The daily growth index of CA25 was 1.92 compared to 1.86 in the control group. The viscerosomatic index of CA25 was 36.7% lower than that of the control group. After 8 weeks, the protein content decreased from 19.7 to 17.5% in the CA25 group. The ascidian tunic extract content in the viscera was much higher than in muscle (0.13 vs. 0.03 mg/100 g) after 8 weeks. At the start, the n-3 fatty acid content of the experimental fish was 25.2% in muscle and 23.7% in viscera. The CLA content in muscle in the CA25 group was 2.1% after 4 weeks and 2.3% after 8 weeks. By contrast, the CLA content in viscera did not change after 8 weeks.

사료내 Conjugated Linoleic Acid 첨가가 육계의 성장 및 체지방 대사에 미치는 영향 (Influence of Dietary Conjugated Linoleic Acid on Growth Performance and Body Fat Metabolism in Broiler Chickens)

  • The current study was conducted to investigate the effects of dietary conjugated linoleic acid(CLA) on growth performance and body fat metabolism in broiler chickens. A total of 48 male birds aged 3 days were randomly allotted into three dietary groups; CORN(com oil 1.5%), CLA I (com oil 0.75 %+ CLA 0.75 %) and CLA II(CLA 1.5%) groups. After feeding commercial diet ad libitum for 3 weeks, eight selected birds on the basis of body weight were housed, two birds in a cage, and continuously given ad libitum corresponding experimental diet for another 2 weeks. As a result, dietary addition of CLA did not influence on body weight, gain and feed conversion rote. The relative weights of the liver and deposited fats(abdominal and thigh fat' pads) were not also affected by the dietary treatments. Serum glucose, triglyceride and cholesterol levels markedly( P < 0.05) decreased in CLA II compared with those in CORN group. However, serum nonesterified fatty acid(NEFA) was not altered by dietary CLA. Serum leptin level was tended to be decreased by dietary CLA without statistical difference. The diet supplemented with CLA caused a significant(P< 0.05) decrease in hepatic total lipid and NEFA without changing triglyceride level. Also, feeding dietary CLA at the level of 1.5% reduced leptin mRNA expression in the liver and abdominal fats compared with feeding com oil, In conclusion, our results suggest that dietary 1.5% CLA may affect, at least in part, lipid metabolism in the liver of broiler chickens.

  • The purpose of this study was to examine whether conjugated linoleic acid and selenium supplementation in broiler chicken diets would be effective, enhance indices of immune status and body weight, and modulate serum lipid concentration. Forty Hyline brown chickens, 1 weeks of age, were divided into 5 groups of 8 chickens. Chickens were fed the experimental diets supplemented with 1% CLA (conjugated linoleic acid; Group 1), 1% CLA + selenium (Group 2), 1% safflower-seed-oil as LA (Group 3), 1% safflower-seed-oil as LA + selenium (Group 4) or nothing (Control) for 4 weeks. After 4 weeks, serum, liver, spleen and abdominal fat were taken. Measurement of total immunoglobulin were executed using sandwich ELISA. Weight ratio of liver to body showed that the group fed with CLA were significantly higher than the group fed with CLA + selenium. Weight ratios of spleen and fat to body showed no significantly differences. In concentrations of serum total cholesterol and HDL-cholesterol, the group fed with CLA showed significantly higher values than that fed with CLA + selenium. In concentrations of serum triglyceride and LDL-cholesterol there were no significantly differences between the treatment groups. In conclusion, supplementation of CLA with selenium protected hepatomegaly and reduced level of serum total cholesterol and HDL-cholestererol in chickens.

Supplementation of Conjugated Linoleic Acid with γ-Oryzanol for 12 Weeks Effectively Reduces Body Fat in Healthy Overweight Korean Women

  • The enhancement of the human body fat reduction of conjugated linoleic acid (CLA) with the supplementation of $\gamma$-oryzanol (OZ) was investigated on overweight Korean women (n=51, BMI> 23). Subjects were divided into 4 groups of control, CLA, glyceride form of CLA (GCLA), and CLA plus OZ (CLA-OZ). The soft-gel capsule (500 mg) was used to deliver control (500 mg olive oil), CLA (500 mg CLA), GCLA (500 mg GCLA) and CLA-OZ (500 mg CLA plus 50 mg OZ). Three capsules were taken twice a day for 12 weeks. The CLA-OZ supplementation reduced 1.35% body fat that was 0.34% enhancement against CLA supplementation. As considered subject variations, CLA-OZ reduced body fat ranged from 7.9% to -2.7%, equivalent to 5.6 kg loss to 0.7 kg gain in body fat mass, against CLA. The CLA-OZ reduced body weight and body mass index (BMI), relative to control, but the reductions by CLA-OZ were not different from those by CLA and GCLA. All biochemical markers analyzed for safty were not significantly different within or between groups and were within the normal range. The CLA-OZ supplementation significantly reduced blood pressure, as compared to the supplementation of CLA, GCLA and control. These results suggest that OZ could be a useful ingredient to mix with CLA for the reduction of human body fat.

지방 급원을 달리한 식이에 첨가된 CLA가 지방조직과 Stearoyl-Co A Desaturase 1(SCD 1) 발현에 미치는 영향 (The Effect of Conjugated Linoleic Acid(CLA) Supplemented to Different Fat Sources on Fat Depositions and Stearoyl-CoA Desaturase l(SCDl) Gene Expression in Mice)

  • This study investigated the effects of conjugated linoleic acid(CLA) on the fat deposition, triglyceride levels and the expression of stearoyl-CoA desaturase 1(SCD1) in the livers of male ICR mice that were fed with either soybean oil or beef tallow supplemented with CLA. Mice weighing $25{\sim}30$ g were divided into four groups; soybean oil(SBO), and SBO supplemented with 1% CLA(SBOC), beef tallow(BT) and BT supplemented with 1% CLA(BTC). Each group consisted of 10 mice that were fed the experimental diets for 4 weeks. The experimental diets consisted of 64% carbohydrate, 20% protein, and 16% fat in terms of their contributions to total calories. All other nutrients were identical in the diets. Triglyceride measurements were completed using a kit. Fatty acid compositions were analyzed in the liver using gas chromatography. The levels of SCD1 expression were analyzed by RT-PCR in the liver. No significant differences were found for food intake level, body weight and food efficiency among the experimental groups. However, the weights of epididymal fat pads and plasma triglyceride levels were significantly lower in SBOC and BTC(p<0.05) compared to the SBO and BT groups. These effects were similar in the CLA supplemented groups. The expression level of SCD1 gene and ${\Delta}9$ desaturase index were not significantly different, regardless of the fat used for CLA supplementation. Based on these results, addition of CLA showed decreasing effects on the fat depots weight and the concentration of triglyceride regardless of the fat sources. The SCD1 gene expression and ${\Delta}9$ desaturase index were not influenced by the types of fats with respect to the CLA effects.

바이오디젤유 부산물로 제조한 conjugated linoleic acid(CLA)의 첨가 수준이 젖소의 산유량 및 유성분에 미치는 영향 (Effects on the Levels of Dietary Conjugated Linoleic Acid (CLA) Produced from Bio-Diesel By-Products on the Production and Composition of Dairy Cow Milk)

  • 본 연구는 바이오디젤유 부산물로 제조한 공액리놀레산(Conjugated Linoleic Acid, CLA)을 비유중기 착유우에 첨가 수준을 달리하여 급여하였을 때 산유량, 유성분 및 혈액성상에 미치는 영향을 조사하고자 실시하였다. 공시동물은 비유중기 홀스타인 20두($631{\pm}25.0kg$)로 시험구당 5두를 배치하였으며, 기초사료($NE_L$ 32 Mcal/d, CP 17%)로 농후사료(12kg), 옥수수 사일리지(15kg) 및 톨페스큐+오차드그라스 건초(4kg)를 1일 2회 균등 급여하였다. CLA의 1일 급여수준은 0(대조구), 50(T1), 100(T2) 및 150g(T3)으로 달리하여 네처리군을 설정하고 예비 시험 2주와 본 시험 4주를 수행하였다. 착유우에 CLA의 급여량이 증가함에 따라 산유량과 4% FCM은 점진적으로 증가하였으나, 유지방 함량은 유의적으로 감소하였다(p<0.05). 사료중 CLA 첨가 수준이 증가함에 따라, 우유의 Stearic acid와 Oleic acid 함량은 감소한 반면, total CLA 함량은 증가하였다(p<0.05). 따라서 본 연구에서는 비유중기 착유우 사료에 바이오디젤유 부산물로 제조한 CLA를 첨가하는 것은 유생산량 증가와 유지방 내 CLA 함량 증가에 유리할 것으로 판단된다.

The In vitro Effects of Nano-encapsulated Conjugated Linoleic Acid on Stability of Conjugated Linoleic Acid and Fermentation Profiles in the Rumen

  • This study was aimed to evaluate the stability of conjugated linoleic acids (CLAs) by nano-encapsulation against in vitro ruminal biohydrogenation by microbial enzymatic conversion. CLAs (free fatty acid form of CLA [CLA-FFA], nano-encapsulated CLA-FFA, triglyceride form of CLA [CLA-TG], and nano-encapsulated CLA-TG) were used in the in vitro fermentation experiments. When Butyrivibrio fibrisolvens (B. fibrisolvens) was incubated with CLA-FFAs, the concentrations of cis-9, trans-11 CLA and vaccenic acid (VA) slightly was decreased and increased by nano-encapsulation, respectively. When B. fibrisolvens was incubated with CLA-TG, the concentrations of cis-9, trans-11 CLA and VA decreased, but these were increased when B. fibrisolvens was incubated with nano-encapsulated CLA-TG. The nano-encapsulation was more effective against the in vitro biohydrogenation activity of B.fibrisolvens incubated with CLA-FFA than with CLA-TG. In the in vitro ruminal incubation test, the total gas production and concentration of total volatile fatty acids incubated with nano-encapsulated CLA-FFA and CLA-TG were increased significantly after 24 h incubation (p<0.05). Nano-encapsulated CLA-FFA might, thus, improve the ruminal fermentation characteristics without adverse effects on the incubation process. In addition, nano-encapsulated CLA-FFA increased the population of Fibrobacter succinogenes and decreased the population of B. fibrisolvens population. These results indicate that nano-encapsulation could be applied to enhance CLA levels in ruminants by increasing the stability of CLA without causing adverse effects on ruminal fermentation.

CLA (Conjugated linoleic acid) 급이수준에 따른 역돔의 CLA 축적량 (Accumulation of the Conjugated Linoleic Aacid (CLA) in Tilapia ( Tilapia nilotica) Fed Diets on Various Levels of CLA)

  • 기능성물질인 CLA를 양어사료에 첨가하여 기존의 어류와는 다른 기능성 역돔 생산을 시도하였다. 사육 8주간 매 2주마다 어체내의 CLA축적도를 측정한 결과 8주 후 $1\%$ CLA 첨가구에서는 41.3 mg/g of fat였으며, $10\%$ CLA 첨가구에서는 180.9mg/g of fat으로 역돔의 정상적인 성장을 고려하지 않는다면 $10\%$의 사료내 첨가도 가능한 것으로 생각된다. 그러나 정상적인 성장을 고려하면 CLA의 적정 첨가량은 $1\%$로 나타났으며, 그 이상의 첨가시에는 성장 저하 현상을 나타내었다. 합성된 CLA는 총 6 종류의 이성체로 구성되어 있었고, 그 중 c9, t11 및 t10, c12 이성체가 $85\sim90\%$를 이루고 있었으며, 첨가된 CLA 함량에 따라 축적되는 양도 달라졌다. 사육 8주 후, 육 및 간 중의 지방산 조성은 n-9, 3 계열은 거의 화를 나타내지 않았으나, n-6계열은 첨가된 CLA의 함량에 따라 육 및 간에 축적되는 양은 $5\sim27\%$로, $1\%$ CLA 첨가구가 가장 높았고, $10\%$ CLA 첨가구가 가장 낮은 함량비를 나타내었다.
