Comparative Studies on the Acute Toxicities of Whole Solids and Solids Aqueous Extracts Based on the Inhibition of Bacterial Bioluminescence Production

  • The aim of this investigation was to demonstrate a rapid bioluminescence bioassay for comparison of the toxicity of whole solids and the aqueous extracts of various environmental solid samples. With regard to the toxicities, those for the soil extracts were mostly found to be lower than those of whole soils, which may have been caused by un-extracted pollutants or dilution during the extraction process. Solid samples from dam-reservoir sediments and municipal refuses were also tested. The toxicities of the solid extracts (0-34%; refuses and sediments) were much lower than those of the whole solids (13-91%). The bioluminescence inhibition test indicated that the harmful effects of the contaminated solids samples were greater than those of the solid extracts.

3D-QSAR Study of Competitive Inhibitor for Acethylcholine Esterase (AChE) Nerve Agent Toxicity

  • The cholinesterase-inhibiting organophosphorous (OP) compounds known as nerve agents are highly toxic. The principal toxic mechanism of OP compounds is the inhibition of acethylcholine esterase (AChE) by phosphorylation of its catalytic site. The reversible competitive inhibition of AChE may prevent the subsequent OP intoxication. In this study, three-dimensional quantitative structure-activity relationship (3D-QSAR) was performed to investigate the relationship between the 29 compounds with structural diversity and their bioactivities against AChE. In particular, predictive models were constructed using the comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). The results indicate reasonable model for CoMFA ($q^{2}=0.453,\;r^{2}=0.697$) and CoMSIA ($q^{2}=0.518,\;r^{2}=0.696$). The presence of steric and hydophobic group at naphtyl moiety of the model may lead to the design of improved competitive inhibitors for organophosphorous intoxication.

  • 연기독성생성물로 인한 인명피해가 주요한 화재피해로 보고되고 있다. 화재로 인한 연기독성을 평가하기 위해 국내외의 다양한 시험이 있으며, 국내에서는 KS F 2271의 가스유해성시험으로 건축마감재료의 유해성을 평가하고 있다. 현행 가스유해성시험은 발연가스를 설치류(실험용 흰 쥐)에 노출시켜 행동시간을 측정하여 연소가스 독성을 평가한다. 본 연구에서는 가연성 고분자소재(우레탄 및 고무바닥재)를 대상으로 가스유해성시험을 수행하고 이때 발생되는 가스를 FT-IR로 정량분석하였다. 정량분석결과는 BS6853에서 규정하는 기준값과 비교하여 독성지수(R)를 산정하였다. 동물시험과 독성지수의 상대적 비교를 통해 두 시험법의 상관성을 분석하여 다양한 독성평가를 시도하고자 하였다.

담수조류의 벼재배용 농약 6종에 대한 감수성 비교 (Comparison of the Sensitivity of Freshwater Algae to 6 Pesticides for Paddy Rice)

  • 벼재배용 농약의 담수 조류에 대한 독성과 감수성을 조사하고자 4종의 담수 조류를 이용하여 생장저해시험을 실시하였다. 시험농약은 벼재배용으로 사용되는 butachlor 5% GR, butachlor 33% EW, isoprothiolane 12% GR, isoprothiolane 40% EC, diazinon 3% GR, diazinon 34% EC이며, 시험농약을 담수 녹조류 3종(Selenastrum capricornutum, Scenedesmus subspicatus, Chlorella vulgris)과 규조류 1종(Nitzschia palea)에 72시간 노출시켜 그 생장률을 조사하였고 생장저해율을 이용하여 50% 영향농도($EC_{50}$)를 산출하였다. 시험 결과, 모든 시험농약에서 유제와 유탁제가 입제보다 담수 조류에 대한 독성이 높았으며, 특히 유제의 경우 농약 원제보다도 높은 독성이 나타났다. 이러한 결과로 농약 제제의 특성에 따라 수조류에 대한 독성이 높아지거나 낮아지고 있음을 알 수 있었다. 그리고 시험종간 감수성에 있어서는 대부분의 시험농약에서 녹조류인 S. capricornutum와 S. subspicatus는 비슷한 수준의 감수성을 나타내고 있는 반면 C. vulgris의 감수성은 확연히 낮았다. 규조류인 N. palea의 농약에 대한 감수성은 S. capricornutum와 S. subspicatus에 비해 다소 낮거나 비슷하였으나, C. vulgris에 비해 감수성이 높게 나타났다.

건강보험 급여 한약제제 56종 처방의 계량적 문헌 근거 현황 분석 (Analysis of the current status of quantitative literature evidence for the prescription of 56 herbal medicines covered by health insurance)

  • Objectives: The purpose of this study is to analyze the current state of quantitative literature evidence for the prescription of 56 herbal medicines covered by health insurance that have been studied in Korea for the past 30 years, to evaluate the reliability of the evidence, and to find out the research direction of herbal medicine prescription in the future. Methods: 56 kinds of herbal medicine prescriptions were searched in domestic literature search databases OASIS, DBpia, and overseas PubMed, classified into chemistry, toxicity, cells, animals, clinical cases, and clinical trial studies, and built into an EBM pyramid structure. Results: When classified according to research contents, there were 61 cases (7.5%) of physicochemical analysis to identify constituent substances, 80 cases (9.8%) of toxicity evaluation, and 672 cases (82.7%) of efficacy evaluation. The efficacy evidence was classified according to the evidence-based medical pyramid structure: 196 cell trials (29.1%), 372 animal trials (55.4%), 89 case and case reporting series (13.3%), 7 comparative case studies (1.1%), and 8 randomized control clinical trials (1.2%). In the pyramid composition, the basis for the validity of 56 kinds of herbal medicines prescribed was 568 cases (84.5%) in cell and animal units, which could not be said to be highly reliable. There was no relationship between the ranking of quantitative literature evidence for herbal medicine prescriptions and the ranking of salary administration. Conclusions: In an era that continues to require scientific evidence for herbal medicine, traditional herbal medicine should secure the basis for safety validity even for the 10th most frequent prescription among 56 herbal medicine prescriptions for consumers. In particular, traditional herbal medicine should increase the quantitative and qualitative level of case reports on related herbal medicine prescriptions, focusing on each clinical society, and move toward comparative case studies and randomized clinical trial so that traditional herbal medicine is positioned as Evidence-based medicine.

독성 반응곡선을 이용한 수계 주요 오염물질의 혼합독성평가 (Mixture Toxicity Test of Ten Major Chemicals Using Daphnia magna by Response Curve Method)

  • 기존의 방류수 모니터링에서는 개별 오염물질들의 농도를 기준으로 독성을 평가하였다. 그러나 많은 연구자들에 의해서 오염물질들이 공존하는 상황에서 나타나는 독성은 그들 간의 상호작용을 통해서 혼합독성의 형태로 나타난다고 보고되고 있다. 본 연구에서는 GC/MS 분석을 통해 방류수 중에 존재하는 주요 독성 기여 오염 물질들을 분석하고, Independent Action(IA), Concentration Addition(CA), Effect Summation(ES) 모델을 사용하여 방류수의 혼합독성을 상호 비교 평가하였다. GC/MS로 분석된 오염물질을 대상으로 D. magna 기준 독성 평가를 실시하였고, 10가지의 주요 독성 기여 오염물질을 선별하였다. Chloroneb, butylbenzylphthalate, pendimethaline, di-n-butylphthalate, di-iso-butylphthalate, diazinon, isofenphos, 2-chlorophenol, 2,4,6-trichlorophenol 과 p-octylphenol을 주요 오염물질로 선정하여 혼합독성 평가를 실시하였다. 혼합독성 평가 결과는 IA 예측모델과 매우 높은 상관성($r^2\;=\;0.8475$)을 나타내었다. ES와 CA 모델은 IA 모델과 비교하여 혼합독성 결과와 매우 낮은 상관성을 나타내었으며, 특히 ES는 실측값을 5배나 과도하게 예측하였다. 이러한 결과를 통해서 전남지역 방류수에 존재하는 주요 오염물질들의 혼합독성은 IA 모델을 통해 예측이 가능할 것으로 판단된다.

칠레이리응애에 대한 농약의 선택독성과 장미에서 천적과 농약의 혼용에 의한 점박이응애의 방제효과 (Selective Toxicity of Pesticides to the Predatory Mite, Phytoseiulus persimilis and Control Effects of the Two-spotted Spider Mite, Tetranychus urticae by Predatory Mite and Pesticide Mixture on Rose)

  • 장미의 병해충 방제에 등록되어 있는 34종, 등록되어 있지 않은 5종, 그리고 보조제 3종, 충 42종의 농약에 대한 칠레이리응애(알, 약충, 성충)와 점박이응애(알, 성충)의 선택독성과 칠레이리 응애에 의한 점박이응애 밀도억제효과를 조사하였다. 살비제 중 acequinocyl, bifenazate, fenbutatin oxide, spirodiclofen은 칠레이리응애에 대해서 독성이 낮았으나, 점박이응애에 대해서는 독성이 높았다. 살충제는 acetamiprid, imidacloprid, spinosad, thiamethoxam, acetamiprid+etofenprox가 칠레이리응애와 점박이응애에 대해서 독성이 없거나 적었다. 살균제 중에는 azoxystrobin, kresoximmethyl, myclobutanil, nuarimol, triadimefon, triflumizole, oxadixyl+mancozeb이 칠레이리응애와 점박이응애에 대해서 영향이 없었다. 농약보조제인 cover와 siloxane은 칠레이리응애에 대해서 독성이 컸으나, spreader는 영향이 없었고, 점박이응애에 대해서는 독성이 없거나 낮았다. 장미시설하우스에 서 칠레이리응애에 의한 점박이응애 밀도억제효과를 조사하기 위하여, 점박이응애의 밀도가 잎당 65.3마리였을 때, 칠레이리응애를 주당 30마리 방사하였다. 방사 후 11일에 점박이응애 밀도가 잎당 3.8마리로 크게 감소하였고, 방사 후 20일에는 잎당 0마리로 밀도억제효과가 높게 나타났다. 조사기간 동안 흰가루병 방제로 살균제 4회(kresoxim-methyl, myclobutanil, nuarimol, triflumizole)와 총채벌레 방제약제를 1회(spinosad)처리하였으나, 칠레이리응애 밀도에는 영향이 없었다. 이상의 결과로 보아 장미 병해충의 종합관리체계에서 칠레이리응애에 독성이 적은 약제와 칠레이리응애를 함께 이용할 수 있을 것이다.

Fluoranthene 독성에 기인하는 비정상적 어류행동의 신경생화학적 분석 (Neurobiochemical Analysis of Abnormal Fish Behavior Caused by Fluoranthene Toxicity)

  • Fluoranthene, a common polycyclicaromatic hydrocarbon (PAH), exhibits phototoxicity which may affect aquatic organisms. The eventual goal of this study is to develop a biomarker of Japanese medaka (Oryzias latipes) used in monitoring hazardous chemicals in the ecosystem. In this study we investigated neural toxicity of fluoranthene in Japanese medaka (Oryzias latipes) along with comparative analysis of corresponding behavioral response. The untreated individuals shooed normal behavioral characteristics (i. e., smooth and linear movements). The treated fish, however, showed stopping and abrupt change of orientation (100 ppb), and severely reduced locomotive activity and enhanced surfacing activity (1,000 ppb). Treatment of the medaka fish with fluoranthene caused a significant suppresson of acetycholine esterase (AChE) activities in the body portion but not in the head portion. When fish were exposed to 1,000 ppb of fluoranthene for 24 hr, the body AChE activities decreased from 126.${\pm}$31.89 (nmoles substrate hydrolyzed per min per mg protein) to 49.51${\pm}$11.99. Expressions of tyrosine hydroxylase (TH) protein in the different organs from both head and body portions were comparatively analyzed using an immunohistochemical technique. Five organs of the medaka fish showing a strong TH protein expression were the olfactory bulb, hypothalamus, optic lobe, pons and myelencephalon regions. This study provides molecular and neurobehavioral bases of a biomonitoring system for toxic chemicals using fish as a model organism.

Neurobiochemical Analysis of Abnormal Fish Behavior Caused by Copper and Fluoranthene Toxicity

  • The goal of this study is to develop a biomarker used in monitoring abnormal behaviors of Japanese medaka (Oryzias latipes) as a model organism caused by hazardous chemicals. Japanese medaka was treated by copper and fluoranthene of appropriate sublethal concentrations after starvation for 48 hr. In this study we investigated neural toxicity of copper and fluoranthene in Japanese medaka (Oryzias latipes) along with comparative analysis of corresponding behavioral responses. The untreated individuals showed common behavioral characteristics (i.e., smooth and linear movements). Locomotive activity of the fish was monitored using an image processing and automatic data acquisition system. When treated with copper (100 ppb), the fish showed shaking patterns more frequently. As the concentration of copper increased to 1,000 ppb, activity decreased, and the fish showed an erratic movement. The treated with fluoranthene, however, showed stopping and abrupt change of orientation (100 ppb), and severely reduced locomotive activity and enhanced surfacing activity (1,000 ppb).

우황청심원 환제 및 액제의 급성경구비교독성시험 (Comparative Acute Toxicity Studies on Woo Whang Chung Shim Won Suspension and Pill in Mice)

  • Acute toxic effects of the Woo Whang Chung Shim Won suspension and pills were studied with the doses of 60, 74, 90, and 110 ml/kg in mice. In this study, we daily examined number of deaths, clinical signs, body weights, and pathological findings for 7 days after administration of Woo Whang Chung Shim Won. All mice given the highest dose (110ml/kg) died at 24 hour after administration of Sam-Sung Woo Whang Chung Shim Won suspension (Byunbang, SS-BS and Wonbang, SS-WS), one male and two female mice given 90 ml/kg dose of SS-BS died, and a few mice given middle dose (74 ml/kg) of SS-WS died. In control group (110 ml/kg) treated with vehicle (SS-BS and SS-WS), three out of 5 males and two out of 5 females mice died during the study. However, in animals treated with Kwang-Dong Woo Whang Chung Shim Won suspension (KD-S) and pill (KD-P), deaths were not observed. In the clinical signs, increase of drooling and decrease of spontaneous motor activities were observed in the highest dose group (110 ml/kg). No significant dose-related change in body weight was observed. The results suggest that the toxic effect of SS-BS and SS-WS may be atttributed to the solution for the Woo Whang Chung Shim Won suspension.
