In Vivo Antitumor Activity of Hydrophilic Arginine-Conjugated Linoleic Acid Complex

  • Although conjugated linoleic acid (CLA) exerted potent antitumor activities in several animal models, application of CLA as a bioactive ingredient has been limited due to its hydrophobicity. This study was designed to determine the antitumor activity of arginine-CLA complex (Arg-CLA), a hydrophilic form of CLA. Mouse forestomach cancer was induced by gavage with benzo(a)pyrene (B(a)p) for 4-weeks prior to Arg-CLA (0.2 and 0.5%) feeding. Complete necropsies were performed to determine the number, size and locations of all the forestomach tumors at 20 weeks post-B(a)P administration. All mice in the B(a)P group developed tumors, and tumor incidences were decreased by 31 % and 44% in 0.2% and 0.5% Arg-CLA-fed groups, respectively, whereas no decrease was observed when Arg or com oil was given alone. Our results suggest that Arg-CLA suppresses mouse forestomach cancer.

Staphylococcus haemolyticus Lipase; High-Level Expression in Escherichia coli and Activation of Nonionic Detergent

  • A high level of Staphylococcus haemolyticus L62 lipase was expressed in an Escherichia coli transformant. The expressed lipase activity in the cell-free extract was 70,800 U/l, which corresponded to 30% of the total cellular protein. Pre-mixing of the l62 lipase with some nonionic detergents enhanced its hydrolytic activity towards olive oil: Tween detergents activated the L62 lipase by 3 fold. Gel filtration chromatography of the Tween-80-L62 lipase mixture demonstrated a polymerized complex (∼180 kDa) formed exclusively between Tween-80 and the L62 lipase. The lipase enzyme in the complex showed a higher specific activity towards most triacylglycerols than the intact L62 lipase. The activity enhancement towards each substrate was quite different depending on the acyl chain length; the activity towards tributyrin, trilinolein, and trilinolenin was much more enhanced than the towards the medium and the long-chain saturated triglycerides.

Dietary supplementation with combined extracts from garlic (Allium sativum), brown seaweed (Undaria pinnatifida), and pinecone (Pinus koraiensis) improves milk production in Holstein cows under heat stress conditions

  • Objective: This study was conducted to examine the effects of a mixture of pinecone oil, garlic, and brown seaweed extracts (PGBE) on milk production traits as well as physiological and ethological parameters in Holstein cows during the summer season (24 May to 03 July 2015, Korea). Methods: Among the extract combinations tested, we found that the level of 2,2'-azino-bis (3-ethylberzothiazoline-6-sulphonic acid) cation radical scavenging activity of the 0.16% PBGE complex at ratio of 1:1:1 (vol/vol) was comparable to that of the control (ascorbic acid; 1 mg/mL). Additionally, the PBGE complex reduced lipopolysaccharide-induced COX-2 expression in bovine mammary epithelial cells. Based on these findings, 40 lactating Holstein cows were used to measure the effects of PBGE complex at ratio of 1:1:1 (vol/vol) on milk production, immune response, metabolites, and behavior patterns by dividing the cows into two groups fed diets containing PGBE complex (n = 20; 0.016%/kg feed dry matter basis) or not containing PGBE complex (control, n = 20) for 40 d. Results: Results showed that PGBE complex did not influence milk composition, eating and ear surface temperature patterns, immune response, or metabolic parameters but promoted average milk yield throughout the experimental period. Additionally, a tendency of higher total antioxidant capacity and glutathione in the PGBE group was observed compared to the those in the control. When the temperature-humidity index (THI) exceeded 72 (average THI = 73.8), PGBE complex-fed cows experiencing heat stress showed increased milk yield and a tendency of increased rumination compared to the control. Conclusion: We suggest that incorporation of a combined mixture of 0.016% PGBE (1:1:1 ratio, vol/vol) to diet has the potential to improve milk yield and health status of cows under mild to moderate heat stress, denoting that it might be useful as an alternative anti-stressor in the diet of dairy cows under hot conditions.

Oil Production Evaluation for Hybrid Method of Low-Salinity Water and Polymer in Carbonate Oil Reservoir (탄산염암 저류층에 저염수주입공법과 폴리머공법의 복합 적용에 따른 오일 생산량 평가)

  • Low-salinity water based polymerflooding (LSPF) is one of promising enhanced oil recovery (EOR) method that has the synergetic effect of combining polymer injection method and low-salinity water injection method. In order to maximize EOR efficiency, it is essential to design low-salinity water appropriately considering the properties of polymer. In this aspect, the main purpose of this study is to investigate the effect of pH and $SO_4{^{2-}}$ ion which one of PDI (Potential Determining Ion) on oil production when applying LSPF to carbonate oil reservoir. First, the stability and adsorption of polymer molecule were analyzed in different pH of injection water and $SO_4{^{2-}}$ concentration in injection water. As a result, regardless of pH and $SO_4{^{2-}}$ concentration, when $SO_4{^{2-}}$ ion was contained in injection water, the stability of polymer solution was obtained. However, from the result of polymer retention analysis, in neutral state of injection water, since $SO_4{^{2-}}$ interfered the adsorption of polymer, the adsorption thickness of polymer was thinner as $SO_4{^{2-}}$ concentration was higher. On the other hand, when injection water was acidic as pH 4, the amount of polymer adsorption increased with the injection of polymer solution, so the mobility of polymer solution was greatly lowered. From the results of wettability alteration due to low-salinity water effect, in the case of neutral injection water injected, as $SO_4{^{2-}}$ concentration was increased, more oil which attached on rock surface was detached, altering wettability from oil-wet to water-wet. On the other hand, in acidic condition, due to complex effect of rock dissolution and polymer adsorption, wettability of the entire core system was less altered relatively to neutral condition. Therefore, it was evaluated that better EOR efficiency was obtained when injecting low-salinity water based polymer solution containing high concentration of $SO_4{^{2-}}$ with neutral condition, enhancing the oil production up to 12.3% compared to low-salinity water injection method.

Enhanced Nasal Absorption of Ketoconazole by Inclusion with Cyclodextrin (시클로덱스트린과의 포접에 의한 케토코나졸의 비점막 흡수증가)

  • Inclusion complexes of ketoconazole(KT) with ${\alpha}^_$, ${\beta}^_$cyclodextrin(CD) and $dimethy1-{\beta}-cyclodextrin$ (CD) and $dimethy1-{\beta}-cyclodextrin(DM{\beta}CD)$ as nasal absorption enhancer were prepared in 1: 2 molar ratios by freeze-drying and solvent evaporation methods. In order to compare with the intrinsic absorptivity of KT in the jejunum(J) and the nasal cavity(N), the in situ simultaneous perfusion method was employed. The in situ recirculation study revealed that KT-CD inclusion complexes with the greater stability constant and the faster dissolution rate proportionally increased the absorption of KT in the J and N of rats. The rank order of apparent KT permeability$(P_{app}\;:\;cm/sec\;{\time}\;1O^{-5}{\pm}S.E.)$, corrected by surface area of absorption, was $5.10{\pm}0.3(N,\; KT-DM{\beta}CD)$ )> $4.13{\pm}0.4(N,\;KT-{\beta}-CD)$ )> $3.52{\pm}0.2(N,\;KT-{\alpha}-CD)$ )> $2.76{\pm}0.3(J,\; KT-DM{\beta}CD)$ )> $2.61{\pm}0.5(J,\;KT-{\beta}-CD)$ )> $2.42{\pm}0.4(J,\;KT-{\alpha}-CD)$ at pH 4.0. The in crease in permeability of $KT-DM{\beta}CD$ inclusion complex was 2.6 folds in the J and 4.5 folds in the N when the perfusing solution was changed from the buffer(pH 4.0) to saline. The absorption rate of $KT-DM{\beta}CD$ inclusion complex after nasal administration was more rapid than those of ketoconazole alone and $KT-DM{\beta}CD$ inclusion complex after oral administration to rats. In comparision with an oral administration of ketoconazole suspension in corn oil, the relative bioavailability was calculated 137.3% for the oral and 195.0% for nasal $KT-DM{\beta}CD$ inclusion complex in rats. The present results suggest that $KT-DM{\beta}CD$ inclusion complex may serve as a potential nasal absorption enhancer for the nasal delivery of ketoconazole.

Development of Source Profiles and Estimation of Source Contribution for VOCs by the Chemical Mass Balance Model in the Yeosu Petrochemical Industrial Complex (여수석유화학산단 내 VOCs에 대한 오염원 분류표의 개발 및 CMB 모델에 의한 기여도 산정)

  • The purposes of this study were to characterize the local levels of VOCs (volatile organic compounds), to develop source profiles of VOCs, and to quantify the source contribution of VOCs using the CMB (chemical mass balance) model. The concentration of VOCs had been measured every 6-day duration in the SRO monitoring site in the Yeosu Petrochemical Industrial Complex from September 2000 to August 2002. The total of 35 target VOCs, which were included in the TO-14 designated from the U.S. EPA, was selected to be monitored in the study area. During a 24-h period, the ambient VOCs were sampled by using canisters placing about 10 ~ 15 m above the ground level. The collected canisters were then analyzed by a GC-MS in the laboratory. Aside from ambient sampling at the SRO site, the VOCs had been intensively and massively measured from 8 direct sources and 4 general sources in the study area. The results obtained in the study were as follows; first, the annual mean concentrations of the target VOCs were widely distributed regardless of monitoring sites in the Yeosu Petrochemical Industrial Complex. In particular, the concentrations of BTX (Benzene, Toluene, Xylene), vinyl chloride were higher than other target compounds. Second, based on these source sample data, source profiles for VOCs were developed to apply a receptor model, the CMB model. Third, the results of source apportionment study for the VOCs in the SRO Site were as follows; The source of petrochemical plant was apportioned by 31.3% in terms of VOCs mass. The site was also affected by 16.7% from wastewater treatment plant, 14.0% from iron mills, 8.4% from refineries, 4.4% from oil storage, 3.8% from automobiles, 2.3% from fertilizer, 2.3% from painting, 2.2% from waste incinerator, 0.6% from graphic art, and 0.4% from gasoline vapor sources.

Fusion of 3D seismic exploration and seafloor geochemical survey for methane hydrate exploration (메탄 하이드레이트 탐사를 위한 3 차원 탄성파 탐사와 해저면 지구화학탐사의 융합 기술)

  • The MH21 Research Consortium has conducted a high-resolution 3D seismic survey and a seafloor geochemical survey, to explore methane hydrate reservoirs in the eastern Nankai Trough, offshore Japan. Excellent geological information about shallow formations was obtained from the high-resolution 3D seismic survey, which was designed to image the shallow formations where methane hydrates exist. The information is useful in constructing a geological and geochemical model, and especially to understand the complex geology of seafloor, including geochemical manifestations and the structure of migration conduits for methane gas or methane-bearing fluid. By comparing methane seep sites observed by submersibles with seismic sections, some significant relationships between methane hydrate reservoirs, free gas accumulations below the seafloor, and seafloor manifestations are recognised. Bathymetric charts and seafloor reflection amplitude maps, constructed from seismic reflections from the seafloor, are also useful in understanding the relationships over a vast area. A new geochemical seafloor survey targeted by these maps is required. The relationships between methane hydrate reservoirs and seafloor manifestations are becoming clearer from interpretation of high-resolution 3D seismic data. The MH21 Research Consortium will continue to conduct seafloor geochemical surveys based on the geological and geochemical model constructed from high-resolution 3D seismic data analysis. In this paper, we introduce a basis for exploration of methane hydrate reservoirs in Japan by fusion of 3D seismic exploration and seafloor geochemical surveys.

Competitive Displacement of Methylcellulose from Oil-Water Interface by Various Emulsifiers (저분자량 유화제 첨가에 의한 계면 흡착 메칠셀룰로오스의 경쟁이탈 특성 연구)

  • Competitive displacement of methylcellulose (MC) absorbed at the oil-water interface was investigated by interfacial composition, surface shear viscosity, or surface tension measurements. It was found that all emulsifiers could competitively displace the interfacial MC from the oil-water interface but their behaviors were different from each other. With Tween 20 added to MC emulsion (1 wt% MC, 10 wt% n-tetradecane, 20 mM bis-tris, pH 7), MC load was steadily decreased with increasing concentrations of the emulsifier, as confirmed by surface shear viscosity measurements; moreover, there was complete MC displacement from the emulsion droplet surface at high concentration (0.1 wt%). The oil-soluble Span 80 was found to show a synergism with MC at the interface, which resulted in higher MC load at relatively low emulsifier concentrations ($\leq$0.05 wt%). At a higher emulsifier concentration (0.1 wt%) limited MC displacement was observed. These results were well supported by surface shear viscosity measurements. With water-soluble SDS, MC load was decreased with increasing concentrations of the emulsifier. Unlike Tween 20, however, it was found that at high concentrations (> 0.1 wt%), there was still some MC remaining at the droplet surface. Surface tension measurements are suggestive of an interfacial complex between MC and SDS.

Coating Layer Behavior Analysis of Al-Si Coated Boron Steel in Hot Bending Process

  • Nowadays, the usage of high strength steel has been growing in automobile industry mainly as structural parts since for its lightweight and high strength properties the oil crisis happened. Owing to poor formability, complex-shaped high-strength steel components are invariably produced through hot press forming. The high-strength steel sheets are in so me instances used with an Al-Si-coating with a view to prevent scaling of components during hot press forming. How ever, friction and fracture characteristics of Al-Si-coated high-strength steel during hot press forming process have not yet been investigated. In this paper, the formed parts which were formed in hot bending process were investigated by using EDS. SEM and nano indenter in order to analysis the coating layer behavior.

  • The essential oils are fragrant products whose complex compositions are obtained from various parts of plants by dry or steam distillation. Plants with variable biological activities have been explored worldwide. The presence of a large number of phenols, terpenes and other aromatic compounds make essential oils more precise in their mode of action. Because of this, they are known to possess many biological activities like antimicrobial, antioxidant and anti-inflammatory etc. In this article, we will review the published literature summarizing the chemistry of essential oils and their important biological activities.
