국내 아마란스(Amaranthus spp.)에 발생하는 주요 해충의 발생소장과 수량 감소율 (Seasonal Occurrence of Insect Pests and Related Yield Loss in Amaranth Crop in South Korea)

  • 2019~2020년 동안 강원도 강릉지역의 아마란스 포장에서 진딧물류 3종(잠두진딧물, 목화진딧물, 복숭아혹진딧물), 노린재류 12종(애긴노린재, 각시장님노린재, 풀밭장님노린재 등), 나방류 4종(흰띠명나방, 파밤나방, 담배거세미나방, 도둑나방) 등 총 7과 18종의 해충을 확인하였다. 이 가운데 발생량이 많아 경제적으로 큰 피해를 주는 종류는 잠두진딧물, 애긴노린재, 각시장님노린재, 파밤나방, 흰띠명나방 등 5종이었다. 진딧물류는 4월 하순부터 발생하기 시작하여 6월 초순에 최대 발생을 보였다. 애긴노린재는 5월 중순부터 발생하여 8월 하순에 최대 발생량을 보이다가 9월 초순부터 밀도가 감소하였고, 각시장님노린재는 9월 중순부터 밀도가 급격히 늘어나서 10월 하순까지 지속적으로 발생하는 경향을 보였다. 흰띠명나방과 파밤나방은 8월 중순부터 유충 발생이 시작되어 10월 중순까지 아마란스에 피해를 주었다. 각각의 해충에 대한 방제구와 무방제구의 수량을 비교한 결과, 진딧물류와 노린재류에 의한 종실 수량 감소율은 각각 51.9%, 69.8%였다. 또한 흰띠명나방과 파밤나방에 의한 잎 생체량 감소율은 각각 72.5%, 36.5%였다.

통초(通草), 목통(木通) 신속 감별용 ITS 염기서열 기반 SCAR 마커 및 Multiplex-SCAR 분석법 개발 (Development of ITS sequence based SCAR marker and multiplex-SCAR assay for the rapid authentication of Tetrapanacis Medulla and Akebiae Caulis)

  • Objectives : Tetrapanacis Medulla and Akebiae Caulis are one of the most frequently adulterated herbal medicines because of their confusability of terms in the ancient writings and the similarity of morphological features of dried herbal products. The major adulterant is Aristolochia manshuriensis (Guanmutong) which has a serious safety concern with its toxicity. To ensure the safety and quality of the two herbal medicines, it is necessary to discriminate the toxic adulterant from authentic species. The aim of this study is to develop SCAR markers and to establish the multiplex-SCAR assay for discrimination of four plant species related to Tetrapanacis Medulla and Akebiae Caulis. Methods : ITS regions of fifteen samples of four species (Tetrapanax papyrifer, Fatsia japonica, Aristolochia manshuriensis, and Akebia quinata) collected from different sites were amplified and sequenced. Fifteen obtained ITS sequences were aligned and analysed for the detection of species-specific sequence variations. The SCAR markers were designed based on the sequence alignments and then, multiplex-SCAR assay enhancing rapidity was optimized. Results : ITS sequences clearly distinguished the four species at the species level. The developed SCAR markers and multiplex-SCAR assay were successfully discriminated four species and detected the adulteration of commercial product samples by comparison of the amplified DNA fragment sizes. Conclusions : These SCAR markers and multiplex-SCAR assay are a rapid, simple, and reliable method to identify the authentic Tetrapanacis Medulla and Akebiae Caulis from adulterants. These genetic tools will be useful to ensure the safety and to standardize the quality of the two herbal medicines.

몇 가지 키위 품종의 대목에 따른 접목 활착 특성 (Grafting Characteristics of Several Kiwifruit Cultivars as Affected by Rootstocks)

  • 최근 다양한 키위 신품종이 육성, 보급되고 있다. 과수의 증식법은 무성번식의 한 종류인 접목이 많이 이용되고 있으며, 대목은 실생 대목을 많이 이용하고 있다. 키위 품종도 증식법으로 실생대목을 활용한 접목이 주로 이용되고 있다. 많은 키위 종 중에 상업적으로 재배되는 종은 크게 A. chinensis, A. deliciosa 2종이다. 최근 식이편의성을 위해 다래(A. arguta) 교잡종이 육성되는 등 종간 교배를 통한 품종이 육성되고 있다. 종이 다를 경우 접목 불친화성이 발현될 수 있기에 최근 육성된 골드품종과 교잡종의 대목에 따른 접목친화성을 알아보고자 하였다. 접목 50일 후 신초 신장량을 조사한 결과, 골드 3품종은 같은 종인 '괴밀(Kuimi)'을 대목으로 이용한 처리에서 1미터 정도로 가장 양호했고 종이 다른 대목에서는 신초신장이 억제되는 경향이었다. 다래(A. arguta) 교잡종 '스키니그린'은 모든 대목에서 1미터 정도자라 친화성이 좋은 것으로 보였다. 하지만, 이종간의 접목에서는 몇 년 뒤에 불친화성이 발현되기도 하기에 장기적인 추가 연구가 필요할 것으로 본다.

방풍통성산 제제의 성분 및 생리활성 비교 연구 (Comparative Study of Characterizing Components and Biological Activities of Bangpungtongseong-san Formulation)

  • The purpose of this study was to investigate the quality of two different commercial Bangpungtongseong-san (BTS) extract granules (BTS-2 and BTS-3) by comparing with BTS decoction (BTS-1). The contents of characterizing components and biological activities of two different commercial BTS extract granules were compared with those of the BTS decoction. The contents of characterizing components were analyzed with HPLC. The antioxidative effects were determined by measuring 2,2-diphenyl-1-picrylhygrazyl (DPPH) radical scavenging and superoxide dismutase (SOD)-like activity. Also, we compared the effects on lipid accumulation and reactive oxygen species (ROS) production during differentiation of 3T3-L1 preadipocytes. The contents of five components except liquiritin and sennoside A were higher in BTS-1. The DPPH radical scavenging and SOD-like activity were higher in BTS-1. BTS-1 significantly inhibited lipid accumulation during differentiation of 3T3-L1 preadipocytes and showed stronger effects than BTS-2, BTS-3. In addition BTS-1 showed stronger inhibition effects on ROS production during differentiation of 3T3-L1 preadipocytes than BTS-2, BTS-3. These results indicate that BTS decoction has strong biological activities than commercial BTS extract granules. It is also consistent with the contents of characterizing components.

Varietal characteristics of new white button mushroom 'Seolwon' in Agaricus bisporus

  • Commonly known as the button mushroom, Agaricus bisporus is one of the most widely cultivated mushroom species of edible fungi. In the breeding of new button mushroom, Seolwon was developed by crossing two homokaryons. Because of the predominantly pseudohomothallic life cycle, only a small percentage of homokaryotic meiospores are produced, which do not fruit. Homokaryotic cultures derived from these types of single spores produce a vegetative mycelium that contain a variable number of genetically identical nuclei per cell. After crossing two homokaryons, hybrids were cultivated on a small scale and on a commercial scale at a farm. The spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of Seolwon on CDA was better at $25^{\circ}C$ when it was compared with that of Seolgang. The mature cap shape of new strain Seolwon is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 39.7 mm on average. In comparison with white strain Seolgang, the strain had a yield that was 11% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 9.7% firmer with a good shelf life. Days of fruiting body were 1-2 days later than those of Seolgang. The physical characteristics such as springiness, chewiness, adhesiveness, gumminess were better than that of Seolgang.

  • 담배거세미나방, 파밤나방, 배추좀나방은 중요 경제 작물의 재배, 수확 시에 해를 입히는 해충들이다. 이들 해충의 방제를 위해 Bacillus thuringiensis를 이용한 Bt제는 여러 해 동안 화학 살충제의 대체 방제 수단으로 이용되고 있다. 국내에 시판 중인 미생물 살충제의 방제 효율 증진을 위해 Bt제의 추천 농도 이상과 제품 간 혼합에 따른 효과 평가를 실내 생물검정을 통해 수행하였다. 본 연구에 사용된 5 제품 모두 추천농도에서 배추좀나방 2령 유충에 높은 방제 효과를 보였으나, 파밤나방과 담배거세미나방에는 낮은 살충율을 보였다. 추천농도의 2배액과 4배액 처리 및 Bt 제품들의 혼합 처리에서도 파밤나방과 담배거세미나방에서는 방제 상승 효과는 없었다.

쌍끌이 기선저인망 어선의 어업용 어군탐지기 활용을 위한 기초연구 (Preliminary study for the application of a commercial echosounder installed a pair trawler)

  • For scientific research, a number of acoustic surveys using commercial echosounders equipped in fishing vessels were conducted throughout the world; however, few studies were performed in South Korea. Hence, this research is an preliminary study for presenting the application of a sounder from a fishing vessel. The fishing operations using a pair trawler (7 Cheonghae) was conducted in the Northwest-Western sea of Jeju Island from 20 to 23 April, 2016. Substantial impulse noises and attenuated signals were eliminated by the latest algorithms. Acoustic signals were grouped into the fish aggregations and long layer-like signals. The fish aggregations appeared between 30 and 60 m, and long layer-like signals showed the diurnal vertical migration. Energetic, morphological and positional properties of the fish aggregations and layer-like signals were described. The fish aggregations appeared mainly between sunrise and sunset; however layer-like signals tended to be presented regardless of time in consideration of the time series analysis. On the basis of the consignment sales, Scomberomorus niphonius, the target species of F/V 7 Cheonghae, was the highest catch with 4,280 kg (74.6%) and might have appeared in fish aggregations and layer forms.

Antioxidant and Neuroprotective Effects of Doenjang Prepared with Rhizopus, Pichia, and Bacillus

  • A new type of doenjang was manufactured by mixing soaked soybean, koji (Rhizopus oryzae), cheonggukjang (Bacillus amyloliquefaciens MJ1-4 and B. amyloliquefaciens EMD17), and Pichia farinosa SY80 as a yeast, salt, and water, followed by fermentation with koji that was made by fermenting whole wheat with R. oryzae. The mixed culture doenjang was designed to have a more palatable flavor and stronger biological activities than the conventional product. The extract of mixed culture doenjang showed higher antioxidant activity than the commercial doenjang as evaluated by the ferric reducing antioxidant power assay although it was not significantly different from the commercial product in 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activities. Further, the mixed culture doenjang reduced intracellular reactive oxygen species levels and protected cells from glutamate-induced cytotoxicity more efficiently in human hippocampal HT22 neuroblastoma cells than the commercial doenjang. In conclusion, a newly-developed mixed culture doenjang had a strong antioxidant activity in vitro and cultured cell model systems, exhibited a potential to prevent oxidative stress-associated disorders although animal and clinical studies are needed to confirm its in vivo efficacy.

Survey of Major Viruses in Commercial Nursery Trees of Major Pear Cultivars in Korea

  • Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), and Apple scar skin viroid (ASSVd) are economically important viruses that infect pear tree species worldwide. To evaluate the prevalence of these viruses in Korea, we investigated infection degree of three viruses and one viroid for the commercial nursery trees of the pear cultivars, Niitaka, Chuwhang, Wonwhang, and Whasan in 2017 and 2018. The results showed that the infection ratio of ACLSV, ASPV, ASGV, and ASSVd for the scion of pear cultivar Niitaka was 10%, 45%, 77%, and 50%, respectively. From the scion of pear cultivar Chuwhang, infection ratios of ASPV, ASGV, and ASSVd were found to be 70%, 50%, and 60%, respectively. From the scion of pear cultivar Whasan, infection ratios of ACLSV, ASPV, ASGV and ASSVd were found to be 40%, 60%, 93%, and 20%, respectively. From the root stock of pear cultivar Wonwhang, infection ratios of ACLSV, ASPV, ASGV, and ASSVd showed 28%, 57%, 100%, and 14%, respectively. ASGV had the highest recorded infection rate, and ACLSV was characterized by the lowest infection rate. The mixed infection ratio of Niitaka, Chuwhang, Whasan, and Wonwhang was 45%, 60%, 70%, and 85%, respectively.

당귀(Angelica spp.)의 기원분석에 관한 분자생물학적 연구 현황 및 향후과제 (Current status on the development of molecular markers for differentiation of the origin of Angelica spp.)

  • 당귀는 우리나라를 포함하여 중국과 일본 등 아시아국가에서 유용하게 이용되는 한약재이다. 그러나 국가마다 그 기원을 달리하기 때문에 혼 오용이 심하고 국제시장에서 혼란을 불러일으킬 소지가 다분하다. 따라서 오래전부터 형태학적, 세포유전학적 분석과 지표성분을 이용한 화학적 판별 마커의 개발에 관한 연구가 많이 진행되어 왔다. 또한 최근에는 다양한 재배환경과 수확 후 가공 및 처리방법에도 비교적 안전한 유전자 단편의 염기서열 비교분석을 통한 분자생물학적 기술을 적용한 판별기술의 개발에 관한 연구결과 들이 발표되고 있다. 그러나 아직까지 이들 기술의 실용화를 통한 현장 적용에는 한계가 있으며 보다 많은 후속연구가 수행되어야 한다. 이에 본 논문에서는 현재까지의 연구결과를 바탕으로 얻어진 문제점을 논하고 향후 필요한 추가 연구 과제들에 관하여 기술하였다.
