Ligand Binding Properties of the N-Terminal Domain of Riboflavin Synthase from Escherichia coli

  • Riboflavin synthase from Escherichia coli is a homotrimer of 23.4 kDa subunits and catalyzes the formation of one molecule each of riboflavin and 5-amino-6-ribitylamino- 2,4(1H,3H)-pyrimidinedione by the transfer of a 4-carbon moiety between two molecules of the substrate, 6,7- dimethyl-8-ribityllumazine. Each subunit comprises two closely similar folding domains. Recombinant expression of the N-terminal domain is known to provide a $C_2$-symmetric homodimer. In this study, the binding properties of wild type as well as two mutated proteins of N-terminal domain of riboflavin synthase with various ligands were tested. The replacement of the amino acid residue A43, located in the second shell of riboflavin synthase active center, in the recombinant N-terminal domain dimer reduces the affinity for 6,7-dimethyl-8-ribityllumazine. The mutation of the amino acid residue C48 forming part of activity cavity of the enzyme causes significant $^{19}F$ NMR chemical shift modulation of trifluoromethyl derivatives of 6,7-dimethyl-8-ribityllumazine in complex with the protein, while substitution of A43 results in smaller chemical shift changes.

  • Primary uraninite and secondary uranium minerals such as torbernite, metatorbernite, tyuyamunite, metatyuyamunite, autunite and metaautunite have been identified from various types of uranium ores. Uranium minerals occur as accessory minerals in both the primary and secondary ores. Low·grade uranium ores consist of various kinds of primary and secondary minerals. Major constituent minerals of primary uranium ores are graphite. quartz. Ba-feldspar and sericite/muscovite, and accessories are calcite, chlorite, fluorapatite, barite, diopside, sphene, rutile, biotite, laumontite, heulandite, pyrite, sphalerite and chalcopyrite, and secondary minerals consist of kaolinite, gypsum and goethite. Uraninite grains occur as microscopic very fine-grained anhedral to euhedral disseminated particles in the graphitic matrix, showing well·stratified or zonal distribution of uranium on auto-radiographs of low-grade uranium ores. Some uraninite grains are closely associated with very fine-grained pyrite aggregates, showing an elliptical form parallel to the schistosity. Some uraninite grains include extremely fine-grained pyrite particle. Sphalerite and pyrite are often associated with uraninite in graphite-fluorapatite nodule. The size of uraninite is $2{\mu}m$ to $20{\mu}m$ in diameter. Low-grade uranium ores are classified into 5 types on the basis of geometrical pattern of mineralization. They are massive, banded, nodular, quartz or sulfide veinlet-rich and cavity filling types. Well-developed alternation of uranium-rich and uranium-poor layers, concentric distribution of uranium in graphite-fluorapatite nodule and geopetal fabrics due to the load cast of the nodule suggest that the uranium was originally deposited syngenetically. Uraninite crystals might have been formed from organo-uranium complex during diagenesis and recrystallized by metamorphism. Secondary uranium minerals such as torbernite, tyuyamunite and autunite have been formed by supergene leaching of primary ores and subsequent crystallization in cavities.

Residual stresses and viscoelastic deformation of an injection molded automotive part

  • Injection molding is one of the most common operations in polymer processing. Good quality products are usually obtained and major post-processing treatment is not required. However, residual stresses which exist in plastic parts affect the final shape and mechanical properties after ejection. Residual stresses are caused by polymer melt flow, pressure distribution, non-uniform temperature field, and density distribution. Residual stresses are predicted in this study by numerical methods using commercially available softwares, $Hypermesh^{TM},\;Moldflow^{TM}\;and\;ABAQUS^{TM}$. Cavity filling, packing, and cooling stages are simulated to predict residual stress field right after ejection by assuming an isotropic elastic solid. Thermo-viscoelastic stress analysis is carried out to predict deformation and residual stress distribution after annealing of the part. Residual stresses are measured by the hole drilling method because the automotive part selected in this study has a complex shape. Residual stress distribution predicted by the thermal stress analysis is compared with the measurement results obtained by the hole drilling method. The molded specimen has residual stress distribution in tension, compression, and tension from the surface to the center of the part. Viscoelastic deformation of the part is predicted during annealing and the deformed geometry is compared with that measured by a three dimensional scanner. The viscoelastic stress analysis with a thermal cycle will enable us to predict long term behavior of the injection molded polymeric parts.

  • By the recently developed GMP(Glass Molding Press) process, aspheric glass lenses are widely used in many optical applications such as digital cameras, optical data storages and electrical devices etc. The GMP process can economically produce complex shaped glass lenses with high precision and good repeatability. This study deals the optimization of molding conditions for aspheric glass lenses in progressive GMP process through Design Of Experiment(Taguchi method). Tree main factors for molding conditions were selected based on pressure, temperature and cooling time at 1st cooling stage. From the analysis of experiments which were preformed with 3-cavity glass mold, it was revealed that the cooling time was the most sensitive parameter for form accuracy(PV) in progressive GMP process.

THE ATTITUDE STABILITY ANALYSIS OF A RIGID BODY WITH MULTI-ELASTIC APPENDAGES AND MULTI-LIQUID-FILLED CAVITIES USING THE CHETAEV METHOD

  • The stability problem of steady motion of a rigid body with multi-elastic appendages and multi-liquid-filled cavities, in the presence of no external forces or torque, is considered in this paper. The flexible appendages are modeled as the clamped -free-free-free rectangular plates, or/and as the discrete mass- spring sub-system. The motion of liquid in every single ellipsoidal cavity is modeled as the uniform vortex motion with a finite number of degrees of freedom. Assuming that stationary holonomic constraints imposed on the body allow its rotation about a spatially fixed axis, the equation of motion for such a systematic configuration can be very complex. It consists of a set of ordinary differential equations for the motion of the rigid body, the uniform rotation of the contained liquids, the motion of discrete elastic parts, and a set of partial differential equations for the elastic appendages supplemented by appropriate initial and boundary conditions. In addition, for such a hybrid system, under suitable assumptions, their equations of motion have four types of first integrals, i.e., energy and area, Helmholtz' constancy of liquid - vortexes, and the constant of the Poisson equation of motion. Chetaev's effective method for constructing Liapunov functions in the form of a set of first integrals of the equations of the perturbed motion is employed to investigate the sufficient stability conditions of steady motions of the complete system in the sense of Liapunov, i.e., with respect to the variables determining the motion of the solid body and to some quantities which define integrally the motion of flexible appendages. These sufficient conditions take into account the vortexes of the contained liquids, the vibration of the flexible components, and coupling among the liquid-elasticity solid.

First Record of Two Cold-Water Jellyfishes Aurelia limbata and Parumbrosa polylobata (Scyphozoa: Semaeostomeae: Ulmaridae) in Korean Coastal Waters

  • Most scyphomedusae jellyfishes recorded in Korean waters are temperate and subtropical species. In the present study, two cold-water jellyfishes from Korean waters are first described. Scyphomedusae were collected from the coasts of Gangneung and Ulsan of the East Sea, and Boryeong, and the eastern area of the Yellow Sea from June 2006 to May 2015. Scyphomedusae collected in Ulsan and Gangneung were identified morphologically as Aurelia limbata Brandt, 1835. Their umbrella was 250-500 mm in width and 30-50 mm in height; the exumbrella was white and the subumbrella was dark brown in color. The vascular system was complex with anastomosed branches. Individuals of this species lived at about 6.6-9.9℃ and 30-50 m depth in Korean waters. Scyphomedusae collected from the center of the Yellow Sea and from Boryeong were identified as Parumbrosa polylobata Kishinouye, 1910. Their umbrella was disc-shaped, 80-200 mm in width and 20-40 mm in height. Its milky white gonads could be seen through the transparent exumbrella. The stomach cavity was round and flat with four interradial cavities connected by subgenital cavities, and the four oral arms were bifurcated and spear-head shaped. Individuals were found at about 6.4-10.0℃ and 40-90 m depth in Korean waters. The identities of both species were confirmed by molecular analysis using nuclear ribosomal DNA sequences.

The Relationship between Oral Health and Stroke in Adults Based on the 6th (2015) Korea National Health and Nutrition Examination Survey

  • Stroke is the second cause of death worldwide, although the survival period is increasing after the occurrence of stroke, severe physical disability is caused with aftereffect. Oral inflammation is not limited to the oral cavity, it can cause malignant changes in other tissues and organs. In previous studies, we confirmed the relationship between tooth loss and stroke due to periodontal inflammation. The purpose of this study was to investigate the relationship between oral health and stroke such as oral hygiene behavior, tooth loss and periodontal disease among Korean adults over 40 years of age. This study was analyzed using the 6th Korea National Health and Nutrition Examination Survey (KNHANES) data. A total of 3,389 adults over 40 years of age were analyzed as final subjects. Socioeconomic statuses and oral health status was analyzed using a complex sample analysis technique. Logistic regression was used to analyze the relationship of oral health and stroke, and 95% confidence intervals were computed using SPSS. When the prevalence of stroke according to oral hygiene behavior was checked, the prevalence of strokes was lower in subjects who had a lot of brushings per day and subjects who used oral hygiene products (p<0.05). The risk of stroke was 2.17 times (95% confidence interval, 1.43~3.28) higher in the group with less than 19 remaining teeth, but it was not statistically significant as a result of adjusting for age and sex, income level, education level, drinking and smoking (p>0.05). Loss of teeth was found to be associated with the risk factor of stroke. Therefore, loss of teeth due to periodontal disease is an additional issue that should be considered as a risk factor for stroke.

  • The purpose of this paper is to evaluate the wind energy resources with high spatial resolution in Sunghak and Guduck mountains in Busan Metropolitan area under the various atmospheric stabilities. The numerical model used in this research is A2C (Atmosphere to CFD), mainly applied to assess the regional scale and microscale meteorological phenformin. Wind under the strong atmospheric stability moves around mountain side smoothly due to the strong potential energy. On the other hand, the cavity region on the lee side of mountain tends to be created and expanded as the atmospheric stability decrease. Annually the average distribution of wind power density, turbulence kinetic energy, and vertical wind shear help to explain quantitatively that wind resource near the northern side of Guduck mountain top is more suitable to establish wind energy complex than that in any other regions in the target area.

The Temporal and Spatial Expression of the Cytokeratin in Keratinocytes during Cutaneous Wound Healing on the Amphibian(Bombina orientalis) (양서류 피부 상처회복과정 중 각질화세포 cytokeratin의 분포)

  • The system of wound healing is very complex biological processing that includes inflammatory, reepithelialization, and matrix construction. For identification of the transitional pathway of the keratinocytes, we have employed immunohistochemical analysis using cytokeratin antibody after wounding. Epithelium in skin of the frog(Bombina orientalis) was examined with transmission electron microscopy. Cytokeratin was expressed in normal basal and gland cavity cells. At 3-hour basal layer cells were strong positive, however cells of the upper layer were negative reaction. Day1 and 2 after post-wounding, regenerating epithelial cell layer was positive reaction, especially basal layer cells were strong positive. At day 10 after wounding, the degree of positive reaction to basal cells of regenerating epithelial tissue was equal to day 7 wound tissue. At day of 19th, basal and spinous layer cells were strong positive reaction. Regenerating epithelial cells were positive but some basal cells were strong positive at day 27. From this result, we identified that the migration of the keratinocytes in amphibian skin wounds is initiated from basal layer fells and the keratinocytes migrate into basal and middle of the wound area.

A Case study on the construction of a long tunnel in the youngdong railroad (Mt. Dongbaek-Dokye) (영동선 동백산-도계간 장대터널 시공사례 연구)

  • This paper presents a case study on the construction of a long tunnel named as "Solan tunnel", which connects between Mt. Dongbaek station and Dokye station in the Youngdong Railroad. The tunnel will be the longest tunnel with length of 16.4 km in Korea when completed. The tunnel site is located in a complex geological region with faults, cavities and coal measures. In construction of adit No. 2, geophysical investigation methods such as electrical resistivity method and GPR(Ground Penetration Radar) were used to detect faults, cavities and coal measures in advance with some success. The geophysical investigation results and in-situ boring data were used as feedback to improve tunnel reinforcement design. Also, the tube umbrellas of grouted steel pipes were found to have a good reinforcement and grouting effects in zones of faults, cavities. In zones of coal measures, swellex rockbolts with mortar grouting were verified as successful.
