Development of Analytical Method and Monitoring for Bifenazate in Commercial Agricultural Products (농산물 중 Bifenazate의 분석법 개선 및 모니터링)

  • The analytical method for bifenazate was developed using a HPLC (UVD). Also, analytical condition of LC/MS was set up for bifenazate. We validated the method for the precision and the reproducibility. The correlation coefficient of bifenazate ranged from 0.05 to 2.5 mg/kg was 1.0. Limit of quantitation (LOQ) was 0.01 mg/kg. To measure recoveries from agricultural products such as foxtail millet (cereal grains), kidney bean (beans), orange (fruits), perilla leaves (vegetables) and oak mushroom (mushrooms), bifenazate was spiked. Mean recoveries of bifenazate for each sample were 82.7~104.1% at the level of 0.1 mg/kg and 73.1~104.3% at the level of 0.5 mg/kg. The relative standard deviations (n=3) were 0.2~9.7%. Pesticide residues for bifenazate were investigated in 16 commodities (rice, foxtail millet, buckwheat, kidney bean, peanut, sesame, orange, grapefruit, kiwifruit, spinach, perilla leaves, leek, garlic stem, garlic, ginger and oak mushroom) collected from 22 provinces in 2009. Bifenazate was analyzed using analytical method by HPLC from 304 samples, and residue was not detected.

Structural Behavior of Steel Wire Truss Deck with Continuous Lattices to the Longitudinal Direction (길이방향으로 연속된 래티스를 가지는 철선 트러스데크의 구조 거동)

  • A truss deck system that has replaced the slab form conventional method has become widely used in the construction of reinforced concrete structures as well as steel structures. The current commercial products, however, have some problems. The discontinuity between the lattice wires on the joint of the bottom wire induces vierendeel behavior, which increases the deflection of the system. In this research, a new truss deck system with continuous lattice wires on the level of the bottom wire was developed to reduce the system's vierendeel behavior and to improve its deformation capacity. To investigate the system's structural behavior, an experimental test and an analysis were performed. The main parameters of the test and analysis were the longitudinal shape and spacing of the lattices. To simulate the loading condition in the construction field, uniform construction loads were directly applied on the deck plates of the analysis model and the test specimens. The results of such analysis and test revealed that the longitudinal shape of the lattice wires is a major factor affecting the structural behavior of a steel wire truss deck. Thus, continuous lattice wires could result in decreased vierendeel behavior in the steel wire truss deck. It was also found that the truss deck system with lattices spaced longer than in the conventional products could be effectively used without increasing the member stresses.

  • The effect of irradiation on the shelf-life of commercial ham products and its quality were investigated during eight weeks storage at $30^{\circ}C$ following 3-8kGy irradiation. Total bacterial count of unirradiated group was proliferated from. $5.4{\times}10^{3}$ per gram of sample to $10^{6}$ after two weeks storage but the bacterial count in irradiated group was propagated to the same level after three to five weeks storage and the growth of coliform group was shown negative. The freshness of stored ham was better in irradiated group than in unirradiated one and the changes in components related to the quality of ham were almost the same between unirradiated and irradiated group. Discoloration and irradiation odor were positive in 8kGy irradiated group according to the sensory test, but 5 kGy irradiated group was not only good in texture but also in the extension of shelf-life to more than two times as compared with that of unirradiated one.

Research on Pesticides Residue in Commercial Agricultural Products in 2009 (2009년 유통 농산물 중 잔류농약 실태조사)

  • Pesticide residues were investigated in 16 commodities (rice, foxtail millet, buckwheat, kidney bean, peanut, sesame, orange, grapefruit, kiwifruit, spinach, perilla leaves, leek, garlic stem, garlic, ginger and oak mushroom) collected from 22 provinces (Seoul, Busan, Incheon, Daegu, Daejeon, Gwangju, Ulsan, Suwon, Seongnam, Goyang, Bucheon, Yongin, Cheongju, Jeonju, Jeju, Cheonan, Changwon, Pohang, Gumi, Jinju, Wonju and Yeosu) in 2009. Pesticides (172 kinds) were analyzed using multiresidue method by GC/MS/MS from 510 samples, and phenthoate in kiwifruit was violated by exceeding MRL. The intake assessment for 24 kinds of pesticide residues including the detected pesticides at multi pesticide residue monitoring (bifenthrin etc.) were carried out. The result showed that the ratio of EDI (estimated daily intake) to ADI (acceptable daily intake) was 0.000007~0.458% which means that the detected pesticide residues were in a safe range so that residual pesticides in the agricultural products in Korea.

Cultivation of Saccharomyces cerevisiae using Defatted Rice Bran Hydrolyzed in Near-critical Water as a Culture Medium (탈지미강의 아임계수 가수분해 생성물을 배지로 이용한 Saccharomyces cerevisiae의 배양)

  • The hydrolysis of defatted rice bran using near-critical water was performed, and the feasibility of consequent hydrolyzate as a growth medium was investigated by the cultivation of Saccharomyces cerevisiae. The near-critical water hydrolysis was carried out through a series of batch experiments, and the contents of total carbohydrates, disaccharides, and monosaccharides, total organic carbon (TOC), total nitrogen (TN), pH of products were measured. The growth rate of Saccharomyces cerevisiae was measured with optical density. The yield of total carbohydrates, TOC, and TN increased with temperature below $240^{\circ}C$, however, decreased above $240^{\circ}C$. The decrease of yields above $240^{\circ}C$ was caused by the formation of organic acids, and it agreed with the change of pH of products. The yield of glucose was a maximum at $200^{\circ}C$ and it decreased dramatically at higher temperature. The growth rate of Saccharomyces cerevisiae cultivated in the hydrolyzate was similar with that in the commercial medium under certain conditions. The growth rate was correlated with the content of glucose in hydrolyzate.

The Cytotoxic Effect of Oral Wet Wipes on Gingival Cells (시판 중인 구강청결티슈의 세포 독성 관찰)

  • Wet wipes are being increasingly used because of their convenience. Particularly, oral wet wipes are useful for regular cleaning of a baby's mouth after birth. Therefore, the consumption of oral wet wipes has increased over the past few years and a variety of products are commercially available. However, product information on safety is not sufficiently provided and still raises doubts regarding adverse effects. To confirm the safety of wet wipes as an oral hygiene item and provide information for their use, we investigated the cytotoxicity of oral wet wipes and verified the underlying mechanism. The anti-bacterial effect of oral wet wipes was analyzed using the disk diffusion method. The cytotoxic effects of oral wet wipes were observed based on morphological changes using microscopy and determined using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay in gingival epithelial cells and gingival fibroblasts. Evaluation of apoptosis by oral wet wipes was explored using propidium iodide flow cytometric analysis and a terminal deoxynucleotidyl transferase (TdT)-mediated deoxyuridine triphosphate (dUTP) nick-end labeling (TUNEL) assay. Apoptosis-related molecules were also analyzed using western blotting. Five types of oral wet wipes were tested, and two products from Fisher-Price and Dr. Kennedy revealed strong cytotoxic effects on gingiva epithelial cells and gingiva fibroblasts, although they also showed intense anti-bacterial effects on oral bacteria. Cell cycle arrest in the G2/M phase and apoptosis were observed based on treatment of extracts from Fisher-Price and Dr. KENNEDY. Relatively high TUNEL levels, reduction of proliferating cell nuclear antigen and cyclin-dependent kinase 4 expression, and fragmentation of poly (ADP-ribose) polymerase were also elucidated. These results suggest that commercial oral wet wipes could exert cytotoxic influences on oral tissue, although there are anti-bacterial effects, and careful attention is required, especially for infants and toddlers.

  • Yeonsan Ogae has been known as supporting health and high efficacy of treatment. In recent days, as the efficacy of functional peptides has known, the optimization of oligo peptides production and its characteristics from Ogae viscera has been performed. Response surface method was used to perform the optimizaion of enzyme hydrolysis. The range of processes was temperature (40, 50 and $60^{\circ}C$), pH(6.0, 7.0 and 8.0), and enzyme(1, 2 and 3%). The degree of hydrolysis, amono acids, molecular weight of products were analyzed. The optimum process of enzyme hydrolysis were determined as temperature $58^{\circ}C$, pH 7.5, and enzyme concetration 3%. At optimum conditions, the degree of hydrolysis after 2 h reaction was 75-80%. The total amino acids of amino acid and were 386.15 mg/100 g and 155.26 mg/100 g, respectively. The molecular weight of products by using Maldi-TOF was ranged from 300 to 1,000 Da.

Anti-inflammatory Effect of Polysaccharide Derived from Commercial Kanjang on Mast Cells (비만세포에서 시판 간장 유래 다당류의 항염증 효과)

  • Soy sauce is a traditional fermented seasoning in several oriental countries, such as Korea and Japan, and recently it has been reported to have biological activities. In Korean soy sauce, soybeans and wheat are the two main raw materials. Polysaccharides that originate from the cell wall of soybeans are resistant to enzymatic hydrolysis. These polysaccharides remain in the soy sauce even after fermentation and are termed Kanjang polysaccharides (KPS). In this study, polysaccharides were obtained from dialysate of different soy sauces labeled as A~T and manufactured by fermentation or the acid-hydrolyzate method. We investigated anti-inflammatory activities by examining the effects of these KPS on proinflammatory cytokine release and mRNA expression in mast cells. Histamine and ${\beta}$-hexosaminidase release were strongly decreased by the KPS treatment in RBL-2H3 cells. Treatment with KPS clearly reduced mRNA expression and the release of the proinflammatory cytokines interleukin (IL)-6, IL-8, and tumor necrosis factor-alpha (TNF-${\alpha}$) in PMACI-stimulated HMC-1 cells. In particular, KPS derived from fermented Kanjang products showed a significant anti-inflammation effect on mast cells compared to the acid-hydrolyzed Kanjang products. This study suggests that KPS appear to be effective in suppressing allergic inflammatory reactions.

A Study on the Preparation of Electrolytic Manganese Dioxide (전해 이산화망간 제조에 관한 연구)

  • With the intention of obtaining technical data for the industrial production of ${\gamma}-MnO_2$ for dry cell depolarizer by electrolytic oxidation of acidic manganese sulfate solution made from domestic rhodochrosite, optimum conditions of ore leaching, purification of leached solution and electrolytic oxidation of divalent manganes to tetravalent were investigated using simulated micro pilot plant having a production capacity of 4 kg of $MnSO_4$ per day. The nature and quality of the products were investigated by means of chemical analysis, DTA, X-ray diffraction and electron microscopy. The cell activity of $MnO_2$were examined by cell discharging character measurements. The optimum electrolysis conditions were as follow: Temperature of the electrolyte, above $90^{\circ}C$; current density, 0.7${\sim}A/dm^2$; anode materials, graphite or lead ; concentration of electrolyte, $MnSO_4 50{\sim}150g/l $ g/l and $H_2SO_4/MnSO_4 = 0.15{\sim}0.25$. Under the best condition the current efficiency was 99% and the products were almost pure ${\gamma}-MnO_2$. The cell discharging character were good and almost the same as that of regular grade commercial electrolytic manganese dioxide.

Gas Liquid Chromatographic Analysis of Fatty Acids in Ginseng Products (Gas Liquid Chromatography에 의한 인삼(人蔘) 제품(製品) 중의 지방산(脂肪酸) 분석(分析))

  • Six commercial ginseng products, red ginseng, white ginseng I and II, red ginseng extract, white ginseng extract I and II were extracted with ethyl ether. Total fatty acid composition of the extracted free lipids was analyzed by gas liquid chromatography and digital integrator was used to measure the area of each peak. Crude lipid contents of red and white ginsengs were similiar. Those of red ginseng extract, white ginseng extract I and II were 1.10, 1.13 and 0.40%, respectively. It was found that 22 kinds of fatty acids existed in red and white ginsengs. Among them, 16 kinds of even numbered fatty acids were identified. Linoleic acid in red and white ginsengs was the most abundant. The contents of that in red ginseng, white ginseng I and II were 63.33, 45.55 and 41.06%, respectively. The next most abundant acid was palmitic acid, the contents of which were 11.30, 14.4 and 18.10% for red ginseng, white ginseng I and II, respectively. Major fatty acids for red ginseng extract and white ginseng extract I were linoleic and palmitic acids in the same order of magnitude. Linoleic and palmitic acids for red ginseng extract were 15.93 and 15.71 %, respectively, while linoleic and palmitic acids for white ginseng extract I were 21.94 and 19.15%, respectively. However, white ginseng extract II contained only 9.21% of linoleic acid and 16.13% of palmitic acid which was the major fatty acid.
